The objective of this work was to analyze the epicuticular n-alkane profile of domesticated and wild peanut genotypes. Foliar epicuticular n-alkanes of four Arachis hypogaea genotypes and two wild species - A. monticola and A. stenosperma - were analyzed by gas chromatography. Chemical relationships between them were evaluated using the Dice coefficient and UPGMA method. Two clusters were formed: one with four A. hypogaea genotypes and the other with the two wild species. There is more similarity between the BR1 and LIGO-PE06 genotypes and between the BRS 151 L-7 and BRS Havana genotypes.

A goiaba é um fruto muito perecível. Assim, objetivou-se avaliar os efeitos de ceras à base de carnaúba na conservação pós-colheita de goiabas Pedro Sato sob condição ambiente. Utilizaram-se cinco ceras comerciais: Citrosol AK (18%), Citrosol M (10%), Fruit wax (18 a 21%), Meghwax ECF-100 (30%) e Cleantex wax (18,5 a 20,5%), as quais foram aplicadas manualmente, na proporção de 0,15 a 0,20mL por fruta. Frutas sem aplicação de cera foram utilizadas como controle. O delineamento experimental foi inteiramente casualizado com 6 tratamentos, 4 repetições e 5 frutas por parcela. As goiabas foram caracterizadas imediatamente após a colheita e avaliadas aos 2, 4 e 6 dias após a aplicação dos tratamentos. As ceras exerceram pouca influência nos teores de sólidos solúveis totais, acidez total titulável e ácido ascórbico, porém, foram eficientes em retardar o amadurecimento, reduzir a perda de massa e a incidência de podridões. A cera Meghwax ECF-100 apresenta potencial para utilização em goiabas, porém há necessidade de ser avaliada em maior diluição, para evitar alterações indesejáveis.

Graviolas 'Morada', provenientes de pomar comercial localizado em Limoeiro do Norte, Ceará, foram colhidas na maturidade fisiológica com o objetivo de avaliar o efeito da aplicação pós-colheita de 1-meticiclopropeno (1-MCP) e cera na conservação, durante o armazenamento refrigerado. Os frutos foram armazenados por 0; 4; 8; 11; 13 e 15 dias, a 15,4±1,1ºC e 86,0±7,3% UR. Os tratamentos pós-colheita foram os seguintes: controle, 200 nL L-1 de 1-MCP (SmartFresh™), pulverização com a cera Fruit wax® e pulverização com Fruit wax® seguida de aplicação de 200 nL L-1 de 1-MCP. O delineamento experimental foi o inteiramente casualizado, em fatorial 4x6 (tratamentos pós-colheita x tempo de armazenamento), com quatro repetições. As variáveis analisadas foram: cor da casca e da polpa, a partir da luminosidade (L), cromaticidade (C) e ângulo de cor (Hº); perda de massa; pH; acidez titulável (AT); sólidos solúveis totais (SST); açúcares solúveis totais (AST) e açúcares redutores (AR). Os tratamentos cera e cera+1-MCP mantiveram estáveis os valores de L da casca e da polpa até o 8º e o 4º dia de armazenamento, respectivamente. O aumento em SST foi temporariamente atrasado pelos tratamentos pós-colheita. A AT, pH, AST e AR não foram influenciados pela cera e pelo 1-MCP. O uso da cera diminuiu a perda de massa em 23%, quando comparado ao controle.

A alta perecibilidade do maracujá-amarelo reduz sua vida de prateleira limitando sua comercialização. Este trabalho teve por objetivo aumentar a conservação pós-colheita do maracujá-amarelo pelo uso de cera e saco plástico poliolefínico. Os frutos, após colhidos, selecionados, lavados e desinfestados com hipoclorito a 1%, foram submetidos aos tratamentos: 1-imersão em cera de carnaúba (Fruit wax®), diluída na proporção 1:4 (m/v); 2 embalagem em saco plástico poliolefínico com 0,015 mm; 3 associação entre a imersão na cera de carnaúba e embalagem plástica; 4 testemunha, onde os frutos foram imersos em água com hipoclorito a 1%. Foram feitas análises da porcentagem de perda de matéria fresca, do teor relativo de água na casca, de sólidos solúveis, da acidez titulável, da relação sólidos solúveis/acidez titulável da polpa, das porcentagens de casca e polpa, e da relação casca/polpa. Houve menor porcentagem de perda de matéria fresca ao utilizar os tratamentos 2 e 3 ao longo do período de armazenamento. Entretanto, não houve diferença entre os tratamentos com relação à matéria fresca do fruto, casca e polpa, havendo redução destas variáveis durante o período de armazenamento. Também não houve diferença com relação às porcentagens de casca e de polpa, e relação polpa/casca, sendo que a primeira reduziu, e as duas seguintes aumentaram no armazenamento. Em geral, houve redução dos sólidos solúveis e da acidez.

The survival of micropropagated plants during and after acclimatization is a limiting process to plant establishment. There is little information on how the anatomy of vegetative organs of Ficus carica can be affected by culture conditions and acclimatization. The present research aimed to study the effects of time on culture medium and substrates during the acclimatization of fig tree plantlets produced in vitro, characterizing some leaf anatomy aspects of plantlets cultured in vitro and of fig trees produced in field. Plantlets previously multiplied in vitro were separated and transferred into Wood Plant Medium (WPM) where they were kept for 0, 15, 30, 45 and 60 days. Different substrates were tested and studies on leaf anatomy were performed in order to compare among plantlets grown in vitro, plantlets under 20, 40 and 60 days of acclimatization, and field grown plants. Keeping plantlets for 30 days in WPM allowed better development in Plantmax during acclimatization. Field grown plants presented higher number of stomata, greater epicuticular wax thickness and greater leaf tissue production compared to in vitro ones. The leaf tissues of in vitro plantlets show little differentiation and have great stomata number compared with acclimatized plants, which reduce the number of stomata during the acclimatization process.

The fractIons of dichloromethane extracts of leaves from andiroba (Carapa guianensis - Meliaceae), caapi (Banisteriopsis caapi - Malpighiaceae), cocoa (Theobroma cacao - Sterculiaceae), Brazil nut (Bertholletia excelsa - Lecytidaceae), cupuaçu (Theobroma grandiflorum - Sterculiaceae), marupá (Simaruba amara - Simaroubaceae) and rubber tree (Hevea brasiliensis - Euphorbiaceae), were analyzed by HT-HRGC and HT-HRGC-MS. Esters of homologous series of fatty acids and long chain alcohols, phytol, amyrines and tocopherols were characterized. The characterization of the compounds was based mainly in mass spectra data and in addition by usual spectrometric data (¹H and 13C NMR, IR).

To identify formulations of biological agents that enable survival, stability and a good surface distribution of the antagonistic agent, studies that test different application vehicles are necessary. The efficiency of two killer yeasts, Wickerhamomyces anomalus (strain 422) and Meyerozyma guilliermondii (strain 443), associated with five different application vehicles, was assessed for the protection of postharvest papayas. In this study, after 90 days of incubation at 4ºC, W. anomalus (strain 422) and M. guilliermondii (strain 443) were viable with all application vehicles tested. Fruits treated with different formulations (yeasts + application vehicles) had a decreased severity of disease (by at least 30%) compared with untreated fruits. The treatment with W. anomalus (strain 422) + 2% starch lowered disease occurrence by 48.3%. The most efficient treatments using M. guilliermondii (strain 443) were those with 2% gelatin or 2% liquid carnauba wax, both of which reduced anthracnose by 50% in postharvest papayas. Electron micrographs of the surface tissues of the treated fruits showed that all application vehicles provided excellent adhesion of the yeast to the surface. Formulations based on starch (2%), gelatin (2%) and carnauba wax (2%) were the most efficient at controlling fungal diseases in postharvest papayas.

This study aims to evaluate the prognostic value of microscopic parameters of asymptomatic leaves of Clusia hilariana Schltdl. subjected to particulate deposition of iron (2.14 mg cm-2 day-1) for 45 consecutive days. Samples of young and expanded leaves without symptoms were collected and subjected to light and scanning electron microscopy techniques. The height of the epidermal cells on both surfaces of the leaf and the thickness of the hypodermis, the chlorophyll parenchyma, and the leaf blade were measured. Micromorphological injury occurred in the abaxial surface of young leaves and on both surfaces of expanded leaves. Erosion of the epicuticular wax and cuticle rupture were frequent on the adaxial surface, while on the abaxial surface of both leaves there was a loss of sinuosity on the anticlinal wall of the epidermal cells, stomatal deformity and obstruction. Micromorphometric alterations were seen in all leaf tissues except in the height of epidermic cells, probably due to the thick cuticle and prominent cuticular flanges. The highest difference in thickness of the leaf blade was seen in young leaves of plants subjected to SPMFe, indicating greater sensibility to particulate iron in comparison to the expanded leaves. The micromorphological and micromorphometric alterations in the leaf blade of Clusia hilariana Schltdl. showed the prognostic potential of these tools on the evaluation of impacts caused by the deposition of particulate matter, especially in the 'Restinga' natural vegetation, where the exposure is increasing due to the presence of iron ore industry in their surroundings.

ABSTRACT This paper aims at evaluating the shelf life of mini tomatoes (Lycopersicum esculentum Mill.) cultivar ‘Sweet Grape’, grown in hydroponics, and stored under environmental and refrigerated conditions inside different packages. We adopted a completely randomized design, in which treatments were combinations of storage conditions: environment (e) and refrigerated (r) with packaging: polyvinyl chloride film (PVC); low-density polyethylene (LDPE); biofilm of tomato fruit of Solanum lycocarpum A.St.-Hil (lobeira) (TFB); cassava starch biofilm (CSB); carnauba wax (Copernicia prunifera) (CW), and without packaging - control (C). Physicochemical and sensory tests were carried out at the beginning (day zero), and at 8, 19, and 33 days of storage (DS). Fruit stored inside PVCr, LDPEe, LDPEr, and CWr had an acceptable shelf life of 33 days. The use of cassava and tomato starches were not effective in controlling fruit fresh weight loss. ‘Sweet Grape’ tomato postharvest conservation was enhanced under refrigerated conditions. The sensory evaluation results revealed that CWr treatment most pleased appraisers, while PCV had the highest rejection rate.

Policosanol is a mixture of higher aliphatic primary alcohols isolated from sugar cane wax, whose main component is octacosanol. An inhibitory effect of policosanol on platelet aggregation and cerebral ischemia in animal models has been reported. Thus, the objective of the present study was to evaluate the effect of policosanol on cerebral ischemia induced by unilateral carotid ligation and bilateral clamping and recirculation in Mongolian gerbils. Policosanol (200 mg/kg) administered immediately after unilateral carotid ligation and at 12- or 24-h intervals for 48 h significantly inhibited mortality and clinical symptoms when compared with controls, whereas lower doses (100 mg/kg) were not effective. Control animals showed swelling (tissue vacuolization) and necrosis of neurons in all areas of the brain studied (frontal cortex, hippocampus, striatum and olfactory tubercle), showing a similar injury profile. In the group treated with 200 mg/kg policosanol swelling and necrosis were significantly reduced when compared with the control group. In another experimental model, comparison between groups showed that the brain water content of control gerbils (N = 15) was significantly higher after 15 min of clamping and 4 h of recirculation than in sham-operated animals (N = 13), whereas policosanol (200 mg/kg) (N = 19) significantly reduced the edema compared with the control group, with a cerebral water content identical to that of the sham-operated animals. cAMP levels in the brain of control-ligated Mongolian gerbils (N = 8) were significantly lower than those of sham-operated animals (N = 10). The policosanol-treated group (N = 10) showed significantly higher cAMP levels (2.68 pmol/g of tissue) than the positive control (1.91 pmol/g of tissue) and similar to those of non-ligated gerbils (2.97 pmol/g of tissue). In conclusion, our results show an anti-ischemic effect of policosanol administered after induction of cerebral ischemia, in two different experimental models in Mongolian gerbils, suggesting a possible therapeutic effect in cerebral vascular disorders.

The effect of D002, a defined mixture of higher primary alcohols purified from bee wax, on in vivo and in vitro lipid peroxidation was studied. The extent of lipid peroxidation was measured on the basis of the levels of thiobarbituric acid reactive substances (TBARS). When D002 (5-100 mg/kg body weight) was administered orally to rats for two weeks, a partial inhibition of the in vitro enzymatic and non-enzymatic lipid peroxidation was observed in liver and brain microsomes. Maximal protection (46%) occurred at a dose of 25 mg/kg. D002 behaved differently depending on both the presence of NADPH and the integrity of liver microsomes, which suggests that under conditions where microsomal metabolism was favored the protective effect of D002 was increased. D002 (25 mg/kg) also completely inhibited carbon tetrachloride- and toluene-induced in vivo lipid peroxidation in liver and brain. Also, D002 significantly lowered in a dose-dependent manner the basal level of TBARS in liver (19-40%) and brain (28-44%) microsomes. We conclude that the oral administration of D002 (5, 25 and 100 mg/kg) for two weeks protected rat liver and brain microsomes against microsomal lipid peroxidation in vitro and in vivo. Thus, D002 could be useful as a dietary natural antioxidant supplement. More studies are required before these data can be extrapolated to the recommendation for the use of D002 as a dietary antioxidant supplement for humans.

Policosanol is a mixture of higher aliphatic alcohols purified from sugar cane wax, with cholesterol-lowering effects demonstrable in experimental models and in patients with type II hypercholesterolemia. The protective effects of policosanol on atherosclerotic lesions experimentally induced by lipofundin in rabbits and rats and spontaneously developed in stumptail monkeys have been described. The present study was conducted to determine whether policosanol administered orally to rabbits with exogenous hypercholesterolemia also protects against the development of atherosclerotic lesions. Male New Zealand rabbits weighing 1.5 to 2 kg were randomly divided into three experimental groups which received 25 or 200 mg/kg policosanol (N = 7) orally for 60 days with acacia gum as vehicle or acacia gum alone (control group, N = 9). All animals received a cholesterol-rich diet (0.5%) during the entire period. Control animals developed marked hypercholesterolemia, macroscopic lesions and arterial intimal thickening. Intima thickness was significantly less (32.5 ± 7 and 25.4 ± 4 µm) in hypercholesterolemic rabbits treated with policosanol than in controls (57.6 ± 9 µm). In most policosanol-treated animals, atherosclerotic lesions were not present, and in others, thickness of fatty streaks had less foam cell layers than in controls. We conclude that policosanol has a protective effect on the atherosclerotic lesions occurring in this experimental model.

A produção de camarões no Brasil é expressiva com condições propícias para expansão. Apesar de ser bem apreciado em termos culinários e ser uma fonte rica de proteínas, o camarão é apontado como um alimento de alto conteúdo de colesterol. Considerando que o nível de colesterol sangüíneo humano é dependente não só do teor de colesterol, mas também da quantidade de gordura e do tipo de ácidos graxos na dieta, um estudo integrado destes três constituintes foi realizado em camarão rosa (Penaeus brasiliensis), tamanho médio proveniente de São Paulo. A extração e a determinação do teor de lipídios totais foram realizadas de acordo com método de Folch, Less & Stanley. O método para determinação de colesterol por cromatografia líquida de alta eficiência, com coluna C18 e detector por conjunto de diodos, foi estabelecido no nosso laboratório. Este método mostrou-se eficiente, rápido e simples. A composição de ácidos graxos foi obtida por cromatografia gasosa com coluna capilar de sílica fundida com DB-WAX. Os teores de colesterol e lipídios totais para camarão rosa médio foram 127 ± 9mg/100 g e 1,0 ± 0,1 g/100 g, respectivamente. Foram detectados oitenta e sete ácidos graxos, sendo 20:5w3 (EPA), 16:0, 22:6w3 (DHA), 18:0, 18:1w9, 16:1w7, 20:4w6 e 18:1w7 os principais. O teor de colesterol encontrado no camarão analisado é alto. Por outro lado, o teor de gordura é baixo e os níveis de ácidos graxos poliinsaturados, especialmente EPA e DHA, são altos.

O presente trabalho objetivou prolongar a conservação póscolheita de pêssegos, armazenando-os à temperatura ambiente. Inicialmente selecionou-se uma microemulsão à base de fécula de mandioca e cera de abelha. Posteriormente ela foi testada, aplicando-a na superfície dos frutos em comparação com "Fruit wax" (cera comercial), com o intuito de se verificar o efeito dos diferentes tratamentos na composição química, física e físico-química dos mesmos. Utilizaram-se pêssegos 'Biuti' colhidos manualmente em 14/01/1999, ao atingirem o ponto de maturação fisiológica. Do lote colhido foram selecionados 120 frutos sendo os mesmos analisados quanto a perda de massa fresca, taxa respiratória, textura, sólidos solúveis totais, acidez total titulável e pH, a cada 3 dias. Os frutos receberam os tratamentos: Testemunha, "Fruit Wax", Fécula e Microemulsão. Os tratamentos "Fruit Wax" e "Microemulsão" proporcionaram melhor eficiência em relação à perda de massa fresca que os frutos dos tratamentos Testemunha e Fécula. Quanto à taxa de respiração, verificou-se picos da ordem de 40mg de CO2.kg-1.h-1 . Quanto aos açúcares, verificou-se que a sacarose foi o açúcar encontrado em maior quantidade, com apenas traços de glicose e frutose em algumas amostras. Quanto aos teores de sólidos solúveis totais, os frutos tratados com "Fruit Wax" apresentaram valores inferiores aos do tratamento Testemunha. O efeito da Microemulsão mostrou-se similar ao da cera "Fruit Wax" em todos os atributos e, superior ao dos tratamentos Testemunha e Fécula na redução da perda de massa fresca.

The application of technologies to extend the postharvest life of mangosteen fruit was studied and compared to storage at 25 °C/70-75%R.H (25 °C control treatment). The fruits were packed in expanded polystyrene (EPS) trays (5 fruits/tray). Five treatments were carried out at 13 °C/ 90-95% RH: application of carnauba wax coating, lecithin + CMC (carboxymethyl cellulose) coating, 50 µm LDPE (low density polyethylene) film coating, 13 µm PVC (Polyvinyl chloride), and non-coated sample (13 °C control treatment). Physicochemical analyses were performed twice a week. A statistical design was completely randomized with 8 repetitions for each treatment plus the control treatment. The results were submitted to variance analysis, and the averages compared by the Tukey test at 5% probability. Among the quality parameters analyzed, more significant differences were observed for weight loss, texture, and peel moisture content. The results showed that the maximum storage period for mangosteen at 25 °C is two weeks; while storage at13 °C can guarantee the conservation of this fruit for 25 days. Therefore, the treatment at 13 °C/90-95% RH without the use of coatings and films was more effective and economical.
