Trypanosoma cruzi trypomastigotes excrete-secrete a complex mixture of antigenic molecules. This antigenic mixture denominated trypomastigote excreted-secreted antigens contains a 150-160 kDa band that shows excellent performance in Chagas' disease diagnosis by immunoblotting. The present study partially characterized by two-dimensional gel electrophoresis the immunoreactivity against the 150-160kDa protein using sera samples from chagasic patients in different phases of the disease. Trypomastigote excreted-secreted antigen preparations were subjected to high-resolution two-dimensional (2D) gel electrophoresis followed by immunoblotting with sera from chagasic and non-chagasic patients. The 150-160kDa protein presented four isoforms with isoelectric focusing ranging from 6.2 to 6.7. The four isoforms were recognized by IgM from acute phase and IgG from chronic phase sera of chagasic patients. The 150-160kDa isoform with IF of approximately 6.4 became the immunodominant spot with the progression of the disease. No cross-reactivity was observed with non-chagasic or patients infected with Leishmania sp. In this study we provide basic knowledge that supports the validation of trypomastigote excreted-secreted antigens for serological diagnosis of Chagas' disease.

INTRODUCTION: This work was an epidemiological investigation of the risk of Trypanosoma cruzi transmission in the rural Quilombola community of Furnas do Dionízio, State of Mato Grosso do Sul, Brazil. METHODS: Of the 71 animals examined, seven were captured (two opossums, Didelphis albiventris; four rats, Rattus rattus; and one nine-banded armadillo, Dasypus novemcinctus) and 64 were domestic (one canine, Canis familiaris; five pigs, Sus scrofa; two bovines, Bos taurus; five caprines, Capra sp.; and 51 ovines, Ovis aries). Parasitological tests were performed to detect parasites in the blood and to identify the morphology of flagellates. These methods included fresh examinations, buffy coat tests and blood cultures. Molecular analysis of DNA for identification of trypanosomatids was performed by polymerase chain reaction (PCR) with primers S35 and S36. RESULTS: The parasitological tests showed flagellates in an opossum and two cattle. The molecular tests showed DNA from T. cruzi in an opossum and a pig. Triatoma sordida was the only triatomine species found in the community, and it colonized households (four specimens) and the surrounding areas (124 specimens). Twenty-three specimens tested positive for flagellates, which were subsequently identified as T. cruzi by PCR. CONCLUSIONS: Data analysis demonstrated that T. cruzi has a peridomestic life cycle that involves both domestic and wild mammals.

Introduction: The aim of this study was to evaluate the serological cross-reactivity between Leishmania sp. and other canine pathogens. Methods: Positive serum samples for Ehrlichia canis, Babesia canis, Toxoplasma gondii, Neospora caninum and Trypanosoma cruzi were tested using three serological methods enzyme linked immunosorbent assay (ELISA), indirect immunofluorescent antibody test (IFAT) and Kalazar Detect™, for canine visceral leishmaniasis. Results: Of the 57 dog samples tested, 24 (42.1%) tested positive using one of the three serological methods: 10/57 (17.5%) for ELISA, 11/57 (19.3%) for IFAT and 3/57 (5.3%) for Kalazar Detect™. Conclusions: Our results demonstrated that the presence of other infectious agents may lead to cross-reactivity on leishmaniasis serological tests.

ABSTRACTINTRODUCTION: The transmission cycle of Trypanosoma cruzi in the Brazilian Pantanal region has been studied during the last decade. Although considerable knowledge is available regarding the mammalian hosts infected by T. cruzi in this wetland, no studies have investigated its vectors in this region. This study aimed to investigate the presence of sylvatic triatomine species in different habitats of the Brazilian Pantanal region and to correlate their presence with the occurrences of vertebrate hosts and T. cruzi infection.METHODS: The fieldwork involved passive search by using light traps and Noireau traps and active search by visual inspection. The light traps were placed at five selected points along forested areas for seven nights during each of the nine excursions. At each point where a light trap was set, eight Noireau traps were placed in palm trees and bromeliads.RESULTS: In all, 88 triatomine bugs were collected: two and one individuals from light traps and Noireau traps, respectively; three from peridomestic areas; 23 in coati nests; and 59 in thornbird nests. In this study, active search in microhabitats showed higher efficiency than passive search, since 95% of the triatomine bugs were caught in nests. Further, triatomine bugs were only found to be infected by T. cruzi in coati nests.CONCLUSIONS: Coati nests might act as a point of convergence and dispersion for triatomine bugs and mammal hosts infected by T. cruzi, thereby playing an important role in the sylvatic cycle of T. cruziin the Pantanal region.

Lankesterella alencari n. sp. a Sporozoa that occur in the blood and CNS of the South American frog Leptodactylus acellatus is described. Since the tissue forms of this parasite have been previously reported as belonging to the genus Toxoplasma, we attempted in fection of 2 species of amphibia (Bufo marinus an dLeptodactylus ocellatus) with a Toxoplasma strain of human origen; inoculation was by intraperitoneal injection of parasite-containing ascitic fluid from infected mice. Attempt of experimental inoculation of the parasite found in the CNS of L. ocellatus in a highly susceptible host (mice) was unsuccessful. These results suggest that Toxoplasma does not occur naturally in the amphibia; be related to Toxoplasma is excluded. The following genera of haematozoa found in brazilian amphibia have been considered briedfly: Haemobartonella, Cytamoeba, Dactylosoma, Hepatozoon and Trypanosoma.

To identify wild hosts of Trypanosoma cruzi, surveys were conducted in the subandean valleys of Jujuy Province, Argentina, between June 1986 and March 1987. Seventy two mammals from 13 different species were examined by xenodiagnosis. Fifty two of them were mostly roedents trapped at the localities of Maimará, León and Tilcara, and the remainder had been kept in captivity at the Estación Biológica Experimental, in Jujuy. Trypanosoma cruzi infection was detected only in 2 Octodontomys gliroides (2 pos./8 exam. 25%) from all 72 examined mammals. Isolates were called Octodontomys Argentina 1 and 2 (OA1 and OA2). Both infected animals were caught at the archaelogical ruin of Pucará, at Tilcara. Repeated searches for triatomines in the ruin itself and in neighbour houses rendered negative results. Groups of mice inoculated with either OA1 or OA2 isolates became infected between 7 (OA1) to 12 days (OA2) postinoculation PI. Parasitemia peaks were observed between day 12th - 14th PI. Scarce amastigote nests were found in myocardium and skeletal muscle. Mortality was observed only for mice inoculated with OA1. Isoenzyme patterns of OA1 and OA2 were identical to one found in dogs and slightly different from that of human parasites in Argentina. Bones from Octodontomys sp., were recently found in a cave, dated 10200-8600 BC, in Pumamarca, near Tilcara, Jujuy. There are evidences that O. gliroides cohabited with man in ancient times and was associated to the domestic cycle of T. cruzi transmission, playing a role like that of domestic cavies. in Bolivia.

Leptoconops nosopheris sp. n. (Diptera: Ceratopogonidae) is described from a blood-filled female biting midge in Early Cretaceous Burmese amber. The new species is characterized by a very elongate terminal flagellomere, elongate cerci, and an indistinct spur on the metatibia. This biting midge contained digenetic trypanosomes (Kinetoplastida: Trypanosomatidae) in its alimentary tract and salivary glands. These trypanosomes are described as Paleotrypanosoma burmanicus gen. n., sp. n., which represents the first fossil record of a Trypanosoma generic lineage.

Aiming to identify new sources of bioactive secondary metabolites, we isolated 82 endophytic fungi from stems and barks of the native Brazilian tree Caesalpinia echinata Lam. (Fabaceae). We tested their ethyl acetate extracts in several in vitro assays. The organic extracts from three isolates showed antibacterial activity against Staphylococcus aureus and Escherichia coli [minimal inhibitory concentration (MIC) 32-64 μg/mL]. One isolate inhibited the growth of Salmonella typhimurium (MIC 64 μg/mL) and two isolates inhibited the growth of Klebsiella oxytoca (MIC 64 μg/mL), Candida albicans and Candida tropicalis (MIC 64-128 μg/mL). Fourteen extracts at a concentration of 20 μg/mL showed antitumour activities against human breast cancer and human renal cancer cells, while two isolates showed anti-tumour activities against human melanoma cancer cells. Six extracts were able to reduce the proliferation of human peripheral blood mononuclear cells, indicating some degree of selective toxicity. Four isolates were able to inhibit Leishmania (Leishmania) amazonensis and one isolate inhibited Trypanosoma cruzi by at least 40% at 20 μg/mL. The trypanocidal extract obtained from Fusarium sp. [KF611679] culture was subjected to bioguided fractionation, which revealed beauvericin as the compound responsible for the observed toxicity of Fusarium sp. to T. cruzi. This depsipeptide showed a half maximal inhibitory concentration of 1.9 μg/mL (2.43 μM) in a T. cruzi cellular culture assay.

A busca por modelos experimentais constitui passo fundamental para o avanço da medicina. OBJETIVO: Demonstrar, através da dissecção com técnicas microcirúrgicas, as estruturas anatômicas do osso temporal do primata Callithrix sp. FORMA DE ESTUDO: Experimental. MÉTODO: Dissecção de ossos temporais de Callithrix sp e documentação fotográfica. RESULTADOS: Identificamos as principais estruturas do osso temporal (orelhas externa, média e interna, e nervo facial). CONCLUSÃO: O primata não-humano Callithrix sp representa aparentemente um modelo viável para o estudo do osso temporal uma vez que apresenta alta similaridade anatômica com humanos.

A associação de bactérias a plantas tem sido estudada como uma possível tecnologia emergente, para fitorremediação de contaminantes, entre eles os herbicidas, que, por sua recalcitrância, ameaçam a qualidade do ambiente. O objetivo deste trabalho foi verificar a tolerância de mucuna-anã (Stizolobium deeringianum Bort) e mucuna-preta (Stizolobium aterrimum Piper & Tracy), inoculadas e não inoculadas com Rhizobium sp., ao herbicida atrazina. Os tratamentos foram: plantas com inoculante + 0,1 g/m², 0,2 g/m² atrazina e sem atrazina (T1, T2 e T3, respectivamente), sem inoculante + 0,1 g/m², 0,2 g/m² atrazina e sem atrazina (T4, T5 e T6, respectivamente). O delineamento experimental utilizado foi inteiramente casualizado, com três repetições. Foram avaliados germinação, sobrevivência, número de nódulos, altura, biomassa verde, biomassa seca da parte aérea, após o crescimento das plantas por 50 dias em casa de vegetação. Nos tratamentos com inoculante, avaliou-se a porcentagem de germinação de plantas bioindicadoras (Bidens pilosa L.). Mucuna-preta e mucuna-anã demonstraram maior tolerância ao herbicida quando associadas ao Rhizobium. Os valores de sobrevivência de mucuna-preta, nas doses 0,1 e 0,2 g/m² de atrazina (T1 e T2), foram de 34 a 24% superiores aos observados nas mesmas doses, mas sem o inoculante (T4 e T5). Para mucuna-anã, T1 e T2 foram de 17 e 8% superiores a T4 e T5, respectivamente. As alturas médias de mucuna-anã em T1, T2 e T3 foram mais elevadas que em T4, T5 e T6, reforçando a importância do simbionte à resistência ao herbicida. Os resultados encontrados para as variáveis altura, biomassa verde e seca para mucuna-preta não apresentaram diferença estatística entre os tratamentos com e sem inoculante, mostrando uma resistência natural à atrazina e a possibilidade de atuar como planta remediadora. A germinação de B. pilosa indica uma possível degradação da atrazina no solo com ambas as espécies de mucunas inoculadas com Rhizobium sp.

Apesar da existência de contatos entre planalto e litoral brasileiros ser atualmente quase um consenso entre os arqueólogos nacionais, não há muita certeza de como teria se dado tal contato e qual seria o fluxo entre interior e costa. O vale do Ribeira de Iguape (SP) é uma das raras regiões do Sul-Sudeste do país onde tal comunicação seria bastante facilitada devido a peculiaridades de sua geomorfologia. Neste trabalho, apresentamos os resultados de uma análise craniométrica comparativa entre 12 esqueletos provenientes de sambaquis fluviais do vale do Ribeira datados entre 6.000 e 1.200 anos AP e 225 esqueletos oriundos de diversas séries pré-históricas brasileiras do interior e do litoral. Ao contrário do que se observa no início do Holoceno nesse vale, não há qualquer afinidade biológica entre os ribeirinhos mais tardios e os paleoíndios de Lagoa Santa ou qualquer outra série interiorana. Os grupos fluviais (ambos os sexos) associam-se aos sambaquis da costa de São Paulo e do Paraná, mostrando que houve realmente um contato considerável entre a planície costeira e o planalto, ao menos no estado de São Paulo a partir da segunda metade do Holoceno.

Observou-se que o Trypanosoma cruzi não se multiplica na sanguessuga (Haementeria lutzi Pinto); os tripanosomas sugados degeneram após algum tempo; outros permanecem aparentemente normais, porém 48 horas após a ingestão infectante acabam morrendo. Observou-se ainda que os tripanosomas parasitas da rã (T. rotatorium e T. leptodactyli) bem como o T. hogei, parasita da serpente Rachidelus brazili, não se multiplicam no intestino dos triatomíneos. O mais resistente (o T. leptodactyli), permanece vivo até 72 horas após a ingestão infectante, porém as outras duas espécies (T. rotatorium e T. hogei) não resistem mais de 24 horas após serem sugadas pelos triatomíneos.

Se analizan los factores que interfieren en la esterilidad inducida químicamente en R. Prolixus mediante aplicaciones tópicas en distintas dosis de metepa. Se sugiere profundizar el estudio de la susceptibilidad individual para conocer la composición de la población frente al quimioesterilizante y la probable aparición de resistencia. Se investiga además la acción del metepa sobre T. cruzi en triatominos infectados experimentalmente, obteniéndose resultados negativos.

Recovery of Trypanosoma forattinii from natural infected Oryzomys capito velutinus from Trinidad, is reported. Negative results were obtained with attempts to infect experimentally, several laboratories animals, silvatyc rodents of the same ecological area and several species of triatomids bugs. Positive results were obtained only by using O. capito velutinus born in laboratory.

Foi pesquisado no 2.° semestre de 1969, o índice de infecção tuberculosa nos escolares de nível primário da sede municipal de Laranjal Paulista, SP, com faixa etária de 7 a 15 anos. Utilizou-se PPD RT23 com 2UT (0,04 mcg) de acordo com a 2.ª recomendação do Serviço Nacional de Tuberculose, sendo encontrado nível de 8% de reatores fortes o que sugere necessidade de intensificação local do controle da tuberculose. Houve diferenças significativas a 5% quanto à cor e grupo etário, predominando nos "não brancos" e nos de 11 a 15 anos, o mesmo não ocorrendo, entretanto, quanto ao sexo. Os reatores e seus conviventes foram encaminhados ao Dispensário de Tuberculose. Foi também pesquisada a viragem tuberculínica pelo BCG oral e intradérmico, comparados com grupo controle, encontrando-se resultados significantes. O resultado verificado no grupo que tomou BCG por via intradérmica foi quase duas vezes maior que os observados no grupo de BCG oral. Não houve diferenças quanto ao sexo e grupo etário, mas o grupo "não branco" mostrou-se mais reativo. Não foram definidas estatisticamente as reações pós-vacinais e a expectativa populacional foi considerada dentro da esperada para atividades desse tipo.
