The mosquito Aedes aegypti is the main vector of dengue in Venezuela. The genetic structure of this vector was investigated in 24 samples collected from eight geographic regions separated by up to 1160 km. We examined the distribution of a 359-basepair region of the NADH dehydrogenase subunit 4 mitochondrial gene among 1144 Ae. aegypti from eight collections. This gene was amplified by the polymerase chain reaction and tested for variation using single strand conformation polymorphism analysis. Seven haplotypes were detected throughout Venezuela and these were sorted into two clades. Significant differentiation was detected among collections and these were genetically isolated by distance.

This work, dedicated to the study of nesting habits of the species of the Neotropical genus Partamona Schwarz, is a sequence to the taxonomic revision recently published elsewhere. A total of 214 nests and nest aggregations of 18 species [Partamona epiphytophila Pedro & Camargo, 2003; P. testacea (Klug, 1807); P. mourei Camargo, 1980; P. vicina Camargo, 1980; P. auripennis Pedro & Camargo, 2003; P. combinata Pedro & Camargo, 2003; P. chapadicola Pedro & Camargo, 2003; P. nhambiquara Pedro & Camargo, 2003; P. ferreirai Pedro & Camargo, 2003; P. pearsoni (Schwarz, 1938); P. gregaria Pedro & Camargo, 2003; P. batesi Pedro & Camargo, 2003; P. ailyae Camargo, 1980; P. cupira (Smith, 1863); P. mulata Moure in Camargo, 1980; P. seridoensis Pedro & Camargo, 2003; P. criptica Pedro & Camargo, 2003; P. helleri (Friese, 1900)] were studied , including data about habitat, substrate, structural characteristics, construction materials and behavior. The descriptions of the nests are illustrated with 48 drawings. Partial data of the nests of P. bilineata (Say, 1837), P. xanthogastra Pedro & Camargo, 1997, P. orizabaensis (Strand, 1919), P. peckolti (Friese, 1901), P. aequatoriana Camargo, 1980, P. musarum (Cockerell, 1917) and P. rustica Pedro & Camargo, 2003 are also presented. Nests of P. grandipennis (Schwarz, 1951), P. yungarum Pedro & Camargo, 2003, P. subtilis Pedro & Camargo, 2003, P. vitae Pedro & Camargo, 2003, P. nigrior (Cockerell, 1925), P. sooretamae Pedro & Camargo, 2003 and P. littoralis Pedro & Camargo, 2003 are unknown. The species of Partamona build notable nest entrance structures, with special surfaces for incoming / exiting bees; some of them are extremely well-elaborated and ornamented, serving as flight orientation targets. All species endemic to western Ecuador to Mexico with known nesting habits (P. orizabaensis, P. peckolti, P. xanthogastra, P. bilineata, P. aequatoriana and P. musarum) build their nests in several substrates, non-associated with termitaria, such as cavities and crevices in walls, among roots of epiphytes and in bases of palm leaves, in abandoned bird nests, under bridges, and in other protected places, except P. peckolti that occasionally occupies termite nests. In South America, on the eastern side of the Andes, only P. epiphytophila and P. helleri nest among roots of epiphytes and other substrates, non-associated with termitaria. All other species studied (P. batesi, P. gregaria, P. pearsoni, P. ferreirai, P. chapadicola, P. nhambiquara, P. vicina, P. mourei, P. auripennis, P. combinata, P. cupira, P. mulata, P. ailyae, P. seridoensis, P. criptica and P. rustica) nest inside active termite nests, whether epigeous or arboreous. The only species that builds obligate subterranean nests, associated or not with termite or ant nests (Atta spp.) is P. testacea. Nests of Partamona have one vestibular chamber (autapomorphic for the genus) closely adjacent to the entrance, filled with a labyrinth of anastomosing pillars and connectives, made of earth and resins. One principal chamber exists for food and brood, but in some species one or more additional chambers are filled with food storage pots. In nests of P. vicina, there is one atrium or "false nest", between the vestibule and the brood chamber, which contains involucral sheaths, cells and empty pots. All structures of the nest are supported by permanent pillars made of earth and resins (another autapomorphy of the genus). The characters concerning nesting habits were coded and combined with morphological and biogeographic data, in order to hypothesize the evolutive scenario of the genus using cladistic methodology. The phylogenetic hypothesis presented is the following: (((((P. bilineata (P. grandipennis, P. xanthogastra)) (P. orizabaensis, P. peckolti)) (P. aequatoriana, P. musarum)) P. epiphytophila, P. yungarum, P. subtilis, P. vitae) (((((P. testacea (P. mourei, P. vicina)) (P. nigrior (P. auripennis, P. combinata))) (P. ferreirai (P. pearsoni (P. gregaria (P. batesi (P. chapadicola, P. nhambiquara)))))) ((((P. ailyae, P. sooretamae) P. cupira, P. mulata) P. seridoensis) P. criptica, P. rustica, P. littoralis)) P. helleri))). One area cladogram is presented. Dates of some vicariance / cladogenesis events are suggested. For bilineata / epiphytophila group, which inhabits the Southwestern Amazonia and the Chocó-Mexican biogeographical components, the origin of ancestral species is attributed to the Middle Miocene, when the transgressions of the Maracaibo and Paranense seas isolated the tropical northwestern South America from the eastern continental land mass. The next cladogenic event in the history of the bilineata / epiphytophila group is attributed to the Plio-Pleistocene, when the Ecuadorian Andes reached more than 3000 m, and the ancestral species was fragmented in two populations, one occupying the western Andes (ancestral species of the bilineata subgroup) and other the southwestern Amazon (ancestral species of the epiphytophila subgroup). Other aspects of the history of Partamona are also discussed.

Neotropical Meliponini: the genus Partamona Schwarz, 1939 (Hymenoptera, Apidae). The systematics and biogeography of Partamona Schwarz, a Neotropical genus of stingless bees (Meliponini, Apinae, Apidae), are revised. Seventeen new species are described: P. epiphytophila sp. nov., P. subtilis sp. nov., P. nhambiquara sp. nov., P. batesi sp. nov., P. yungarum sp. nov., P. vitae sp. nov., P. ferreirai sp. nov., P. gregaria sp. nov., P. auripennis sp. nov., P. nigrilabris sp. nov., P. combinata sp. nov., P. chapadicola sp. nov., P. seridoensis sp. nov., P. littoralis sp. nov., P. criptica sp. nov., P. rustica sp. nov. and P. sooretamae sp. nov. Partamona pseudomusarum Camargo, 1980, is considered as junior synonym of P. vicina Camargo, 1980. Types of P. grandipennis (Schwarz, 1951), P. xanthogastra Pedro & Camargo, 1996-1997, P. pearsoni (Schwarz, 1938), P. ailyae Camargo, 1980, P. pseudomusarum, P. vicina, P. mulata Moure in Camargo, 1980, P. aequatoriana Camargo, 1980, P. mourei Camargo, 1980, P. peckolti, (Friese, 1901), P. testacea (Klug, 1807), P. helleri (Friese, 1900) and P. musarum (Cockerell, 1917) were examined. Lectotypes of P. orizabaensis (Strand, 1919), and P. cupira (Smith, 1863) are designated. An identification key for the species and drawings of morphological characters are presented. A phylogenetic hypothesis, based mainly on morphological characters is proposed. Four groups are defined, considering the shape of mandible of workers and sternum VII of males: bilineata / epiphytophila group (western Amazon to México), including P. bilineata (Say), P. grandipennis, P. xanthogastra P. orizabaensis P. peckolti P. epiphytophila sp. nov., P. subtilis sp. nov., P. nhambiquara sp. nov., P. batesi sp. nov., P. yungarum sp. nov. and P. vitae sp. nov.; musarum group (Central Brazil, north of South America to Central America), including P. musarum, P. aequatoriana, P. vicina, P. mourei, P. pearsoni, P. ferreirai sp. nov., P. gregaria sp. nov. and P. testacea; nigrior group (Central Brazil to northeast of South America) including P. nigrior (Cockerell, 1925), P. auripennis sp. nov., P. nigrilabris sp. nov., P. combinata sp. nov., P. chapadicola sp. nov., P. seridoensis sp. nov. and P. littoralis sp. nov., and cupira group (southeastern and Central Brazil), including P. cupira, P. mulata, P. ailyae, P. sooretamae sp. nov., P. criptica sp. nov., P. rustica sp. nov. and P. helleri. Some geographic distribution patterns, congruent with that of other Meliponini bees, are commented.

The objective of this work was to monitor the maintenance of Citrus tristeza virus (CTV) protective isolates stability in selected clones of 'Pêra' sweet orange (Citrus sinensis), preimmunized or naturally infected by the virus, after successive clonal propagations. The work was carried out in field conditions in the north of Paraná State, Brazil. Coat protein gene (CPG) analysis of 33 isolates collected from 16 clones of 'Pêra' sweet orange was performed using single strand conformational polymorphism (SSCP). Initially, the isolates were characterized by symptoms of stem pitting observed in clones. Then viral genome was extracted and used as template for the amplification of CPG by reverse transcription polimerase chain reaction (RTPCR). RTPCR products electrophoretic profiles were analyzed using the Jaccard coefficient and the UPGMA method. The majority of the clones had weak to moderate stem pitting symptoms and its CTV isolates showed alterations in the SSCP profiles. However, the stability of the protective complex has been maintained, except for isolates from two analised clones. Low genetic variability was observed within the isolates during the studied years.

The chemotherapy agents against cancer may be classified as "cell cycle-specific" or "cell cycle-nonspecific". Nevertheless, several of them have their biological activity related to any kind of action on DNA such as: antimetabolic agents (DNA synthesis inhibition), inherently reactive agents (DNA alkylating electrophilic traps for macromolecular nucleophiles from DNA through inter-strand cross-linking - ISC - alkylation) and intercalating agents (drug-DNA interactions inherent to the binding made due to the agent penetration in to the minor groove of the double helix). The earliest and perhaps most extensively studied and most heavily employed clinical anticancer agents in use today are the DNA inter-strand cross-linking agents.

The generation of reactive oxygen species (ROS) may be both beneficial to cells, performing functions in intracellular signaling and detrimental, modifying cellular biomolecules. ROS can cause DNA damage, such as base damage and strand breaks. Organisms respond to chromosome insults by activation of a complex and hierarchical DNA-damage response pathway. The extent of DNA damages determines cell fate: cell cycle arrest and DNA repair or cell death. The ATM is a central protein in the response to DNA double-strand breaks by acting as a transducer protein. Collected evidences suggest that ATM is also involved in the response to oxidative DNA damage.

To investigate oxidative lesions and strand breaks induction by singlet molecular oxygen (¹O2), supercoiled-DNA plasmid was treated with thermo-dissociated DHPNO2 and photoactivated-methylene blue. DNA lesions were detected by Fpg that cleaves DNA at certain oxidized bases, and T4-endoV, which cleaves DNA at cyclobutane pyrimidine dimers and apurinic/apyrimidinic (AP) sites. These cleavages form open relaxed-DNA structures, which are discriminated from supercoiled-DNA. DHPNO2 or photoactivated-MB treatments result in similar plasmid damage profile: low number of single-strand breaks or AP-sites and high frequency of Fpg-sensitive sites; confirming that base oxidation is the main product for both reactions and that ¹O2 might be the most likely intermediate that reacts with DNA.

A method to detect Apple stem grooving virus (ASGV) based on reverse transcription polymerase chain reaction (RT-PCR) was developed using primers ASGV4F-ASGV4R targeting the viral replicase gene, followed by a sandwich hybridisation, in microtiter plates, for colorimetric detection of the PCR products. The RT-PCR was performed with the Titan™ RT-PCR system, using AMV and diluted crude extracts of apple (Malus domestica) leaf or bark for the first strand synthesis and a mixture of Taq and PWO DNA polymerase for the PCR step. The RT-PCR products is hybridised with both a biotin-labelled capture probe linked to a streptavidin-coated microtiter plate and a digoxigenin (DIG)-labelled detection probe. The complex was detected with an anti-DIG conjugate labelled with alkaline phosphatase. When purified ASGV was added to extracts of plant tissue, as little as 400 fg of the virus was detected with this method. The assay with ASGV4F-ASGV4R primers specifically detected the virus in ASGV-infected apple trees from different origins, whereas no signal was observed with amplification products obtained with primers targeting the coat protein region of the ASGV genome or with primers specific for Apple chlorotic leaf spot virus (ACLSV) and Apple stem pitting virus (ASPV). The technique combines the power of PCR to increase the number of copies of the targeted gene, the specificity of DNA hybridization, and the ease of colorimetric detection and sample handling in microplates.

Citrus tristeza virus (CTV) isolates from 35 grapefruit accessions belonging to Citrus Active Germplasm Bank of the "Instituto Agronômico de Campinas" located at the "Centro APTA Citros Sylvio Moreira", Cordeirópolis, São Paulo state, Brazil, were characterized and evaluated through symptoms in the trees, biological indexing, immunological diagnosis with different monoclonal antibodies and SSCP analysis (single-strand conformation polymorphism) of the coat protein gene. Symptomatology indicated that, in general, the group of plants with smaller canopy volume and severe stem pitting differed significantly from the group that presented greater vegetative development and mild to moderate stem pitting. However, the isolates from most of the accessions induced mild reaction on Mexican lime. The serological evaluation through the DAS-ELISA using monoclonal antibodies did not reveal any association between virus titer in the plant tissue and symptoms. The reaction with different monoclonal antibodies and the distinct electrophoresis patterns obtained through SSCP showed that there is a high degree of diversity among the isolates that infect these grapefruit accessions. High complexity within the same isolate was also observed in the SSCP profiles. This finding indicates that the CTV isolates from these plants are a complex mixture of CTV haplotypes. Similar SSCP banding patterns were observed among some plants with strong stem pitting symptoms, and among some plants with weak or moderate stem pitting symptoms.

O objetivo do trabalho foi caracterizar isolados de Phytophthora da acácia-negra (Acacia mearnsii) provenientes do Sul do Brasil, com base em características fenotípicas tais como morfologia, crescimento micelial, características das culturas, compatibilidade sexual e patogenicidade, e em perfis de polimorfismo de conformação de fita simples (SSCP = Single Strand Conformation Polymorphism) da região ITS-gene 5.8S do rDNA. Os isolados apresentaram esporângios com papilas proeminentes, arranjo dos esporângios irregularmente simpodial, culturas heterotálicas com presença de anterídios anfígenos, presença de clamidósporos e crescimento micelial em temperatura acima de 35°C, permitindo a classificação dos 12 isolados como P. nicotianae. Todos os isolados foram patogênicos, causando necrose em ramos de acácia-negra, sem formação de goma, com diferenças significativas de agressividade (p=0,05). Duas populações podem ser distinguidas em P. nicotianae da acácia negra pela análise PCR-SSCP do rDNA; no entanto essa separação não apresenta aparente correlação com características fenotípicas.

A gomose dos citros é considerada uma doença de grande importância para a citricultura no Brasil e em nível mundial. A etiologia desta doença compreende um complexo de espécies de Phytophthora. Embora importante, pouco se conhece sobre a gomose nas regiões produtoras de citros no Estado do Paraná. Por isso, este trabalho teve como objetivo identificar espécies de Phytophthora associadas à gomose em pomares de citros no Paraná. Nas regiões Norte, Noroeste e Vale do Ribeira foram retiradas amostras de raízes de plantas com sintomas de gomose e também de solo da rizosfera. Em laboratório, empregando pêra cv. D'anjou como isca e meio de cultivo batata-dextrose-ágar, foram obtidos 21 isolados de Phytophthora spp. Todos os isolados infectaram mudas de limão 'Cravo', reproduzindo os sintomas de gomose e também apresentaram crescimento micelial a 8º C e a 36º C, com exceção do isolado PR20 para 36º C . "In vitro", esses isolados foram heterotálicos, sendo 20 compatíveis ao tipo padrão A2 e um compatível ao tipo padrão A1. Vinte isolados formaram esporângios persistentes e papilados, com 25,5 - 62,0 µm de comprimento (C) e 27,9 - 49,6 µm de largura (L) e a relação C/L foi de 1,38:1. Um isolado (PR20) apresentou esporângios medindo 40,3 - 55,8 µm de comprimento e 27,9 - 37,2 µm de largura, formando esporângios persistentes, papilados ou bipapilados e de formas distorcidas, não formando clamidósporos. A temperatura ótima para crescimento desse isolado foi entre 20 a 28º C, enquanto para os demais foi de 24 a 32º C, tendo estes produção abundante de clamidósporos globosos de diâmetro variando entre 21,7 a 43,4 µm. De acordo com as características morfofisiológicas apresentadas, dos 21 isolados analisados, 20 pertenceram à espécie P. nicotianae e um à espécie P. citrophthora. A análise de sequências de genes da região ITS1-5.8S-ITS2 do rDNA e usando o teste de "Single-Strand Conformation Polymorphism" (SSCP), confirmou P. nicotianae e P. citrophthora como as duas espécies de Phytophthora associadas à gomose em pomares de citros no estado do Paraná.

Clonal cleaning, followed by pre-immunization with protective complexes of Citrus tristeza virus(CTV), allowed the commercial cultivation of Pêra sweet orange, a variety that has great importance for Brazilian citriculture but is sensitive to the virus. The use of mild protective isolates in other citrus varieties, even those more tolerant to CTV, can also be of interest to prevent the spread of severe isolates. The aim of this study was to characterize, by means of SSCP (Single Strand Conformational Polymorphism) analysis of the coat protein gene, CTV isolates present in plants of the sweet orange cultivars Pêra, Hamlin and Valencia propagated from four budwood sources: 1) old lines, 2) nucellar lines, 3) shoot-tip-grafted lines, and 4) shoot-tip-grafted lines pre-immunized with the mild CTV protective isolate 'PIAC'. We also evaluated the correlation of the obtained SSCP patterns to stem pitting intensity, tree vigor and fruit yield. SSCP results showed low genetic diversity among the isolates present in different trees of the same variety and same budwood source and, in some cases, in different budwood sources and varieties. Considering tristeza symptoms, lower intensity was noted for plants of new, shoot-tip-grafted and pre-immunized shoot-tip-grafted lines, compared to old lines of the three varieties. The observed SSCP patterns and symptomatology suggested that more severe CTV complexes infect the plants of old lines of all three varieties. The protective complex stability was observed in the SSCP patterns of CTV isolates of some shoot-tip-grafted and pre-immunized clones. It was concluded that the changes detected in other electrophoretic profiles of this treatment did not cause loss of the protective capacity of CTV isolate 'PIAC' inoculated in the pre-immunization.

Este estudo teve como objetivo a determinação da umidade de equilíbrio de painéis de madeira numa câmara de climatização e comparação dos seus valores com os obtidos através da equação de Nelson. O trabalho foi desenvolvido na Unidade Experimental de Produção de Painéis de Madeira da Universidade Federal de Lavras. Foram utilizadas amostras de aglomerado, compensado multilaminado e sarrafeado, chapa dura, OSB (Oriented Strand Board) , MDF (Medium Density Fiberboard), HDF (High Density Fiberboard), madeira-cimento e madeira-plástico, sendo esses produtos de escala industrial e experimental. As dimensões dos corpos-de-prova foram de 2,60 cm de largura por 2,60 cm de comprimento. As espessuras foram aquelas mais utilizadas comercialmente de cada produto, sendo utilizadas cinco repetições por painel. Os corpos-de-prova foram pesados e levados para uma câmara de climatização, onde foram submetidos a diferentes condições de umidade relativa (90, 80, 70, 60, 50 e 40%) e uma temperatura constante de 30 ºC. Determinaram-se a histerese e a umidade de equilíbrio, que variaram de 40 a 90%. Os resultados indicaram que a equação de Nelson se mostrou eficiente na estimativa da umidade de equilíbrio e que não existe em média diferença de histerese entre os produtos estudados, mostrando que todos apresentam a mesma estabilidade dimensional. Observou-se, também, que o revestimento dos painéis não afetou a umidade de equilíbrio.

Este trabalho teve como objetivo avaliar as propriedades de chapas de 0riented Strand Board (OSB), fabricadas com flocos de madeira de Eucalyptus grandis, Eucalyptus urophylla e Eucalyptus cloeziana e oriundas dos Municípios de Ponte Alta e Três Marias, no Estado de Minas Gerais. As massas específicas básicas das três espécies de eucaliptos das duas regiões foram, respectivamente: Ponte Alta (0,55; 0,61; e 0,70 g/cm³) e Três Márias (0,56; 0,58; e 0,69 g/cm³). Quando necessário, para manter as massas específicas das chapas próximas de 0,70 g/cm³ foram acrescentadas às partículas de madeira de eucalipto partículas de madeira de Pinus elliottii, oriundo da cidade de Viçosa, com massa específica de 0,45 g/cm³. Os flocos foram gerados nas dimensões médias de 90,00 x 20,00 x 0,46 mm. O adesivo utilizado foi o fenol-formaldeído, na proporção de 8% de sólidos, em relação à massa seca de partículas. Parte dos flocos de eucaliptos foram acetilados. As chapas foram prensadas à temperatura de 170 °C e 32 kgf/cm² de pressão. As propriedades das chapas foram determinadas segundo as normas da ABNT NBR 14810-3 (2002) e ASTM-D 1037 (1991). Os resultados foram comparados utilizando-se as normas ANSI/A - 208.1 (1993) e CSA 0437-93 (1993). As chapas contendo partículas acetiladas foram mais estáveis e adsorveram menos umidade. Na tração perpendicular, observou-se que as chapas contendo 100% de flocos acetilados apresentaram resultados inferiores ao estipulado pela norma CSA O437-0/93 (1993). A resistência ao arrancamento de parafuso, módulo de ruptura (paralelo e perpendicular) e compressão longitudinal (perpendicular), foi reduzida pela acetilação nas chapas contendo 100% dos flocos acetilados. As espécies que apresentaram, numericamente, as maiores médias para resistência mecânica foram: Eucalyptus grandis não acetilado (dureza Janka) e Eucalyptus cloeziana misturado com Pinus sp (módulo de ruptura). Somente a resistência à compressão longitudinal foi afetada pela região de origem da madeira. Os painéis fabricados com madeira de Eucalyptus urophylla, oriunda do Município de Três Marias, tiveram médias inferiores aos das chapas feitas com a mesma espécie, porém oriundas do Município do Rio Doce.

Os painéis reconstituídos à base de madeira ganharam espaço antes ocupado pela madeira maciça, o que tem gerado volume expressivo de resíduos. A indústria moveleira contribui para a geração desses resíduos, que se apresentam como alternativa viável para a exploração de sua aplicação na produção artesanal de pequenos objetos. Este trabalho teve por objetivo produzir pequenos objetos com painéis e com mistura de resíduos de painéis de madeira, a fim de comparar suas qualidades. A partir de 10 projetos gráficos, três artesãos confeccionaram 10 pequenos objetos, utilizando painéis de MDF (Medium Density Fiberboard), OSB (Oriented Strand Board) e compensado. Cada artesão produziu nove exemplares de cada objeto. Um quarto artesão confeccionou os mesmos objetos com resíduos de três empresas do polo moveleiro da cidade de Ubá, MG, com três repetições, sendo comparados os atributos de qualidade dos objetos. Concluiu-se que todos os painéis são adequados para a confecção dos objetos e alguns se destacam em relação a outros, conforme o atributo avaliado. Os objetos confeccionados com a mistura de resíduos de painéis de madeira tiveram acabamento uniforme e não apresentaram diferenças significativas em relação aos produzidos com painéis com relação aos atributos funcionalidade, peso, proporção e simetria. A mistura de resíduos de diferentes painéis influenciou negativamente a avaliação dos atributos acabamento, beleza e cor. Um objeto produzido a partir de resíduos do mesmo painel pode ser mais harmonioso e atrativo.
