Angiostrongylus costaricensis has a broad geographic distribution spanning from North to South America and the infections of vertebrates with this nematode can result in abdominal complications. Human infections are diagnosed by histological or serological methods because the isolation of larvae from feces is not feasible, as most parasites become trapped in intestinal tissues due to intense eosinophilic inflammation. Because A. costaricensis is difficult to maintain in the laboratory, an immunodiagnostic IgG enzyme-linked immunosorbent assay (ELISA) using antigens from the congeneric Angiostrongylus cantonensis species was evaluated against a panel of serum samples from patients who were histologically diagnosed with A. costaricensis infections. Sera from uninfected individuals and individuals infected with other parasites were used as controls. The sensitivity and specificity of the assay were estimated at 88.4% and 78.7%, respectively. Because the use of purified or cloned antigens has not been established as a reliable diagnostic tool, the use of heterologous antigens may provide a viable alternative for the development of an ELISA-based immunodetection system for the diagnosis of abdominal angiostrongyliasis.

Culex is the largest genus of Culicini and includes vectors of several arboviruses and filarial worms. Many species of Culex are morphologically similar, which makes their identification difficult, particularly when using female specimens. To aid evolutionary studies and species distinction, molecular techniques are often used. Sequences of the second internal transcribed spacer (ITS2) of ribosomal DNA (rDNA) from 16 species of the genus Culex and one of Lutzia were used to assess their genomic variability and to verify their applicability in the phylogenetic analysis of the group. The distance matrix (uncorrected p-distance) that was obtained revealed intragenomic and intraspecific variation. Because of the intragenomic variability, we selected ITS2 copies for use in distance analyses based on their secondary structures. Neighbour-joining topology was obtained with an uncorrected p-distance. Despite the heterogeneity observed, individuals of the same species were grouped together and correlated with the current, morphology-based classification, thereby showing that ITS2 is an appropriate marker to be used in the taxonomy of Culex.

OBJECTIVE To verify the adequacy of the professional nursing staff in the emergency room of a university hospital and to evaluate the association between categories of risk classification triage with the Fugulin Patient Classification System. METHOD The classification of patients admitted into the emergency room was performed for 30 consecutive days through the methodology proposed by Gaidzinski for calculating nursing requirements. RESULTS The calculation determines the need for three registered nurses and four non-registered nursing for each six hour shift. However, only one registered nurse and four non-registered nurse were available per shift. There was no correlation between triage risk classification and classification of care by the Fugulin Patient Classification System. CONCLUSION A deficit in professional staff was identified in the emergency room. The specificity of this unit made it difficult to measure. To find the best strategy to do so, further studies should be performed.

The use of machinery in agricultural and forest management activities frequently increases soil compaction, resulting in greater soil density and microporosity, which in turn reduces hydraulic conductivity and O2 and CO2 diffusion rates, among other negative effects. Thus, soil compaction has the potential to affect soil microbial activity and the processes involved in organic matter decomposition and nutrient cycling. This study was carried out under controlled conditions to evaluate the effect of soil compaction on microbial activity and carbon (C) and nitrogen (N) mineralization. Two Oxisols with different mineralogy were utilized: a clayey oxidic-gibbsitic Typic Acrustox and a clayey kaolinitic Xantic Haplustox (Latossolo Vermelho-Amarelo ácrico - LVA, and Latossolo Amarelo distrófico - LA, respectively, in the Brazil Soil Classification System). Eight treatments (compaction levels) were assessed for each soil type in a complete block design, with six repetitions. The experimental unit consisted of PVC rings (height 6 cm, internal diameter 4.55 cm, volume 97.6 cm³). The PVC rings were filled with enough soil mass to reach a final density of 1.05 and 1.10 kg dm-3, respectively, in the LVA and LA. Then the soil samples were wetted (0.20 kg kg-1 = 80 % of field capacity) and compacted by a hydraulic press at pressures of 0, 60, 120, 240, 360, 540, 720 and 900 kPa. After soil compression the new bulk density was calculated according to the new volume occupied by the soil. Subsequently each PVC ring was placed within a 1 L plastic pot which was then tightly closed. The soils were incubated under aerobic conditions for 35 days and the basal respiration rate (CO2-C production) was estimated in the last two weeks. After the incubation period, the following soil chemical and microbiological properties were detremined: soil microbial biomass C (C MIC), total soil organic C (TOC), total N, and mineral N (NH4+-N and NO3--N). After that, mineral N, organic N and the rate of net N mineralization was calculated. Soil compaction increased NH4+-N and net N mineralization in both, LVA and LA, and NO3--N in the LVA; diminished the rate of TOC loss in both soils and the concentration of NO3--N in the LA and CO2-C in the LVA. It also decreased the C MIC at higher compaction levels in the LA. Thus, soil compaction decreases the TOC turnover probably due to increased physical protection of soil organic matter and lower aerobic microbial activity. Therefore, it is possible to conclude that under controlled conditions, the oxidic-gibbsitic Oxisol (LVA) was more susceptible to the effects of high compaction than the kaolinitic (LA) as far as organic matter cycling is concerned; and compaction pressures above 540 kPa reduced the total and organic nitrogen in the kaolinitic soil (LA), which was attributed to gaseous N losses.

Since different pedologists will draw different soil maps of a same area, it is important to compare the differences between mapping by specialists and mapping techniques, as for example currently intensively discussed Digital Soil Mapping. Four detailed soil maps (scale 1:10.000) of a 182-ha sugarcane farm in the county of Rafard, São Paulo State, Brazil, were compared. The area has a large variation of soil formation factors. The maps were drawn independently by four soil scientists and compared with a fifth map obtained by a digital soil mapping technique. All pedologists were given the same set of information. As many field expeditions and soil pits as required by each surveyor were provided to define the mapping units (MUs). For the Digital Soil Map (DSM), spectral data were extracted from Landsat 5 Thematic Mapper (TM) imagery as well as six terrain attributes from the topographic map of the area. These data were summarized by principal component analysis to generate the map designs of groups through Fuzzy K-means clustering. Field observations were made to identify the soils in the MUs and classify them according to the Brazilian Soil Classification System (BSCS). To compare the conventional and digital (DSM) soil maps, they were crossed pairwise to generate confusion matrices that were mapped. The categorical analysis at each classification level of the BSCS showed that the agreement between the maps decreased towards the lower levels of classification and the great influence of the surveyor on both the mapping and definition of MUs in the soil map. The average correspondence between the conventional and DSM maps was similar. Therefore, the method used to obtain the DSM yielded similar results to those obtained by the conventional technique, while providing additional information about the landscape of each soil, useful for applications in future surveys of similar areas.

ABSTRACT In recent years, geotechnologies as remote and proximal sensing and attributes derived from digital terrain elevation models indicated to be very useful for the description of soil variability. However, these information sources are rarely used together. Therefore, a methodology for assessing and specialize soil classes using the information obtained from remote/proximal sensing, GIS and technical knowledge has been applied and evaluated. Two areas of study, in the State of São Paulo, Brazil, totaling approximately 28.000 ha were used for this work. First, in an area (area 1), conventional pedological mapping was done and from the soil classes found patterns were obtained with the following information: a) spectral information (forms of features and absorption intensity of spectral curves with 350 wavelengths -2,500 nm) of soil samples collected at specific points in the area (according to each soil type); b) obtaining equations for determining chemical and physical properties of the soil from the relationship between the results obtained in the laboratory by the conventional method, the levels of chemical and physical attributes with the spectral data; c) supervised classification of Landsat TM 5 images, in order to detect changes in the size of the soil particles (soil texture); d) relationship between classes relief soils and attributes. Subsequently, the obtained patterns were applied in area 2 obtain pedological classification of soils, but in GIS (ArcGIS). Finally, we developed a conventional pedological mapping in area 2 to which was compared with a digital map, ie the one obtained only with pre certain standards. The proposed methodology had a 79 % accuracy in the first categorical level of Soil Classification System, 60 % accuracy in the second category level and became less useful in the categorical level 3 (37 % accuracy).

This paper evaluates the presence of organochlorinated pesticides in the water supply system of the city of Bauru, State of São Paulo, Brazil. Soil samples were collected in agricultural areas and water samples at several points of the water source (river) and the distribution net. The results of the analyses of the water from the source were compared with the Brazilian classification system for inland waters (CONAMA/86) and the results of analyses of treated water were compared with Brazilian standards for finished drinking water.

This paper describes an approach for the colour-based classification of RGB (red-green-blue) images, acquired using a common scanner, of commercial carbonated soft drinks. Mean histograms of image colour channels were evaluated for the PCA classification of 29 brands of Guaraná, Cola, and orange flavors. Loadings for principal component axes resulted in different patterns for sample grouping on score plots according to RGB histograms. pH, sorbic acid and sucrose measurements were also correlated to the analyzed brands through PCA score plots of the digitalized images.

Currently, benznidazole (BNZ) is a unique therapeutic alternative available in Brazil to treat Chagas disease. Despite its traditional medical use, little is known about the chemical nature of this drug. A detailed study of the physicochemical properties of BNZ was performed using multiple assays. Thermal, diffractometric, morphological and reological drug profiles were obtained. The partition coefficient and solubility results allowed this drug to be classified as a class IV drug according to the biopharmaceutical classification system. This information will be useful for the development of more effective BNZ formulations and for establishing the quality profile of BNZ.

An analytical method by liquid chromatography has been proposed and validated to study the apparent solubility of ibuprofen in biorelevant dissolution media. The main properties of the studied media were pH values of 5.0 and 6.5 and the presence or absence of some natural surfactant agents. The parameters evaluated were specificity, linearity, precision, accuracy, and detection and quantification limits, as well as the drug stability under the analysis conditions. The developed method was useful to determine the apparent solubility of this drug as a function of temperature and surfactants concentration to demonstrate the validity of the Biopharmaceutics Classification System.

Este artigo aborda uma contribuição às classificações geotécnicas tradicionais de solos Unified Soil Classification System (USC), Transportation Research Board System (TRB) e a Metodologia Miniatura, Compactado, Tropical (MCT) para um caso aplicado à rodovia municipal não pavimentada VCS 346, no Município de Viçosa, MG, considerando-se várias amostras deformadas de solos coletadas no subleito da referida via. Os principais objetivos deste estudo residem na contribuição de um banco de dados geotécnicos sobre classificações de solos tropicais, bem como na verificação de possíveis agrupamentos entre os sistemas geotécnicos tradicionais de classificação e a metodologia MCT. O programa de laboratório envolveu a realização dos seguintes ensaios: (i) granulometria conjunta; (ii) limites de Atterberg (LL e LP); (iii) massa específica dos grãos do solo; (iv) compactação Mini-MCV; e (v) perda de massa do corpo-de-prova por imersão em água. Os resultados possibilitaram verificar que, após a classificação das amostras, vários agrupamentos entre os sistemas de classificações geotécnicas tradicionais coincidiram com aqueles já apresentados na metodologia MCT e, considerando a pedogênese da região de Viçosa, MG, para o grupo NG', houve a contribuição dos grupos SC, SM e CL do Sistema USC e do grupo A-4 do Sistema TRB.

OBJETIVO: estudar o uso de medicamentos por gestantes atendidas durante o pré-natal em unidades básicas do Sistema Único de Saúde (SUS) na cidade de Natal, rio Grande do Norte, Brasil. MÉTODOS: foram entrevistadas 610 grávidas, entre o primeiro e o terceiro trimestre de gestação, que compareceram para consulta pré-natal em unidades de saúde localizadas nos quatro distritos sanitários de Natal, entre maio e julho de 2006. Os dados foram coletados com entrevistas estruturadas, baseando-se em perguntas uso-orientadas e medicamento-orientadas. Os fármacos foram classificados de acordo com o Anatomical Therapeutic Chemical Classification System (ATC) e segundo critérios de risco para a gestação da Food and Drug Administration (FDA). Utilizou-se teste do chi2 para análise dos dados. RESULTADOS: eram utilizados 1.505 medicamentos, obtendo-se uma média de 2,4 drogas por mulher. O uso de pelo menos um fármaco na gravidez foi relatado por 86,6% das gestantes. As classes mais utilizadas foram os antianêmicos (35,6% dos medicamentos), analgésicos (24,9%), drogas para distúrbios gastrintestinais (9,1%) e vitaminas (7%). De acordo com a classificação do FDA, dos medicamentos empregados 42,7% pertencem a categoria A de risco; 27,1% à categoria B, 29,3% à categoria C; 0,3 à categoria D e nenhum à categoria X. Foram usados, no primeiro trimestre da gestação, 43,6% dos fármacos. Observou-se maior uso de medicamentos quanto maior a escolaridade e a renda familiar da mulher. A automedicação ocorreu em 12,2% dos medicamentos; esse índice foi maior no primeiro trimestre de gravidez e em gestantes de baixa escolaridade e multigestas. CONCLUSÕES: as gestantes de Natal estão sendo expostas a uma variedade de medicamentos, cuja segurança na gravidez ainda é incerta, o que exige prescrição criteriosa para evitar possíveis danos ao feto.

OBJETIVO: analisar a evolução clínica de três pacientes grávidas com malária grave internadas em unidade de terapia intensiva de um hospital localizado em Porto Velho (RO). MÉTODOS: foi realizado estudo descritivo em três gestantes, portadoras de malária por Plasmodium falciparum, internadas em unidade de terapia intensiva em Porto Velho, no período de 2005 a 2006. As variáveis categóricas utilizadas foram os critérios de classificação da Organização Mundial de Saúde para classificação de malária grave e os índices Acute Physiology and Chronic Health disease Classification System II (APACHE II) e Sepsis Related Organ Failure Assessment (SOFA) preditores de morbidade e gravidade das doenças em unidade de terapia intensiva. RESULTADOS: a malária adquirida pelas gestantes, caracterizada pela infecção por Plasmodium falciparum na forma grave da doença, resultou em óbito para as três pacientes e seus conceptos. CONCLUSÕES: embora a casuística seja pequena, a importância deste estudo reflete a repercussão da malária grave em gestantes, bem como a necessidade de um acompanhamento pré-natal mais criterioso e atento à identificação precoce do início das complicações da malária em gestantes.

Azospirillum amazonense revealed genomic organization patterns of the nitrogen fixation genes similar to those of the distantly related species A. brasilense. Our work suggests that A. brasilense nifHDK, nifENX, fixABC operons and nifA and glnB genes may be structurally homologous to the counterpart genes of A. amazonense. This is the first analysis revealing homology between A. brasilense nif genes and the A. amazonense genome. Sequence analysis of PCR amplification products revealed similarities between the amino acid sequences of the highly conserved nifD and glnB genes of A. amazonense and related genes of A. brasilense and other bacteria. However, the A. amazonense non-coding regions (the upstream activator sequence region and the region between the nifH and nifD genes) differed from related regions of A. brasilense even in nitrogenase structural genes which are highly conserved among diazotrophic bacteria. The feasibility of the 16S ribosomal RNA gene-based PCR system for specific detection of A. amazonense was shown. Our results indicate that the PCR primers for 16S rDNA defined in this article are highly specific to A. amazonense and can distinguish this species from A. brasilense.

This study aimed to evaluate the effect of preoperative imaging techniques on the success and complication rates of ureteroscopy. We performed a retrospective analysis of 736 patients (455 males and 281 females), with a mean age of 45.5±15.2 years (range, 1-88 years), who underwent rigid ureteroscopic procedures for removal of ureteral stones. Patients were divided into 4 groups according to the type of imaging modality used: group I, intravenous urography (n=116); group II, computed tomography (n=381); group III, computed tomography and intravenous urography (n=91), and group IV, ultrasonography and abdominal plain film (n=148). Patients’ demographics, stone size and location, prior shock wave lithotripsy, lithotripsy technique, operation time, success rate, and rate of intraoperative complications were compared among the groups. There were no significant differences in success and complication rates among the groups. The stone-free rate after primary ureteroscopy was 87.1% in group I, 88.2% in group II, 96.7% in group III, and 89.9% in group IV (P=0.093). The overall incidence of intraoperative complications was 11.8%. According to the modified Satava classification system, 6.1% of patients had grade 1, 5.1% had grade 2, and 0.54% had grade 3 complications. Intraoperative complications developed in 12.1% of patients in group I, 12.6% of patients in group II, 7.7% of patients in group III, and 12.2% of patients in group IV (P=0.625). Our findings clearly demonstrate that ureteroscopic treatment of ureteral stones can be safely and effectively performed with no use of contrast study imaging, except in doubtful cases of anatomical abnormalities.
