Chloride poisoning is known as having an inhibitor effect in the activity of metal catalysis. In this work in situ infrared spectroscopy (FTIR) of adsorbed carbon monoxide and x-ray photoelectron spectroscopy (XPS) were used to investigate the effect of chloride presence in the electronic metal density in the d subshell of palladium dispersed on alumina. The chloride poisoning effect was interpreted as an electronic effect since a weak back-bonded Pd-CO was formed due to the decrease in the electronic density of the d subshell of palladium, which could be also detected by the higher Pd 3d5/2 binding energy in the chloride presence. A similar poisoning effect was also observed for chloride free Pd/Al2O3 reduced at 500 ºC, and it was interpreted based on the interaction of metal with the alumina surface. The use of molybdena/alumina binary system as support, yield a contrary effect due to the metal-support interaction.

A new solid phase extraction (SPE) method has been developed for the selective separation and preconcentration of Cu (II) ions in food and water samples prior to its flame atomic absorption spectrometry determination. The method is based on the adsorption of the Cu(II) - 2-{[4-Amino-3-(4-methylphenyl-5-oxo-4,5-dihydro-1H-1,2,4-triazol-1-yl]acetyl}-N-phenyl hydrazinecarbothioamide complex on Amberlite XAD-8 resin. The metal complex retained on the resin was eluted with 7.5 mL of 2.0 mol L-1 HCl in acetone. The optimum conditions for the SPE of Cu(II) ions were investigated, and the method was subsequently applied to sea water, stream water, rice, tea, and tobacco samples for the determination of Cu(II) levels.

A presença das biovares I e/ou II de Ralstonia solanacearum em uma lavoura de batatas (Solanum tuberosum) tem influência direta no sucesso das medidas adotadas para controlar a murcha bacteriana. As biovares diferem entre si em relação à agressividade, latência e sobrevivência. Assim, um experimento de campo foi conduzido em uma área naturalmente infestada em duas épocas de cultivo com os objetivos de verificar (1) a incidência de biovares I e/ou II, (2) a relação entre biovar e época de plantio e (3) a relação entre biovar e cultivar de batata. Os isolados obtidos de plantas das cultivares Achat, Baronesa, Elvira, Macaca, Monte Bonito e Trapeira foram identificados como biovar I ou II através da PCR, utilizando os oligonucleotídeos iniciadores T3A e T5A. Ambas as biovares foram encontradas na área naturalmente infestada. De 73 isolados de R. solanacearum, 94,5% foram identificados como biovar II e 5,5% como biovar I. A biovar II foi isolada dos cultivos de primavera e de outono, independente da cultivar, mas a I apenas do cultivo de primavera e de plantas assintomáticas das cultivares Achat e Macaca. A maior população da biovar I nestas duas cultivares pode ser uma evidência da possível relação entre biovar e cultivar.

A influência de digeridos e suspensões de açúcar no comportamento térmico de As usando Pd(NO3)2, Pd(NO3)2 + Mg(NO3)2, and Ni(NO3)2 como modificadores químicos foi investigada. Influência de 0,2%, 5%, 10% e 35% (v/v) HNO3 na calibração (1,00 - 10,0 µg As L-1) também foi feita. Um volume de 20 µL de amostra mais 5 µL Pd ou 5 µL Pd + 3 µL Mg, ou 20 µL Ni foi injetado no atomizador do GF AAS. Para cada modificador, temperaturas de pirólise e atomização foram avaliadas por meio de curvas de pirólise e de atomização, respectivamente. Para suspensão de açúcar, temperaturas de pirólise e atomização (Tp, Ta) obtidas foram: sem modificador (400° C, 2000° C); Pd (1400° C, 2200° C); Pd/Mg (1400° C, 2200° C) e Ni (1600° C, 2200° C). Os valores para digeridos de açúcar foram: sem modificador (400° C, 2200° C); Pd (1400° C, 2200° C); Pd/Mg (1400° C, 2200° C) e Ni (600° C, 2200° C). Paládio foi selecionado como o melhor modificador químico. O tempo de vida do tubo de grafte foi de aproximadamente 350 queimas, o RSD (n = 12) para uma amostra típica contendo 5,52 µg As L-1 foi < 2,2% e o limite de detecção foi 2,4 pg As. Recuperações entre 80 e 92% foram encontradas.

O objetivo deste trabalho foi empregar a análise exploratória de dados, no caso, a técnica de análise de componentes principais (PCA) como ferramenta na avaliação de modificadores químicos na determinação direta e simultânea de Al, As, Cu, Fe, Mn e Ni em álcool etílico combustível por espectrometria de absorção atômica em forno de grafite (GFAAS). Os modificadores químicos avaliados foram: Pd(NO3)2 + Mg(NO3)2; W/Rh; W+ co-injeção de Pd(NO3)2 + Mg(NO3)2 e para cada modificador foram utilizadas trintas amostras de álcool etílico combustível. Como dados experimentais foram utilizados os resultados dos testes de adição e recuperação dos analitos frente aos diferentes modificadores químicos estudados. O emprego da técnica de PCA possibilitou a separação dos tipos de modificadores em função do intervalo de recuperação do analito. Dentre os modificadores avaliados, W+ co-injeção de Pd(NO3)2 + Mg(NO3)2 apresentou-se como a espécie de maior correlação positiva, pois apresenta os maiores teores de recuperação, e sendo assim, foi o escolhido para o desenvolvimento de metodologia para determinação direta e simultânea de Al, As, Cu, Fe, Mn e Ni em álcool etílico combustível por GFAAS.

O trabalho teve por objetivo avaliar artroscopicamente a articulação do joelho de ovinos hígidos em diferentes idades, com o intuito de determinar, de acordo com os portais estabelecidos, a capacidade de observação das estruturas. Utilizaram-se 18 animais da raça Santa Inês, divididos em três grupos experimentais eqüitativos, conforme a idade cronológica, sendo: Grupo I, 6-8 meses (peso médio de 25 kg); Grupo II, 2 anos (peso médio de 50 kg); e Grupo III, 3,5-5 anos (peso médio de 55 kg). Foram estabelecidos três portais: (1) portal artroscópico primário, craniolateral; (2) portal para fluxo de drenagem; e (3) portal instrumental, craniomedial. Identificaram-se com facilidade a patela, sulco e bordas trocleares, côndilos medial e lateral do fêmur, e tendão do músculo extensor longo dos dedos. Após a remoção da maior parte do coxim gorduroso infrapatelar com emprego do "shaver" foram visibilizados, ainda com dificuldade, os ligamentos cruzados e os meniscos. Para a identificação dos primeiros foram necessárias movimentos de flexão e extensão e para os meniscos manobras de varo e valgo. O grau de dificuldade foi inversamente proporcional ao tamanho da articulação. Sendo assim, foi possível concluir que os portais artroscópicos utilizados possibilitaram a identificação das principais estruturas articulares, independente da idade, e o método em si teve por vantagem a rápida recuperação pós-operatória dos animais.
Anti-tumor necrosis factor-a for the treatment of steroid-refractory acute graft-versus-host disease

Allogeneic stem cell transplantation has been increasingly performed for a variety of hematologic diseases. Clinically significant acute graft-versus-host disease (GVHD) occurs in 9 to 50% of patients who receive allogeneic grafts, resulting in high morbidity and mortality. There is no standard therapy for patients with acute GVHD who do not respond to steroids. Studies have shown a possible benefit of anti-TNF-a (infliximab)for the treatment of acute GVHD. We report here on the outcomes of 10 recipients of related or unrelated stem cell transplants who received 10 mg/kg infliximab, iv, once weekly for a median of 3.5 doses (range: 1-6) for the treatment of severe acute GVHD and who were not responsive to standard therapy. All patients had acute GVHD grades II to IV (II = 2, III = 3, IV = 5). Overall, 9 patients responded and 1 patient had progressive disease. Among the responders, 3 had complete responses and 6 partial responses. All patients with cutaneous or gastrointestinal involvement responded, while only 2 of 6 patients with liver disease showed any response. None of the 10 patients had any kind of immediate toxicity. Four patients died, all of them with sepsis. Six patients are still alive after a median follow-up time of 544 days (92-600) after transplantation. Considering the severity of the cases and the bad prognosis associated with advanced acute GVHD, we find our results encouraging. Anti-TNF-a seems to be a useful agent for the treatment of acute GVHD.

Chronic neurodegenerative processes have been identified in the rat forebrain after prolonged survival following hyperthermia (HT) initiated a few hours after transient global ischemia. Since transient global ischemia and ischemic penumbra share pathophysiological similarities, this study addressed the effects of HT induced after recirculation of focal brain ischemia on infarct size during long survival times. Adult male Wistar rats underwent intra-luminal occlusion of the left middle cerebral artery for 60 min followed by HT (39.0-39.5°C) or normothermia. Control procedures included none and sham surgery with and without HT, and middle cerebral artery occlusion alone. Part I: 6-h HT induced at recirculation. Part II: 2-h HT induced at 2-, 6-, or 24-h recirculation. Part III: 2-h HT initiated at recirculation or 6-h HT initiated at 2-, 6- or 24-h recirculation. Survival periods were 7 days, 2 or 6 months. The effects of post-ischemic HT on cortex and striatum were evaluated histopathologically by measuring the area of remaining tissue in the infarcted hemisphere at -0.30 mm from bregma. Six-hour HT initiated from 6-h recirculation caused a significant decrease in the remaining cortical tissue between 7-day (N = 8) and 2-month (N = 8) survivals (98.46 ± 1.14 to 73.62 ± 8.99%, respectively). When induced from 24-h recirculation, 6-h HT caused a significant reduction of the remaining cortical tissue between 2- (N = 8) and 6-month (N = 9) survivals (94.97 ± 5.02 vs 63.26 ± 11.97%, respectively). These data indicate that post-ischemic HT triggers chronic neurodegenerative processes in ischemic penumbra, suggesting that similar fever-triggered effects may annul the benefit of early recirculation in stroke patients over the long-term.

Besides other physiological functions, adenosine-5'-triphosphate (ATP) is also a neurotransmitter that acts on purinergic receptors. In spite of the presence of purinergic receptors in forebrain areas involved with fluid-electrolyte balance, the effect of ATP on water intake has not been investigated. Therefore, we studied the effects of intracerebroventricular (icv) injections of ATP (100, 200 and 300 nmol/µL) alone or combined with DPCPX or PPADS (P1 and P2 purinergic antagonists, respectively, 25 nmol/µL) on water intake induced by water deprivation. In addition, the effect of icv ATP was also tested on water intake induced by intragastric load of 12% NaCl (2 mL/rat), acute treatment with the diuretic/natriuretic furosemide (20 mg/kg), icv angiotensin II (50 ng/µL) or icv carbachol (a cholinergic agonist, 4 nmol/µL), on sodium depletion-induced 1.8% NaCl intake, and on food intake induced by food deprivation. Male Holtzman rats (280-320 g, N = 7-11) had cannulas implanted into the lateral ventricle. Icv ATP (300 nmol/µL) reduced water intake induced by water deprivation (13.1 ± 1.9 vs saline: 19.0 ± 1.4 mL/2 h; P < 0.05), an effect blocked by pre-treatment with PPADS, but not DPCPX. Icv ATP also reduced water intake induced by NaCl intragastric load (5.6 ± 0.9 vs saline: 10.3 ± 1.4 mL/2 h; P < 0.05), acute furosemide treatment (0.5 ± 0.2 vs saline: 2.3 ± 0.6 mL/15 min; P < 0.05), and icv angiotensin II (2.2 ± 0.8 vs saline: 10.4 ± 2.0 mL/2 h; P < 0.05), without changing icv carbachol-induced water intake, sodium depletion-induced 1.8% NaCl intake and food deprivation-induced food intake. These data suggest that central ATP, acting on purinergic P2 receptors, reduces water intake induced by intracellular and extracellular dehydration.

This study reports preliminary results of virus neutralizing antibody (VNA) titers obtained on different days in the course of human anti-rabies immunization with the 2-1-1 schedule (one dose is given in the right arm and one dose in the left arm at day 0, and one dose is apllied on days 7 and 21), recommended by WHO for post-exposure treatment with cell culture vaccines. A variant schedule (double dose on day zero and another on day 14) was also tested, both employing suckling mouse brain vaccine. A complete seroconversion rate was obtained after only 3 vaccine doses, and almost all patients (11 of 12) presented titers higher than 1.0 IU/ml. Both neutralizing response and seroconversion rates were lower in the group receiving only 3 doses, regardless of the sample collecting day. Although our results are lower than those found with cell culture vaccines, the geometry mean of VNA is fully satisfactory, overcoming the lower limit recommended by WHO of 0.5 IU/ml. The 2-1-1 schedule could be an alternative one for pre exposure immunization, shorter than the classical 3+1 regimen (one dose on days 0, 2, 4 and 30) with only three visits to the doctor, instead of four.

The purpose of this study was to determine the seroprevalence of human immunodeficiency virus (HIV-1/2), human T-cell lymphotropic virus (HTLV-I/II), hepatitis B virus (HBV), hepatitis C virus (HCV), Treponema pallidum and Trypanosoma cruzi among 63 male prisoners in Manhuaçu, Minas Gerais, Brazil and to compare this with data from eligible blood donors. The positive results were as follows: 11/63 (17.5%) for HBV, 5/63 (7.4%) for syphilis, 4/63 (6.3%) for HCV, 3/63 (4.8%) for Chagas' disease, 2/63 (3.2%) for HIV-1/2 and 1/63 (1.6%) for HTLV-I/II. The seroprevalence in prisoners was higher than among blood donors, mainly for antibodies to HIV-1/2, HCV and HBV. This is probably due to low social economic level, illiteracy, higher proportion with a prior history of intravenous drug use and/or unsafe sexual behavior. Therefore, these prisoners constitute a high risk group and routine screening and counseling are recommended.

Foram continuados os estudos da parte experimental do método da 2,2'-dipiridil cetoxima, cuja finalidade é o estabelecimento de uma técnica adequada para a determinação do cobalto em material vegetal. Estudos sobre o preparo, concentração e conservação do reativo, sobre a influência do pH na formação e extração do composto colorido e sobre o sistema tampão, já foram efetuados na etapa inicial. Nesta oportunidade, procurou-se estudar o solvente; sua seleção, a sensibilidade, o tempo de extração, a influência do contato entre as fases e a estabilidade da cor do composto.

Of the 110 dentists who had presented seroconversion 50 days after the intradermal application of three 2 µg doses of the Belgian recombinant vaccine against hepatitis B (HB), administered eight years before at an interval of one month between the 1st and 2nd doses and of five months between the 2nd and 3rd doses, 51 were included for the assessment of the persistence of immunity. None of the dentists had hepatitis or had received HB vaccine during this period. All subjects were submitted to serological tests for the detection of the following markers of hepatitis B virus (HBV) infection: HBsAg, anti-HBc, HBeAg, anti-HBe, and anti-HBs, with no HBsAg, anti-HBc, HBeAg or anti-HBe being detected. A microparticle enzyme immunoassay (MEIA) revealed the presence of anti-HBs at protective titers (> 10 mIU/ml) in 42 dentists (82.4%), with the anti-HBs titer being higher than 100 mIU/ml in 36 of them (70.6%) (good responders), between 10 and 100 mIU/ml in 6 (11.8%) (poor responders), and lower than 10 mIU/ml in 9 (17.6%) (non-responders). According to clinical data and serological tests, none of the dentists had presented disease or latent HBV infection during the eight years following the first vaccination. A 2 µg booster dose was administered intradermally to eight dentists with anti-HBs titers lower than 10 mIU/ml (non-responders) and to six dentists with titers ranging from 10 to 100 mIU/ml (poor responders); the determination of anti-HBs one month later demonstrated the occurrence of seroconversion in the eight non-responders and an increase in anti-HBs titer in the six poor responders. In summary, the present results demonstrated the prolonged persistence of protection against HBV infection and the development of immunologic memory provided by vaccination against HB - with intra-dermal application of three 2 µg doses of the Belgian recombinant vaccine at 0, 1, and 6 months - carried out eight years before in 51 dentists.

gamma-Hydroxy-alpha-diazo-beta-ketoesters are key intermediates in the chemistry of penicilin-based antibiotics and natural products. The method developed here for the synthesis of ethyl 2-diazo-4-hydroxy-3-oxo-butanoate 17 (in two steps from the diazo mercurial 2) compares very favorably with those reported in the literature for similar compounds. The Rh2(OAc)4-mediated intramolecular OH-insertion reaction of the diazo hydroxy ester 17 was investigated, furnishing the oxetan-3-one-2-carboxilate 18 in good yield. When the diazo ester lacks a free hydroxyl group as in the case of the phenoxy diazo ester 11 an intramolecular CH-insertion takes place, affording the 2H-chromene 20 in almost quantitative yield. The behavior of other functionalized diazo esters towards Rh2(OAc)4 was also investigated.
