Monocytes/macrophages are important targets for dengue virus (DENV) replication; they induce inflammatory mediators and are sources of viral dissemination in the initial phase of the disease. Apoptosis is an active process of cellular destruction genetically regulated, in which a complex enzymatic pathway is activated and may be trigged by many viral infections. Since the mechanisms of apoptotic induction in DENV-infected target cells are not yet defined, we investigated the virus-cell interaction using a model of primary human monocyte infection with DENV-2 with the aim of identifying apoptotic markers. Cultures analyzed by flow cytometry and confocal microscopy yielded DENV antigen positive cells with rates that peaked at the second day post infection (p.i.), decayed afterwards and produced the apoptosis-related cytokines TNF-α and IL-10. Phosphatidylserine, an early marker for apoptosis, was increased at the cell surface and the Fas death receptor was upregulated at the second day p.i. at significantly higher rates in DENV infected cell cultures than controls. However, no detectable changes were observed in the expression of the anti-apoptotic protein Bcl-2 in infected cultures. Our data support virus modulation of extrinsic apoptotic factors in the in vitro model of human monocyte DENV-2 infection. DENV may be interfering in activation and death mechanisms by inducing apoptosis in target cells.

Astrocytes play a vital role in neuronal protection, homeostasis, vascular interchange and the local immune response. Some viruses and parasites can cross the blood-brain barrier and infect glia. Trypanosoma cruzi, the aetiological agent of Chagas disease, can seriously compromise the central nervous system, mainly in immune-suppressed individuals, but also during the acute phase of the infection. In this report, the infective capacity of T. cruzi in a human astrocyte tumour-derived cell line was studied. Astrocytes exposed to trypomastigotes (1:10 ratio) produced intracellular amastigotes and new trypomastigotes emerged by day 4 post-infection (p.i.). At day 6 p.i., 93% of the cells were infected. Using flow cytometry, changes were observed in both the expression of major histocompatibility complex class I and II molecules and the chemokine secretion pattern of astrocytes exposed to the parasite. Blocking the low-density lipoprotein receptor on astrocytes did not reduce parasite intracellular infection. Thus, T. cruzi can infect astrocytes and modulate the immune response during central nervous system infection.

Eicosanoids affect the immunity of several pathogen/insect models, but their role on the Anopheles gambiae response to Plasmodium is still unknown. Plasmodium berghei-infected mosquitoes were injected with an eicosanoid biosynthesis inhibitor, indomethacin (IN), or a substrate, arachidonic acid (AA), at day 7 or day 12 post-infection (p.i.). Salivary gland invasion was evaluated by sporozoite counts at day 21 p.i. IN promoted infection upon sporozoite release from oocysts, but inhibited infection when sporozoites were still maturing within the oocysts, as observed by a reduction in the number of sporozoites reaching the salivary glands. AA treatment had the opposite effect. We show for the first time that An. gambiae can modulate parasite survival through eicosanoids by exerting an antagonistic or agonistic effect on the parasite, depending on its stage of development.

O objetivo deste trabalho foi avaliar o impacto da redução do inóculo no controle da podridão-'olho-de-boi' (POB) causada por Cryptosporiopsis sp. em maçãs e minimização de perdas. Os experimentos foram realizados em Vacaria-RS, nos ciclos 2006/07 e 2007/08. Na fase de dormência de macieiras Pink Lady®, foram aplicados os seguintes tratamentos erradicantes (dose p.i.): calda sulfocálcica (3,0%); oxicloreto de cobre (0,5%); hidróxido de cobre (0,3%); óxido cuproso (0,3%) e testemunha sem pulverização. A redução do inóculo foi avaliada em solução oriunda da lavagem de ramos, gemas floríferas e frutos, semeados em meio semisseletivo. Na colheita, a incidência da POB foi avaliada em quatro categorias de frutos: caídos sob a área de projeção da copa; nas plantas; infecções latentes, e após três meses de armazenamento em câmara frigorífica. Houve relação linear positiva entre os níveis de inóculo de Cryptosporiopsis sp. em gemas e os danos por POB (R² = 0,948; P = 0,001). O uso da calda sulfocálcica possibilitou a maior redução de inóculo nas gemas, com redução em 50% dos danos totais ocasionados por POB na safra de 2006/07. Nos frutos, o inóculo apresentou um incremento linear ao longo do ciclo, sendo relativamente maior em 2007/08, onde os tratamentos erradicantes possibilitaram controle superior a 40% nos danos com benefício econômico pela redução do inóculo.

A infecção genital de vacas pelo herpesvírus bovino tipo 1.2 (BoHV-1.2) pode resultar em vulvovaginite e infertilidade temporária. Após a infecção aguda, o BoHV-1 estabelece infecção latente, que pode cursar com episódios periódicos de reativação. O presente trabalho descreve os aspectos virológicos e clínico-patológicos da vulvovaginite aguda e infecção latente resultantes da inoculação de bezerras com uma amostra de BoHV-1.2 isolada de casos de balanopostite em touros. A inoculação do vírus em quatro bezerras pela via genital (10(8.1)TCID50/animal) resultou em replicação viral na mucosa genital e no desenvolvimento de vulvovaginite moderada a severa. Os animais inoculados excretaram o vírus nas secreções genitais até o dia 10 pós-inoculação (p.i.) com título máximo de 10(7.3)TCID50/mL. Foram observados congestão e edema da mucosa vulvovestibular, e formação de pequenas vesículas e pústulas. Durante a progressão clínica, as vesículas e pústulas aumentaram de tamanho e eventualmente se tornaram coalescentes e recobertas por um exsudato fino de coloração amarelada. Estes sinais foram observados a partir do dia 2 p.i. e aumentaram progressivamente de severidade até os dias 5-8 p.i. A administração de dexametasona no dia 55 p.i. resultou em excreção viral nas secreções genitais dos quatro animais por até 10 dias. A reativação da infecção latente foi acompanhada de recrudescência clínica, porém com sinais menos severos e com menor duração do que na infecção aguda. O DNA viral latente foi detectado por PCR, aos 36 dias pós-reativação (p.r.), nos seguintes tecidos: gânglio sacrais: pudendo (4/4); genitofemoral e retal caudal (3/4) e obturador (4/4) e em alguns linfonodos regionais. Estes resultados demonstram que o isolado SV-56/90 é virulento para fêmeas soronegativas, após inoculação genital, e pode ser utilizado em estudos de patogenia e de desafio vacinal.

Este estudo teve por objetivo avaliar o proteinograma e os teores de cobre, ferro e zinco no soro sangüíneo de ovelhas com mastite induzida por cepa de campo de Staphylococcus aureus. Foram utilizadas 10 ovelhas da raça Santa Inês, primíparas, recém-paridas, com aproximadamente dois anos de idade e bom estado nutricional. Inoculou-se na metade direita da glândula mamária 1,0x10(4) UFC/mL da bactéria, enquanto que a metade esquerda serviu como controle. Os animais foram acompanhados diariamente e a partir do diagnóstico clínico de mastite, procedeu-se colheita do material para realização do proteinograma sérico em gel de poliacrilamida contendo dodecil sulfato de sódio (SDS-PAGE) e para determinação do teor plasmático de fibrinogênio e das concentrações séricas de cobre, ferro e zinco em 16 momentos a saber: antes da inoculação (controle) e 12h, 24h, 36h, 48h, 60h, 72h, 84h, 96h, 108h, 120h, 132h, 168h, 180h, 288h e 336h após a inoculação (p.i.). Todas as ovelhas apresentaram quadro clínico de mastite, com perda da funcionalidade da glândula mamária. O proteinograma permitiu a identificação de 23 proteínas, cujos pesos moleculares (PM) variaram de 26.000 a 185.000 dáltons (Da), incluindo proteínas de fase aguda, IgG e IgA. Notou-se aumento significativo nas concentrações de haptoglobina e ceruloplasmina, assim como de IgG e IgA. Não se constatou alteração nos teores de antitripisina e de glicoproteína ácida .Verificou-se diminuição nos teores de ferro e zinco e elevação na concentração de cobre. Constatou-se correlação positiva entre o teor plasmático de fibrinogênio e as concentrações séricas de ceruloplasmina (r=0,74), a haptoglobina (r=0,62) e IgA(r=0,62). Estes resultados mostram a importância das proteínas de fase aguda ceruloplasmina e haptoglobina como indicadores auxiliares da infecção intramamária de ovelhas, assim como ratifica a relevância do fibrinogênio como marcador inflamatório em razão de sua alta correlação com as proteínas especificas. As alterações nas concentrações séricas de Cu, Fe e Zn sugerem a ação de mediadores inflamatórios, estimulados por S. aureus.

A infecção dos felinos pelo Vírus da Imunodeficiência Felina (FIV) resulta no desenvolvimento da síndrome de imunodeficiência dos felinos. Gengivite, perda de peso, linfadenomegalia generalizada, anemia, insuficiência renal crônica, complicações neurológicas, diarréia crônica e infecções bacterianas são encontradas frequentemente. A fase aguda da infecção pode ser assintomática, retardando o estabelecimento do diagnóstico e a implantação de medidas profiláticas para restringir o contágio e a transmissão do agente aos felinos suscetíveis. Com a finalidade de estudar as características clínicas da fase aguda da infecção, dez felinos jovens, sem definição racial, com oito meses de idade foram inoculados por via endovenosa com 1mL de sangue venoso de um gato portador do FIV subtipo B. A confirmação da infecção foi obtida através de teste sorológico em quatro e oito semanas pós-inoculação (p.i.) e por nested-PCR. Foram realizados hemogramas semanais, exame ultrassonográfico do abdômen quinzenais e exame oftalmológico mensal, durante doze semanas p.i. Discreta tendência a linfopenia na segunda semana p.i. e a neutropenia entre a quinta e sétima semana p.i., febre intermitente em alguns gatos, linfadenomegalia e hepato-esplenomegalia entre a quarta e a 12ª semana p.i. foram as alterações clínicas observadas. Apenas um gato apresentou uveíte unilateral direita. A fase aguda da infecção transcorreu com alterações clínicas inespecíficas. A linfadenomegalia e a hepato-esplenomegalia observadas no decorrer da infecção, refletindo hiperplasia linfóide, sugerem a necessidade de se realizar o teste sorológico para o FIV, em todos os gatos que se apresentarem com essas alterações, o que permitirá o diagnóstico precoce da infecção e a adoção de medidas profiláticas no sentido de minimizar a propagação da infecção.

Com o objetivo de se verificar a ação do butylate, aplicado isoladamente e em mistura com atrazine, no controle de plantas daninhas da cultura do milho, foi instalado um experimento de campo em solo fino areno-argiloso. Foram utilizados os seguintes tratamentos: butylate a 2,80; 3,60 e 4,32 kg/ha (p.p.i.); butylate + atrazine a 3,24 + 0,80; 3,24 + 1,20 e 3,60 + 0,96 kg/ha (p.p.i.); atrazine a 3,00 kg/ha e atrazine + metolachlor a 1,40 + 2,10 kg/ha, ambos aplicados em pré-emergência e empregados como herbicidas padrão para a cultura. As plantas daninhas encontradas foram: tiririca - Cyperus rotundus L., carurú comum - .:maranthus viridis L., capim de colchão - Digitaria sanguinalis (L.) Scop. e capim pé-de-galinha Eleusine indica (L.) Gaertn. Butylate nas três doses apresentou-se bem contra C. rotundus e E. indica; nas doses de 3,60 e 4,32 kg foram obtidos bons resultados sobre D. sanguinalis. Butylate + atrazine controlou, nas três doses, todas as espécies incidentes, o mesmo ocorrendo com a mistura atrazine + metolachlor. Atrazine foi mais eficiente para A. viridis e E. indica. Nas condições em que foi conduzido o experimento nenhum dos herbicidas foi prejudicial para a cultura.

Weeds have a negative influence on several fruit tree characteristics, such as yield, making it difficult to management practices in orchards. Alternative weed management methods, aiming to reduce the use of herbicides, have become attractive since herbicides are costly and cause environmental degradation. The use of cultivars with greater competitive ability against weeds has attracted international attention. The objective of this work was to evaluate the floristic composition and growth of weeds under the canopies of irrigated custard apple tree progenies. Twenty halfsibling progenies around three years of age were evaluated in a random block design with five replicates and four plants per plot. A circle with a 0.5 m² area was established around the trunk of each plant. Floristic composition, fresh matter, and dry matter mass of the above-ground part of the weeds, were evaluated in this area. Root collar and canopy diameters, as well as leaf area of the progenies were also evaluated. Fifty-eight weed species were recorded. The five weed families with the most species were Leguminosae, Convolvulaceae, Euphorbiaceae, Malvaceae and Sterculiaceae, in decreasing order. The number of weed species per plot ranged from 6 to 18, but there was no difference between the mean percentages of different weeds under the canopies of the progenies. The lowest weed fresh and dry matter masses occurred in progenies JG1 and SM8, respectively. There were no differences between progenies with regard to root collar diameter and leaf area; however, one of the lowest weed dry matter yields was observed under the canopy of progeny FE4, which showed the largest canopy diameter.

Herbicides have simplified weed control, but the use of herbicides, besides being costly, resulted in the selection of herbicide-resistant weed biotypes and has become an environmental contamination factor. Herbicide use reduction is one of the goals of modern agriculture, with several alternatives being investigated, including intercropping. The objective of this study was to evaluate the effects of cowpea and corn cultivar intercropping on weed control and corn green-ear (immature ears with 80% humidity grains) and grain yield. A completely randomized block design with split-plots and four replications was used. AG 1051, AG 2060 and PL 6880 corn cultivars (assigned to plots) were submitted to the four treatments: no weeding, two hoe-weeding (22 and 41 days after planting), and intercropping with cowpea (BR 14 and IPA 206 cultivars, with indeterminate growth). The cowpea was planted (with corn planting) between the corn rows, in pits 1.0 m apart, with two plants per pit. The corn cultivars did not differ from each other as to weed density (WD), fresh above-ground weed biomass (WB), green-ear yield and grain yields. Higher WD and WB mean values were found in no weeding subplots; lower mean values in two hoe-weeding subplots; and intermediate mean values in intercropped subplots, indicating that cowpea plants had, to a certain extent, control over weeds. The no-weeded plots and the intercropped plots had lower green-ear and grain yields. Although the cowpea cultivars had a certain control over weeds (mean reductions of 22.5 and 18.3%, in terms of green matter density and weight of the above-ground part of weeds, respectively), they also competed against the corn plants, leading to yield reduction (mean reductions of 17.0 and 32% in green ear and grain yield, respectively). The cowpea cultivars did not produce grain, certainly due to the strong competition exerted by the corn and weeds on cowpea plants.

The objectives of this work were to evaluate the floristic composition and dry biomass of weeds under the canopy of seven perennial species adapted to the Semi-Arid region of Brazil, and correlate these characteristics with growth traits of the perennial species. The following perennial species were evaluated in two experiments (E1 and E2): mesquite (Prosopis juliflora), jucá (Caesalpinia ferrea), white popinac (Leucaena leucocephala), mofumbo (Combretum leprosum), neem (Azadirachata indica), sabiá (Mimosa caesalpiniaefolia) and tamarind (Tamarindus indica). In E1, the seven species were evaluated in a random block design with four replicates and nine plants per plot. In E2, evaluation comprised four species (mesquite, jucá, white popinac, and tamarind) in a random block design with eight replicates and nine plants per plot. A circle with an area of 1.77 m² was established around the trunk of each plant, two years after they were transplanted to the permanent location. The weeds collected within this circle were cut even with the ground, classified and weighed. At this time, plant height, and crown and stem diameters were evaluated in all trees of each plot. In E1 there were no differences between tree species as to weed frequency under their canopies; however, weed growth was smaller under the canopy of sabiá trees. Mesquite and sabiá had the greatest plant height and crown diameter means, but only sabiá had the greatest stem diameter. In E2, the perennial species were not different with regard to weed frequency and growth under their canopies, but mesquite had the greatest growth, as measured by plant height (with significant results for jucá as well) and crown and stem diameter.

The reduction in herbicide use is one of the greatest interests for modern agriculture and several alternatives are being investigated with this objective, including the adoption of cultivars that suppress weeds. The objective of this study was to verify if maize cultivars develop differently, in competition with weeds, to produce green ears and grain. Randomized complete block design was used, with split-plots and five replications. Cultivars DKB 390, DKB 466, DKB 350, AG 7000, AG 7575 and Master, were evaluated in the plots, without weeding and two weedings (at 22 and 41 days after sowing) in sub plots. Twenty-one species were identified in the experimental area, the most frequent being Gramineae (Poaceae), Euphorbiaceae, Leguminosae (Fabaceae) and Convolvulaceae species. There was no difference in the dry biomass above-ground part of the weeds in the plots of the evaluated cultivars. The cultivars behaved similarly in treatments with or without hoeing, except for plant height and ear height evaluations. Without hoeing, plant height increased in cultivar DKB 390, while plant height and ear height decreased in cultivar AG 7575. In the other cultivars, these traits did not change under weed control. The presence of weeds decreased the values of all traits employed to assess green corn yield, with the exception of the total number of green ears and grain yield.

Although labor is intensive, evaluating the growth of crops may allow a better understanding of crop performance, including the reasons why certain cultivars can compete better with weeds. This study aims at evaluating growth, green ear yield, and grain yield in corn when in competition with weeds. Cultivars AG 1051 and BRS 106 were grown with (two hoeings, at 20 and 40 days after sowing) or without weed control. In order to evaluate crop growth, six collections of the above-ground part and the root system of corn were performed, every 15 days, with the first collection made 30 days after sowing. A randomized complete block design was adopted, with split-split plots (weed control in plots, cultivars in subplots, and collections in sub-subplots) and ten replicates. Eighteen weed species were found in the experiment area. Increased values of corn leaf area, above-ground part and root system, due to plant age function, were smaller in non-hoed plots than in hoed plots and were dependent upon cultivar. The lack of weed control increased dry matter of weeds aboveground part and decreased green ear yield and grain yield. Cultivar AG 1051 had higher increases in leaf area, above-ground part of the plant and root system, due to plant age function, and controlled weeds better than cultivar BRS 106. In addition, cultivar AG 1051 was superior to other cultivars with respect to most traits used for green corn yield and grain yield assessment.

The aim of this study was to determine the weed strip control (WSC) required for adequate coffee growth after transplanting. A non-irrigated, field-planted (spaced 3.80 x 0.70 m) crop was used. The experimental design was a randomized block, with four replicates. The treatments were arranged in a 9 x 18 split-plot design to test the WSC of 0, 15, 30, 45, 60, 90, 120, 150, and 190 cm, which involved continuously hand-weeding at each side of the coffee row, and 18 coffee growth measurements. Multiple regression analyses were carried out relating to growth-variables as a function of both WSC and growth-evaluation times. Brachiaria decumbens was the main weed accomplishing 88.5% of the total weed dry mass. The minimum width of the WSC increases as the crop ages after transplanting. Assuming reductions of 2% and 5% in the maximum coffee growth, the recommended WSC was 75 and 52 cm at 4 months after transplanting (MAT), 104 and 85 cm at 6 MAT, 123 and 105 cm at 9 MAT, 134 and 116 cm at 12 MAT, 142 and 124 cm at 15 MAT, and 148 and 131 cm at 18 MAT, respectively. It was concluded that integrated weed management in young coffee crops must focus on the weed control only in a minimum range along coffee rows, which increases with coffee plant age, keeping natural vegetation in the inter-rows.

Gliricidia (Gliricidia sepium) seedlings are usually beneficial to corn crops when planted between corn rows. The objective of this work was to assess the effects of corn intercropped with gliricidia and "sabiá" (Mimosa caesalpiniifolia), a species native to the Brazilian northeastern region, on weed control and corn green ear and grain yields. The experiment was carried out at Estação Experimental da Universidade Federal Rural do Semi-Árido - UFERSA (Mossoró, State of Rio Grande do Norte, Brazil). The experimental design consisted of randomized complete blocks (multifactorial design) with five replications, arranged in split-plots. The plots consisted of corn cultivars AG1051 and BM 2022; subplot treatments (six) were no-hoeing, twice-hoeing (at 20 and 40 days after sowing) and intercropping with gliricidia and "sabiá", either directly sown or transplanted, simultaneously with corn sowing. The intercropped leguminous plants were spaced 0.40 m from each other, and directly seeded or transplanted (30-day-old seedlings) in between two 1 m-spaced corn rows. Twenty three weed species were identified during the experiment. Gliricidia seedlings were superior to "sabiá" seedlings with regard to plant height and survival rate. The highest corn green ear and grain yields were found for twice-hoed subplots, while the lowest yield was found for no-hoed or intercropped subplots. However, grain yield values in intercropped treatments did not differ from grain yield values in hoed plots. In addition, marketable husked green ear mean weights did not differ between twice-hoed subplots and subplots directly seeded with gliricidia and "sabiá". Such results indicated that corn benefited from the intercropping system, but intercropping with gliricidia and "sabiá" transplanted resulted in lower benefits than with the direct sowing of those species.
