A new orally absorbable triazole (Schering 39304) with a long serum half-life in man (60 hours), was tried in a murine model of progressive paracoccidioidomycosis and compared with itraconazole, another triazole which has proven effective in this mycosis. Only 15% of the infected, untreated mice survived while 53 to 75% of the animals receiving itraconazole survived. Mice treated with Schering 39304 exhibited higher (86 - 100%) survival rates. Statistically, the 5 mg/kg Sch 39304 was superior to the 50 mg/kg itraconazole dose. Lung cultures showed that 20 mg/kg/day of Sch achieved sterilization of the infectious foci. These results indicate that the new triazole will have a place in the treatment of paracoccidioidomycosis

The recommended treatment for latent tuberculosis (TB) infection in adults is a daily dose of isoniazid (INH) 300 mg for six months. In Brazil, INH was formulated as 100 mg tablets. The treatment duration and the high pill burden compromised patient adherence to the treatment. The Brazilian National Programme for Tuberculosis requested a new 300 mg INH formulation. The aim of our study was to compare the bioavailability of the new INH 300 mg formulation and three 100 mg tablets of the reference formulation. We conducted a randomised, single dose, open label, two-phase crossover bioequivalence study in 28 healthy human volunteers. The 90% confidence interval for the INH maximum concentration of drug observed in plasma and area under the plasma concentration vs. time curve from time zero to the last measurable concentration “time t” was 89.61-115.92 and 94.82-119.44, respectively. The main limitation of our study was that neither adherence nor the safety profile of multiple doses was evaluated. To determine the level of INH in human plasma, we developed and validated a sensitive, simple and rapid high-performance liquid chromatography-tandem mass spectrometry method. Our results showed that the new formulation was bioequivalent to the 100 mg reference product. This finding supports the use of a single 300 mg tablet daily strategy to treat latent TB. This new formulation may increase patients’ adherence to the treatment and quality of life.

Infiltration is the passage of water through the soil surface, influenced by the soil type and cultivation and by the soil roughness, surface cover and water content. Infiltration absorbs most of the rainwater and is therefore crucial for planning mechanical conservation practices to manage runoff. This study determined water infiltration in two soil types under different types of management and cultivation, with simulated rainfall of varying intensity and duration applied at different times, and to adjust the empirical model of Horton to the infiltration data. The study was conducted in southern Brazil, on Dystric Nitisol (Nitossolo Bruno aluminoférrico húmico) and Humic Cambisol (Cambissolo Húmico alumínico léptico) soils to assess the following situations: simulated rains on the Nitisol from 2001 to 2012 in 31 treatments, differing in crop type, sowing direction, type of soil opener on the seeder, amount and type of crop residue and amount of liquid swine manure applied; on the Cambisol, rains were simlated from 2006 to 2012 and 18 treatments were evaluated, differing in crop, seeding direction and crop residue type. The constant of the water infiltration rate into the soil varies significantly with the soil type (30.2 mm h-1 in the Nitisol and 6.6 mm h-1 in the Cambisol), regardless of the management system, application time and rain intensity and duration. At the end of rainfalls, soil-water infiltration varies significantly with the management system, with the timing of application and rain intensity and duration, with values ranging from 13 to 59 mm h-1, in the two studied soils. The characteristics of the sowing operation in terms of relief, crop type and amount and type of crop residue influenced soil water infiltration: in the Nitisol, the values of contour and downhill seeding vary between 27 and 43 mm h-1, respectively, with crop residues of corn, wheat and soybean while in the Cambisol, the variation is between 2 and 36 mm h-1, respectively, in soybean and corn crops. The Horton model fits the values of water infiltration rate into the soil, resulting in the equation i = 30.2 + (68.2 - 30.2) e-0.0371t (R2 = 0.94**) for the Nitisol and i = 6.6 + (64.5 - 6.6) e-0.0537t (R2 = 0.99**) for the Cambisol.

Two high performance liquid chromatography (HPLC) methods for the quantitative determination of indinavir sulfate were tested, validated and statistically compared. Assays were carried out using as mobile phases mixtures of dibutylammonium phosphate buffer pH 6.5 and acetonitrile (55:45) at 1 mL/min or citrate buffer pH 5 and acetonitrile (60:40) at 1 mL/min, an octylsilane column (RP-8) and a UV spectrophotometric detector at 260 nm. Both methods showed good sensitivity, linearity, precision and accuracy. The statistical analysis using the t-student test for the determination of indinavir sulfate raw material and capsules indicated no statistically significant difference between the two methods.

Ilex paraguariensis (yerba-mate) is used as a beverage, and its extract requires adequate quality control methods in order to guarantee quality and safe use. Strategies to develop and optimize a chromatographic method to quantify theobromine, caffeine, and chlorogenic acid in I. paraguariensis extracts were evaluated by applying a quality by design (QbD) model and ultra high-performance liquid chromatography (UHPLC). The presence of these three phytochemical markers in the extracts was evaluated using UHPLC-MS and was confirmed by the chromatographic bands in the total ion current traces (m/z of 181.1 [M+H]+, 195.0 [M+H]+, and 353.0 [M−H]−, respectively). The developed method was then transferred to a high-performance liquid chromatography (HPLC) platform, and the three phytochemical markers were used as external standards in the validation of a method for analyses of these compounds in extracts using a diode array detector (DAD). The validated method was applied to quantify the chlorogenic acid, caffeine, and theobromine in the samples. HPLC-DAD chromatographic fingerprinting was also used in a multivariate approach to process the entire data and to separate the I. paraguariensis extracts into two groups. The developed method is very useful for qualifying and quantifying I. paraguariensis extracts.

This paper presents an experimental research about the behavior of two-phase flows in inclined pipes. The inclination angle varied from 5° to 45° and the slurry solid concentration varied up to 15%. It was concluded that the head losses of the downward sloping pipe flow are always lower than the head losses of the horizontal flow and these are always lower than the head losses of the upward sloping pipe flow, regardless the concentration and inclination angle. It was possible to develop empirical equations to calculate the head losses of the horizontal flow and the upward and downward sloping pipe flows.

This article discusses, from the standpoint of cellular biology, the deterministic and indeterministic androgenesis theories. The role of the vacuole and of various types of stresses on deviation of the microspore from normal development and the point where androgenetic competence is acquired are examined. Based on extensive literature review and data on wheat studies from our laboratory, a model for androgenetic capacity of pollen grain is proposed. A two point deterministic model for in vitro androgenesis is our proposal for acquisition of androgenetic potential of the pollen grain: the first switch point would be early meiosis and the second switch point the uninucleate pollen stage, because the elimination of cytoplasmatic sporophytic determinants takes place at those two strategic moments. Any abnormality in this process allowing the maintenance of sporophytic informational molecules results in the absence of establishment of a gametophytic program, allowing the reactivation of the embryogenic process
