O presente estudo objetivou descrever as lesões pulmonares macro e microscópicas associadas ao parasitismo por Sebekia oxycephala em 100 espécimes de jacarés-açu (Melanosuchusniger), abatidos na Reserva de Desenvolvimento Sustentável Mamirauá, situada no Estado do Amazonas, Brasil. Durante a avaliação macroscópica, exemplares dos parasitos encontrados no tecido pulmonar foram coletados individualmente em AFA (Álcool etílico - Formalina - Ácido acético glacial) e formol a 5% para avaliação parasitológica e classificação taxonômica. Amostras de tecido pulmonar de todos os animais, independentemente da ocorrência de lesões macroscópicas, foram fixadas em formol 10% e incluídas em parafina. Secções histológicas coradas por Hematoxilina-Eosina destas amostras foram avaliadas por meio de microscopia óptica. Macroscopicamente, 4 dos 100 animais (4%) apresentaram espécimes de pentastomídeos no parênquima pulmonar. Os parasitos foram classificados taxonomicamente como pertencentes à espécie Sebekia oxycephala. Nenhuma alteração macroscópicas foi observada, porém, microscopicamente, lesões pulmonares foram encontradas em 37% dos casos, sendo que, lesões inflamatórias associadas ao parasitismo corresponderam a 75,6% dos mesmos (28/37). Nestes, segmentos degenerados e ovos de S. oxycephala encontravam-se envolvidos por cápsula de tecido conjuntivo fibroso e infiltrado inflamatório predominantemente composto por células gigantes do tipo corpo estranho. Três espécimes apresentaram espessamento de septos alveolares e sete exemplares continham infiltrado inflamatório granulocítico multifocal no parênquima pulmonar. As lesões associadas ao parasitismo, de modo geral, apresentaram intensidade discreta e parecem não representar uma causa importante de doença pulmonar entre a população estudada. Esta é a primeira descrição de lesões pulmonares em M. niger associadas ao parasitismo por S. oxycephala na Amazônia brasileira.

In this work, a protocol for the formulation of an enzyme concentrated product to be applied in fruit juice treatment is described. Downstream processing conditions for the recovery and concentration of pectinases produced by the new strain Aspergillus niger LB-02-SF in solid state cultivation were assessed. The solid-liquid ratio in the extraction step of pectinases recovery from the cultivated media was evaluated and the highest activity was obtained with a solid-liquid ratio of 1:10. The crude extract was concentrated by ultrafiltration and the total pectinase (TP) activity was 73.6-fold concentrated in relation to the crude extract, and a final TP titer of 663 U mL–1 was obtained with 73.7% of recovery yield. KCl and different glycerol concentrations were added to the concentrated extract and the stability of pectinases during the storage at 5°C for 59 weeks was tested. The formulation with 50% w/w glycerol was applied to the treatment of apple and grape juices and the results of these tests were statistically comparable to those obtained with two high-quality commercial preparations.

A fístula oroantral é a comunicação patológica entre cavidade oral e seio maxilar, quase sempre decorrentes de traumatismos durante procedimentos dentários. OBJETIVO: Apresentar experiência de 25 casos. CASUÍSTICA E MÉTODOS: Realizado estudo retrospectivo de pacientes com FOA no período de 1996 a 2000. O diagnóstico incluiu exame otorrinolaringológico, endoscopia nasal ou da fístula, tomografia computadorizada das cavidades paranasais, pesquisa bacteriológica, fúngica e análise patológica. RESULTADOS: Encontrados 25 casos, sendo 10 de segundo molar, 8 de primeiro molar, 6 de segundo pré-molar e 1 de canino. Todos foram operados pela técnica de Caldwell-Luc, reavivamento das bordas da fístula, meatotomia média e rotação de retalho mucoso geniano. DISCUSSÃO: Nas fístulas de alto débito (n=14), colocou-se enxerto ósseo da própria parede anterior do seio. Todos, exceto um, tiveram resultado cirúrgico bom. A cultura bacteriológica (n=19) demonstrou estreptococos pneumoniae (13), haemophilus influenzae (6), moraxella catarrhalis (2), estafilococos aureus (2). Encontrado aspergilus niger em um caso que apresentava imagem radiológica de bola fúngica. CONCLUSÕES: Após 30 dias, os resultados foram bons em todos, exceto um dos casos. Este foi reoperado com colocação de enxerto ósseo, inicialmente não utilizado, tendo sucesso. Após 6 meses, todos os 23 pacientes localizados não apresentavam problemas.

The objective of this study was to evaluate the effect of the fungus Aspergillus niger strain CCT4355 in the release of nutrients contained in two types of rock powder (diabase and phonolite) by means of in vitro solubilization trials. The experimental design was completely randomized in a 5 x 4 factorial design with three replications. It was evaluated five treatments (phonolite dust + culture medium; phonolite dust + fungus + culture medium; diabase powder + culture medium; diabase powder + fungus + culture medium and fungus + culture medium) and four sampling dates (0, 10, 20 and 30 days). Rock dust (0.4% w/v) was added to 125 mL Erlenmeyer flasks containing 50 mL of liquid culture medium adapted to A. niger. The flasks were incubated at 30°C for 30 days, and analysis of pH (in water), titratable acidity, and concentrations of soluble potassium, calcium, magnesium, zinc, iron and manganese were made. The fungus A. niger was able to produce organic acids that solubilized ions. This result indicates its potential to alter minerals contained in rock dust, with the ability to interact in different ways with the nutrients. A significant increase in the amount of K was found in the treatment with phonolite dust in the presence of the fungus. The strain CCT4355 of A. niger can solubilize minerals contained in these rocks dust.

Pela importância da garantia de esterilidade em produto médico-hospitalares e pela predominância da forma tubular dentre estes produtos, conduziu-se o estudo de monitores biológicos em que os corpos de prova apresentavam diferentes comprimentos e diâmetros. O sensor biológico foi constituído de esporos de Bacillus subtilis var. niger adquirido do mercado nacional sob tiras de papel. Após esterilização por processo industrial, seguiu-se a recuperação dos esporos sobreviventes através de inoculação dos suportes em caldo tioglicolato, caldo caseína-soja e este adicionado de azul de bromotimol. A capacidade promotora de crescimento destes 3 meios de cultura não apresentou diferenças. A eficácia esterilizante foi dependente do tamanho de corpos de prova. A determinação periódica do teor residual de gás nos corpos de prova, bem como a monitoração ambiental industrial indicaram a importância da legislação em vigor, devendo ser obedecida a fim de garantir inocuidade aos pacientes e operadores.

In view of the lack of researches on otomycoses in Brazil, we have tried to study their incidence, their clinical characteristics and the predisponent factors During one year, 22 suspected cases were seen, 20 of them corresponded to otomycosis infections. The most frequent species were Aspergillus niger (35%) and Candida albicans (20%). The genus Aspergillus represented 75% of the isolates. Itching and hyperaemia (70%), otalgia (65%), hipoacusia (50%) were the commonest signs. Lack of cerumen (70%) chronic otitis (30%) previous antibiotic therapy and eczema (25%) were the most outstanding predisponent factors.

The purpose of the present study is to compare the tomato juice agar, a well known medium employed to observe ascospore formation, with niger seed agar, casein agar and sunflower seed agar, applied to a differentiation between C. dubliniensis and C. albicans. After 48 hours of incubation at 30 ºC all 26 (100%) C. dubliniensis isolates tested produced chlamydospores on tomato juice agar as well as in the other three media evaluated. However, when we inoculated all media with C. albicans, the absence of chlamydospores became resulting in the following percents: tomato juice agar (92.47%), niger seed agar (96.7%), casein agar (91.39%), and sunflower seed agar (96.7%). These results indicate that tomato juice agar is another medium which can also be used in the first phenotypic differentiation between C. dubliniensis and C. albicans.

Cryptococcus neoformans is an encapsulated yeast, etiological agent of cryptococcosis. The species is commonly associated with pigeon droppings and plant materials. The aim of the present work was to verify the presence of the yeast in pigeon droppings, and to identify the isolates obtained in serotypes and mating types (MAT). Ten samples of pigeon droppings were collected in the rural area of the city of Alfenas, Brazil. Samples were inoculated in agar Niger medium for fungal isolation and 22 isolates with characteristics of C. neoformans were obtained. The serotypes and MAT were determined by multiplex PCR using specific primers. Serotypes were also determined by using the Kit Crypto Check. Among the 22 samples evaluated, eight were identified as C. neoformans by classic identification tests. These samples were characterized as serotype A by the Kit Crypto check and as serotype A MAT alpha by the multiplex PCR. The present study reinforces the evidence that pigeon droppings are a reservoir for C. neoformans and confirms the prevalence of C. neoformans var. grubii (Aalpha) among environmental isolates. It also demonstrates that multiplex PCR is an acceptable alternative for serotype analysis because it reduces the costs for each reaction and analyses serotype and MAT simultaneously.

SUMMARYCryptococcosis is a severe systemic mycosis caused by two species of Cryptococcus that affect humans and animals: C. neoformans and C. gattii. Cosmopolitan and emergent, the mycosis results from the interaction between a susceptible host and the environment. The occurrence of C. neoformanswas evaluated in 122 samples of dried pigeon excreta collected in 49 locations in the City of Cuiabá, State of Mato Grosso, Brazil, including public squares (n = 5), churches (n = 4), educational institutions (n = 3), health units (n = 8), open areas covered with asbestos (n = 4), residences (n = 23), factory (n = 1) and a prison (n = 1). Samples collected from July to December of 2010 were seeded on Niger seed agar (NSA). Dark brown colonies were identified by urease test, carbon source assimilation tests and canavanine-glycine-bromothymol blue medium. Polymerase chain reaction primer pairs specific for C. neoformans were also used for identification. Cryptococcus neoformans associated to pigeon excreta was isolated from eight (6.6%) samples corresponding to six (12.2%) locations.Cryptococcus neoformans was isolated from urban areas, predominantly in residences, constituting a risk of acquiring the disease by immunocompromised and immunocompetent individuals.

The identification of the fungal species belonging to the healthy microflora in animals is a precondition for the recognition of pathological processes causing them. The aim of this study was to investigate the presence of potentially pathogenic fungi in the feces of wild birds collected in Screening Centers. Samples were collected from the feces of 50 cages with different species of birds. The samples were processed according to the modified method STAIB and the plates incubated at 32 °C for up to ten days with daily observation for detection of fungal growth. The isolation of the following species was observed: Malassezia pachydermatis, Candida albicans, C. famata, C. guilliermondii, C. sphaerica, C. globosa, C. catenulata, C. ciferri, C. intermedia, Cryptococcus laurentii, Trichosporon asahii, Geotrichum klebahnii, Aspergillus spp., A. niger and Penicillium spp. Knowing the character of some opportunistic fungi is important in identifying them, facilitating the adoption of preventive measures, such as proper cleaning of cages, since the accumulation of excreta may indicate a risk for both health professionals and centers for screening public health.

Criptococose é considerada a infecção fúngica sistêmica oportunista mais comum em pacientes com AIDS. Nestes pacientes tem predominado como agente etiológico Cryptococcus neoformans var. neoformans, e muito raramente relata-se C. neoformans var. gattiii, mesmo nas regiões onde se verifica a sua prevalência. Foram estudados 50 pacientes com lesões de criptococose meningoencefálica associada com AIDS. Os isolados foram identificados através de características microscópicas e macroscópicas exibidas em meios de ágar Sabouraud, ágar niger e Christensen. As variedades de C. neoformans foram determinadas pela reação de coloração obtida em meio de L-canavanina glicina-azul de bromotimol (CGB). Em todos os pacientes examinados foram isolados C. neoformans, sendo identificados C. neoformans var. neoformans em 47 isolados e C. neoformans var. gattii em 3. Os resultados encontrados mostram que a criptococose em pacientes com AIDS pode também ser causada por C. neoformans var. gatti, apesar de haver predominância de C. neoformans var neoformans nesta população.

Com intuito de identificar a microbiota fúngica em condicionadores de ar nas unidades de terapia intensiva de hospitais públicos e particulares de Teresina-PI, coletou-se material sólido de dez UTIs, isolando 33 espécies pertencentes às Moniliaceae e Dematiaceae, sendo primeira referência para o Piauí. Registrou-se elevada freqüência de Aspergillus niger Van Tieghem (60%); Aspergillus fumigatus Fres (50%); Trichoderma koningii Oudem (50%), Aspergillus flavus Link: Fr (40%). A validade da limpeza dos condicionadores de ar ultrapassou em todas as UTIs, a quantidade de unidades formadoras de colônia estava além do permitido pela Portaria 176/00 do Ministério da Saúde. É importante que os profissionais estejam munidos de equipamento de proteção individual, além de adotar medidas de controle de infecção hospitalar, sensibilizar para a existência de infecções fúngicas, melhorar ventilação de ar, possibilitando arejamento do ambiente e limpar periodicamente os condicionadores de ar, conscientizando os profissionais de saúde da importância destes fungos no ambiente hospitalar.

INTRODUÇÃO: Onicomicoses são comuns em pacientes imunocomprometidos embora espécies emergentes tenham sido verificadas, modificado o perfil epidemiológico desta micose. Assim, o objetivo desta pesquisa é avaliar o perfil clínico e micológico da onicomicose em pacientes com infecção pelo HIV/AIDS. MÉTODOS: Amostras clínicas foram coletadas, processados para exame direto e a cultura mantida a temperatura de 30°C e 37ºC durante 15 dias. RESULTADOS: Dos 100 pacientes, 32 apresentavam onicomicose. Os agentes isolados foram Candida albicans, C. parapsilosis, C. tropicalis, C. guilliermondii, Trichophyton rubrum, T. mentagrophytes, Fusarium solani, Scytalidium hialinum, S. japonicum, Aspergillus niger, Cylindrocarpon destructans e Phialophora reptans. CONCLUSÕES: Onicomicoses em HIV/AIDS apresentam variadas manifestações clínicas e podem ser causadas por fungos emergentes. As peculiaridades apresentadas pelos diferentes agentes de origem fúngica justificam a necessidade de identificação ao nível da espécie, com a finalidade de orientar uma melhor abordagem terapêutica e minimizar a exposição desses pacientes a condições de risco de uma infecção disseminada.

INTRODUCTION: Melanin production by species of Cryptococcus is widely used to characterize C. neoformans complex in mycology laboratories. This study aims to test the efficacy of methyldopa from pharmaceutical tablet as a substrate for melanin production, to compare the production of melanin using different agar base added with methyldopa, and to compare the melanin produced in those media with that produced in Niger seed agar and sunflower seed agar by C. neoformans, C. laurentii, and C. albidus. Two isolates of each species, C. neoformans, C. laurentii, and C. albidus, and one of Candida albicans were used to experimentally detect conditions for melanin production. METHODS: The following media were tested: Mueller-Hinton agar (MHA), brain and heart infusion agar (BHIA), blood agar base (BAB), and minimal medium agar (MMA), all added with methyldopa, and the media Niger seed agar (NSA) and sunflower seed agar (SSA). RESULTS: All isolates grew in most of the culture media after 24h. Strains planted on media BAB and BHIA showed growth only after 48h. All isolates produced melanin in MMA, MHA, SSA, and NSA media. CONCLUSIONS: Methyldopa in the form pharmaceutical tablet can be used as a substrate for melanin production by Cryptococcus species; minimal medium plus methyldopa was more efficient than the BAB, MHA, and BHIA in the melanin production; and NSA and SSA, followed by MMA added with methyldopa, were more efficient than other media studied for melanin production by all strains studied.

Introduction The incidence of opportunistic fungal infections has increased in recent years and is considered an important public health problem. Among systemic and opportunistic mycoses, cryptococcosis is distinguished by its clinical importance due to the increased risk of infection in individuals infected by human immunodeficiency virus. Methods To determine the occurrence of pathogenic Cryptococcus in pigeon excrement in the City of Araraquara, samples were collected from nine environments, including state and municipal schools, abandoned buildings, parks, and a hospital. The isolates were identified using classical tests, and susceptibility testing for the antifungal drugs (fluconazole, itraconazole, voriconazole, and amphotericin B) independently was also performed. After collection, the excrement samples were plated on Niger agar and incubated at room temperature. Results A total of 87 bird dropping samples were collected, and 66.6% were positive for the genus Cryptococcus. The following species were identified: Cryptococcus neoformans (17.2%), Cryptococcus gattii (5.2%), Cryptococcus ater (3.5%), Cryptococcus laurentti (1.7%), and Cryptococcus luteolus (1.7%). A total of 70.7% of the isolates were not identified to the species level and are referred to as Cryptococcus spp. throughout the manuscript. Conclusions Although none of the isolates demonstrated resistance to antifungal drugs, the identification of infested areas, the proper control of birds, and the disinfection of these environments are essential for the epidemiological control of cryptococcosis.
