Hepatic viscerotomy of paraffin-preserved old specimens, collected in the period from 1934 to 1967, were analyzed by immunohistochemical assays to detect hepatitis B, hepatitis D, dengue and yellow fever virus antigens. The material belongs to the Yellow Fever Collection, Department of Pathology, Instituto Oswaldo Cruz, Rio de Janeiro, Brazil and the cases were diagnosed at that time according to clinical aspects and histopathological findings reporting viral hepatitis, yellow fever, focal necrosis and hepatic atrophy. From the 79 specimens, 69 were collected at the Labrea Region and the other 10 in different other localities in the Amazon Region. The five micra thick histological slices were analyzed for the presence of hepatitis B surface antigen (HBsAg) and hepatitis B core antigen (HBcAg) by immunoperoxidase technique. An immunofluorescence assay was applied to the detection of hepatitis D, yellow fever and dengue virus antigens. Nine (11.4%) histological samples were HBsAg reactive and 5 (6.3%) were HBcAg reactive. The oldest reactive sample was from 1934. Viral antigens related to the other pathologies were not detected in this study. Our results confirm that the methodology described may be used to elucidate the aetiology of hepatitis diseases even after a long time of conservation of the specimens.

Biomphalaria amazonica Paraense, 1996 was collected from a permanent pond in the outskirts of the Bolivian city of Santa Cruz. Identification of the collected specimens was made by comparison with the original description of the species and with topotypic material in the collection of Instituto Oswaldo Cruz. Phylogenetic analysis confirmed that these Bolivian specimens belong to B. amazonica.

This study estimated the prevalence and distribution of human papillomavirus (HPV) types among women with cervical intraepithelial neoplasia (CIN) grade III and invasive cervical cancer from Goiás (Brazil Central Region). Seventy-four cases were analyzed and consisted of 18 CIN III, 48 squamous cell carcinomas, 4 adenocarcinomas, 1 adenosquamous carcinoma and 3 undifferentiated carcinomas. HPV-DNA sequences were examined in formalin-fixed and paraffin-embedded tissues using primers from L1 region GP5+/GP6+. Polymerase chain reaction products were typed with dot blot hybridization using probes for HPV 16, 18, 31, 33, 45, 54, 6/11, 42/43/44, 51/52, 56/58. The prevalence of HPV was estimated to be 76% (56/74). HPV 16 was the most frequently found type, followed by HPV 33, 18 and 31. The prevalence of untyped HPV was 6%; 79% percent of the squamous cell carcinoma cases and 61% percent of the CIN III were positive for HPV and the prevalence rate of HPV types was the same for the total number of cases. According to other studies, HPV type 16 is the most prevalent virus in all Brazilian regions, but there is variation regarding to other types. Type 18 is the second most prevalent HPV in North, Southeast and South Brazil regions and types 31 and 33 are the second most prevalent HPV in Northeast and Central Brazil, respectively.

Twenty-two vertically human immunodeficiency virus type 1 (HIV-1) infected Brazilian children were studied for antiretroviral drug resistance. They were separated into 2 groups according to the administration of antiretroviral therapy into those who presented disease symptoms or without symptoms and no therapy. Viral genome sequencing reactions were loaded on an automated DNA sampler (TruGene, Visible Genetics) and compared to a database of wild type HIV-1. In the former group 8 of 12 children presented isolates with mutations conferring resistance to protease inhibitors (PIs), 7 presented isolates resistant to nucleoside reverse transcriptase inhibitors (NRTIs) and 2 presented isolates resistant to non-nucleoside reverse transcriptase inhibitors (NNRTIs). Ten children were included in the antiretroviral naïve group. Eight were susceptible to NRTIs and all of them were susceptible to PIs; one presented the V108I mutation, which confers low-level resistance to NNRTIs. The data report HIV mutant isolates both in treated and untreated infants. However, the frequency and the level of drug resistance were more frequent in the group receiving antiretroviral therapy, corroborating the concept of selective pressure acting on the emergence of resistant viral strains. The children who presented alterations at polymorphism sites should be monitored for the development of additional mutations occurring at relevant resistance codons.

Lymnaea cousini Jousseaume, 1887 was collected in Mucubaji, Merida State, Venezuela, from a permanent pond located at a very high altitude (3760 m). Identification of the collected specimens was made by comparison with the original description of the shell by Jousseaume and the description of the renal organ and reproductive system of topotypic specimens by Paraense.

Several anatomical parameters of the reproductive system have been used to distinguish Lymnaea cubensis from L. viatrix, the snail hosts of fascioliasis in South America and the Caribbean area. Three samples have been collected in the type localities of L. cubensis (Cuba), L. viatrix var. A ventricosa (Argentina, Río Negro Lower Valley), and L. viatrix var. B elongata (Peru, Lima), respectively. Only one parameter, the relative lengths of the penis sheath and preputium, showed significant differences between L. viatrix var. ventricosa and the two other taxa. None of the studied parameters separated L. cubensis from L. viatrix var. elongata.

Until recently, Toxoplasma gondii was considered to be clonal with little genetic variability. In this paper we summarize recent genotyping data from chickens in Brazil, and pigs, lambs and white-tailed deer in the USA, to demonstrate the high genetic diversity and geographical distribution of T. gondii. A total of 149 T. gondii isolates from 13 geographical areas of Brazil and 182 T. gondii isolates from pigs, 53 isolates from sheep and 15 isolates from fetal white-tailed deer from USA were genotyped using the 10 RCFP-PCR genetic markers SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1 and Apico. Genotyping of 149 T. gondii isolates from free range chickens in Brazil identified 58 genotype groups. No clonal type II lineage was found. Of the 253 isolates from animals from USA, 18 genotypes were identified, predominantly type II. These studies indicate a higher genetic diversity than previously recognized.

The oocyst wall of coccidian parasites is a robust structure that is resistant to a variety of environmental and chemical insults. This resilience allows oocysts to survive for long periods, facilitating transmission from host to host. The wall is bilayered and is formed by the sequential release of the contents of two specialized organelles - wall forming body 1 and wall forming body 2 - found in the macrogametocyte stage of Coccidia. The oocyst wall is over 90% protein but few of these proteins have been studied. One group is cysteine-rich and may be presumed to crosslink via disulphide bridges, though this is yet to be investigated. Another group of wall proteins is rich in tyrosine. These proteins, which range in size from 8-31 kDa, are derived from larger precursors of 56 and 82 kDa found in the wall forming bodies. Proteases may catalyze processing of the precursors into tyrosine-rich peptides, which are then oxidatively crosslinked in a reaction catalyzed by peroxidases. In support of this hypothesis, the oocyst wall has high levels of dityrosine bonds. These dityrosine crosslinked proteins may provide a structural matrix for assembly of the oocyst wall and contribute to its resilience.

The aim of this study was to determine the incidence of congenital toxoplasmosis (CT) and to assess the performances of prenatal and neonatal diagnoses. From 1994-2005, in Toulouse University Hospital, France, amniocentesis was performed on 352 pregnant women who were infected during pregnancy. All women were treated with spiramycin and pyrimethamine-sulfadoxine when prenatal diagnosis was positive. Among the 275 foetuses with follow-up, 66 (24%) were infected. The transmission rates of Toxoplasma gondii were 7%, 24% and 59% in the first, second and third trimesters, respectively. The sensitivity and specificity of PCR on amniotic fluid (AF) were 91% and 99.5%, respectively. One case was diagnosed by mouse inoculation with AF and six cases were diagnosed by neonatal or postnatal screening. The sensitivity and specificity of PCR on placentas were 52% and 99%, respectively. The sensitivity of tests for the detection of specific IgA and IgM in cord blood was 53% and 64%, respectively, and specificity values were 91% and 92%. In conclusion, PCR performed on AF had the highest levels of sensitivity and specificity for the diagnosis of CT. This permits an early diagnosis of most cases and should be recommended.

An extensive malacological survey was carried out between 2005-2009 in order to clarify the exact number of lymnaeid species which may be intermediate hosts of Fasciola hepatica in Venezuela. Four species were discovered during this survey, including two local species: Lymnaea cubensis and Lymnaea cousini and two exotic species: Lymnaea truncatula and Lymnaea columella. The most common local species was L. cubensis which was found at 16 out of the 298 sampling sites. This species has a large distribution area throughout the Northern part of Venezuela and was encountered from sea level to an altitude of 1,802 m in state of Trujillo. The second local species L. cousini was collected at only two sites of the Andean Region at altitudes of 3,550 m and 4,040 m, respectively. The European L. truncatula was found at 24 sites all located in the states of Mérida and Táchira at an altitude varying between 1,540-4,000 m. The respective distribution areas of L. cubensis and L. truncatula do not appear to overlap, but more detailed malacological surveys are needed. The fourth lymnaeid species, L. columella was collected in a canal from Mérida at an altitude of 1,929 m and in an irrigation canal from the state of Guárico, at an altitude of 63 m. The role of these four lymnaeid species in the transmission of fascioliasis in Venezuela is discussed.

O presente trabalho objetivou estudar a diversidade da virulência de isolados de Pyricularia grisea coletados na Estação Experimental do IAC, Mococa, estado de São Paulo. A composição das raças do fungo e sua compatibilidade com genes de resistência foram estudadas utilizando-se cultivares diferenciais de arroz (Oryza sativa) do Japão. Cinqüenta isolados monospóricos foram obtidos das panículas das cultivares IAC 201 e IAC 4440 afetadas com brusone. As raças JP 137 e JP 177 de P. grisea na cultivar de arroz de sequeiro IAC 201 e a raça JP 200 na cultivar IAC 4440, arroz irrigado, foram identificadas. Foi observada uma baixa freqüência de ocorrência de raças fisiológicas no local de seleção de linhagens de melhoramento. Enquanto todos os 25 isolados provenientes de IAC 4440 foram compatíveis somente ao gene de resistência pi-ta², os isolados de IAC 201 foram compatíveis para sete dos nove genes das diferenciais japonesas. Os resultados mostraram que somente um gene de resistência (pi-z t) foi efetivo a todos os isolados de P. grisea coletados das cultivares de arroz IAC 201 e IAC 4440.

OBJETIVO: Avaliar a eficácia e as complicações da utilização da janela pericárdica (JP) transdiafragmática no diagnóstico de lesão cardíaca por ferimento penetrante. MÉTODO: No período de janeiro de 1994 a dezembro de 2004 a JP foi indicada em 245 casos com suspeita de trauma cardíaco por ferimento penetrante. Em 38 deles (15,5%) a JP foi realizada através de acesso transdiafragmático, constituindo a população do presente estudo. RESULTADOS: O mecanismo de lesão foi ferimento por projétil de arma de fogo (FPAF) em 26 casos (68,4%) e por arma branca (FAB) em 12 casos (31,6%). Na maioria dos pacientes (27 casos - 71%) os ferimentos eram múltiplos. A média de RTS foi de 7,32 e a média de PATI foi de 9,8. A JP foi considerada positiva em seis casos (15,8%) dos quais cinco (83%) por FPAF e um (17%) por FAB. Houve um paciente, vítima de múltiplos FPAF, com JP positiva e toracotomia em que não foi identificada lesão cardíaca. A sensibilidade do método foi de 97,4% e a especificidade de 100%. Em 12 (31,6%) pacientes houve associação de lesão com perfuração de víscera oca. Houve um único caso (2,6%) de complicação diretamente relacionada ao procedimento, em uma vítima de múltiplos FPAF com lesão perfurada de fígado, estômago e cólon, que evoluiu com pericardite, necessitando posteriormente de drenagem pericárdica, com boa resolução. A mortalidade foi de 7,9% (três casos), um dos quais em pós-operatório de reparo de lesão cardíaca por coagulopatia/acidose. CONCLUSÕES: A JP trandiafragmática é um procedimento que permite o rápido diagnóstico de lesão cardíaca em pacientes nos quais a prioridade é a laparotomia. Trata-se de método de fácil realização e alta sensibilidade.

OBJETIVO: Analisar epidemiologicamente a utilização da janela pericárdica(JP) no diagnóstico de lesão cardíaca em um hospital universitário de trauma de Curitiba. MÉTODOS: Estudo observacional, retrospectivo, de análise dos prontuários de pacientes que foram submetidos a pericardiotomia por trauma contuso ou penetrante, no período de seis anos, no serviço de Urgência e Emergência do Hospital Universitário Cajuru. RESULTADOS: 120 pacientes foram submetidos à Janela Pericárdica no período acima referido. A faixa etária variou de 15 a 80 anos, sendo a maior prevalência entre os 20 a 30 anos (49,7%), 105(87,5%) pacientes eram homens e 15(12,5%) mulheres. Os traumas fechados foram 14(11,67%) e penetrantes 105(87,5%). Dos penetrantes, 41 foram por ferida de arma branca, 60 por ferida de arma de fogo e quatro por ambas. Quanto à localização das lesões: 47,5% foram precordiais, 34,16% em transição tóraco-abdominal, 5,0% em ambas e 13,33% em outras localizações. Das JP realizadas, 72,5% foram negativas e 27,5% positivas. Dentre as positivas, as lesões cardíacas encontradas foram: átrio direito 21,2%, ventrículo direito 30,3%, ventrículo esquerdo 24,2%, aorta ascendente 3%, nenhuma lesão 21,2%. Houve 35 óbitos: 18 deles até 24hs e 17 após 24hs. CONCLUSÃO: A janela pericárdica foi mais realizada em homens jovens com ferimentos penetrantes por arma de fogo, em sua maioria com lesão do ventrículo direito como principal achado, concordando com a literatura revisada.

AbstractAscorbic acid, carotenoids and polyphenols stand out among the orange juice natural antioxidants. The winemaking process affects their bioavailability and bioactivity. Antioxidant activities (AA) were estimated in different process conditions to asses those properties. The AA and their correlation with ascorbic acid, total phenolics and carotenoids content were calculated. The variables and levels analyzed were: pasteurized and natural must (PJ and NJ), pH 3.5 and 4.0 and fermentation temperatures at 10°C and 20°C. Statistically significant differences (α=0.05) were found among bioactive compounds concentrations. Antioxidant compounds concentration was higher in raw material than in orange wine. Juice pasteurization caused the major losses while subsequent vinification affects them to a lesser extent. Highest antioxidants retention was measured in wines from JN fermented at pH 3.5 and 10 °C (JN-3.5-10) followed by wines from JP and fermented at the same conditions (JP-3.5-10). AA determined by DPPH showed a positive and close correlation with FRAP, while ABTS showed a low correlation with former assays. Juice pasteurization process and fermentation temperature influenced bioactive compound reduction which correlates with the AA variation.
