Visual implant elastomer (VIE) has recently been employed to investigate different aspects of earthworm ecology. However, a number of fundamental questions relating to the detection and positioning of the tag, its persistence and potential effects on earthworms remain unknown. Seven earthworm species belonging to three ecological groupings, with different pigmentation and burrowing behaviour, were tagged using different coloured VIE. External inspection after two days, one week and 1, 10 and 27 months were followed by preservation, dissection and internal inspection. Tags could be seen in living specimens to 27 months, and dissection revealed that in most cases they were lodged in the coelomic cavity, held in place by septa. However, over longer time periods (more than two years), the chlorogogenous tissue tended to bind to the tags and made external observation increasingly difficult. Migration of the VIE material towards the posterior of the earthworm and potential loss of the tag were only observed on rare occasions, and a recovery rate in excess of 98% was recorded. By introducing a reasonable amount of VIE into segments, just after the clitellum, this technique can become a valuable tool in earthworm ecology and life history studies, particularly in short-medium term laboratory and field experiments.

Over the last 60 years, planting densities for apple have increased as improved management systems have been developed. Dwarfing rootstocks have been the key to the dramatic changes in tree size, spacing and early production. The Malling series of dwarfing rootstocks (M.9 and M.26) have been the most important dwarfing rootstocks in the world but are poorly adapted in some areas of the world and they are susceptible to the bacterial disease fire blight and the soil disease complex, apple replant disease which limits their uses in some areas. Rootstock breeding programs in several parts of the world are developing improved rootstocks with resistance to fire blight, and replant disease, and improved cold hardiness and yield efficiency. A second important trend has been the increasing importance of new cultivars. New cultivars have provided opportunities for higher prices until they are over-produced. A new trend is the "variety club" in which variety owners manage the production and marketing of a new unique cultivar to bring higher prices to the growers and variety owners. This has led to many fruit growers being unable to plant or grow some new cultivars. Important rootstock and cultivar genes have been mapped and can be used in marker assisted selection of future rootstock and cultivar selections. Other important improvements in apple culture include the development of pre-formed trees, the development of minimal pruning strategies and limb angle bending which have also contributed to the dramatic changes in early production in the 2nd-5th years after planting. Studies on light interception and distribution have led to improved tree forms with better fruit quality. Simple pruning strategies and labor positioning platform machines have resulted in partial mechanization of pruning which has reduced management costs. Improved plant growth regulators for thinning and the development of a thinning prediction model based on tree carbohydrate balance have improved the ability to produce the optimum fruit size and crop load. Other new plant growth regulators have also allowed control of shoot growth, control of preharvest fruit drop and control of fruit softening in storage after harvest. As we look to the future, there will be continued incremental improvement in our understanding of plant physiology that will lead to continued incremental improvements in orchard management but there is likely to be dramatic changes in orchard production systems through genomics research and genetic engineering. A greater understanding of the genetic control of dwarfing, precocity, rooting, vegetative growth, flowering, fruit growth and disease resistance which will lead to new varieties and rootstocks which are less expensive to grow and manage.

Coumarins are special metabolites well distributed in the Angiospermae, either in Monocotyledoneae or Dicotyledoneae. Simple coumarins, the most widespread type, is found in all coumarin-producing families, such as: Apiaceae, Rutaceae, Asteraceae, Fabaceae, Oleaceae, Moraceae e Thymelaeaceae. The other types, linear- and angular furanocoumarins, linear- and angular pyranocoumarins, lignocoumarins, bis- and triscoumarins, are of more restricted circumscription. Among the families with occurrence numbers (NO) > 100, the more advanced ones are specialized in the production of only one or two coumarin types, while the primitive families are very well diversified in types. Calculations of percentual numbers of occurrence (%NO) show relevant meaning of coumarin-types in the taxonomic positioning of the producing taxa.

This review focuses the development of electrochemical detection systems coupled to capillary electrophoresis. Conductometric, amperometric, voltametric, and potentiometric modes of detection are reviewed. The positioning of the electrodes, interferences of high electric field, and the materials employed in the fabrication and modification of the electrodes are discussed. The advantages of the use of electrochemical detection with capillary electrophoresis, regarding to the sensitivity and selectivity, is exemplified with a large number of applications. Also, the use of electrochemical detection systems in microchip technology is addressed.

A simple low-cost flow cell was developed, built and optimized in order to observe dynamic interfacial tension of continuous flow systems. Distinct materials can be used in one of the cell walls in order to observe the intermolecular forces between the flowing liquid and the chemical constitution of the walls. The fluorescence depolarization was evaluated using Rhodamine B as fluorescent probe seeded in ethylene glycol. The effects of the positioning angles on the data acquired across the cell are reported. The reproducibility of the data was evaluated with a spectrometer assembled in-house and the relative standard deviation was below 3%.

Teaching classes and events regarding the molecular aspects of drug-receptor interactions is not an easy task. The ligand stereochemistry and the spatial arrangement of the macromolecular targets highly increase the complexity of the process. In this context, the use of alternative and more playful approaches could allow students to gain a more thorough understanding of this important topic in medicinal chemistry. Herein, we describe a practical teaching approach that uses computational strategies as a tool for drug-receptor interaction studies performed for angiotencsin converting enzyme inhibitors (ACEi). Firstly, the students learn how to find the crystallographic structure (enzyme-ligand complex). Then, they proceed to the treatment of crude crystallographic data. Thereafter, they learn how to analyze the positioning of the drug on the active site of the enzyme, looking for regions related to the molecular recognition. At the end of the study, students can summarize the molecular requirements for the interaction and the structure-activity relationships of the studied drugs.

Objetivou-se caracterizar a variabilidade espacial da incidência de Colletotrichum truncatum (Schwein) em sementes colhidas de soja e determinar o melhor modelo e método de semivariograma que represente a incidência de C. truncatum dessas sementes. O experimento foi realizado em condições de campo na safra 2009/10, em 3 parcelas de 9,9 x 10 m, com sementes inoculadas com C.truncatum. O inóculo correspondeu a 0,8; 1,6 e 2,4% do total semeado. Foram demarcadas 3 malhas com receptores GNSS, totalizando 112 pontos em cada parcela distanciados a 1,3 m na linha. No final do ciclo da soja, realizaram-se a colheita, a secagem e a análise sanitária das sementes pelo método 'blotter test', referente aos 336 pontos demarcados. Quatro modelos de semivariogramas foram ajustados aos dados coletados utilizando os métodos mínimos quadrados ordinários (OLS), mínimos quadrados ponderados (WLS), máxima verossimilhança (ML) e máxima verossimilhança restrita (REML). A validação cruzada foi empregada para escolha final do modelo e método do semivariograma. Em seguida, efetuaram-se a krigagem e o desvio padrão da krigagem. Os mapas de krigagem ilustraram a transmissão da semente para a semente e a sua variância. Verificou-se estrutura de dependência espacial da transmissão de C. truncatum via semente. O melhor modelo de semivariograma foi o esférico e o melhor ajuste foi o REML. Houve alcance de 0,95, 4,03 e 7,05 m para as parcelas com 0,8, 1,6 e 2,4% de sementes inoculadas respectivamente. Quanto maior o inóculo primário da parcela, maior foi a transmissão para as sementes próximas à fonte de inóculo.

Foram identificados, mapeados e caracterizados os geoambientes do Parque Estadual do Ibitipoca (PEI), Minas Gerais, com o objetivo de subsidiar o manejo ecológico da área. Para este fim, realizaram-se coletas de solos, em pontos georreferenciados por GPS (Global Positioning System), fotointerpretações a partir de ortofotos e uso de mapas planialtimétricos, além de intenso reconhecimento de solos no campo. Oito geoambientes foram identificados e caracterizados: (1) Patamares com Espodossolos, 2) Cristas Ravinadas, 3) Escarpas, 4) Grotas, 5) Mata Baixa com Candeia, 6) Mata Alta sobre Xisto, 7) Topos Aplainados e 8) Rampas com Vegetação Aberta. Em cada ambiente a vegetação associada é fortemente condicionada pela profundidade do solo e pelo tempo de permanência de água no sistema. Os ambientes de mata, tanto sobre xistos quanto sobre quartzitos, sofrem menor estresse hídrico, seja por melhores condições físicas do solo e maior retenção de água, seja pela presença de ambiente mais ombrófilo e úmido, como nas Grotas. Os geoambientes florestados possuem concentrações de P e K mais elevadas do que nos ambientes campestres abertos. No geoambiente de Mata Baixa com Candeia, a pobreza química do substrato parece ser o impedimento à ocorrência de uma floresta mais densa. Os Campos de Altitude ocorrem nas altitudes mais elevadas no PEI, sendo desenvolvidos sobre solos mais profundos do que sob campos rupestres, onde há freqüente exposição da rocha ou solos muito rasos.

Sistemas de piloto automático vêm sendo utilizados com relativa frequência na agricultura brasileira. Na citricultura, o uso dessa tecnologia vem difundindo-se na operação de abertura de sulcos para o transplantio de mudas. O presente trabalho teve como objetivos avaliar a acurácia de um piloto automático e avaliar possíveis ganhos gerados pela utilização dessa tecnologia de orientação automática em relação ao sistema convencional de abertura de sulcos na implantação de lavoura de citros. A primeira etapa consistiu em georreferenciar as plantas no campo, três meses após o transplantio, com sulcos feitos com as duas tecnologias. Na segunda etapa do trabalho, foi conduzida a avaliação da capacidade operacional da operação de abertura de sulcos, com e sem a utilização de piloto automático coletando dados de tempos e de movimentos com um receptor de GPS. Foi obtido um erro médio de desalinhamento de sulcos de 0,08 m no sistema convencional e de 0,04 m com o uso do piloto automático. A operação de abertura de sulcos utilizando o piloto automático apresentou eficiência de campo de 73,4% e no sistema convencional a eficiência foi de 77,6%. Porém, com piloto automático, foi possível trabalhar com velocidades maiores, o que resultou numa capacidade de campo efetiva maior para este sistema em relação ao sistema convencional de abertura de sulcos (4,65 ha h-1 e 4,09 ha h-1). O usuário reportou custo da operação de abertura de sulcos menor para o sistema com piloto automático quando comparado com o sistema convencional, especialmente devido à redução de demanda de mão de obra.

The objective of this study consisted on mapping the use and soil occupation and evaluation of the quality of irrigation water used in Salto do Lontra, in the state of Paraná, Brazil. Images of the satellite SPOT-5 were used to perform the supervised classification of the Maximum Likelihood algorithm - MAXVER, and the water quality parameters analyzed were pH, EC, HCO3-, Cl-, PO4(3-), NO3-, turbidity, temperature and thermotolerant coliforms in two distinct rainfall periods. The water quality data were subjected to statistical analysis by the techniques of PCA and FA, to identify the most relevant variables in assessing the quality of irrigation water. The characterization of soil use and occupation by the classifier MAXVER allowed the identification of the following classes: crops, bare soil/stubble, forests and urban area. The PCA technique applied to irrigation water quality data explained 53.27% of the variation in water quality among the sampled points. Nitrate, thermotolerant coliforms, temperature, electrical conductivity and bicarbonate were the parameters that best explained the spatial variation of water quality.

The implementation of local geodetic networks for georeferencing of rural properties has become a requirement after publication of the Georeferencing Technical Standard by INCRA. According to this standard, the maximum distance of baselines to GNSS L1 receivers is of 20 km. Besides the length of the baseline, the geometry and the number of geodetic control stations are other factors to be considered in the implementation of geodetic networks. Thus, this research aimed to examine the influence of baseline lengths higher than the regulated limit of 20 km, the geometry and the number of control stations on quality of local geodetic networks for georeferencing, and also to demonstrate the importance of using specific tests to evaluate the solution of ambiguities and on the quality of the adjustment. The results indicated that the increasing number of control stations has improved the quality of the network, the geometry has not influenced on the quality and the baseline length has influenced on the quality; however, lengths higher than 20 km has not interrupted the implementation, with GPS L1 receiver, of the local geodetic network for the purpose of georeferencing. Also, the use of different statistical tests, both for the evaluation of the resolution of ambiguities and for the adjustment, have enabled greater clearness in analyzing the results, which allow that unsuitable observations may be eliminated.

In the present case (77 years-old woman), the diagnosis on an extramucosal lesion by endosonography was leiomyoma or schwanoma. Radiological exam of the upper digestive tract with barium and abdominal computed tomography confirmed the site of the lesion at the level of the lesser curvature. The operative technique followed the steps of the laparoscopic partial gastric resection (wedge resection) for gastric mesenchymal tumours, described elsewhere. The Endo-GIA stapler was introduced through the 12mm port in the right upper quadrant. Proper positioning of the stapler over the lesser curvature and a satisfactory margin of tissue around the mass were attained. Nine sequential firings of the Endo-GIA 30 were needed to completely surround the mass. Histopathological diagnosis was a spindle-cell tumour measuring 3cm in diameter. Mitotic index was measured at almost null. The neoplastic cells were strongly reactive for vimentine and CD34 and negative for the immunohistochemical markers S-100 protein, muscle actin, desmin and Ag linked to VIII factor. There was a slight reaction with keratin (+/+++). The XIIIA factor reaction revealed less than 5% of dendritic elements. These data favour a vascular cell origin better than smooth muscle cell origin. In conclusion it was a gastric hemangiopericytoma. Follow-up showed no recurrence at seven years.

BACKGROUND: A new procedure for the treatment of esophageal fistula, mainly associated to the ebb esophagojejunal in patients submitted to the total gastrectomy and reconstruction with loop jejunal Rouxen-Y anastomosis is present. METHODS: The method is based in the use of "probe standard enteral prolongated with drain to laminate adapted in extremity", which results in advanced positioning inside the jejunum, making the administration of enteral nutrition possible and impeding ebb esophagojejunal. RESULTS: The authors discuss the theoretical advantages of the procedure and they suggest that the treatment of esofagic fistula with probe prolonged enteral would be suitable in the treatment of the fistula esophagojejunal by preventing the ebb esophagojejunal, which would result in smaller period of duration of the fistula esophagojejunal and it would prevent the high mortality rate. CONCLUSIONS: Preliminary studies demonstrated that this is a technically easy, low cost procedure through the endoscopic use. A prospective evaluation for morbility and mortality related to the method is needed.

The authors present the four-arm single docking full robotic surgery to treat low rectal cancer. The eight main operative steps are: 1- patient positioning; 2- trocars set-up and robot docking; 3- sigmoid colon, left colon and splenic flexure mobilization (lateral-to-medial approach); 4-Inferior mesenteric artery and vein ligation (medial-to-lateral approach); 5- total mesorectum excision and preservation of hypogastric and pelvic autonomic nerves (sacral dissection, lateral dissection, pelvic dissection); 6- division of the rectum using an endo roticulator stapler for the laparoscopic performance of a double-stapled coloanal anastomosis (type I tumor); 7- intersphincteric resection, extraction of the specimen through the anus and lateral-to-end hand sewn coloanal anastomosis (type II tumor); 8- cylindric abdominoperineal resection, with transabdominal section of the levator muscles (type IV tumor). The techniques employed were safe and have presented low rates of complication and no mortality.

Supporting patients with acute respiratory distress syndrome (ARDS), using a protective mechanical ventilation strategy characterized by low tidal volume and limitation of positive end-expiratory pressure (PEEP) is a standard practice in the intensive care unit. However, these strategies can promote lung de-recruitment, leading to the cyclic closing and reopening of collapsed alveoli and small airways. Recruitment maneuvers (RM) can be used to augment other methods, like positive end-expiratory pressure and positioning, to improve aerated lung volume. Clinical practice varies widely, and the optimal method and patient selection for recruitment maneuvers have not been determined, considerable uncertainty remaining regarding the appropriateness of RM. This review aims to discuss recent findings about the available types of RM, and compare the effectiveness, indications and adverse effects among them, as well as their impact on morbidity and mortality in ARDS patients. Recent developments include experimental and clinical evidence that a stepwise extended recruitment maneuver may cause an improvement in aerated lung volume and decrease the biological impact seen with the traditionally used sustained inflation, with less adverse effects. Prone positioning can reduce mortality in severe ARDS patients and may be an useful adjunct to recruitment maneuvers and advanced ventilatory strategies, such noisy ventilation and BIVENT, which have been useful in providing lung recruitment.
