Flower scent analysis of Encyclia Vespa (vell.) Dressler & G. E. Pollard and E. Fragrans (Sw.) Lemée

The volatile constituents obtained from the pentane extract, using simultaneous distillation-extraction of the flowers of Encyclia vespa and E. fragrans were analysed by GC/ MS. The main volatile components identified in the flowers of E. vespa were terpinen-4-ol (20.3%), verbenone (14.8%), trans-verbenol (13.6%) and x-pinene (11.8%). The major volatiles of the flowers of E. fragrans were terpinen-4-ol (18.3%), (2Z,6E)-farnesol (15.4%) and trans-verbenol (10.2%).

Lauraceae é uma das famílias mais importantes na floresta amazônica. Diversas de suas espécies apresentam em sua composição substâncias com pronunciadas atividades biológicas. Nesse trabalho, os extratos obtidos em etanol de galhos e folhas de 20 espécies de Lauraceae foram coletados na Reserva Florestal Ducke e avaliados quant o às atividades antioxidante frente ao radical livre 2,2-difenil-1-picrilhidrazila(DPPH) e inibidora da enzima acetilcolinesterase. O extrato dos galhos de Licaria martinianal apresentou a maior capacidade de sequestro do radical DPPH (6,96 µg.mL-1),mas os extratos de Ocotea iminor foram os mais ativos na inibição da enzima acetilcolinesterase, apresentando também pronunciada atividade antioxidante. O perfil de atividade antioxidante e anticolinesterásico destes extratos está sendo relatado pela primeira vez neste trabalho.

Plantas de duas formas botânicas de Catharanthus roseus, de flores lilases e de flores brancas foram cultivadas em soluções nutritivas deficientes em N, P, K, Ca, Mg, S e B, e em solução completa, a fim de se obter o quadro sintomatológico das deficiências, assim como os níveis analíticos de nutrientes nas folhas, caules, raízes e flores. Manifestaram-se sintomas de deficiência claros para todos os nutrientes estudados. Nas plantas de flores lilases, as concentrações de nutrientes na matéria seca de folhas de plantas normais e deficientes foram, respectivamente, para cada nutriente estudado: N(%): 3,53-1,20; P(%): 0,35-0,11; K(%): 2,45-0,76; Ca(%): 1,77-0,81; Mg(%): 0,55-0,46; S(%):0,21-0,12; B(ppm): 382-37. Nas plantas de flores brancas, estas concentrações foram: N(%): 3,78-0,92; P(%): 0,38-0,09; K(%): 2,60-0,86; Ca(%): 1,37-1,15; Mg(%): 0,56-0,44; S(%): 0,10-0,07; B(ppm):372-39.

This study sought the characterization of rotaviruses in a trial with a tetravalent rhesus-human rotavirus vaccine in Belém, Brazil in children who received three doses of vaccine or placebo in the 1st, 3rd and 5th months of life. Rotavirus electropherotypes, subgroups, G serotypes, G, [P] and [P],G genotypes were determined in 93.3%, 95.9%, 93.3%, 73.3%, 95.5% and 92.2% of isolates, respectively. Serotypes G1, G2 and G4 were detected in 58.9%, 30% and 4.4% of the cases, respectively. Rotavirus genotype G5 was detected for the first time in Northern region in 4.4% of the infections. Rotavirus genotypes P[8], P[4], P[6] and P[8+6] were detected in 54.5%, 26.7%, 12.2%, and 2.2% of the cases, respectively. The predominant genotypes were P[8],G1 and P[4],G2 with 53% and 26.6% of the infections, respectively. Unusual strains accounted for 20.5% including P[4],G1, P[6],G1, P[6],G4, P[6],G5, P[8],G2, P[8],G5. Mixed infections involving P[8+6],G2 and P[8+6],G1 were also noted. The neonatal P[6] strains associated with diarrhea were detected among children aged 9-24 months. To our knowledge, this study represents the first in Brazil to analyse, on molecular basis, rotavirus genotypes from children participating in a rotavirus vaccine trial. These results are of potential importance regarding future rotavirus vaccination strategies in Brazil.

Maghemite (g-Fe2O3) is the most usually found ferrimagnetic oxide in red basalt-derived soils. The variable degrees of ionic substitution of Fe3+ for different metals (e.g. Ti4+, Al3+, Mg2+, Zn2+, and Mn2+) and non-metals in the maghemite structure influence some cristallochemical features of this iron oxide. In this study, synthetic Zn-substituted maghemites were prepared by co-precipitation in alkaline aqueous media of FeSO4.7H2O with increasing amounts of ZnSO4.7H2O to obtain the following sequence of Fe3+ for Zn2+ substitutions: 0.0, 0.025, 0.05, 0.10, 0.15, 0.20, and 0.30 mol mol-1. The objective of this work was to evaluate the cristallochemical alterations of synthetic Zn-substituted maghemites. The dark black synthetic precipitated material was heated to 250 °C during 4 h forming a brownish maghemite that was characterized by chemical analysis as well as X ray diffraction (XRD), specific surface area and mass-specific magnetic susceptibility. The isomorphic substitution levels observed were of 0.0013, 0.0297, 0.0590, 0.1145, 0.1764, 0.2292 and 0.3404 mol mol-1, with the formation of a series of maghemites from Fe2Zn0O3 to Fe(1.49)Zn(0.770)O3 . The increase in Fe3+ for Zn2+ substitution, [Zn mol mol-1] increased the dimension a0 of the cubic unit cells of the studied maghemites according to the regression equation: a0 = 0.8343 + 0.02591Zn (R² = 0.98). On the other hand, the mean crystallite dimension and mass-specific magnetic susceptibility of the studied maghemites decreased with increasing isomorphic substitution.

Tucumã é uma palmeira amazônica. A polpa dos frutos é comestível sem cozimento e muito apreciada como recheio de sanduíches na culinária regional, possui alto valor nutritivo e destaque no teor de vitamina A. O presente estudo teve como objetivo avaliar aspectos da comercialização dos frutos e da polpa de tucumã em feiras e mercados de Manaus. Durante o estudo de um ano, foram levantadas as procedências e preços dos frutos e estimada a quantidade comercializada em seis feiras e mercados de Manaus. Os dados de procedência, quantidade, preço e outras informações relacionadas foram obtidos através da aplicação mensal de um questionário semiestruturado aos comerciantes. O preço real e sua evolução histórica, desde 1995, foram deduzidos por comparação com estudos anteriores. Entre maio de 2011 e abril de 2012, os frutos foram provenientes de 20 diferentes localidades a uma distância de até mil km de Manaus. Os locais mais citados foram os municípios de Itacoatiara, Terra Santa, Rio Preto da Eva, Autazes e a bacia do Rio Madeira. A alternância das procedências garantiu o abastecimento contínuo ao longo do ano. Entretanto, uma redução do número de procedências e da quantidade de frutos comercializados foi verificada entre setembro e novembro. A sazonalidade da oferta foi refletida nos preços dos frutos e da polpa. Durante os 12 meses, foi comercializado um total de 367,8 toneladas de frutos, com 30,7 toneladas de média mensal. Cerca da metade (53%) desta quantia foi descascada durante a jornada de trabalho dos feirantes e comercializada apenas a polpa. Neste estudo e baseado em informações anteriores, tucumã foi sempre um dos frutos mais caros dos mercados de Manaus. Entre 1995 e 2012, os valores reais aumentaram substancialmente, tanto da saca (230%), com cerca de 41 kg, como da dúzia de frutos (316%) e do kg da polpa (253%). Os resultados indicam que o mercado de tucumã continua em expansão. Aparentemente, a demanda é maior do que a oferta e, dessa forma, há necessidade de aumento da produção.

A lectin present in the marine red alga Pterocladiella capillacea was purified and characterised by extraction of soluble proteins (crude extract) in 20 mM Tris-HCl buffer, pH 7.5. Among the analysed erythrocytes (human blood group A, B and O and the animals ox, goat, chicken and rabbit) the lectin agglutinated specifically rabbit erythrocytes. The hemagglutinating activity assay showed that the lectin was not dependent on divalent cations and was shown to be inhibited by the glycoproteins avidin and mucin. The purification procedure was conduced by precipitation of the crude extract with 80% saturation ammonium sulfate (F0/80) followed by affinity chromatography on guar-gum column. The lectin of P. capillacea was purified 14.5 fold and had a recovery of 27.4% of the original total specific activity present in the crude extract. The absence of carbohydrate suggested that the lectin is not a glycoprotein. The molecular mass of P. capillacea lectin, determined by gel filtration, was 5.8 kDa. SDS-PAGE in the presence of ß-mercaptoethanol gave one band, indicating that the native lectin is a monomeric protein. The activation energy of denaturation process (D G') was calculated to be 106.87 kJ . mol-1 at 70 ºC.

Foi estudada a biologia reprodutiva de Melocactus glaucescens e M. paucispinus (Cactaceae) no Município de Morro do Chapéu, Chapada Diamantina, Bahia, sendo abordados aspectos da fenologia, biologia floral, polinização e sistema reprodutivo. Foram registrados os períodos de floração e frutificação, visitantes florais, freqüência e tipo de visitas, além de estratégia e comportamento dos visitantes às flores. Foram realizadas polinizações experimentais para verificar o sistema reprodutivo das espécies. As duas espécies de Melocactus estudadas apresentaram sobreposição de floração ao longo do período de estudo. Os atributos florais de ambas as espécies são típicos da síndrome da ornitofilia: cores atrativas, estrutura tubulosa e produção de néctar com baixa concentração de solutos, entre 20% e 30%. O beija-flor Chlorostilbon aureoventris Boucier & Mulsant (1948) foi o visitante mais freqüente, com 82% e 89% do total de visitas para M. paucispinus e M. glaucescens, respectivamente. Outras espécies de beija-flores e borboletas também visitaram as flores das espécies. A sobreposição no período de floração e a similaridade na composição da guilda de polinizadores destas e demais espécies simpátricas de Melocactus favorece a hibridação no gênero, como foi observado na região. Melocactus glaucescens apresentou auto-incompatibilidade e alogamia, enquanto M. paucispinus é auto-compatível e autogâmica, porém com menor frutificação em autopolinização do que em polinização cruzada, possivelmente devido à ocorrência de depressão endogâmica. Polinizações interespecíficas indicam a ocorrência de inter-compatibilidade entre M. paucispinus e M. concinnus Buining & Brederoo, sustentando hipóteses correntes de hibridação entre estas espécies.

We examined the radioprotective effect of aminothiol 2-N-propylamine-cyclo-hexanethiol (20-PRA) on a human leukemic cell line (K562) following various radiation doses (5, 7.5 and 20 Gy) using a source of 60Co g-rays. At 5 Gy and 1 nM 20-PRA, a substantial protective effect (58%) was seen 24 h after irradiation, followed by a decrease at 48 h (11%). At the high radiation dose (20 Gy) a low protective effect was also seen (35%). In addition, the antitumorigenic potential of 10 nM 20-PRA was shown by the inhibition of crown gall formation induced by Agrobacterium tumefaciens. The radioprotective potency of 20-PRA is 105-106 times higher than that of the aminothiol WR-1065 (N-(2-mercaptoethyl)-1,3-diaminopropane) whose protective effect is in the 0.1 to 1.0 mM range.

We report the antinociceptive activity, determined by the writhing, formalin and hot-plate tests in mice, of crude (F0/60), lectin and carbohydrate fractions isolated by ammonium sulfate precipitation (0 to 60%) from Bryothamnion seaforthii and B. triquetrum, species of red algae. Not only fraction F0/60 but also lectins from both species significantly inhibited acetic acid-induced abdominal contractions after intraperitoneal or oral administrations. In the formalin test, lectins (1 and 5 mg/kg, ip, and 5 to 20 mg/kg, po) inhibited the 1st and 2nd phases (5 and 20 min, respectively), but the effect occurred predominantly on the 2nd phase. The effects of the lectins were totally or partially reversed by naloxone (2 mg/kg, sc) in the 1st and 2nd phases, respectively. Experiments performed with lectins in the absence and presence of avidin (1 mg/kg, ip) and D-mannose (1 mg/kg, ip) showed that avidin did not interfere with the effect of B. seaforthii lectin but partially reversed the effect of B. triquetrum lectin. D-Mannose completely reversed the effects of both species. F0/60 fractions from both algae significantly increased the latency time in response to thermal stimuli, and naloxone reversed antinociception, indicating the involvement of the opioid system in both the peripheral and central effects of the fractions. In the writhing test, the carbohydrate fractions were the most active, inhibiting the contractions by 71 and 79% (B. triquetrum) and by 46 and 69% (B. seaforthii) at doses of 1 and 5 mg/kg, ip, respectively. Sulfated carbohydrate fractions of B. seaforthii and B. triquetrum, containing only about 5% protein as contaminants, are probably responsible for the antinociceptive effects of these red algae.

The aim of the present study was to characterize the interactions of antagonist G (H-Arg-D-Trp-NmePhe-D-Trp-Leu-Met-NH 2)-targeted sterically stabilized liposomes with the human variant small cell lung cancer (SCLC) H82 cell line and to evaluate the antiproliferative activity of encapsulated doxorubicin against this cell line. Variant SCLC tumors are known to be more resistant to chemotherapy than classic SCLC tumors. The cellular association of antagonist G-targeted (radiolabeled) liposomes was 20-30-fold higher than that of non-targeted liposomes. Our data suggest that a maximum of 12,000 antagonist G-targeted liposomes were internalized/cell during 1-h incubation at 37ºC. Confocal microscopy experiments using pyranine-containing liposomes further confirmed that receptor-mediated endocytosis occurred, specifically in the case of targeted liposomes. In any of the previously mentioned experiments, the binding and endocytosis of non-targeted liposomes have revealed to be negligible. The improved cellular association of antagonist G-targeted liposomes, relative to non-targeted liposomes, resulted in an enhanced nuclear delivery (evaluated by fluorimetry) and cytotoxicity of encapsulated doxorubicin for incubation periods as short as 2 h. For an incubation of 2 h, we report IC50 values for targeted and non-targeted liposomes containing doxorubicin of 5.7 ± 3.7 and higher than 200 µM doxorubicin, respectively. Based on the present data, we may infer that receptors for antagonist G were present in H82 tumor cells and could mediate the internalization of antagonist G-targeted liposomes and the intracellular delivery of their content. Antagonist G covalently coupled to liposomal drugs may be promising for the treatment of this aggressive and highly heterogeneous disease.

In order to evaluate the performance of a 1-h 75-g oral glucose tolerance test (OGTT) for the diagnosis of gestational diabetes mellitus (GDM), a cohort of 4998 women, 20 years or older, without previous diabetes being treated in prenatal care clinics in Brazil answered a questionnaire and performed a 75-g OGTT including fasting, 1-h and 2-h glucose measurements between their 24th and 28th gestational weeks. Pregnancy outcomes were transcribed from medical registries. GDM was defined according to WHO criteria (fasting: ≥126 mg/dL; 2-h value: ≥140 mg/dL) and macrosomia as a birth weight equal to or higher than 4000 g. Areas under the receiver operator characteristic curve (AUC) were compared and diagnostic properties of various cut-off points were evaluated. The AUCs for the prediction of macrosomia were 0.606 (0.572-0.637) for the 1-h and 0.589 (0.557-0.622) for the 2-h plasma glucose test. Similar predictability was demonstrable regarding combined adverse outcomes: 0.582 (0.559-0.604) for the 1-h test and 0.572 (0.549-0.595) for the 2-h test. When the 1-h glucose test was evaluated against a diagnosis of GDM defined by the 2-h glucose test, the AUC was 0.903 (0.886-0.919). The cut-off point that maximized sensitivity (83%) and specificity (83%) was 141 mg/dL, identifying 21% of the women as positive. A cut-off point of 160 mg/dL, with lower sensitivity (62%), had higher specificity (94%), labeling 8.6% as positive. Detection of GDM can be done with a 1-h 75-g OGTT: the value of 160 mg/dL has the same diagnostic performance as the conventional 2-h value (140 mg/dL). The simplification of the test may improve coverage and timing of the diagnosis of GDM.

Polymorphisms of hormone receptor genes have been linked to modifications in reproductive factors and to an increased risk of breast cancer (BC). In the present study, we have determined the allelic and genotypic frequencies of the ERα-397 PvuII C/T, ERα-351 XbaI A/G and PGR PROGINS polymorphisms and investigated their relationship with mammographic density, body mass index (BMI) and other risk factors for BC. A consecutive and unselected sample of 750 Brazilian BC-unaffected women enrolled in a mammography screening program was recruited. The distribution of PGR PROGINS genotypic frequencies was 72.5, 25.5 and 2.0% for A1A1, A1A2 and A2A2, respectively, which was equivalent to that encountered in other studies with healthy women. The distribution of ERα genotypes was: ERα-397 PvuII C/T: 32.3% TT, 47.5% TC, and 20.2% CC; ERα-351 XbaI A/G: 46.3% AA, 41.7% AG and 12.0% GG. ERα haplotypes were 53.5% PX, 14.3% Px, 0.3% pX, and 32.0% px. These were significantly different from most previously published reports worldwide (P < 0.05). Overall, the PGR PROGINS genotypes A2A2 and A1A2 were associated with fatty and moderately fatty breast tissue. The same genotypes were also associated with a high BMI in postmenopausal women. In addition, the ERα-351 XbaI GG genotype was associated with menarche ≥12 years (P = 0.02). ERα and PGR polymorphisms have a phenotypic effect and may play an important role in BC risk determination. Finally, if confirmed in BC patients, these associations could have important implications for mammographic screening and strategies and may be helpful to identify women at higher risk for the disease.
