Introduction Leprosy is a chronic disease that affects skin and peripheral nerves. Disease complications include reactional episodes and physical impairment. One World Health Organization (WHO) goal of leprosy programs is to decrease the number of grade 2 impairment diagnoses by 2015. This study aims to evaluate clinical factors associated with the occurrence of leprosy reactions and physical impairment in leprosy patients. Methods We conducted a retrospective study of data from medical records of patients followed in two important centers for the treatment of leprosy in Aracaju, Sergipe, Brazil, from 2005 to 2011. We used the chi-square test to analyze associations between the following categorical variables: gender, age, operational classification, clinical forms, leprosy reactions, corticosteroid treatment, and physical impairment at the diagnosis and after cure. Clinical variables associated with multibacillary leprosy and/or reactional episodes and the presence of any grade of physical impairment after cure were evaluated using the logistic regression model. Results We found that men were more affected by multibacillary forms, reactional episodes, and grade 2 physical impairment at diagnosis. Leprosy reactions were detected in a total of 40% of patients and all were treated with corticosteroids. However, physical impairment was observed in 29.8% of the patients analyzed at the end of the treatment and our multivariate analysis associated a low dose and short period of corticosteroid treatment with persistence of physical impairments. Conclusions Physical impairment should receive an increased attention before and after treatment, and adequate treatment should be emphasized.

Introduction Molecular biology procedures to detect, genotype and quantify hepatitis C virus (HCV) RNA in clinical samples have been extensively described. Routine commercial methods for each specific purpose (detection, quantification and genotyping) are also available, all of which are typically based on polymerase chain reaction (PCR) targeting the HCV 5′ untranslated region (5′UTR). This study was performed to develop and validate a complete serial laboratory assay that combines real-time nested reverse transcription-polymerase chain reaction (RT-PCR) and restriction fragment length polymorphism (RFLP) techniques for the complete molecular analysis of HCV (detection, genotyping and viral load) in clinical samples. Methods Published HCV sequences were compared to select specific primers, probe and restriction enzyme sites. An original real-time nested RT-PCR-RFLP assay was then developed and validated to detect, genotype and quantify HCV in plasma samples. Results The real-time nested RT-PCR data were linear and reproducible for HCV analysis in clinical samples. High correlations (> 0.97) were observed between samples with different viral loads and the corresponding read cycle (Ct - Cycle threshold), and this part of the assay had a wide dynamic range of analysis. Additionally, HCV genotypes 1, 2 and 3 were successfully distinguished using the RFLP method. Conclusions A complete serial molecular assay was developed and validated for HCV detection, quantification and genotyping.

INTRODUCTION: Human immunodeficiency virus type 1 (HIV-1) has spread worldwide, with several subtypes and circulating recombinant forms. Brazil has an incidence of 20.5 HIV-1/acquired immunodeficiency syndrome (AIDS) patients per 100,000 inhabitants; however, the Southernmost State of Rio Grande do Sul (RS) has more than twice the number of HIV-1-infected people (41.3/100,000 inhabitants) and a different pattern of subtype frequencies, as previously reported in studies conducted in the capital (Porto Alegre) and its metropolitan region. This study examined HIV-1/AIDS epidemiological and molecular aspects in the countryside of Rio Grande do Sul. METHODS: Socio-demographic, clinical and risk behavioral characteristics were obtained from HIV-1-positive adult patients using a structured questionnaire. HIV-1 subtypes were determined by nested-polymerase chain reaction (PCR) and sequencing of the pol and env genes. RESULTS: The study sample included 149 (55% women) patients with a mean age of 41.8 ± 11.9 years. Most (73.8%) patients had a low education level and reported heterosexual practices as the most (91.9%) probable transmission route. HIV-1 subtypes were detected in 26 patients: 18 (69.2%) infected with subtype C, six (23.1%) infected with subtype B and two (7.7%) infected with BC recombinant forms. CONCLUSIONS: These data highlight the increasing number of HIV-1 subtype C infections in the countryside of South Brazil.

OBJETIVO: Avaliar a segurança e eficácia da braquiterapia intracoronariana usando o sistema Beta-CathTM na prevenção da recorrência de restenose intra-stent (RIS), por meio da análise dos resultados clínicos, angiográficos e pelo ultra-som intracoronariano (USIC). MÉTODO: Foram submetidos à angioplastia com cateter-balão, seguida de beta-radiação intracoronariana com o sistema Beta-CathTM (90Sr/Y) 30 pacientes com RIS em artérias coronárias nativas e, posteriormente, avaliados. RESULTADOS: Incluíram-se lesões reestenóticas complexas (77% do tipo difuso-proliferativo) com extensão elevada (18,66±4,15 mm). O sucesso da braquiterapia foi de 100%. A dose média utilizada foi de 20,7±2,3 Gy, liberada em um período médio de 3,8±2,1 min. No seguimento tardio, o diâmetro luminal mínimo (DLM) intra-stent diminuiu discretamente (1,98±0,30mm para 1,84±0,39 aos 6 meses, p=0,13), com uma perda tardia de 0,14±0,18 mm. O DLM intra-segmentar foi significativamente menor do que o intra-stent (1,55±0,40mm vs.1,84±0,39mm, p=0,008), associando-se à perda tardia (0,40±0,29mm vs. 0,14±0,18mm; p=0,0001). No USIC, observou-se discreto incremento do tecido neointimal em 6,8±14,3 mm³ aos 6 meses (p=0,19) e a percentagem de obstrução volumétrica aumentou em 4,7±7,5%. A reestenose binária e a revascularização do vaso-alvo recorreram em 17% dos casos; houve 1 caso (3%) de oclusão tardia, associada a infarto do miocárdio. A sobrevida livre de eventos foi de 80%. CONCLUSÃO: O manejo da reestenose intra-stent com a beta-radiação intracoronariana mostrou-se procedimento seguro e eficaz, com alta taxa de sucesso imediato, representando uma opção terapêutica para a inibição da hiperplasia neointimal.

OBJETIVO: Avaliar a importância da interação entre a integrina Mac-1 dos leucócitos (a Mb 2) e a glicoproteína (GP) Iba das plaquetas para o recrutamento de leucócitos após a lesão vascular e o efeito da neutralização da interação Mac-1-GPIba sobre a proliferação celular e a hiperplasia neointimal desencadeadas por lesão vascular. MÉTODOS: Um peptídeo denominado M2 ou anticorpo anti-M2 foi desenvolvido para bloquear a interação Mac-1-GPIba . Esse peptídeo foi injetado e comparado com anticorpo-controle em camundongos C57B1/6J submetidos a lesão vascular da artéria femoral com corda-guia. Um, cinco ou 28 dias após a lesão vascular, as artérias femorais foram retiradas para a realização de morfometria e imuno-histoquímica. RESULTADOS: O bloqueio da interação Mac-1-GPIba promoveu uma redução estatisticamente significativa do número de leucócitos na camada média no primeiro dia após a lesão vascular (controle: 7,9±5,0% do total de células versus anti-M2: 2,0±1,6%, p=0,021), bem como determinou uma diminuição estatisticamente significativa do acúmulo de leucócitos na neoíntima em cinco e 28 dias (controle: 42,3±12,9% versus anti-M2: 24,6±10,8%, p=0,047 e controle: 7,9±3,0% versus anti-M2: 3,3±1,3%, p=0,012; respectivamente). A proliferação celular na camada média do vaso em cinco dias pós-lesão foi reduzida com o bloqueio da interação Mac-1-GPIba (controle: 5,0±2,9% do total de células versus anti-M2: 1,8±0,5%; p=0,043), assim como houve diminuição significativa da proliferação celular na camada íntima do vaso em 28 dias (controle: 3,8±1,7% versus anti-M2: 2,0±1,2%; p=0,047). O bloqueio da interação Mac-1-GPIba também determinou uma redução estatisticamente significativa do espessamento intimal em 28 dias pós-lesão (controle: 10.395±3.549 µm² versus anti-M2: 4.561±4.915 µm²; p=0,012). CONCLUSÃO: O recrutamento de leucócitos após a lesão vascular é dependente da interação Mac-1-GPIba e a neutralização dessa interação inibe a proliferação celular e a formação neointimal.

The subfamily Corinninae is characterized and diagnosed. Two synapomorphies are hypothesized for the subfamily, both regarding the male palpal reservoir, which is primarily coiled and presents a sclerotized distal sector. Seventeen genera are recognized, six of which are new: Abapeba (type species Corinna lacertosa Simon), Erendira (type species Corinna pallidoguttata Simon), Septentrinna (type species Corinna bicalcarata Simon), Simonestus (type species Diestus validus Simon), Tapixaua (type species T. callida sp. nov.) and Tupirinna (type species T. rosae sp. nov.). The genera Creugas Thorell, Falconina Brignoli and Paradiestus Mello-Leitão are revalidated. Diestus Simon and Lausus Simon are newly synonymized with Corinna C. L. Koch. Chemmis Simon is included in the synonymy of Megalostrata Karsch. Hypsinotus L. Koch is removed from the synonymy of Corinna and included in the synonymy of Creugas. Thirteen new species are described: Septentrinna yucatan and S. potosi from Mexico; Tupirinna rosae from Venezuela and Brazil; Tapixaua callida from Brazil and Peru; Abapeba hoeferi, A. rioclaro, A. taruma, Corinna ducke, C. colombo, C. mourai, C. recurva and Parachemmis manauara from Brazil; Creugas lisei from Brazil, Argentina and Uruguay. Twenty seven species are redescribed. Fifty eight new combinations are presented: from Chemmis, Septentrinna steckleri (Gertsch); from Corinna, Abapeba abalosi (Mello-Leitão), A. cleonei (Petrunkevitch), A. echinus (Simon), A. grassima (Chickering), A. guanicae (Petrunkevitch), A. lacertosa (Simon), A. luctuosa (F. One new name, Creugas berlandi, is erected for Corinna bellatrix Schmidt. Males of Creugas cinnamius, Corinna kochi, Methesis semirufa Simon, Paradiestus aurantiacus, Septentrinna steckleri and Xeropigo smedigari, the females of Paradiestus giganteus, Septentrinna bicalcarata and the adult female of S. steckleri are described for the first time.

Duas espécies de Eustala Simon, 1895 são descritas do Rio Grande do Sul, Brasil: Eustala belissima sp. nov. e Eustala crista sp. nov., representadas por ambos os sexos. A fêmea de E. itapocuensis Strand, 1916 e os machos de E. nasuta Mello-Leitão, 1939, E. perfida Mello-Leitão, 1947 e E. secta Mello-Leitão, 1945, são descritos pela primeira vez. Novas ocorrências do Brasil são listadas para Eustala illicita (O. P.-Cambridge, 1889) e E. sagana (Keyserling, 1893).

In this paper seven new species of Latonigena Simon, 1893 are described, including the first descriptions of males of this genus. Latonigena beni sp. nov. is described from Bolivia and Brazil and six species are described exclusively from Brazil: L. colombo sp. nov. from Paraná; L. lami sp. nov. from Paraná and Rio Grande do Sul; L. santana sp. nov., L. sapiranga sp. nov. and L. taim sp. nov. from Rio Grande do Sul; L. turvo sp. nov. from Tocantins, Santa Catarina and Rio Grande do Sul. Latonigena africana Tucker, 1923 is transferred to Trichothyse Tucker, 1923 with basis on the morphological characters.

The male of Latonigena auricomis Simon, 1893 is described for the first time and the female is redescribed. New records are provided for Argentina, Brazil and Uruguay. Notes on the natural history and a potential distribution model of the species are presented in the Neotropical Region.

A new species of Tmesiphantes Simon, 1892, T. aridai sp. nov., is described and illustrated based on three males and three females collected at the Floresta Nacional do Tapajós, Santarém, state of Pará, Brazil. Males can be distinguished from T. perp Guadanucci & Silva, 2012 by the palpal bulb with an inconspicous tegular basal projection, but presenting a very slender embolus with shorter keels not extending to the tip, and from T. nubilus Simon, 1892 by the tibial apophysis with two similarly sized branches, the prolateral one with a strong spine on the retrolateral margin. Females resemble T. nubilus by the aspect of the seminal receptacle but the constriction near the apex is less evident and apex shape is irregular. The new species represents the first record of Tmesiphantes from the Amazonian region, bringing the total number of species to eight.

The genus Pycnothele Chamberlin, 1917 is revised. The Brazilian species Pycnothele perdita Chamberlin, 1917, P. singularis (Mello-Leitão, 1934) and P. auronitens (Keyserling, 1891) are redescribed, diagnosed and illustrated. The females of P. auronitens and P. singularis are described for the first time and P. auripila (Mello-Leitão, 1946) from Uruguay, currently a junior synonym of P. auronitens, is revalidated. Pycnothele piracicabensis (Piza, 1938) is transferred to Rachias Simon, 1892, with which it shares the morphology of the sexual structures, restoring the original combination. Five new species are proposed for Brazil: Pycnothele rubra sp. nov., P. jatai sp. nov. and P. araraquara sp. nov., from state of São Paulo, P. arapongas sp. nov., from state of Paraná, and P. gauderio sp. nov., from state of Rio Grande do Sul. After this revision the genus includes ten Neotropical species. Maps with distribution of all species is presented for the first time and the occurrence of species of the genus is enhanced for the Brazilian state of Paraná.

ABSTRACT A new species of Phrixotrichus Simon, 1889, P. pucara sp. nov., is described and illustrated based on a male from Pucará river, Neuquén province, Argentina. Male can be distinguished from all other species of the genus by the presence of a long strong spine on inner face of prolateral branch of tibial apophysis; also, it differs from P. scrofa (Molina, 1788) and P. vulpinus (Karsch, 1880) by a serrated prolateral keel of the male palpal bulb. Male resembles P. jara Perafán & Pérez-Miles, 2014 but can be distinguished by the uniform color on dorsal cephalothorax and by the palpal organ morphology being wider on the bulb base and embolus shorter and thicker, with the tip of embolus not so directed retrolaterally and prolateral keel bearing a serrated edge with three teeth. Additionally, P. vulpinus is reported for the first time for Argentina along with new distributional data.
