The authors carried out joint analyses of data referring to six experiments with varieties of sugar cane, carried out by SEGALLA and ALVAREZ in six locations in the State of S. Paulo, Brasil. The analyses showed that for cane or sugar yield, either for plant-cane or for plant-cane together with the first two ratoons, the best five varieties were CB 40-69, CB 41-76, CB 40-13, CB 40-19 and Co 419. The yield of sugar cane/for all varieties studied is given below, in metric tons produced in plant cane and the first two ratoons. Varieties Yield of sugar cane (tons/hectare) CB 40-69 205.2 CB 41-76 204.5 CB 40-13 199.4 CB 40-19 192.4 Co 419 192.1 CB 38-30 182.1 CB 41-70 181.5 Co 413 177.5 CB 38-22 174.4 CB 36-14 172.8 Co 290 166.6 CB 41-35 147.9 The least significant difference by Tukey's test, at the 5% level of probability, is A = 28.3 metric tons/hectare.

At the 2nd. Department of Zootechny of the E. S. A. L. Q., in Piracicaba, between 1953 and 1955 an experiment of sugar cane varieties was carried out, with the objective of discovering varieties to substitute "Taquara" (the variety most widely used) and Co 290 (the most recommended). The former was condemned as being too susceptible to cane smut and the latter showes signs if degeneracy. In the experiment, 8 varieties were used with 3 replications in randomized blocks, in 3 rows each. The cane was crop not in the same period, but when they were at comparable ripeness (70 cm of apparent culm). They were crop twice during the year, with a sharp hoe near the soil. The summary of the results and the statistical analyses are shown in tables 1 to 3, showing the possibility of there being 3 groups: A superior one composed of Co 419, a median one, in decreasing order of production, composed of Kassoer, CB 40-69. Co 413, IAC 36-25 and POJ 161 and an inferior group composed of Co 290 and Taquara. There is a possibility that POJ 161 belongs to the last group. Nevertheless, this variety is not recommend because of its susceptibility to smut. As Kassoer is more healthy, vigorous and enduring than Co 419 and other varieties, it is shown recommendable. IAC 36-25 is being recommended presently for forage since its productions is lower than Kassoer, placing 5th productivity, although statistical significance was not detected. As our final conclusions, Co 419, Kassoer, CB 40-69, Co 413 and IAC 36-25 can be planted as forage while POJ 161, Co 290 and Taquara should not. The last two were exactly those used as forage reserve in the 2nd. Department at the beginning of the experiment.

The authors carried out 3 experiments on the sampling of sugar cane for technological determinations, one with each of the varieties Co 419, CB 40-69 and CB 41-58, in Piracicaba, State of São Paulo, Brasil. The main intent of the project was to compare 2 methods of sampling, namely: 1) Method A, where the sample is a hill (CATANI et al, 1959) or, more generally, 20 stalks all together in a randomly selected point of the furrow; 2) Method B, where 20 stalks are taken, from 20 points evenly spread but on the whole plot. Coefficients of variation for 20 stalk samples Variety Characteristic 20 stalks per hill 1 stalk per hill Brix 4.8% 1.9% Pol 6.4% 2.5% CB 40-69 Coefficient of purity 2.1% 0.83% Available sucrose 7.3% 2.7% Weight 6.6% 6.9% Brix 5.3% 1.8% Pol 7.6% 2.6% Co 419 Coefficient of purity 2.9% 1.0% Available sucrose 8.6% 3.0% Weight 21.2% 6.5% Brix 2.8% 1.4% Pol 4.1% 1.9% CB 41-58 Coefficient of purity 1.8% 0.8% Available sucrose 5.0% 2.2% Weight 10.9% 6.2% For the 3 varieties studied and for the data on Brix, pol, coefficient of purity, available sucrose and weight, analyses of variance were carried out. Further computations led to the following coefficients of variation. For available sucrose, which is probably the most important characteristic studied, the average coefficient of variation for the 3 varieties was 2.7%, for the case of method B, that is, 20 stalk samples, one stalk per hill. Assuming this coefficient of variation, in a trial with 5 treatments and 6 replications, in randomised blocks, the least significant difference among treatment means, at the 5% level, would be 4.7% of available sucrose by Tukey's test, and 3.3% by the t test. For the case of method A the average coefficient of variation is 7.0% and, in similar conditions, the least significant difference would be 15.1% by Tukey's test, and 12.1% by the t test. Since differences of available sucrose among treatments in experiments with fertilizers seldom are higher than 3 or 4% of the mean (PIMENTEL GOMES & CARDOSO, 1958), method B with a 20 stalk sample per plot gives more or less the minimum amount of cane to be sampled for technological determinations. In experiments with varieties, however, where differences may be assumed to be higher, a sample of 10 to 20 stalks one per hill, can be enough.

We have used monoclonal antibodies specific for acetylated and non-acetylated alpha-tubulin to localize microtubules containing acetylated alpha-tubulin in all developmental forms of the life cycle of Trypanosoma cruzi. This was demonstrated using immunofluorescence and by transmission electron microscopy of thin sections, negative stained cells, and replicas of whole Triton X-100 extracted cells immunolabeled with antibody-gold complex. The antibody specific for acetylated alpha-tubulin (6-11B-1) binds to the flagellar, as well as to the sub-pellicular microtubules. The extent of labeling of the sub-pellicular microtubules with the monoclonal antibody recognized alpha-acetylated tubulin was smaller than that observed with the antibody which recognizes all tubulin isoforms. In relation to the developmental forms, the extent of labeling of the microtubules with antibody 6-11B-1 was larger in epimastigote and trypomastigote than in amastigote forms. Incubation of the parasites for 1 h at 0º C or in the presence of either colchicine or vinblastine did not interfere with the sub-pellicular microtubules. These observations, in agreement with those reported for Trypanosoma brucei brucei (Schneider et al., 1987; Schulze et al., 1987; Sasse per cent Gull, 1988) indicate that the sub-pellicular microtubules of trypanosomatids represent stable microtubules containing acetylated alpha-tubulin (or the alpha 3-tubulin isotype).

Eosinophils, along with mast cells are key cells involved in the innate immune response against parasitic infection whereas the adaptive immune response is largely dependent on lymphocytes. In chronic parasitic disease and in chronic allergic disease, IL-5 is predominantly a T cell derived cytokine which is particularly important for the terminal differentiation, activation and survival of committed eosinophil precursors. The human IL-5 gene is located on chromosome 5 in a gene cluster that contains the evolutionary related IL-4 family of cytokine genes. The human IL-5 receptor complex is a heterodimer consisting of a unique a subunit (predominantly expressed on eosinophils) and a beta subunit which is shared between the receptors for IL-3 & GM-CSF (more widely expressed). The a subunit is required for ligand-specific binding whereas association with the beta subunit results in increased binding affinity. The alternative splicing of the alphaIL-5R gene which contains 14 exons can yield several alphaIL-5R isoforms including a membrane-anchored isoform (alphaIL-5Rm) and a soluble isoform (alphaIL-5Rs). Cytokines such as IL-5 produce specific and non-specific cellular responses through specific cell membrane receptor mediated activation of intracellular signal transduction pathways which, to a large part, regulate gene expression. The major intracellular signal transduction mechanism is activation of non-receptor associated tyrosine kinases including JAK and MAP kinases which can then transduce signals via a novel family of transcriptional factors named signal transducers and activators of transcription (STATS). JAK2, STAT1 and STAT 5 appear to be particularly important in IL-5 mediated eosinophil responses. Asthma is characterized by episodic airways obstruction, increased bronchial responsiveness, and airway inflammation. Several studies have shown an association between the number of activated T cells and eosinophils in the airways and abnormalities in FEV1, airway reactivity and clinical severity in asthma. It has now been well documented that IL-5 is highly expressed in the bronchial mucosa of atopic and intrinsic asthmatics and that the increased IL-5 mRNA present in airway tissues is predominantly T cell derived. Immunocytochemical staining of bronchial biopsy sections has confirmed that IL-5 mRNA transcripts are translated into protein in asthmatic subjects. Furthermore, the number of activated CD 4 + T cells and IL-5 mRNA positive cells are increased in asthmatic airways following antigen challenge and studies that have examined IL-5 expression in asthmatic subjects before and after steroids have shown significantly decreased expression following oral corticosteroid treatment in steroid-sensitive asthma but not in steroid resistant and chronic severe steroid dependent asthma. The link between T cell derived IL-5 and eosinophil activation in asthmatic airways is further strengthened by the demonstration that there is an increased number of alphaIL-5R mRNA positive cells in the bronchial biopsies of atopic and non-atopic asthmatic subjects and that the eosinophil is the predominant site of this increased alphaIL-5R mRNA expression. We have also shown that the subset of activated eosinophils that expressed mRNA for membrane bound alpha IL5r inversely correlated with FEV1, whereas the subset of activated eosinophils that expressed mRNA for soluble alphaIL5r directly correlated with FEV1. Hence, not only does this data suggest that the presence of eosinophils expressing alphaIL-5R mRNA contribute towards the pathogenesis of bronchial asthma, but also that the eosinophil phenotype with respect to alphaIL-5R isoform expression is of central importance. Finally, there are several animal, and more recently in vitro lung explant, models of allergen induced eosinophilia, late airway responses(LARS), and bronchial hyperresponsiveness(BHR) - all of which support a link between IL-5 and airway eosinophila and bronchial hyperresponsiveness. The most direct demonstration of T cell involvement in LARS is the finding that these physiological responses can be transferred by CD4+ but not CD8+ T cells in rats. The importance of IL-5 in animal models of allergen induced bronchial hyperresponsiveness has been further demonstrated by a number of studies which have indicated that IL-5 administration is able to induce late phase responses and BHR and that anti-IL-5 antibody can block allergen induced late phase responses and BHR. In summary, activated T lymphocytes, IL5 production and eosinophil activation are particularly important in the asthmatic response. Human studies in asthma and studies in allergic animal models have clearly emphasised the unique role of IL-5 in linking T lymphocytes and adaptive immunity, the eosinophil effector cell, and the asthma phenotype. The central role of activated lymphocytes and eosinophils in asthma would argue for the likely therapeutic success of strategies to block T cell and eosinophil activation (eg steroids). Importantly, more targeted therapies may avoid the complications associated with steroids. Such therapies could target key T cell activation proteins and cytokines by various means including blocking antibodies (eg anti-CD4, anti-CD40, anti-IL-5 etc), antisense oligonucleotides to their specific mRNAs, and/or selective inhibition of the promoter sites for these genes. Another option would be to target key eosinophil activation mechanisms including the aIL5r. As always, the risk to benefit ratio of such strategies await the results of well conducted clinical trials.

The molecular karyotype of nine Trypanosoma rangeli strains was analyzed by contour-clamped homogeneous electric field electrophoresis, followed by the chromosomal localization of ß-tubulin, cysteine proteinase, 70 kDa heat shock protein (hsp 70) and actin genes. The T. rangeli strains were isolated from either insects or mammals from El Salvador, Honduras, Venezuela, Colombia, Panama and southern Brazil. Also, T. cruzi CL-Brener clone was included for comparison. Despite the great similarity observed among strains from Brazil, the molecular karyotype of all T. rangeli strains analyzed revealed extensive chromosome polymorphism. In addition, it was possible to distinguish T. rangeli from T. cruzi by the chromosomal DNA electrophoresis pattern. The localization of ß-tubulin genes revealed differences among T. rangeli strains and confirmed the similarity between the isolates from Brazil. Hybridization assays using probes directed to the cysteine proteinase, hsp 70 and actin genes discriminated T. rangeli from T. cruzi, proving that these genes are useful molecular markers for the differential diagnosis between these two species. Numerical analysis based on the molecular karyotype data revealed a high degree of polymorphism among T. rangeli strains isolated from southern Brazil and strains isolated from Central and the northern South America. The T. cruzi reference strain was not clustered with any T. rangeli strain.

Between 1985 and 2000, epidemiological surveys of the American tegumentary leishmaniasis (ATL) were carried out in several rural and urban communities in Espírito Santo, Brazil. A total of 100 stocks of Leishmania (comprising isolates from both human and canine hosts with ATL) were identified by two methods of molecular characterization, using specific monoclonal antibodies and multilocus enzyme electrophoresis. Parasite isolates from 19 municipalities were found to belong to the same zymodeme and serodeme type as of the Leishmania (Viannia) braziliensis reference strain. In contrast, our genotyping studies have shown intra-specific variation among these parasites (comparisons of the variability of the internal transcribed spacers between the small and large subunits of the rRNA genes of the 22 stocks studiedrevealed at least 11 genotypes). Two main clusters of L. (V.) braziliensis genotypes were observed, representing parasites collected from different endemic regions in the state, where transmission reflects distinct eco-epidemiological features. Infection with this pathogen was associated with the characteristic disease forms, but neither the clinical outcome nor the response to treatment could be related to the genetic polymorphism of the isolates, as defined by using the proposed methodology.
