An in house indirect immmunofluorescence assay ( IFA ) in relation to neutralization (NT) reference test, was assessed as a fast and cheap method to carry out serological surveys for St. Louis Encephalitis virus (SLE). Sera obtained from 213 blood donors were analyzed by both tests. The prevalence of seropositivity obtained with IFA was lower than (30.98%) that observed on NT (41.78%). The relative specificity rate of IFA was 96.77% whereas its relative sensitivity rate was 69.66%. Kappa index showed a good correlation between both tests. The results indicate that neutralization assay is still the serological test with the highest sensitivity and specificity relative rates for detecting antibodies against SLE virus. Nevertheless, the IFA could be useful as an alternative test in order to learn the circulation of the Flavivirus genus in a certain area.

A serological screening was performed in 615 individuals aged 0-87 years, living in the city of Cordoba, Argentina to study the relationship between antibody prevalence for the SLE virus and age. A 13.98% prevalence of neutralizing antibodies was obtained and its relation to age was significantly high (p = 0.045). The highest seroprevalence was noted on individuals over 60 years old (>20%), whereas no subject under 10 was seropositive for this virus. Our results confirm that the agent is endemic in this area and neurological pathology studies should be performed on those individuals aged 60 since they represent the most susceptible group to SLE virus.

This paper reports the isolation of St. Louis encephalitis virus (SLEV) from a febrile human case suspected to be dengue, in São Pedro, São Paulo State. A MAC-ELISA done on the patient's acute and convalescent sera was inconclusive and hemagglutination inhibition test detected IgG antibody for flaviviruses. An indirect immunofluorescent assay done on the C6/36 cell culture inoculated with the acute serum was positive for flaviviruses but negative when tested with dengue monoclonal antibodies. RNA extracted from the infected cell culture supernatant was amplified by RT-PCR in the presence of NS5 universal flavivirus primers and directly sequenced. Results of BLAST search indicated that this sequence shares 93% nucleotide similarity with the sequence of SLEV (strain-MSI.7), confirmed by RT-PCR performed with SLEV specific primers. Since SLEV was identified as the cause of human disease, it is necessary to improve surveillance in order to achieve early detection of this agent in the state of São Paulo and in Brazil. This finding is also an alert to health professionals about the need for more complete clinical and epidemiological investigations of febrile illnesses as in the reported case. SLEV infections can be unrecognized or confused with other ones caused by an arbovirus, such as dengue.

The dengue virus (DENV), which is frequently involved in large epidemics, and the yellow fever virus (YFV), which is responsible for sporadic sylvatic outbreaks, are considered the most important flaviviruses circulating in Brazil. Because of that, laboratorial diagnosis of acute undifferentiated febrile illness during epidemic periods is frequently directed towards these viruses, which may eventually hinder the detection of other circulating flaviviruses, including the Saint Louis encephalitis virus (SLEV), which is widely dispersed across the Americas. The aim of this study was to conduct a molecular investigation of 11 flaviviruses using 604 serum samples obtained from patients during a large dengue fever outbreak in the state of Mato Grosso (MT) between 2011 and 2012. Simultaneously, 3,433 female Culex spp. collected with Nasci aspirators in the city of Cuiabá, MT, in 2013, and allocated to 409 pools containing 1-10 mosquitoes, were also tested by multiplex semi-nested reverse transcription PCR for the same flaviviruses. SLEV was detected in three patients co-infected with DENV-4 from the cities of Cuiabá and Várzea Grande. One of them was a triple co-infection with DENV-1. None of them mentioned recent travel or access to sylvatic/rural regions, indicating that transmission might have occurred within the metropolitan area. Regarding mosquito samples, one pool containing one Culex quinquefasciatus female was positive for SLEV, with a minimum infection rate (MIR) of 0.29 per 1000 specimens of this species. Phylogenetic analysis indicates both human and mosquito SLEV cluster, with isolates from genotype V-A obtained from animals in the Amazon region, in the state of Pará. This is the first report of SLEV molecular identification in MT.

Introduction Saint Louis encephalitis virus (SLEV) primarily occurs in the Americas and produces disease predominantly in humans. This study investigated the serological presence of SLEV in nonhuman primates and horses from southern Brazil. Methods From June 2004 to December 2005, sera from 133 monkeys (Alouatta caraya, n=43; Sapajus nigritus, n=64; Sapajus cay, n=26) trap-captured at the Paraná River basin region and 23 blood samples from farm horses were obtained and used for the serological detection of a panel of 19 arboviruses. All samples were analyzed in a hemagglutination inhibition (HI) assay; positive monkey samples were confirmed in a mouse neutralization test (MNT). Additionally, all blood samples were inoculated into C6/36 cell culture for viral isolation. Results Positive seroreactivity was only observed for SLEV. A prevalence of SLEV antibodies in sera was detected in Alouatta caraya (11.6%; 5/43), Sapajus nigritus (12.5%; 8/64), and S. cay (30.8%; 8/26) monkeys with the HI assay. Of the monkeys, 2.3% (1/42) of A. caraya, 6.3% 94/64) of S. nigritus, and 15.4% (4/26) of S. cay were positive for SLEV in the MNT. Additionally, SLEV antibodies were detected by HI in 39.1% (9/23) of the horses evaluated in this study. Arboviruses were not isolated from any blood sample. Conclusions These results confirmed the presence of SLEV in nonhuman primates and horses from southern Brazil. These findings most likely represent the first detection of this virus in nonhuman primates beyond the Amazon region. The detection of SLEV in animals within a geographical region distant from the Amazon basin suggests that there may be widespread and undiagnosed dissemination of this disease in Brazil.

Introduction Arboviruses are an important public health problem in Brazil, in especially flaviviruses, including the Saint Louis encephalitis virus (SLEV) and the Rocio virus (ROCV), are especially problematic. These viruses are transmitted to humans or other vertebrates through arthropod bites and may cause diseases with clinical manifestations that range from asymptomatic infection, viral hemorrhagic fever to encephalitis. Methods A serological survey of horses from various regions of Brazil using an enzyme-linked immunosorbent assay (ELISA) with recombinant SLEV domain III peptides and ROCV E protein as antigens. Results Overall, 415 (55.1%) of the 753 horses that were screened were seropositive for flavivirus and, among them, monotypic reactions were observed to SLEV in 93 (12.3%) and to ROCV in 46 (6.1%). These results suggested that these viruses, or other closely related viruses, are infecting horses in Brazil. However, none of the studied horses presented central nervous system infection symptoms. Conclusions Our results suggest that SLEV and ROCV previously circulated among horses in northeast, west-central and southeast Brazil.

Desde 1914, com o trabalho inicial de Castellani, pesquisadores de diversos países teem assinado o encontro de toxoplasmose humana, o que foi até recentemente objeto de controvérsias. Como resultado dos trabalhos de Torres, Levaditi & coL, Wolf, Cowen & Paige, Pinkerton & Henderson e Sabin, a questão do parasitismo do homem por Toxoplasma, deve ser encarada considerando-se que teem sido descritos casos duvidosos e casos não duvidosos: a) Duvidosos, são os casos referidos por Castellani (Ceilão, 1914); Fedorovitch (Mar Negro, 1916); Chalmers & Kamar (Sudão, 1920); e Bland (Londres, 1930-31). b) Não duvidosos são os relatados por Janku (Praga, 1923); Torres, (Rio de Janeiro, 1927); Wolf & Cowen (New-York, 1937); Richter (Chicago, 1936: diagnósticado por Wolf & Cowen em 1938); Wolf, Cowen & Paige (New-York, 1939); Hertig (Massachusetts, 1935: diagnósticado por Pinkerton & Weinman em 1940); Pinkerton & Weinman (Lima-Peru, 1940); Sabin (dois casos, Cincinnati, 1941); Pinkerton & Henderson (dois casos, St. Louis, 1941); Paige, Cowen & Wolf (três casos, New-York, 1942); e De Lange (Amsterdam, 1929: diagnósticado por Paige, Cowen & Wolf em 1942). Destes relatos considerados como casos não duvidosos de toxoplasmose humana, 10 representam uma doença congênita (Torres e Paige, Wolf & Cowen) ocorrendo em crianças recemnascidas ou de poucos meses de vida. Como nas vezes em que foi possivel examinar as mães dessas crianças tratava-se de mulheres com aspecto sadio, concluiu-se que a moléstia, embora sumamente grave e mortal, pode apresentar uma forma inaparente. Em dois casos, provas de soro proteção demonstraram essa infecção materna inaparente, pelo encontro de anticorpos neutralizantes para Toxoplasma (Paige, Cowen & Wolf). Nada obstante não ter sido feito um estudo microscópico detalhado em todos os casos, a doença caracteriza-se por uma meningoence falomielite granulomatosa (Wolf & Cowen), associada frequentemente à miocardite e corioretinite. Os sintomas apresentados pelos pacientes, foram: febre, convulsões, distúrbios respiratórios, hidrocefalia, cianose, vômitos, labidade de temperatura, etc., sendo sinal diagnóstico de máxima importância, o encontro de focos de calcificação cerebral profunda e corioretinite em recem nascido (Dyke, Wolf, Cowen, Paige & Caffey) . Em dois casos de toxoplasmose verificados em pacientes de seis e oito anos respectivamente, foi encontrada uma encefalite clinicamente atípica, sendo que um deles sarou (Sabin). Os três casos restantes foram descritos em adultos; num deles as lesões não eram típicas (havia concomitância de infecção por Bartonella bacilliformis) e nos outros dois, a doença assumiu uma "forma exantemática", simulando as febres maculosas (Pinkerton & Henderson), sendo o achado mais importante microscopicamente, uma pneumonia intersticial. Alem da semelhança clínica, tambem os focos de encefalite encontrados nessa "forma exantemática" da toxoplasmose, são semelhantes aos descritos nas...

Observações de laboratório sobre o pilideo Pomacea haustrum (Reeve, 1856) competidor-predador de planorbineos hospedeiros intermediários da esquistossomose mansoni mostraram que: Machos e fêmeas atingem maturidade sexual após um ano de idade, copulando preferencialmente pela manhã e ovipondo à noite. O tempo gasto nestes atos é variável; no caso da oviposição ele depende do número de ovos a serem postos. Os ovos são arredondados, com diâmetro médio de 3mm e coloração rósea que vai se alterando à medida que os embriões se desenvolvem. Com um período de incubaçãod e 15 a 23 dias, condicionado pela temperatura ambiente, eles resistem até 5 a 6 dias imersos em água sem danos aos embriões; independem de luz para eclodir. Os exemplares recém-eclodido têm, em média, 2,4 por 1,7mm de altura e diâmetro, respectivamente. Criados em isolamento crescem e sobrevivem mais que quando criados em grupo. Resistem pelo menos 90 dias fora d'água, mantendo-se neste período dentro das conchas, com o opérculo hermeticamente fechado, em anidrobiose. Nesta fase, podem morrer por ataque de larvas de dípteros.

Algumas observações de campo sobre o pilídeo Pomacea haustrum (Reeve, 1856), predador-competidor de planorbíneos hospedeiros intermediários da esquistossomose mansoni mostraram que: Cópula e oviposição são realizadas, preferencialmente, à noite e de madrugada. As desovas - cujas dimensões e formas dependem do número de ovos e tipos dos suportes - sao ovipostas, na maioria das vezes, de 6 a 10 cm acima do nível da água eclodindo após 9 a 30 dias de incubação; o número médio de ovos por desova foi de 236. A alimentação habitual deste molusco consiste de algas confervóides, além de vegetais aquáticos natantes e submersos; entretanto, e comum a utilização de outros materiais como alimento.Tem como habitat a zona marginal mais rasa das coleções hídricas onde e encontrado, predominando, aparentemente, nos ambientes lênticos. A distribuição espacial esta condicionada por determinadas características dos biótopos. Algumas aves aquáticas - anatídeos, ralídeos, etc.- revelaram-se importantes inimigos naturais, atacando além das desovas exemplares jovens e adultos; em determinadas condições podem constituir fator impeditivo da implantação e colonização de novos biótopos.

The ultrastructure of endogenous stages of Eimeria ninakohlyakimovae was observed in epithelial cells of cecum and colon crypts from a goat experimentally infected with 2.0 x 105 oocysts/kg. The secondary meronts developed above the nucleus of the host cell. The nucleus first divides and merozoites then form on the surface of multinucleated meronts. Free merozoites in the parasitophorous vacuole present a conoid, double membrane, one pair of rhoptries, micronemes, micropore, anterior and posterior polar ring, a nucleus with a nucleolus and peripheral chromatin. The microgamonts are located below the nucleus of the host cell and contain several nuclei at the periphery of the parasite. The microgametes consist of a body, a nucleus, three flagella and mitochondria. The macrogamonts develop below the nucleus of the host cell and have a large nucleus with a prominent nucleolus. The macrogametes contain a nucleus, wall-forming bodies of type I and type II. The young oocysts present a wall containing two layers and a sporont

Dirofilaria immitis is a widespread mosquito-borne parasite that causes dirofilariasis, a commonly diagnosed disease of dogs that is rarely reported in cats and humans. A mosquito survey was conducted in Itacoatiara in the State of Rio de Janeiro, from March 1995 to February 1996, using canine, feline and human baits. A total of 3,667 mosquitoes were dissected for D. immitis larvae, representing 19 species and 10 genera. From those, Ae. scapularis, Ae taeniorhynchus, Cx. quinquefasciatus, Cx. declarator, Cx. saltanensis and Wy. bourrouli were found infected with D. immitis parasites, and among those, only the first three harbored infective larvae. The majority of larvae were found in the Malpighian tubules (889/936), and larval melanization was observed in the two Aedes species. In descending order, the best vectors were Ae. scapularis, Ae. taeniorhynchus, and Cx. quinquefasciatus which alternate seasonally in importance. Cx. quinquefasciatus is suggested to be a vector to cats. The potential transmission of D. immitis parasites by these three vectors to man is discussed.
