The enteropathogenic role of cytotoxic necrotizing factor (CNF)-producing Escherichia coli was investigated by searching cnf genes among 2074 isolates from 200 children with and 200 without acute diarrhea in Brazil. Fourteen (7%) cases versus 10 (5%) control children carried at least one cnf positive isolate (P = 0.50) and most isolates expressed CNF type 1. DNA sequences of virulence factors of extraintestinal pathogenic E. coli (ExPEC) were detected in 78.6% of CNF1-producing isolates. Besides not being associated with human acute diarrhea, the CNF1-producing isolates here identified may represent potential ExPEC transitorily composing the normal intestinal flora.

E-2-chloro-8-methyl-3-[(4'-methoxy-1'-indanoyl)-2'-methyliden]-quinoline (IQ) is a new quinoline derivative which has been reported as a haemoglobin degradation and ß-haematin formation inhibitor. The haemoglobin proteolysis induced by Plasmodium parasites represents a source of amino acids and haeme, leading to oxidative stress in infected cells. In this paper, we evaluated oxidative status in Plasmodium berghei-infected erythrocytes in the presence of IQ using chloroquine (CQ) as a control. After haemolysis, superoxide dismutase (SOD), catalase, glutathione cycle and NADPH + H+-dependent dehydrogenase enzyme activities were investigated. Lipid peroxidation was also assayed to evaluate lipid damage. The results showed that the overall activities of glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase were significantly diminished by IQ (by 53.5% and 100%, respectively). Glutathione peroxidase activity was also lowered (31%) in conjunction with a higher GSSG/GSH ratio. As a compensatory response, overall SOD activity increased and lipid peroxidation decreased, protecting the cells from the haemolysis caused by the infection. CQ shared most of the effects showed by IQ; however it was able to inhibit the activity of isocitrate dehydrogenase and glutathione-S-transferase. In conclusion, IQ could be a candidate for further studies in malaria research interfering with the oxidative status in Plasmodium berghei infection.

Leprosy is an infectious and contagious spectral disease accompanied by a series of immunological events triggered by the host response to the aetiologic agent, Mycobacterium leprae . The induction and maintenance of the immune/inflammatory response in leprosy are linked to multiple cell interactions and soluble factors, primarily through the action of cytokines. The purpose of the present study was to evaluate the serum levels of tumour necrosis factor (TNF)-α and its soluble receptors (sTNF-R1 and sTNF-R2) in leprosy patients at different stages of multidrug treatment (MDT) in comparison with non-infected individuals and to determine their role as putative biomarkers of the severity of leprosy or the treatment response. ELISA was used to measure the levels of these molecules in 30 healthy controls and 37 leprosy patients at the time of diagnosis and during and after MDT. Our results showed increases in the serum levels of TNF-α and sTNF-R2 in infected individuals in comparison with controls. The levels of TNF-α, but not sTNF-R2, decreased with treatment. The current results corroborate previous reports of elevated serum levels of TNF-α in leprosy and suggest a role for sTNF-R2 in the control of this cytokine during MDT.

Human T-cell lymphotropic virus (HTLV) may impact the clinical course of tuberculosis (TB). Both infections are highly endemic in Brazil. The aim of this study was to assess the prevalence of HTLV-1/2 in TB patients in Central-West Brazil and to perform a genetic characterisation of the respective isolates. Of the 402 patients, six (1.49%) were positive for anti-HTLV and five (1.24%; 95% confidence interval: 0.46-3.05) were infected with HTLV-1/2. Genetic characterisation demonstrated that the four HTLV-1 isolates belonged to the Transcontinental subgroup A of the Cosmopolitan subtype a and that the HTLV-2 isolate belonged to subtype a (HTLV-2a/c). The prevalence of HTLV infection observed in this study is higher than that observed in local blood donors and the HTLV-1 and 2 subtypes identified are consistent with those circulating in Brazil.

Este trabalho teve como objetivo avaliar o efeito de indutores de florescimento em três cultivares de abacaxizeiro em Rio Branco-Acre. O delineamento experimental foi em blocos ao acaso, em esquema fatorial 3 x 2 x 2, com quatro repetições, sendo combinadas três cultivares de abacaxi (RBR-1, SNG-2 e SNG-3), dois indutores de florescimento (carbureto de cálcio- CaC2 e etefon-ácido 2-cloroetilfosfônico) e duas épocas de aplicação (aos 10 e 12 meses do plantio). As plantas induzidas com etefon aos 10 meses apresentaram maior percentagem de florescimento (96,25%) quando comparadas com as induzidas com CaC2 (62,92%). A percentagem de florescimento foi significativamente superior nas plantas induzidas aos 12 meses com CaC2 (85,42%) quando comparadas com as induzidas aos 10 meses (62,92%).

A new method for the preparation of 2-chloro-1,3,2-dioxaphospholane (1) and 2-chloro-4,5-benzo-1,3,2-dioxaphospholane (2), are reported. The modifications introduced in the synthetic route improved the yield and facilitated the control of reaction, but the synthesis require longer reaction time. The compounds were characterized by ¹H, 13C{¹H} and31P{¹H} NMR spectroscopy. Due to the complexity of the spin system AA'BB'X (A, A', B, B' = ¹H; X = 31P) of 2, a simulation of the ¹H NMR spectra was done and it's in agreement with the bibliography.

The [3+4] cycloaddition between furan and the oxyallyl cation generated from 1-bromo-1-phenylpropan-2-one (4), resulted in the formation of 2-phenyl-8-oxabicyclo[3.2.1]oct-6-en-3-one (5) in 30% yield. This compound was further converted into 2-phenyl-6,7-exo-isopropylidenedioxi-8-oxabicyclo[3.2.1]oct-2-ene (13) in 35.4% yield. The selective effect of compound (13) and its isomer 3-phenyl-6,7-exo-isopropylidenedioxi-8-oxabicyclo[3.2.1]oct -2-ene (1a) on the radicle growth of Sorghum bicolor L. (sorghum) and Cucumis sativus L. (cucumber) were evaluated. For both plants, compound 13 showed to be more potent than its isomer 1a.

Preparation of the title compounds are described as an alternative nucleophilic aromatic substitution for practices in the graduate laboratory. The low toxicity and disponibility of the reagents make a suitable procedure approach to experiments regarding this aromatic reaction.

A new sensitive and selective procedure for speciation of trace dissolved Fe(III) and Fe(II), using modified octadecyl silica membrane disks and determination by flame atomic absorption spectrometry was developed. A ML3 complex is formed between the ligand and Fe(III) responsible for extraction of metal ion on the disk. Various factors influencing the separation of iron were investigated and the optimized operation conditions were established. Under optimum conditions, an enrichment factor of 166 was obtained for Fe3+ ions. The calibration graph using the preconcentration system for Fe3+ was linear between 40.0 and 1000.0 μg L-1.

Both primary and secondary amines react with 2,4-dinitrochlorobenzene to give derivatives of 1-alkylamino-2,4-dinitrobenzene. These compounds are important intermediates for the synthesis of a diverse range of products. The methodology reported in the present study involves either the room temperature reaction or heating at 70 °C in ethanol in the presence of triethylamine. This transformation occurs via a nucleophilic substitution reaction. The 1-alkylamino-2,4-dinitrobenzene derivatives were obtained in greater than 90% purified yield. The selective reduction of dinitro compounds is an important synthetic strategy for the synthesis of intermediates for dyes, pharmaceuticals and agrochemicals. The use of SnCl2 as a suspension in EtOAc is a promising method for the regio- and chemo-selective reduction of 1-alkylamino-2,4-dinitrobenzenes to 1-alkylamino-2-amino-4-nitrobenzenes. These products are useful intermediates in organic synthesis.

Com a finalidade de verificar a ocorrência e a distribuição de biovares de Ralstonia solanacearum no Estado do Rio Grande do Sul, plantas com sintomas de murcha-bacteriana foram coletadas, na primavera de 1999, em 25 lavouras de batata (Solanum tuberosum) em dez municípios das quatro áreas de produção: Serra do Nordeste, Planalto Superior, Depressão Central e Grandes Lagoas. A determinação da espécie foi feita com teste sorológico através de ELISA e da biovar com base no metabolismo oxidativo de açúcares e álcoois. Noventa e quatro porcento dos 490 isolados foram identificados como biovar 2. A ocorrência da biovar 1 foi registrada na Serra do Nordeste e Depressão Central. A ocorrência das biovares não mostrou relação com a temperatura média local ou cultivar de batata.

Bis-(µ2-oxo)-tetrakis{[1-feniltriazene-1,3-diil)-2-(phenyltriazenil)benzene copper(II) is a tetranuclear complex which shows four Cu(II) ions coordinated by four 1,2-bis(phenyltriazene)benzene bridged ligands, with one diazoaminic deprotonated chain, and two O2- ligands. The complex reduces at E1/2 = -0.95 V vs Fc+/Fc, a two electrons process. Cyclic voltammetric and spectroelectrochemical studies showed a reversible process. When immobilized on carbon paste electrode, the complex electrocatalyses the reduction of O2 dissolved on aqueous solution at -0.3 V vs SCE potential. The obtained current shows linearity with O2 concentration.

Objetivos: avaliar a soroprevalência da infecção causada pelo HSV-2 entre as parturientes do Hospital das Clínicas da Faculdade de Medicina de Ribeirão Preto (HCFMRP-USP) e padronizar técnicas laboratoriais para atender a este propósito. Métodos: foram avaliadas 1.500 amostras de sangue de parturientes atendidas no Centro Obstétrico do Departamento de Ginecologia e Obstetrícia do HCFMRP-USP, entre 1º de janeiro e 31 de outubro de 1996. Para determinar a real prevalência da infecção por HSV-2 foi padronizada a técnica de ELISA, verificando-se que esta não apresentava especificidade suficiente para discriminar os dois tipos virais (75%), delineando a necessidade de utilizar-se técnica de maior poder discriminatório. A técnica padronizada para esta finalidade foi o Western blot, capaz de detectar a proteína viral específica do HSV-2. Resultados: a soroprevalência para infecção herpética, pelos dois tipos virais (HSV-1 e HSV-2), foi de 94,5%, utilizando a técnica de ELISA. Com o emprego da técnica de Western blot, encontrou-se a soroprevalência de 31,9% pelo HSV-2 na população avaliada, quer sintomática ou assintomática. Conclusão: verifica-se elevada prevalência do estado de portadora da infecção pelos HSV, evidenciada pelo alto índice de positividade para os anticorpos contra estes vírus. O teste ELISA não mostrou especificidade suficiente para discriminar os anticorpos anti-HSV-2 dos anti-HSV-1.

Este artigo relata a caracterização de duas amostras citopáticas do vírus da Diarréia Viral Bovina (BVDV-1: IBSP-2; BVDV-2: SV-253) submetidas à atenuação por múltiplas passagens em cultivo celular e exposição à radiação ultravioleta. As amostras foram caracterizadas in vitro (tamanho e morfologia de placas, cinética de replicação e perfil antigênico) e in vivo (atenuação e resposta sorológica em bovinos). A caracterização in vitro de populações clonadas dos vírus obtidas nas diferentes passagens em cultivo celular (0, 1, 10, 20 e 30), demonstrou que o processo de atenuação não afetou negativamente as características antigênicas e fenotípicas das amostras. Não foram observadas alterações significativas no tamanho e morfologia de placas e na cinética de replicação. A reatividade com 48 anticorpos monoclonais demonstrou que o perfil antigênico não doi alterado durante as sucessivas passagens in vitro. A inoculação intramuscular dos clones de vírus obtidos na passagem 30 (IBSP-2: 10(7,3) DICC50; SV-253: 10(6,8) DICC50) em 12 novilhas soronegativas com idade média de 15 meses, não resultou em sinais clínicos, comprovando sua atenuação. Após a inoculação, o vírus foi detectado em leucócitos da maioria dos animais inoculados (10/12) entre os dias 3 e 6 pós-inoculação (pi) e em secreções nasais de três animais (dias 4, 7 e 8pi). No entanto, não ocorreu transmissão dos vírus vacinais aos três animais soronegativos mantidos como sentinelas. Todos os animais vacinados soroconverteram aos 14 dias pós-vacinação (dpv). Títulos moderados a altos de anticorpos neutralizantes foram detectados frente a 5 isolados brasileiros do BVDV-1 (títulos de 80 a > 1280) e quatro isolados do BVDV-2 (títulos de 20 a 640). Em geral, os títulos foram de magnitude superior frente a isolados brasileiros do BVDV-1. Aos 240dpv, os animais receberam uma segunda dose dos vírus vacinais (IBSP-2: 10(7,3) DICC50; SV-253: 10(6,8) DICC50). A revacinação induziu uma resposta secundária na maioria dos animais, resultando em um aumento dos títulos de anticorpos neutralizantes principalmente frente ao BVDV-2. Esses resultados são promissores no sentido da utilização dessas amostras na formulação de vacinas atenuadas para o controle da infecção pelo BVDV no Brasil.
