Um protocolo de PCR multiplex foi estabelecido para a detecção do Banana streak virus (BSV) e do Cucumber mosaic virus (CMV) em bananeiras micropropagadas. Estes vírus são responsáveis por perdas na produção de bananas em todo o mundo. Alguns trabalhos descrevem a integração do BSV no genoma B da bananeira. Contudo, a existência de bananeiras híbridas livres do BSV tem sido demonstrada. Ademais, determinadas estirpes do CMV não são transmitidas mecanicamente sob condições de laboratório, nem tampouco detectadas por testes sorológicos. Como conseqüência, a indexação de matrizes para cultura de tecido algumas vezes se mostra ineficiente. A metodologia apresentada neste trabalho sobrepõe esta dificuldade, pois se baseia na detecção do ácido nucléico viral presente em amostras foliares de bananeira. Na reação, foram usados os oligonucleotídeos BADNA 1A e BADNA 4, para a detecção do BSV, e "CMV senso" e "CMV antisenso" para a detecção do CMV. Após a eletroforese foi verificada a presença de dois fragmentos de DNA amplificados simultaneamente, um dos quais com 597 pb correspondente ao BSV e o outro, com 488 pb, correspondente ao CMV. Este resultado indica que o PCR multiplex pode ser utilizado como uma ferramenta adicional na indexação do BSV e do CMV em bananeiras propagadas por cultura de tecido.

Aqueous extracts of the leaves of Ocimum gratissimum at 10, 25, 40 and 50% (w/v) concentrations induced the production of phytoalexins in soybean cotyledons and sorghum mesocotyls. The aqueous extracts also induced systemic resistance in cucumber to Colletotrichum lagenarium, reflected by reduction in disease incidence and an increase in chitinase production. Modes of action and the existence of possible elicitors of defense response in O. gratissimum leaf extracts are discussed.

Visceral Leishmaniasis (VL) is caused by protozoan of genus Leishmania and transmitted by sand flies of genus Lutzomyia, which has been adapted to the peridomicile environment where dogs are their mainly food source, increasing the risk for human cases. In this study, techniques of geoprocessing and spatial statistics were utilized as a contribution to understanding the epidemiological dynamics of VL in the urban area of Ilha Solteira, SP.

PURPOSE: To determine fetal heart rate (FHR) responses to maternal resistance exercise for the upper and lower body at two different volumes, and after 25 minutes post-exercise.METHODS: Ten pregnant women (22-24 weeks gestation, 25.2±4.4 years of age, 69.8±9.5 kg, 161.6±5.2 cm tall) performed, at 22-24, 28-32 and 34-36 weeks, the following experimental sessions: Session 1 was a familiarization with the equipment and the determination of one estimated maximum repetition. For sessions 2, 3, 4 and 5,FHR was determined during the execution of resistance exercise on bilateral leg extension and pec-deck fly machines, with 1 and 3 sets of 15 repetitions; 50% of the weight load and an estimated repetition maximum. FHR was assessed with a portable digital cardiotocograph. Results were analyzed using Student's ttest, ANOVA with repeated measures and Bonferroni (α=0.05; SPSS 17.0).RESULTS: FHR showed no significant differences between the exercises at 22-24 weeks (bilateral leg extension=143.8±9.4 bpm, pec-deck fly=140.2±10.2 bpm, p=0.34), 28-30 weeks (bilateral leg extension=138.4±12.2 bpm, pec-deck fly=137.6±14.0 bpm, p=0.75) and 34-36 weeks (bilateral leg extension=135.7±5.8 bpm, pec-deck fly=139.7±13.3 bpm, p=0.38), between the volumes(bilateral leg extension at 22-24 weeks: p=0.36, at 28-30 weeks: p=0.19 and at 34-36 weeks: p=0.87; pec-deck fly at 22-24 weeks: p=0.43, at 28-30 weeks: p=0.61 and at 34-36 weeks: p=0.49) and after 25 minutes post-exercise.CONCLUSION: Results of this pilot study would suggest that maternal resistance exercise is safe for the fetus.

Haematobia irritans tem causado muitos danos e preocupações na pecuária mundial, bem como despertado o interesse para diversos estudos a seu respeito. Seu nome está relacionado com o local de permanência nos bovinos. É conhecida como "horn fly" (mosca-dos-chifres) na Europa e nos Estados Unidos da América e mosca-da-paleta na América Latina. Os fatores biológicos podem produzir em bovinos de um único rebanho, diferentes níveis de infestação da mosca. Durante o ano de 1998 em Araçatuba, estado de São Paulo, foram avaliados o número médio de mosca por região ana-tômica, bem como os diferentes níveis de infestação em 60 bovinos da raça Nelore. Os bovinos foram filmados de ambos os lados do corpo para registrar o número de mosca em fitas cassetes. As fitas foram assistidas para a contagem e demarcação da mosca em 15 regiões anatômicas. O maior número de mosca (p<0,05) foi observado nas regiões escapular, interescapular e costal; nos períodos chuvosos observou-se um aumento significativo (p<0,05) na região ventral. As avaliações individuais, demonstraram infestação com menos de 50 moscas em 50% dos bovinos, 50 a 100 moscas em 38% e acima de 100 moscas em 12% dos bovinos.

Blood-sucking diptera are important parasites in bovine production systems, especially regarding confinement conditions. Haematobia irritans, the horn fly, is one of the most troublesome species within bovine production systems, due to the intense stress imposed to the animals. An important aspect while studying the variability within a species is the study of the geographic structure of its populations and, attempting to find out the genetic flow of Brazilian populations of horn fly, the RAPD technique, which is suited for this purpose, has been used. The use of molecular markers generated from RAPD made it possible to identify the geographic origin of samples from different Brazilian geographic regions, as well as to estimate the genotypic flow among the different Brazilian populations of the horn fly.

Imazapyr has been used to control stump sprouting in stand of Eucalyptus plantations, where herbicide is applied to the tree trunk before cutting. The herbicide is applied exclusively on the stump to be killed, but little is known about the final fate of the molecule. Imazapyr exudation via roots of eucalypt grown in soil as the substrate was evaluated under greenhouse conditions. Different herbicide doses (0.000, 0.375, 0.750, 1.125, 1.500, and 3.000 kg ha-1 a.i.) were applied on the aerial parts of 8-month-old Eucalyptus grandis clonal seedlings, cultivated in pots with 18.0 dm³ of soil. Forty days after this treatment, the eucalypt plants were cut and a lateral opening in the containers was made and the plants inclined 90º, with plants sensitive to herbicide presence (sorghum and cucumber) sown into the openings along the exposed soil surface. After 15-day sowing, toxicity symptoms on the shoots as well as the shoot and root system dry biomass of the bio-indicators were evaluated. The results suggest that eucalypt roots do exude imazapyr, and/or its metabolites, at concentrations high enough to cause toxicity to the bio-indicators. Toxicity effects were observed in all plants sown along the exposed soil profile of the container, with higher intensity at higher doses.

Aminocyclopyrachlor and indaziflam are under development in Brazil and there is no information about their behavior in Brazilian soils. This study aimed to evaluate the sensitivity of plant species to these new molecules, trying to select plants that can be used as bioindicators for testing the behavior of these herbicides in the soil. Two experiments were conducted, one for each herbicide. The treatments were arranged in a 8 x 6 factorial design, the factors being represented by eight species used as bioindicators cotton, maize, soybean, sorghum, sunflower, millet, cucumber and beet, and six doses of herbicides (aminocyclopyrachlor - 0, 10 , 20, 30 , 40 and 50 g ha-1 and indaziflam 0 , 20, 40 , 60, 80 and 100 g ha-1). Among the species studied, soybean and beet were quite sensitive to the two new herbicide molecules, being great alternatives for bioassays in order to detect low concentrations of aminocyclopyrachlor and indaziflam in the soil.

A biologia de reprodução de Casearia grandiflora (Flacourtiaceae) foi estudada em um remanescente de mata mesófila do Parque do Sabiá, Uberlândia - MG. A espécie é importante no sub-bosque de matas da região, florescendo durante quase todo o ano, mas com maior abundância nos meses de março, abril e maio. As flores são branco-esverdeadas com cerca de 7 mm de diâmetro, dispostas em capítulos sésseis axilares. Não apresentam odor perceptível e duram apenas 1 dia. Possuem 10 estames férteis livres entre si e unidos à corola na base. Entre os estames existem estruturas pilosas originárias do receptáculo floral. Os estames e estas estruturas formam um cone em torno do pistilo, onde se acumula o néctar. A antese ocorre de forma irregular, principalmente no início da manhã. O néctar é relativamente abundante (4 miL) e com concentração média de 38% de equivalentes de sacarose. O pólen é liberado nas horas mais quentes do dia, com alta viabilidade (96,6%). Neste horário também ocorre a receptividade estigmática. O visitante mais freqüente foi a mosca Ornidia obesa (Syrphidae), que visita as flores durante quase todo o dia, embora tenham sido observados também outros visitantes casuais como abelhas Meliponinae, borboletas e outras moscas não identificadas. Polinizações controladas mostraram que a planta é auto-incompatível e não apomítica. No entanto, foram observados tubos polínicos crescendo até o ovário e penetrando os óvulos em flores autopolinizadas, sugerindo a ocorrência de fenômenos de auto-esterilidade de ação tardia (pós-fertilização) ou depressão de endogamia. A auto-esterilidade em C. grandiflora contrasta com a autogamia observada na maior parte das plantas miófilas e de flores generalistas ocorrendo nas florestas do Brasil central, mas é semelhante aos sistemas de reprodução de outras Flacourtiaceae lenhosas estudadas em florestas Neotropicais.

A line of one-winged Drosophila subobscura was studied. The absent wing is substituted by a bulky structure with macro and microchaetes, showing a thoracic appearance. Genetic crosses showed that there is no way to select for the trait by simple crossing. The number of one-winged flies in the various generations was always low. In one case a wingless fly was also obtained. The trait presents an unknown genetic pattern.

Centromere function requires the proper coordination of several subfunctions, such as kinetochore assembly, sister chromatid cohesion, binding of kinetochore microtubules, orientation of sister kinetochores to opposite spindle poles, and their movement towards the spindle poles. Centromere structure appears to be organized in different, separable domains in order to accomplish these functions. Despite the conserved nature of centromere functions, the molecular genetic definition of the DNA sequences that form a centromere in the yeasts Saccharomyces cerevisiae and Schizosaccharomyces pombe, in the fruit fly Drosophila melanogaster, and in humans has revealed little conservation at the level of centromere DNA sequences. Also at the protein level few centromere proteins are conserved in all of these four organisms and many are unique to the different organisms. The recent analysis of the centromere structure in the yeast S. pombe by electron microscopy and detailed immunofluorescence microscopy of Drosophila centromeres have brought to light striking similarities at the overall structural level between these centromeres and the human centromere. The structural organization of the centromere is generally multilayered with a heterochromatin domain and a central core/inner plate region, which harbors the outer plate structures of the kinetochore. It is becoming increasingly clear that the key factors for assembly and function of the centromere structure are the specialized histones and modified histones which are present in the centromeric heterochromatin and in the chromatin of the central core. Thus, despite the differences in the DNA sequences and the proteins that define a centromere, there is an overall structural similarity between centromeres in evolutionarily diverse eukaryotes.

Insertional mutagenesis is an important tool for functional genomics in Drosophila melanogaster. The insertion site in the KG00562 mutant fly line has been mapped to the CG8709 (herein named DmLpin) locus and to the 3’ of kermit (also called dGIPC). This mutant line presents a high lethality rate resulting from a gain of function. To obtain some insight into the biological role of the mutated locus, we have characterized the mutation and its relation to the high mortality of the KG00562 fly line. In this mutant, we did not detect one of the DmLpin transcripts, namely DmLpinK, but we did detect an unusual 2.3-kb mRNA (LpinK-w). Further investigation revealed that the LpinK-w transcript results from an aberrant splicing between the untranslated first exon of DmLpinK and the mini-white marker gene. Lack of DmLpinK or LpinK-w expression does not contribute to lethality, since heterozygous KG00562/Def7860 animals presented lethality rates comparable to those of the wild type. In contrast, the overexpression of kermit was associated with lethality of the KG00562 fly line. Significantly higher levels of kermit were detected in the Malpighian tubules of KG00562/+ flies that presented higher lethality rates than wild-type or KG00562/Def7860 animals, in which the lethality was rescued. In agreement with a recently reported study, our data support the hypothesis that misexpression of kermit/dGIPC could interfere with Drosophila development, with further investigations being needed in this direction.

A primary interest of image analysis of X-rayed seeds is to identify whether the extent of fill in the embryo cavity is associated with to seed physiological quality. The objective of this research was to verify the accuracy of the freely available Tomato Analyzer (TA) software developed at The Ohio State University to determine the ratio of embryo size over total seed area. Seeds of pumpkin, watermelon, cucumber and cotton were X-rayed and analyzed by the software which defines seed and embryo boundaries and automatically generates numerical values to quantify that ratio. Results showed that the TA has the sensitivity to evaluate the extent of embryo growth within the cucurbits and cotton seeds and is a promising alternative for this assessment in other seed species.

O presente trabalho foi realizado com o objetivo de identificar métodos, baseados na tolerância das sementes ao estresse térmico, eficientes para a avaliação do potencial fisiológico das sementes de pepino. O trabalho foi desenvolvido em três épocas, utilizando-se lotes de sementes dos cultivares Safira e Jóia, abrangendo testes de germinação, emergência de plântulas, envelhecimento acelerado e deterioração controlada. Concluiu-se que o teste de deterioração controlada (com ajuste do teor de água para 24%, período de 48 h a 45 ºC) e de envelhecimento acelerado tradicional e com solução saturada de cloreto do sódio (a 41 ºC por 96 h) é sensível para a avaliação do potencial fisiológico de sementes de pepino.
