Objective The aim of this study was to determine the expression of the immunohistochemical markers p16 and Ki-67 in cervical intraepithelial neoplasms and their influence on the level of agreement among different observers and for the same observer. Methods The study included 184 patients with cervical intraepithelial neoplasms previously confirmed through biopsies performed between 2005 and 2006. Three pathologists reviewed the biopsies by using hematoxylin-eosin staining to reach a consensus on the diagnosis. Subsequently, an immunohistochemical study analyzed the expression of p16 and Ki-67 in such cases. Results The comparison among the reviewing pathologists revealed only moderate agreement (kappa = 0.44). The agreement improved when the differentiation of highgrade lesions (cervical intraepithelial neoplasm - CIN - 3) was analyzed (kappa = 0.59). p16 staining exhibited a high negative predictive value and sensitivity; however, the specificity was low. Overall, both qualitative and quantitative analyses of p16 and a quantitative analysis Ki-67 exhibited low accuracy. The agreement among diagnoses before immunohistochemistry was 0.47. The use of immunohistochemistry increased the agreement to 0.68. Conclusion Our study showed that the agreement among observers using traditional diagnostic criteria of cervical intraepithelial lesions can improve with the use of immunohistochemistry.

A acupuntura, é uma terapia que cada vez mais adquire uma credibilidade no mundo ocidental, a sua indicação pode ser para pacientes anêmicos e imunossuprimidos, e outras afecções. O experimento foi realizado com oito cães adultos, sendo 4 fêmeas e 4 machos. O estudo consistiu em análise das concentrações das células vermelhas e brancas avaliado em 4 tratamentos: controle (T0), acupuntura (T1), eletroacupuntura unilateral (T2) e eletroacupuntura bilateral (T3). Os acupontos utilizados foram intestino grosso 4, intestino grosso 11 e estômago 36. Todos os animais passaram por todos os tratamentos com um intervalo de 7 dias para cada tratamento. Os hemogramas séricos foram verificados 20 minutos antes do início do tratamento (M0), logo após o tratamento (M1), e 60 minutos (M2) e 120 minutos (M3) após o tratamento. O resultado da série vermelha não foi significativo, mas notou-se somente uma diminuição significativa da concentração média corpuscular (CHCM) (p<0,05) no tratamento T1 no M1 em comparação aos outros tratamentos; notou-se também uma redução significativa (p<0,05) na concentração plaquetária de T1, T2 e T3 em comparação ao grupo controle. A eletroacupuntura pode levar a uma trombocitopenia, quando estimulado estes pontos, provavelmente por um estímulo parassimpaticomimético.

Polarization curves experimentally obtained in the electro-dissolution of iron in a 1 M H2SO4 solution using a rotating disc as the working electrode present a current instability region within the range of applied voltage in which the current is controlled by mass transport in the electrolyte. According to the literature (Barcia et. al., 1992) the electro-dissolution process leads to the existence of a viscosity gradient in the interface metal-solution, which leads to a velocity field quantitatively different form the one developed in uniform viscosity conditions and may affect the stability of the hydrodynamic field. The purpose of this work is to investigate whether a steady viscosity profile, depending on the distance to the electrode surface, affects the stability properties of the classic velocity field near a rotating disc. Two classes of perturbations are considered: perturbations monotonically varying along the radial direction, and perturbations periodically modulated along the radial direction. The results show that the hydrodynamic field is always stable with respect to the first class of perturbations and that the neutral stability curves are modified by the presence of a viscosity gradient in the second case, in the sense of reducing the critical Reynolds number beyond which perturbations are amplified. This result supports the hypothesis that the current oscillations observed in the polarization curve may originate from a hydrodynamic instability.

Although labor is intensive, evaluating the growth of crops may allow a better understanding of crop performance, including the reasons why certain cultivars can compete better with weeds. This study aims at evaluating growth, green ear yield, and grain yield in corn when in competition with weeds. Cultivars AG 1051 and BRS 106 were grown with (two hoeings, at 20 and 40 days after sowing) or without weed control. In order to evaluate crop growth, six collections of the above-ground part and the root system of corn were performed, every 15 days, with the first collection made 30 days after sowing. A randomized complete block design was adopted, with split-split plots (weed control in plots, cultivars in subplots, and collections in sub-subplots) and ten replicates. Eighteen weed species were found in the experiment area. Increased values of corn leaf area, above-ground part and root system, due to plant age function, were smaller in non-hoed plots than in hoed plots and were dependent upon cultivar. The lack of weed control increased dry matter of weeds aboveground part and decreased green ear yield and grain yield. Cultivar AG 1051 had higher increases in leaf area, above-ground part of the plant and root system, due to plant age function, and controlled weeds better than cultivar BRS 106. In addition, cultivar AG 1051 was superior to other cultivars with respect to most traits used for green corn yield and grain yield assessment.

In the Western Region of Brazil, it is usual to have two agricultural harvests in the same cropping season. Usually the first crop is soybean, followed by corn. In areas where corn is not planted due to a delayed harvest of soybean, farmers generally do not use winter crops. For these areas, the planting of winter oilseed crops aiming at the production of bio-fuels is one of the best alternatives; in addition, this would help in reducing the occurrence of weed species at the following summer crop. This study aimed to assessing the weed community in distinct winter crops post soybean crop, in terms of species composition, level of infestation and severity of occurrence. The following treatments were evaluated: agriculture under a no-till system with winter fallow, winter oilseed crops (crambe, radish, rapeseed) with no-till agriculture in the summer, and agriculture under a conventional tillage system with winter fallow. Phytosociological evaluations of all treatments were carried out 75 DAE of the oilseed crops, and the diversity indexes of Margalef, Menhinick, Simpson, and Shannon-Weiner were determined. Areas were also grouped by cluster analysis based on UPGMA applied at Jaccard's similarity matrix. Among the treatments with winter coverage, radish was the most efficient crop in suppressing the occurrence of weed species. The area with conventional tillage agriculture and winter fallow allowed for a higher occurrence of troublesome weeds. On the other hand, the area under fallow showed the highest absolute level of infestation. Overall, oilseed crops in the winter contribute to lower levels of infestation by weed species in these areas.

Tropical high altitude grasslands present several species with both microphyllous and highly sclerophyllous leaves, and co-occur in specific soil patches, thus exposed to identical environments. In this article we describe herbivory among co-occurring microphyllous species in a tropical high altitude grassland ecosystem of Serra do Cipó, Minas Gerais state, and we tested the effect of variable anatomic traits on leaf herbivory patterns. Leaf anatomical traits were investigated for Baccharis imbricata Heering , Lavoisiera imbricata DC. and L. subulata Triana (focal species). Herbivory was measured from branches and leaves of individual plants and compared among co-occurring species within one multispecific shrub patch and among L. subulata individuals from this patch and an adjacent monospecific patch. For all present plant species and individuals we estimated the proportion of leaves with different levels of area lost. For the focal species, six leaves were sorted and taken for histological sectioning, in order to allow precise measures of defensive structures. Relative mean leaf area lost differed significantly among the six species found in the multispecific patch. Lavoisiera subulata individuals were more attacked in the multispecific than in the monospecific patch. Leaf margin protection traits in both B. imbricata and L. imbricata showed significant effect against herbivory. Data suggest that some anatomic traits have direct effect against herbivory but their effect are not clearly perceptible among branches within individual plants or among plants within the same species.

Por suas condições estruturais e climáticas as florestas tropicais são ecossistemas onde prolifera uma grande diversidade fúngica. A Amazônia é reconhecida como a maior floresta tropical existente. No entanto, estudos taxonômicos sobre fungos hifomicetos nesta região são escassos. Este trabalho teve como objetivo investigar a ocorrência de hifomicetos associados a partes em decomposição da Arecaceae Euterpe oleracea Mart. (açaizeiro). Entre agosto de 2008 e abril de 2009 foram coletadas partes em decomposição de açaizeiro na Área de Proteção Ambiental Ilha do Combu, Município de Belém, Pará. Como parte dos resultados deste estudo foram encontrados dois novos registros para o Neotrópico (Digitodesmiumrecurvum W. H. Ho, K. D. Hyde & Hodgkiss e Pithomyces karoo Marasas & I. H. Schum.) e 10 para a América do Sul. O presente trabalho contém descrições taxonômicas, comentários, distribuição geográfica e ilustrações para cada um dos novos registros.

Bothrops erythromelas is responsible for many snake bites in northeastern Brazil. In the present study we determined the in vivo distribution of the venom following its subcutaneous injection into mice. B. erythromelas venom and albumin were labeled individually with 131I by the chloramine T method, and separated in a Sephacryl® S-200 column. The efficiency of labeling was 68%. Male Swiss mice (40-45 g), which had been provided with drinking water containing 0.05% KI over a period of 10 days prior to the experiment, were inoculated dorsally (sc) with 0.3 ml (2.35 x 105 cpm/mouse) of 131I-venom (N = 42), 131I-albumin or 131I (controls, N = 28 each). Thirty minutes and 1, 3, 6, 12, 18 and 24 h after inoculation, the animals were perfused with 0.85% NaCl and skin and various organs were collected in order to determine radioactivity content. There was a high rate of venom absorption in the skin (51%) within the first 30 min compared to albumin (20.1%) and free iodine (8.2%). Up to the third hour after injection there was a tendency for venom and albumin to concentrate in the stomach (3rd h), small intestine (3rd h) and large intestine (6th h). Both control groups had more radioactivity in the digestive tract, especially in the stomach, but these levels decreased essentially to baseline by 12-18 h postinjection. In the kidneys, the distribution profiles of venom, albumin and iodine were similar. Counts at 30 min postinjection were low in all three groups (1.37, 1.86 and 0.77, respectively), and diminished to essentially 0% by 12-18 h. Albumin tended to concentrate in muscle until the 3rd h postinjection (1.98%). There was a low binding of labeled venom in the liver (<0.54%), thyroid (<0.11%) and lungs (<0.08%), and no iodinated venom was detected in brain, heart, diaphragm, spleen or bladder. The low venom binding observed in most internal organs, comparable to that of albumin, suggests that B. erythromelas venom does not specifically target most internal organs. That is, the systemic effects of envenomation are mainly due to an indirect action

The treatment of pain before it initiates may prevent the persistent pain-induced changes in the central nervous system that amplify pain long after the initial stimulus. The effects of pre- or postoperative intraperitoneal administration of morphine (2 to 8 mg/kg), dipyrone (40 and 80 mg/kg), diclofenac (2 to 8 mg/kg), ketoprofen (10 and 20 mg/kg), and tenoxicam (10 and 20 mg/kg) were studied in a rat model of post-incisional pain. Groups of 5 to 8 male Wistar rats (140-160 g) were used to test each drug dose. An incision was made on the plantar surface of a hind paw and the changes in the withdrawal threshold to mechanical stimulation were evaluated with Von Frey filaments at 1, 2, 6 and 24 h after the surgery. Tenoxicam was given 12 or 6 h preoperatively, whereas the remaining drugs were given 2 h or 30 min preoperatively. Postoperative drugs were all given 5 min after surgery. No drug abolished allodynia when injected before or after surgery, but thresholds were significantly higher than in control during up to 2 h following ketoprofen, 6 h following diclofenac, and 24 h following morphine, dipyrone or tenoxicam when drugs were injected postoperatively. Significant differences between pre- and postoperative treatments were obtained only with ketoprofen administered 30 min before surgery. Preoperative (2 h) intraplantar, but not intrathecal, ketoprofen reduced the post-incisional pain for up to 24 h after surgery. It is concluded that stimuli generated in the inflamed tissue, rather than changes in the central nervous system are relevant for the persistence of pain in the model of post-incisional pain.

ßS-Globin haplotypes were studied in 80 (160 ßS chromosomes) sickle cell disease patients from Salvador, Brazil, a city with a large population of African origin resulting from the slave trade from Western Africa, mainly from the Bay of Benin. Hematological and hemoglobin analyses were carried out by standard methods. The ßS-haplotypes were determined by PCR and dot-blot techniques. A total of 77 (48.1%) chromosomes were characterized as Central African Republic (CAR) haplotype, 73 (45.6%) as Benin (BEN), 1 (0.63%) as Senegal (SEN), and 9 (5.63%) as atypical (Atp). Genotype was CAR/CAR in 17 (21.3%) patients, BEN/BEN in 17 (21.3%), CAR/BEN in 37 (46.3%), BEN/SEN in 1 (1.25%), BEN/Atp in 1 (1.25%), CAR/Atp in 6 (7.5%), and Atp/Atp in 1 (1.25%). Hemoglobin concentrations and hematocrit values did not differ among genotype groups but were significantly higher in 25 patients presenting percent fetal hemoglobin (%HbF) > or = 10% (P = 0.002 and 0.003, respectively). The median HbF concentration was 7.54 ± 4.342% for the CAR/CAR genotype, 9.88 ± 3.558% for the BEN/BEN genotype, 8.146 ± 4.631% for the CAR/BEN genotype, and 4.180 ± 2.250% for the CAR/Atp genotype (P = 0.02), although 1 CAR/CAR individual presented an HbF concentration as high as 15%. In view of the ethnic and geographical origin of this population, we did not expect a Hardy-Weinberg equilibrium for CAR/CAR and BEN/BEN homozygous haplotypes and a high proportion of heterozygous CAR/BEN haplotypes since the State of Bahia historically received more slaves from Western Africa than from Central Africa.

The in vivo antifungal activity of the naphthoquinone beta-lapachone against disseminated infection by Cryptococcus neoformans was investigated. Swiss mice were immunosuppressed daily with dexamethasone (0.5 mg per mouse) intraperitoneally for 3 days, the procedure was repeated 4 days later, and the animals were then challenged intravenously with C. neoformans (10(6) CFU/mL) 1 week later. Seven days after infection, the mice were divided into groups and treated daily with beta-lapachone (10 mg/kg, iv) for 7 (N = 6) and 14 days (N = 10). Amphotericin B (0.5 mg/kg) was used as comparator drug and an additional group received PBS. Treatment with beta-lapachone cleared the yeast from the spleen and liver, and the fungal burden decreased approximately 10(4) times in the lungs and brain 14 days after infection when compared to the PBS group (P < 0.05). This result was similar to that of the amphotericin B-treated group. Protection was suggestively due to in vivo antifungal activity of this drug and apparently not influenced by activation of the immune response, due to similar leukocyte cell counts among all groups. This study highlights the prospective use of beta-lapachone for treatment of disseminated cryptococcosis.

Infection with Bartonella spp may cause cardiac arrhythmias, myocarditis and endocarditis in humans. The aim of the present study was to evaluate a possible association between Bartonella spp bacteremia and endocarditis, arrhythmia and Chagas cardiomyopathy in patients from Brazil and Argentina. We screened for the presence of bacterial 16S rRNA in human blood by PCR using oligonucleotides to amplify a 185-bp bacterial DNA fragment. Blood samples were taken from four groups of subjects in Brazil and Argentina: i) control patients without clinical disease, ii) patients with negative blood-culture endocarditis, iii) patients with arrhythmias, and iv) patients with chronic Chagas cardiomyopathy. PCR products were analyzed on 1.5% agarose gel to visualize the 185-bp fragment and then sequenced to confirm the identity of DNA. Sixty of 148 patients (40.5%) with cardiac disease and 1 of 56 subjects (1.8%) from the control group presented positive PCR amplification for Bartonella spp, suggesting a positive association of the bacteria with these diseases. Separate analysis of the four groups showed that the risk of a Brazilian patient with endocarditis being infected with Bartonella was 22 times higher than in the controls. In arrhythmic patients, the prevalence of infection was 45 times higher when compared to the same controls and 40 times higher for patients with Chagas cardiomyopathy. To the best of our knowledge this is the first report of the association between Bartonella spp bacteremia and Chagas disease. The present data may be useful for epidemiological and prevention studies in Brazil and Argentina.

O objetivo deste trabalho foi construir modelos em Programação Matemática visando otimizar o processo de fabricação do biscoito cracker, aplicando-se Programação Linear e Não Linear. Através do conhecimento das etapas de produção do biscoito cracker, construiu-se um modelo em Programação Linear que minimiza as perdas de produção e o custo envolvido em seu processo de produção. Desenvolveu-se um modelo em Programação Não Linear para controlar os fatores tempo e pH na fermentação da esponja do cracker. Com a utilização do modelo em Programação Linear determinou-se a interpretação detalhada do custo no processo de produção do biscoito cracker e com o modelo Não Linear reduziu-se o tempo de fermentação da esponja do cracker, resultando em aumento na produtividade.

Organofosforados e carbamatos são compostos utilizados no controle de parasitas em animais e podem gerar resíduos nos produtos alimentícios derivados, representando um risco para o consumidor. O presente estudo objetivou pesquisar a presença de resíduos de organofosforados e carbamatos em leite cru produzido em quatro regiões leiteiras no Brasil e verificar se a presença desses compostos teria alguma relação com a ausência de Listeria monocytogenes e Salmonella spp., anteriormente observada nessas amostras. Entre 209 amostras analisadas, a presença de ao menos um desses compostos foi detectada em 196 (93,8%). Para a avaliação da sua interferência na detecção de L. monocytogenes e Salmonella spp., 28 amostras de leite positivas e negativas para esses compostos foram submetidas à fervura por 10 minutos e adicionadas desses patógenos, monitorando-se sua multiplicação durante armazenamento a 4 °C e a 25 °C. Não houve diferença significativa (p < 0,05) entre as taxas médias de multiplicação de L. monocytogenes e Salmonella spp. nas amostras de leite com diferentes resultados para resíduos de organofosforados ou carbamatos, indicando que esses patógenos não foram afetados pela presença desses resíduos. Entretanto, a alta freqüência de amostras de leite cru positivas para esses compostos é preocupante devido ao grande risco que representam para os consumidores, mesmo após o beneficiamento por tratamento térmico.

O suco de uva contém compostos fenólicos em quantidades importantes e, portanto, seu consumo é desejável como aporte de substâncias antioxidantes naturais. Os objetivos do estudo foram investigar os atributos sensoriais e avaliar a aceitação de sucos de uva comerciais. Foram analisados três tipos de suco de uva comercializados no Brasil: suco integral, suco concentrado e néctar. Utilizaram-se a Análise Descritiva Quantitativa modificada e o Teste de Aceitação com escala hedônica estruturada de nove pontos, incluindo questões sobre consumo e compra. Os resultados apontam que o suco integral apresentou cor e gosto amargo mais intensos; o suco concentrado (reconstituído) apresentou baixa intensidade em todos os atributos; e o néctar de uva apresentou maior intensidade de gosto doce e sabor característico de uva. O teste de aceitação mostrou que o néctar de uva e o suco integral foram os mais aceitos. A maioria dos consumidores relatou consumir quatro ou mais copos suco de uva por mês. O suco integral recebeu maior intenção de compra e o néctar foi a bebida mais consumida. Concluiu-se que o consumidor aprecia o suco integral da uva, com equilibrada intensidade nos atributos sensoriais investigados, todavia, consume com maior frequência o néctar, bebida à qual são adicionadas água e sacarose.
