Epidemiological studies performed in El Salvador between 1955-1972 demonstrated a high frequency of two triatomine vectors (Triatoma dimidiata and Rhodnius prolixus). However, the frequency of R. prolixus decreased by 1976 and this species has not been found since 1995. The main factors influencing the elimination of R. prolixus in El Salvador are discussed in this paper.

The goal of this study was to demonstrate the usefulness of an enzyme-linked immunosorbent assay (ELISA) for the serodiagnosis of pulmonary tuberculosis (PTB) and extrapulmonary TB (EPTB). This assay used 20 amino acid-long, non-overlapped synthetic peptides that spanned the complete Mycobacterium tuberculosis ESAT-6 and Ag85A sequences. The validation cohort consisted of 1,102 individuals who were grouped into the following five diagnostic groups: 455 patients with PTB, 60 patients with EPTB, 40 individuals with non-EPTB, 33 individuals with leprosy and 514 healthy controls. For the PTB group, two ESAT-6 peptides (12033 and 12034) had the highest sensitivity levels of 96.9% and 96.2%, respectively, and an Ag85A-peptide (29878) was the most specific (97.4%) in the PTB groups. For the EPTB group, two Ag85A peptides (11005 and 11006) were observed to have a sensitivity of 98.3% and an Ag85A-peptide (29878) was also the most specific (96.4%). When combinations of peptides were used, such as 12033 and 12034 or 11005 and 11006, 99.5% and 100% sensitivities in the PTB and EPTB groups were observed, respectively. In conclusion, for a cohort that consists entirely of individuals from Venezuela, a multi-antigen immunoassay using highly sensitive ESAT-6 and Ag85A peptides alone and in combination could be used to more rapidly diagnose PTB and EPTB infection.

The humoral immune response plays an important role in the clearance of Giardia lamblia. However, our knowledge about the specific antigens of G. lamblia that induce a protective immune response is limited. The purpose of this study was to identify and characterise the immunogenic proteins of G. lamblia in a mouse model. We generated monoclonal antibodies (moAbs) specific to G. lamblia (1B10, 2C9.D11, 3C10.E5, 3D10, 5G8.B5, 5F4, 4C7, 3C5 and 3C6) by fusing splenocytes derived from infected mice. Most of these moAbs recognised a band of ± 71 kDa (5G8 protein) and this protein was also recognised by serum from the infected mice. We found that the moAbs recognised conformational epitopes of the 5G8 protein and that this antigen is expressed on the cell surface and inside trophozoites. Additionally, antibodies specific to the 5G8 protein induced strong agglutination (> 70-90%) of trophozoites. We have thus identified a highly immunogenic antigen of G. lamblia that is recognised by the immune system of infected mice. In summary, this study describes the identification and partial characterisation of an immunogenic protein of G. lamblia. Additionally, we generated a panel of moAbs specific for this protein that will be useful for the biochemical and immunological characterisation of this immunologically interesting Giardia molecule.

Live attenuated vaccines have recently been introduced for preventing rotavirus disease in children. However, alternative strategies for prevention and treatment of rotavirus infection are needed mainly in developing countries where low vaccine coverage occurs. In the present work, N-acetylcysteine (NAC), ascorbic acid (AA), some nonsteroidal anti-inflammatory drugs (NSAIDs) and peroxisome proliferator-activated receptor gamma (PPARγ) agonists were tested for their ability to interfere with rotavirus ECwt infectivity as detected by the percentage of viral antigen-positive cells of small intestinal villi isolated from ECwt-infected ICR mice. Administration of 6 mg NAC/kg every 8 h for three days following the first diarrhoeal episode reduced viral infectivity by about 90%. Administration of AA, ibuprofen, diclofenac, pioglitazone or rosiglitazone decreased viral infectivity by about 55%, 90%, 35%, 32% and 25%, respectively. ECwt infection of mice increased expression of cyclooxygenase-2, ERp57, Hsc70, NF-κB, Hsp70, protein disulphide isomerase (PDI) and PPARγ in intestinal villus cells. NAC treatment of ECwt-infected mice reduced Hsc70 and PDI expression to levels similar to those observed in villi from uninfected control mice. The present results suggest that the drugs tested in the present work could be assayed in preventing or treating rotaviral diarrhoea in children and young animals.

Benznidazole (BZ) is one of the two drugs used for Chagas disease treatment. Nevertheless therapeutic failures of BZ have been reported, which were mostly attributed to variable drug susceptibility among Trypanosoma cruzistrains. ATP-binding cassette (ABC) transporters are involved in a variety of translocation processes and some members have been implicated in drug resistance. Here we report the characterisation of the first T. cruzi ABCG transporter gene, named TcABCG1, which is over-expressed in parasite strains naturally resistant to BZ. Comparison ofTcABCG1 gene sequence of two TcI BZ-resistant strains with CL Brener BZ-susceptible strain showed several single nucleotide polymorphisms, which determined 11 amino acid changes. CL Brener transfected with TcI transporter genes showed 40-47% increased resistance to BZ, whereas no statistical significant increment in drug resistance was observed when CL Brener was transfected with the homologous gene. Only in the parasites transfected with TcI genes there was 2-2.6-fold increased abundance of TcABCG1transporter protein. The analysis in wild type strains also suggests that the level of TcABCG1transporter is related to BZ natural resistance. The characteristics of untranslated regions of TcABCG1genes of BZ-susceptible and resistant strains were investigated by computational tools.

T-cell based vaccines against human immunodeficiency virus (HIV) generate specific responses that may limit both transmission and disease progression by controlling viral load. Broad, polyfunctional, and cytotoxic CD4+T-cell responses have been associated with control of simian immunodeficiency virus/HIV-1 replication, supporting the inclusion of CD4+ T-cell epitopes in vaccine formulations. Plasmid-encoded granulocyte-macrophage colony-stimulating factor (pGM-CSF) co-administration has been shown to induce potent CD4+ T-cell responses and to promote accelerated priming and increased migration of antigen-specific CD4+ T-cells. However, no study has shown whether co-immunisation with pGM-CSF enhances the number of vaccine-induced polyfunctional CD4+ T-cells. Our group has previously developed a DNA vaccine encoding conserved, multiple human leukocyte antigen (HLA)-DR binding HIV-1 subtype B peptides, which elicited broad, polyfunctional and long-lived CD4+ T-cell responses. Here, we show that pGM-CSF co-immunisation improved both magnitude and quality of vaccine-induced T-cell responses, particularly by increasing proliferating CD4+ T-cells that produce simultaneously interferon-γ, tumour necrosis factor-α and interleukin-2. Thus, we believe that the use of pGM-CSF may be helpful for vaccine strategies focused on the activation of anti-HIV CD4+ T-cell immunity.

RESUMOFertilizantes são importantes para a produção agrícola mundial devido à melhoria de produtividade que proporcionam. Neste artigo, utiliza-se a teoria de opções reais para avaliar a opção de troca de produto final, amônia ou ureia, em uma fábrica de fertilizantes nitrogenados. O método de simulação Monte Carlo foi utilizado para definir o valor da opção de troca na fábrica de fertilizantes, considerando as incertezas nos preços do gás natural (principal matéria-prima), amônia e ureia, assumindo que esses seguissem um movimento de reversão à média. Nos resultados, aponta-se que essa opção é relevante na análise de projetos de fábricas de fertilizantes, podendo ser fundamental, em muitos casos, sua consideração para a viabilidade do projeto.

Com o objetivo de descrever a evolução da disciplina "Administração Aplicada à Enfermagem", no período de 1980 a 1995, estudou-se os planos de curso construídos pelos docentes responsáveis por sua elaboração e condução. Os textos dos itens "Objetivos e Conteúdo" foram analisados sob a ótica da Fenomenologia, sendo submetidos ao processo de análise de discurso proposto por Martins e Bicudo. Os resultados dos procedimentos empregados nessa análise permitiram a identificação de seis categorias de pensamento: Gerenciar em Enfermagem, Bases Ideológicas e Políticas do Gerenciar em Enfermagem, A Comunicação como Instrumento para Gerenciar, Gerenciamento de Recursos Físicos, Gerenciamento de Recursos Materiais e Gerenciamento de Recursos Humanos. As modificações observadas nos planos de curso ao longo do período estudado indicaram a ocorrência de dois momentos marcantes - um no fim da década de 80, com a reformulação das bases ideológicas nas quais se assentava a disciplina e outro ao fim do primeiro lustro da década de 90, com a reorganização curricular,. A identificação desta trajetória possibilitou o acompanhamento e a compreensão do desenvolvimento da disciplina, podendo trazer recursos para a análise de disciplinas correlatas ou da mesma disciplina em outros contextos e experiências.

On Gymnocerus Audinet-Serville, 1835 (Coleoptera, Cerambycidae, Lamiinae, Anisocerini) and related genera. The genus Gymnocerus Audinet-Serville, 1835 is characterized and a key to the related genera is added. New combinations: Gymnocerina cratosomoides (Bates, 1862), comb. nov. (from Gymnocerus) and Badenella gavisa (Lane, 1966), comb. nov. (from Chimboincola). New synonyms proposed: Badenella Lane, 1964 = Chimboincola Lane, 1966 syn. nov.; Gymnocerina cratosomoides (Bates, 1862) = Gymnocerus crassus Bates, 1862 syn. nov. = Gymnocerina sulfurea Lane, 1964 syn. nov. = Gymnocerina subsiciva Lane, 1964 syn. nov. = Gymnocerina spuria Lane, 1964 syn. nov. = Gymnocerina oliveirai Lane, 1964 syn. nov.; and Badenella badeni (Bates, 1875) = Gymnocerus badeni fuscus Franz, 1935 syn. nov. = Badenella laceyi Lane, 1964 syn. nov..

Drosophila nappae sp. nov. , belonging to the subgroup I of the Drosophila tripunctata species group of the subgenus Drosophila, is described from flies of one strain established from several females collected from July 1994 through April 1995 at Morro Santana, Porto Alegre, state of Rio Grande do Sul, Brazil. This species has been misidentified during the past fifty years as Drosophila angustibucca (sensu Frota-Pessoa, 1954; non Duda, 1925, described from Costa Rica). Illustrations of male and female terminalia are also provided.

The flowering phenologies of Sessea brasiliensis and three species of Cestrum (C. amictum, C. schlechtendalii and C. sendtnerianum), all belonging to the Solanaceae, were monitored in a fragment of the montane Atlantic rain forest within São Paulo city from July 1993 through June 1995. A total of 11,021 flies belonging to five species of the flavopilosa group of Drosophila (D. cestri, D. cordeiroi, D. hollisae, D. incompta and D. mariaehelenae), plus 1,244 flies belonging to four species of Zygothrica (Z. dispar, Z. vittimaculosa, Z. vittinubila and Z. aff. vittipunctata) emerged from 33,955 sampled flowers. Cestrum amictum, C. sendtnerianum and Sessea brasiliensis are recorded for the first time as host plants for the species of the Neotropical flavopilosa group ofDrosophila. Additionally, at least ten less abundant taxa belonging also to the Drosophilidae, plus 1,073 microhymenopteran parasitoids (Braconidae, Eucoilidae, and Pteromalidae), emerged from the sampled flowers of S. brasiliensis e Cestrum spp. The present study raised from nine to eleven the number of species of Cestrum previously recorded as breeding sites for these flies. A comprehensive list of species of Solanaceae known to host six species of Drosophila and four species of Zygothrica in southeastern and southern Brazil is included.

As espécies de Ceroplastinae (Hemiptera, Coccoidea, Coccidae) que ocorrem no Estado de São Paulo são revisadas. São estudadas 36 espécies de Ceroplastinae, das quais 9 são sinonimizadas, 8 espécies novas são descritas e 19 são redescritas. Ceroplastes campinensis Hempel, 1901, C. bicolor Hempel, 1901, C. excaericae Hempel, 1912 e C. farmairii (Signoret, 1866), mencionadas para o Estado de São Paulo, não foram examinadas, devido a não localização do material tipo ou de outros exemplares. Nossos estudos indicaram que C. albolineatus Cockerell, 1894 e C. confluens Cockerell & Tinsley, 1898 foram erroneamente citadas por Hempel, 1900 para o estado de São Paulo. Coccus stellifer Westwood, 1871, atualmente Vinsonia stellifera (Westwood, 1871), é transferida para gênero Ceroplastes como Ceroplastes stellifer (Westwood, 1871) n. comb. C. flosculoides Matile-Ferrero, 1993 é registrada pela primeira vez para o país e C. cassiae (Chavannes, 1848), C. deodorensis Hempel, 1937, C. formosus Hempel, 1900 e C. quadratus Green, 1935 são registradas pela primeira vez no Estado de São Paulo. Ceroplastinae é agora representada por 31 espécies no Estado de São Paulo, todas incluídas no gênero Ceroplastes. Ilustrações e uma chave para espécies são incluídas. Novos sinônimos: C. formicarius Hempel = Ceroplastes communis Hempel, 1900 n. sin.; C. janeirensis Gray, 1828 = Ceroplastes psidii (Chavannes, 1848) n. sin. = C. simplex Hempel, 1900 n. sin.; C. cirripediformis Comstock, 1881 = C. cultus Hempel, 1900 n. sin. = C. cuneatus Hempel, 1900 n. sin. = C. rarus Hempel, 1900 n. sin e C. rotundus Hempel, 1900 n. sin.; C. lucidus Hempel, 1900 = C. novaesi Hempel, 1900 n. sin.; C. grandis Hempel, 1900 = C. rhizophorae Hempel, 1918 n. sin. The metropolitan region of Recife, Brazil is endemic for Dirofilaria immitis and has an environment favorable to the development of Culex quinquefasciatus. The goal of this study was to evaluate the vector competence of the Cx. quinquefasciatus RECIFE population for D. immitis transmission. A total of 2,104 females of Cx. quinquefasciatus RECIFE population were exposed to different densities of D. immitis microfilariae blood meals, ranging from 1,820 to 2,900 mf/ml of blood, in a natural membrane apparatus. The results showed a variation between 92.3% and 98.8% of females fed. The exposure of the Cx. quinquefasciatus RECIFE population to different densities of microfilariae did not influence the mortality of the mosquitoes. Infective larvae from D. immitis were observed in the Malpighian tubules beginning on the 12th day, whereas larvae were observed in the head and proboscis beginning on the 13th day following infection. The vector efficiency index (VEI) presented by the mosquitoes ranged from 7.8 to 56.5. The data demonstrates that the Cx. quinquefasciatus RECIFE population has great potential for the transmission of D. immitis, as it allowed the development of the filarid until the infectious stage at the different densities of microfilariae to which it was exposed.

Tortopus Needham & Murphy, 1924 and Tortopsis Molineri, 2010 with six and 10 species, respectively, are sister genera distributed in the Neartic and Neotropical regions. In spite that most species are known from the tropics of South America, only one species of each of these groups have been recorded from Brazil. Here we present distributional data expanding greatly the range of these groups including three new country records for Brazil: Tortopus igaranus Needham & Murphy, 1924, Tortopsis limoncocha Molineri, 2010 and Tortopsis spatula Molineri, 2010. We describe a distinct new species, Tortopus ipixuna sp. nov., from male imagos, that present a unique subrectangular pedestal. Additionally we redescribe the male imago of Tortopus harrisi Traver, 1950 (previously known only from the holotype), and we give the first records for Tortopsis canum Gonçalves, Da-Silva & Nessimian, 2011 from Espírito Santo and for Tortopsis spatula from Ecuador. Some variations in morphological structures of these species are also described, illustrated and discussed.

De modo geral, os sistemas de produção agrícola têm levado os agroecossistemas a uma grande instabilidade, acarretando forte degradação ambiental. Este trabalho visa estudar o potencial da utilização de adubação verde na região de São Carlos (SP) como uma técnica complementar aos sistemas de produção ora existentes, buscando alcançar um incremento qualitativo de seus agroecossistemas. A delimitação da região estudada baseou-se em características climáticas, geomorfológicas e administrativas, a partir de informações oriundas de mapas temáticos, manipulados por meio de um sistema de informações geográficas (SIG). A unidade de planejamento agroecológico para a região de São Carlos é constituída pelos municípios de Analândia, Itirapina, Santa Maria da Serra, São Carlos, São Pedro e Torrinha. Na região, descreveram-se o clima e os tipos de solo, tendo sido estes últimos agrupados de acordo com suas respectivas texturas e fertilidades. A partir disto, gerou-se um mapa com as zonas edáficas, predominantemente álicas e de textura arenosa. Paralelamente, foram listadas espécies de adubo verde com ecofisiologia compatível com as características edafoclimáticas encontradas, agrupando tais espécies em três classes: exigentes, rústicas e muito rústicas. Finalmente, elaborou-se um mapa com as zonas de cultivo de cada grupo de espécies. Concluiu-se que a adubação verde é uma técnica com potencial para melhorar a qualidade dos agroecossistemas da região de São Carlos.
