Twenty-nine isolates of the ectomycorrhiza fungus Pisolithus sp. from different geographical and host origins were tested for their ability to form ectomycorrhizae on Eucalyptus grandis and E. urophylla seedlings under greenhouse conditions. The ectomycorrhiza-forming capacity of isolates varied greatly from one eucalypt species to the other. All isolates from Eucalyptus, nine from Pinus spp. and two isolates from unknown hosts formed mycorrhizae with E. grandis and E. urophylla. Root colonization rates varied from 0 to 5.2 % for all Pinus isolates and those from unknown hosts. Colonization rates for these isolates were lower than those observed for Eucalyptus isolates (0.8 to 89.4 %). Three isolates from unknown hosts formed mycorrhizae with neither Eucalyptus species. The main characteristic for distinguishing Pinus from Eucalyptus isolates was mantle color. These data corroborate previous results obtained in our laboratory indicating that the isolates tested represent at least two distinct different species within the genus Pisolithus.

In the last decades, the use of plant growth-promoting rhizobacteria has become an alternative to improve crop production. Rhizobium leguminosarum biovar trifolii is one of the most promising rhizobacteria and is even used with non-legume plants. This study investigated in vitro the occurrence of plant growth-promoting characteristics in several indigenous R. leguminosarum biovar trifolii isolated from soils in the State of Rio Grande do Sul, Brazil. Isolates were obtained at 11 locations and evaluated for indoleacetic acid and siderophore production and inorganic phosphate solubilization. Ten isolates were also molecularly characterized and tested for antagonism against a phytopathogenic fungus and for plant growth promotion of rice seedlings. Of a total of 252 isolates, 59 produced indoleacetic acid, 20 produced siderophores and 107 solubilized phosphate. Some degree of antagonism against Verticillium sp. was observed in all tested isolates, reducing mycelial growth in culture broth. Isolate AGR-3 stood out for increasing root length of rice seedlings, while isolate ELD-18, besides increasing root length in comparison to the uninoculated control, also increased the germination speed index, shoot length, and seedling dry weight. These results confirm the potential of some strains of R. leguminosarum biovar trifolii as plant growth-promoting rhizobacteria.

Eighteen Pisolithus basidiomes were collected from Eucalyptus plantations in the state of Minas Gerais, Brazil. These basidiomes were characterized morphologically and molecularly. The basidiomes varied in shape, color and size. One of them was found underground, indicating a hypogeous fungus. The main morphological distinctive characteristic was spore ornamentation, which distinguished two groups. One group with short and erect spines was identified as Pisolithus microcarpus, and the other with long and curved spines as Pisolithus marmoratus, after analyzing the cladogram obtained by phylogenetic relationship based on internal transcribed spacer (ITS) regions of the nuclear ribosomal DNA of these isolates.

Oito bactérias endofíticas de plântulas de pimenta-do-reino foram testadas em casa de vegetação, objetivando controlar Fusarium solani f. sp. piperis e avaliar respostas morfofisiológicas das plantas tratadas com os agentes de controle. Plantas com quatro meses de idade tiveram o sistema radicular tratado com as suspensões bacterianas (10(9) ufc mL-1) por 10 minutos, e foram plantadas em vasos com solo natural infestado artificialmente com o patógeno (0,25%). O delineamento experimental foi inteiramente casualizado, com três repetições para avaliar o comportamento fotossintético, a produção e a alocação de biomassa, e cinco repetições para avaliar a mortalidade das plantas. As avaliações foram feitas 120 dias após a instalação do ensaio. O isolado B0 (Methylobacterium radiotolerans) controlou F. solani f. sp. piperis, provocando redução significativa do número de plantas mortas. Na ausência do patógeno, M. radiotolerans funcionou ainda como promotor de crescimento. Os demais isolados testados não apresentaram efeito na diminuição da mortalidade das plantas. A aplicação das bactérias endofíticas não afetou a taxa fotossintética instantânea das plantas, à exceção de B6, que teve efeito negativo. As plantas tratadas com M. radiotolerans priorizaram a alocação de carbono para a parte aérea.

Quatro isolados bacterianos da rizosfera de Drosera villosa var. villosa (B1, B2, B3, B4) e dois isolados de Bacillus thuringiensis (B5 e B6), sendo B6 produtor da toxina bioinseticida Cry1Ab, foram avaliados quanto à capacidade de inibir os fungos fitopatogênicos Fusarium solani f. sp. phaseoli, Fusarium solani f. sp. glycines, Fusarium oxysporum e Colletotrichum sp. A cepa mais efetiva foi B1 que inibiu o crescimento dos quatro fungos até o 26º dia. B. thuringiensis inibiu o crescimento de três destes, o que indica que possui atividade antifúngica e abre um novo campo de estudo para a utilização do B. thuringiensis.

The fungus Metarhizium anisopliae var. acridum, strain CG 423, was tested under field conditions against the gregarious grasshopper Rhammatocerus schistocercoides (Rehn) (Orthoptera: Acrididae). Conidia formulated in a racemic mixture of soybean oil and kerosene were sprayed under field conditions using an ultralow-volume hand-held atomizer Ulva Plus adjusted to deliver 2.9 L/ha. Bands composed of 2nd instar nymphs were treated with either 5.0x10(12) or 1.0x10(13) viable conidia/ha. The number of insects in each band was estimated at day one following spraying and by the end of the field trial (15 to 16 days post-treatment). Reductions in population size reached, in average, 65.8% and 80.4% for bands treated with the higher and lower dosage, respectively. For both dosages, total mortality rates of insects collected at two days post-application, and kept in cages for 14 days under lab conditions, showed no significant differences as compared to that obtained with insects collected immediately after spraying. Healthy insects were fed to native grasses sprayed on the field with 1.0x10(13) viable conidia/ha. Mortality levels of the nymphs fed on grasses collected two and four days post-application were not affected when compared to nymphs fed on grasses collected immediately following application.

Seven soybean cultivars (Bossier, Cristalina, Davis, Kent, Lincoln, Paraná and Uberaba), with different levels of resistance to Cercospora sojina, were crossed in a diallel design to determine the general (GCA) and specific (SCA) combining abilities relative to the inheritance of the resistance. Race 04 of the fungus was inoculated in the parents and in the 21 F1 hybrids in a greenhouse in a completely randomized design, with 12 replications. The reactions to the disease were evaluated 20 days after the inoculation, always on the most infected leaflet. Both GCA and SCA were significant for all the evaluated characters, being inferred that, for the expression of the characters, the additive, dominant and, possibly, epistatic genic actions were important. The largest values of estimated SCA effect (ij) were observed in the hybrid combinations where at least one parent presented high GCA. Cristalina, Davis and Uberaba cultivars showed the largest estimates for GCA effect (i), and from the analysis of ii, the contribution of these parents to heterosis of their hybrids will be towards the reduction of the disease symptoms. Therefore, these cultivars are indicated as parents in breeding programs that seek the development of soybean cultivars with resistance to frogeye leaf spot.

The objective of this work was to identify genomic regions that underlie resistance to Fusarium tucumaniae sp. nov., the causing agent of sudden death syndrome (SDS) in soybean in South America, using a population with a genetic background different from that previously reported for Fusarium virguliforme sp. nov. (F. solani f. sp. glycines), also responsible for SDS in soybean. Although major genes and quantitative trait loci (QTL) for SDS resistance have been identified, little is known about the same disease caused by Fusarium tucumaniae sp. nov., in South America. To identify genetic factors related to resistance to F. tucumaniae and DNA markers associated with them, a QTL analysis was performed using recombinant inbred lines. The map locations of the four loci, here identified, differed from those SDS resistance QTL previously described. It was screened a residual heterozygous line (RHL), which was heterozygous around the most effective QTL, RSDS1, and homozygous for the other genomic regions. The genetic effect of RSDS1 was confirmed using near-isogenic lines (NIL) derived from the RHL. The line which was homozygous for the Misuzudaizu genotype showed resistance levels comparable with that of the line homozygous for the Moshidou Gong 503 genotype.

The objective of this work was to evaluate the ability of several P-solubilizing fungi to solubilize aluminum phosphate and Araxá apatite as well as the synergism between the P-solubilizing fungus, PSF 7, and arbuscular mycorrhizal fungi to promote clover growth amended with aluminum phosphate. Two experiments were carried out, the first under laboratory conditions and the second in a controlled environmental chamber. In the first experiment, PSF 7, PSF 9, PSF 21 and PSF 22 isolates plus control were incubated in liquid medium at 28ºC for eight days. On the 2nd, 4th and 8th day of incubation, pH and soluble P were determined. In the second experiment, clover was sowed in plastic pots containing 300 g of sterilized substrate amended with aluminum phosphate, 3 g L-1, in presence and absence of PSF 7 isolate and arbuscular mycorrhizal fungi. A completely randomized design, in factorial outline 2x2 (presence and absence of PSF 7 and arbuscular mycorrhizal fungi) and five replicates were used. In the first experiment, higher P content was detected in the medium containing aluminum phosphate. PSF 7 is the best fungi isolate which increases aluminum solubilization with major tolerance to Al3+. Clover growth was stimulated by presence of PSF 7 and arbuscular mycorrhizal fungi. There is synergism between microorganisms utilized to improve plant nutrition.

A fim de ampliar as opções ao cultivo da soja, a Universidade Federal de Uberlândia lançou a cultivar UFUS-Imperial, proveniente do cruzamento entre (Msoy 8411xMsoy 8914) x (Emgopa 313xTucano). A cultivar apresentou resistência ao acamamento, à deiscência da vagem e aos patógenos: Fusarium solani, Cercospora sojina, Peronospora manshurica, Xanthomonas campestris pv. glycines e Diaporthe phaseolorum f.sp. meridionalis, e resistência parcial a Septoria glycines, Erysiphe diffusa e Phakopsora pachyrhizi. O rendimento dessa cultivar, em ensaios regionais, foi 45% superior ao da testemunha Msoy 6101, e é indicada para o Estado de Mato Grosso.

The objective of this work was to visualize the association between microcracking and other epidermal chilling injury symptoms, and to identify rots in cucumber fruit (Cucumis sativus L.) by scanning electron microscopy (SEM). Depressed epidermal areas and surface cracking due to damages of subepidermal cells characterized the onset of pitting in cucumber fruit. The germination of conidia of Alternaria alternata, with some of them evident on the fractures in the cultivar Trópico, occurred after damaging on the epidermis. Before, the chilling injury symptoms became visible, Stemphylium herbarum conidia germinated, and mycelium penetrated through the hypodermis using the microcracks as pathway. In the cultivar Perichán 121 the fungus was identified as Botrytis cinerea.

O objetivo deste trabalho foi avaliar o efeito de sistemas de manejo do solo e de cultivos prévios ao plantio do feijoeiro (Phaseolus vulgaris L.) sobre a densidade do solo e as populações de Rhizoctonia spp. e de Fusarium spp. Os cultivos prévios incluíram as leguminosas: guandu-anão (Cajanus cajan), estilosantes (Stylosanthes guianensis cv. Mineirão) e crotalária (Crotalaria spectabilis); e as gramíneas: milheto (Pennisetum glaucum cv. BN-2), sorgo granífero (Sorghum bicolor cv. BR 304), capim-mombaça (Panicum maximum cv. Mombaça), braquiária (Brachiaria brizantha cv. Marandu) e milho (Zea mays) consorciado com braquiária. As culturas utilizadas no cultivo prévio foram semeadas nos verões de 2002, 2003 e 2004, e os plantios de feijoeiro, cultivar BRS Valente, foram realizados nos invernos subseqüentes de 2003, 2004 e 2005, com irrigação por pivô central. Os restos culturais dos cultivos eram incorporados ao solo, no plantio convencional, e ficavam à superfície, no plantio direto. De modo geral, as maiores populações de Fusarium spp. e Rhizoctonia spp. e as maiores densidades de solo foram encontradas no solo cultivado em plantio direto. As maiores populações de Rhizoctonia spp. foram observadas em solos mais adensados. As leguminosas geralmente aumentaram populações desses patógenos e devem ser evitadas como culturas prévias ao cultivo do feijoeiro, em ambos os sistemas de cultivo. Plantios prévios de gramíneas, em geral, são supressores das populações de Rhizoctonia spp. e de Fusarium spp. no solo.

The objectives of this work were to evaluate two greenhouse screening methods for sudden death syndrome (SDS) and to determine which one is best correlated with field resistance of soybean genotypes. The evaluations were done with three sets of genotypes that were classified as partially resistant, intermediate, and susceptible to SDS based on previous field evaluations. These three sets were independently evaluated for greenhouse SDS reactions using cone and tray inoculation methods. Plants were infected using grains of white sorghum [Sorghum bicolor (L.) Moench] infested with Fusarium solani f. sp. glycines. Foliar symptom severity was rated 21 days after emergence. The cone and field SDS ratings were significantly correlated and ranged from 0.69 for set 1 to 0.51 for set 3. Correlations of SDS ratings of genotypes between field and greenhouse tray ratings were significant for set 1 and not significant for set 2. The cone method showed the highest correlation with field results and is recommended to screen soybean genotypes for SDS resistance.

The objective of this work was to set up ideal conditions for conidia mass production of Dicyma pulvinata. Four isolates were compared in terms of their growth and conidia production on various substrates (grains of parboiled rice, common rice, maize and wheat, besides chipped maize and rice husk), temperatures (19, 22, 25, 28 and 31ºC), growth containers (aluminum trays, polypropylene bags and Erlenmeyers) and light regimes (continuous darkness, 6 and 12 hours of light/darkness, and continuous light). Temperature effects on conidia germination capacity were also evaluated. The experiments were done in randomized complete block designs, in factorial arrangements (isolates x treatments - substrates, containers, temperatures and light regimes), with four replicates. In general, parboiled rice and polypropylene bags provided the best development of the fungus. Complete darkness and 6 hours of light increased mycelial growth, whereas continuous light favored sporulation. All tested temperatures favored the cultures of the fungus, except 31ºC. Temperatures between 19 and 25ºC ensure spore germination of more than 76%.

O objetivo deste trabalho foi avaliar o efeito do tratamento de sementes, com o indutor de resistência quitosana, sobre o tombamento de plântulas de beterraba e tomate, e sua relação com alterações bioquímicas e a defesa vegetal. Cada parcela foi representada por 25 sementes. Os tratamentos consistiram da imersão das sementes em quitosana nas concentrações de 0; 0,25; 0,5; 1; 2 e 4%. Posteriormente, as sementes foram semeadas em bandejas com o substrato infectado com Rhizoctonia sp., e mantidas em casa de vegetação por 14 dias. A quitosana induziu a resistência das plântulas de beterraba e tomate e reduziu a incidência de tombamento. As concentrações de 1,1 e 2,5% apresentaram maior eficiência na redução do tombamento, para as culturas da beterraba e tomate, respectivamente. O uso da quitosana induz o aumento na atividade da enzima fenilalanina amônia-liase (FAL) e interfere nas variáveis bioquímicas foliares de proteínas e açúcares totais e redutores.
