The soluble and insoluble cotyledon (SPF-Co and IPF-Co) and tegument (SPF-Te and IPF-Te) cell wall polymer fractions of common beans (Phaseolus vulgaris) were isolated using a chemical-enzymatic method. The sugar composition showed that SPF-Co was constituted of 38.6% arabinose, 23.4% uronic acids, 12.7% galactose, 11.2% xylose, 6.4% mannose and 6.1% glucose, probably derived from slightly branched and weakly bound polymers. The IPF-Co was fractionated with chelating agent (CDTA) and with increasing concentrations of NaOH. The bulk of the cell wall polymers (29.4%) were extracted with 4.0M NaOH and this fraction contained mainly arabinose (55.0%), uronic acid (18.9%), glucose (10.7%), xylose (10.3%) and galactose (3.4%). About 8.7% and 10.6% of the polymers were solubilised with CDTA and 0.01M NaOH respectively and were constituted of arabinose (52.0 and 45.9%), uronic acids (25.8 and 29.8%), xylose (9.6 and 10.2%), galactose (6.1 and 3.9%) and glucose (6.5 and 3.8%). The cell wall polymers were also constituted of small amounts (5.6 and 7.2%) of cellulose (CEL) and of non-extractable cell wall polymers (NECW). About 16.8% and 17.2% of the polymers were solubilised with 0.5 and 1.0M NaOH and contained, respectively, 92.1 and 90.7% of glucose derived from starch (IST). The neutral sugar and polymers solubilization profiles showed that weakly bound pectins are present mainly in SPF-Co (water-soluble), CDTA and 0.01-0.1M NaOH soluble fractions. Less soluble, highly cross-linked pectins were solubilised with 4.0M NaOH. This pectin is arabinose-rich, probably highly branched and has a higher molecular weight than the pectin present in SPF-Co, CDTA and 0.01-0.1M NaOH fractions.

Bacillus sp. SMIA-2 cresceu e secretou proteases quando cultivado em culturas líquidas contendo citrato trissódico como fonte de carbono e nitrato de amônio como fonte de nitrogênio. A atividade máxima da enzima foi alcançada após 8 horas de incubação do microrganismo, com níveis de 7,2 U.mg -1 proteína. A suplementação do meio com 2,0 mM de CaCl2 não proporcionou um aumento da atividade da protease, porém aumentou a sua estabilidade de 2 para 4 horas. A redução da concentração do fosfato de potássio no meio de cultura de 11,0 para 5,0 mM promoveu um acréscimo de 82% (13 U.mg -1 proteína) na atividade da protease. A substituição do nitrato de amônio presente no meio de cultura por 0,1% de soro de queijo aumentou a atividade da protease para 25 U.mg -1 proteína. Nestas condições, o tempo requerido para que a enzima atingisse seu pico de atividade máxima, que antes era de 9 horas, passou para 16 horas. A substituição do citrato trissódico do meio de cultura pela água de maceração de milho (0,5%) não só aumentou a atividade da enzima como também retardou o processo de desativação que é típico da produção de proteases. A atividade máxima da enzima obtida quando estes dois resíduos foram utilizados no meio foi 59,5 U.mg -1 proteína. Após alcançar este valor a enzima se manteve estável por mais de 20 horas o que favorece a sua produção em larga escala.

A indústria alimentícia, buscando atender a um mercado consumidor cada vez mais exigente, vem desenvolvendo embalagens ativas para proporcionar qualidade e segurança aos produtos acondicionados. Neste sentido, esta pesquisa objetivou desenvolver filmes ativos incorporados com agente antimicrobiano e avaliá-los na conservação de manteiga. Os filmes de base celulósica foram incorporados com 0% (controle) e 7% (ácido sórbico). Teste de halo de inibição em presença de fungos mostrou um halo de 3,4 cm. Amostras de manteiga foram fatiadas e inoculadas com 1 x 10(8) UFC.mL-1 de fungos filamentosos e leveduras, previamente isolados de manteiga. As amostras foram envolvidas com o filme ativo, embaladas em papel alumínio e armazenadas sob temperatura de refrigeração. As análises microbiológicas da manteiga foram realizadas após 0, 10 e 20 dias de armazenamento à temperatura de 7 ± 2 ºC. A contagem inicial de fungos filamentosos com leveduras na manteiga foi de 3 x 10(6) UFC.g-1 e após 10 e 20 dias de estocagem observou-se redução de 1 ciclo log (9 x 10(5) UFC.g-1) e 2 ciclos log (8 x 10(4) UFC.g-1) para a manteiga embalada com filme incorporado com 7% de ácido sórbico, respectivamente. Os valores de espessuras foram de 22,5 ± 3 µm e 25 ± 5 µm, de carga máxima: 37,4 ± 7 N e 52,4 ± 7 N, e de alongamento 1,7 ± 0,7% e 2,3 ± 0,7%, para os filmes incorporados com 0 e 7% de ácido sórbico, respectivamente. Pode-se concluir que o filme antimicrobiano apresentou maior resistência e alongamento, além de ter sido eficiente na redução de fungos filamentosos e leveduras em manteiga.

Um novo método espectrofotométrico simples, sensível e com boa seletividade é apresentado para a determinação rápida de mercúrio (II) em nível de traços com 2-mercapto-5-metilbenzilimidazol (MMBI) como um novo reagente espectrofotométrico (lambda max = 320 nm), em um meio aquoso ligeiramente alcalino (tampão Britton-Robinson (BR) 0,04 M, pH 8,0). A reação é instantânea e a absorbância permanece estável por mais de 24 horas. O coeficiente de absorção molar encontrado foi de 2,71 x 10(4) L mol-1 cm-1. A composição estequiométrica do complexo é de 1:1 (Hg:MMBI). Considerável excesso dos íons cobre, zinco, chumbo e cádmio não interfere na determinação. O método desenvolvido permitiu a determinação de mercúrio na faixa de 2 x 10-6 a 4 x 10-5 mol.L-1 com boa precisão e exatidão, e o limite de detecção para Hg foi de 9,9 x 10-7 mol.L-1. O método foi aplicado com sucesso a amostras de peixe e os resultados foram avaliados com o clássico método de espectrometria de absorção atômica (EAA). Os desvios padrão relativos para as amostras analisadas foram de 7,2 e 33% (n = 5), enquanto os erros foram de 1,63 e 11,6. O método mostrou-se sensível, seletivo e foi aplicado para a determinação de mercúrio em amostras de peixe com resultados satisfatórios.

Due to the fact that previous studies on the enzymatic activity of Glutathione peroxidase (GSH-Px) diverge widely in their methodology and results, this study aimed to investigate the influence of different analytical conditions on GSH-Px activity in chicken thighs from broilers that were fed different diets with different sources and concentrations of selenium. GSH-Px activity was evaluated six hours after slaughter and 120 days after frozen storage at -18 ºC. The different analytical conditions included time of pre-incubation (0, 10 and 30 minutes), reaction medium, types of substrate (H2O2 (0.72 mM, 7.2 mM, and 72 mM) and Terc-butil hydroperoxide 15 mM), and different buffer concentrations (buffer 1, potassium phosphate 50 mM pH 7.0 + EDTA 1 mM + mercaptoethanol 1 mM, and buffer 2, tris-HCl 50 mM pH 7.6 + EDTA 1 mM + mercapthanol 5 mM). The results show that the highest GSH-Px activity was observed when enzyme and substrate were in contact at 22 ºC without any pre-incubation, and that, when used at concentrations above 0.72 mM, hydrogen peroxide saturated the GSH-Px enzyme and inhibited its activity. The enzyme presented higher affinity to hydrogen peroxide when compared to terc-butil peroxide, and the addition of a buffer containing mercaptoethanol did not increase GSH-Px enzymatic activity. The activity of GSH-Px was not influenced by the source and concentration of selenium in the diet either. The obtained results allowed the determination of the best temperature of contact between the enzyme and substrate (22 ºC), the optimum concentration, and the type of substrate and buffer to be used. This information is extremely useful for future studies on GSH-Px activity in meat due to the divergence and little information found in the literature.

The effects of carnauba-based wax on the quality of 'Delta Valencia' orange produced in Ceará state, Brazil, were studied. The fruits were coated with carnauba-based wax and refrigerated (7 ± 2 ºC and 85 ± 2% R.H.) for 28 days. The quality attribute parameters assessed were weight loss, peel color (brightness, hue angle, and chromaticity), peel moisture, pH, soluble solids (SS), titratable acidity (TA), SS/TA ratio, ascorbic acid, total soluble sugars, reducing sugars, yellow flavonoids, and polyphenols. The results showed that 'Delta Valencia' oranges grown in the dry climate of Ceará state has excellent quality. The coated fruits lost mass at a lower rate than the the control fruits. No significant loss of soluble solids, titratable acidity, pH, and SS/TA ratio was observed, while ascorbic acid, soluble sugars, reducing sugars, yellow flavonoids, and polyphenols increased during storage in both the coated and control fruits. Carnauba-based wax coated fruits showed no signal of dehydration keeping their shiny green peel up to the end of the storage. The use of coating was crucial for the maintenance of visual quality by reducing mass loss, as well as keeping peel moisture.
Soymilk plain beverages: correlation between acceptability and physical and chemical characteristics

The objective of the research was to relate the physical and chemical characteristics of soymilk plain beverages to its sensory acceptance. Five commercial products and a new product formulated based on the less accepted sample were used. The overall acceptance was evaluated by 102 assessors using a 10-point hybrid hedonic scale. The hedonic scores ranged from 3.8 (sample E) to 7.0 (A). Most assessors (55%) preferred sample A, which was viscous (26.6 cp), dark (L* = 77.7), and slightly acidic (pH = 6.6). Sample C, which had lower solid content and higher acidity, was preferred by 29% of the assessors. These two beverages showed the greatest commercial potential of the products analyzed. The least-accepted sample (E), preferred by 8% of the assessors, had a lighter color (L* = 96.8), lower viscosity (13.5 cp), higher lipid content (2.2 g/100 g), and less protein (1.68 g/100 g) than the other products evaluated. A reformulation of the least preferred product (E) with the addition of maltodextrin and also vanilla and milk flavors increased its acceptance, yielding an average score of 7.2. Overall, it was observed that a soymilk plain beverage with higher viscosity, darker color, and higher protein content best meets consumer expectations.

Stability of minimally processed radicchio (Cichorium intybus L.) was evaluated under modified atmosphere (2% O2, 5% CO2, and 93% N2) on 3, 5, 7 and 10 days of storage at 5°C. The samples were hygienized in sodium hypochlorite or hydrogen peroxide solutions to identify the most effective sanitizing solution to remove microorganisms. Microbiological analysis was conducted to identify the presence of coliforms at 35°C and 45°C, mesophilic microorganisms, and yeast and mold. Physicochemical analyses of mass loss, pH, soluble solids, and total acidity were conducted. The color measurements were performed using a Portable Colorimeter model CR-400. The antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl and 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic methods. The sensory evaluation was carried out using a hedonic scale to test overall acceptance of the samples during storage. The sodium hypochlorite (150 mg.L-1) solution provided greater safety to the final product. The values of pH ranged from 6.17 to 6.25, total acidity from 0.405 to 0.435%, soluble solids from 0.5 to 0.6 °Brix, mass loss from 1.7 to 7.2%, and chlorophyll from 1.068 to 0.854 mg/100g. The antioxidant activity of radicchio did not show significant changes during the first 3 days of storage. The overall acceptance of the sample stored in the sealed package without modified atmosphere was 70%, while the fresh sample was obtained 77% of approval. Although the samples packaged under modified atmosphere had a higher acceptance score, the samples in sealed packages had satisfactory results during the nine days of storage. The use of modified atmosphere, combined with cooling and good manufacturing practices, was sufficient to prolong the life of minimally processed radicchio, Folha Larga cultivar, for up to ten days of storage.

Nile tilapia is one of the major fish species cultivated worldwide and in Brazil. The tilapia fillet yield is between 30-35%, thus around 70% of waste is generated. A portion of this waste can be used to obtain minced fish, and the resulting product can be used as meat raw material to prepare fish snacks. The aim of this study was to produce fish snacks containing different inclusion levels (20, 30, and 40%) of minced fish obtained from Nile tilapia processing waste and evaluate their physicochemical characteristics and sensory acceptance. Protein content, ash, water activity, and hardness increased with increasing inclusion of minced fish. The scores obtained in the sensory evaluation were: flavor acceptance (from 7.2 to 5.7), texture (from 7.4 to 5.3), overall acceptance (from 7.1 to 5.9), and willingness to purchase (from 4.0 to 3.1). This study demonstrates that the inclusion of 20 to 40% of minced fish of Nile tilapia in snacks is well accepted and improves their nutritional value without affecting the physicochemical properties.

O objetivo do trabalho foi avaliar a qualidade fisiológica de sementes de soja convencional e transgênica, das cultivares CD 216 e CD 212RR, submetidas ao glifosato. As sementes foram embebidas nas doses de zero; 1,8; 3,6; 5,4 e 7,2 g.L-1 de equivalente ácido de glifosato durante meia hora. Foram realizados os testes de primeira contagem da germinação, germinação, condutividade elétrica, índice de velocidade de emergência, emergência de plântula, comprimento de raízes e parte aérea, massa de matéria seca total. Concluiu-se que a qualidade fisiológica das sementes das cultivar CD 212RR é reduzida pelo herbicida, decrescendo com o incremento das doses do glifosato. Em função dessa conclusão, enfatiza-se que o devido intervalo de tempo entre a dessecação das invasoras ou das plantas de cobertura deve ser respeitado, para diminuir a transferência do glifosato da planta-alvo para a planta não-alvo, e assim, evitar problemas de germinação e estabelecimento da lavoura de soja.
