Este estudo teve como objetivo determinar a prevalência das dermatopatias tumorais de equinos diagnosticadas no Laboratório de Patologia Veterinária (LPV) da Universidade Federal de Santa Maria (UFSM), Rio Grande do Sul. Para isso, foram revisados os protocolos de biópsia de pele de equinos, arquivados no LPV-UFSM, entre janeiro de 1999 e dezembro de 2009. Em todos os casos, foram considerados os diagnósticos morfológicos que constavam nos protocolos. Durante esse período foram submetidas ao LPV-UFSM 139 amostras cutâneas de equinos. Dessas 139 amostras, 108 (77,6%) eram de tumores cutâneos, neoplásicos ou não; os outros 31 casos consistiam de dermatoses não tumorais. Dos 108 equinos, 53 (49,1%) eram fêmeas e 37 (34,2% %) eram machos, em 18 (16,7%) casos não estava descrito no protocolo o sexo. Quanto à raça, 66 (61,1%) eram de raça pura e 13 (12%) não tinham raça definida; em 29 (26,9%) protocolos não havia a descrição da raça. A raça mais prevalente foi a Crioula (44/108 [40,7%]), o restante dos 22 equinos de raça pura pertencia a outras sete raças diferentes. Quanto à idade, as categorias mais prevalentes em ordem decrescente foram a de 1-5 anos (47/108 [43,5%]) e a de 6-14 anos (21/108 [19,5%]), cavalos com 15 anos ou mais representaram 11,1% (12/108). Apenas um cavalo tinha menos de um ano de idade. Em 27 protocolos não constava a idade. Os tumores mais prevalentes incluíram: sarcoide (62/108 [57,4%]); carcinoma de células escamosas (11/108 [10,2%]); pitiose (9/108 [8,3%]); tecido de granulação (7/108 [6,5%]; e o granuloma eosinofílico (4/108 [3,8%]). Os outros tumores contaram com aproximadamente 14% dos casos.

The aims of the present study were to evaluate the performance of Jersey and Holstein cows under different rainfall conditions (dry and rainy seasons) by monitoring aspects related to subclinical mastitis (somatic cell count, microbiological isolation, type of isolated pathogen), milk quality (lactose, protein, fat, total solids) and production (mean milk production) of both breeds. The study was carried out in a dairy farm located in the state of São Paulo, Brazil. Eight visitations were done to the farm, four in a period of high rainfall and four in a period of low rainfall. Milk samples were collected from 79 Holstein cows and 37 Jersey cows for electronic somatic cell count and determination of the main milk components (protein, fat, total solids, lactose). Milk fat, protein, total solids and production were influenced by breed and the season, with similar tendencies for both breeds in both seasons. Somatic cell count (SCC) showed similar results for both breeds. Holstein cows with intramammary infections (IMI) presented a higher increase in SCC when compared to Jersey cows (P<0.001). In the dry season, 53 animals had IMI in at least one month during the study, which 32 were Holstein and 21 were Jersey cows. In the rainy season, 65 animals had intramammary infection, being 43 Holstein and 22 Jersey cows. The frequency of IMI cases was larger in the rainy season than in the dry season. Jersey cows had a lower chance of showing IMI signs and symptoms than Holstein cows in the rainy season (odds ratio=0.52). The larger number of IMI cases in the rainy season may have led to a lower milk lactose rate for both breeds, thus milk lactose rate can be considered an indicator of IMI status. There was prevalence of contagious pathogens overall in the study. The applied model showed that environmental pathogens were more frequently isolated from the breed Jersey, regardless of the study season. There seems to be differences in the immune response of Jersey and Holstein breeds.

Os arquivos de biópsia do período de 2000 a 2010 do SPV-UFRGS foram revisados e levantados os casos de neoplasias mesenquimais de genitália (útero, cérvix, vagina e vulva) de cadelas. Realizou-se estudo retrospectivo de 43 casos, incluindo caracterização histológica, coloração tricrômico de Masson (TM) e imuno-histoquímica (IHQ). As principais raças acometidas foram Cocker, Poodle e Pastor Alemão, porém o maior número de casos foi observado em cães sem raça definida (SRD). A idade média dessas cadelas foi de 10,6 anos (variação de 3 a 10 anos). Quanto à localização dos tumores, 44,2% estavam na vagina, 27,9% no útero, 23,3% na vulva e 4,6% na cérvix. Histologicamente, 46,5% eram leiomioma, 41,9% fibroleiomioma, 7,0% fibroma e 4,6% leiomiossarcoma. Na coloração de TM, os fibromas apresentaram todas as células neoplásicas coradas em azul (colágeno), os leiomiomas e os leiomiossarcomas apresentaram menos de 50% de colágeno em meio às células neoplásicas musculares e os fibroleiomiomas mais de 50%. Na IHQ, os fibromas apresentaram marcação positiva para vimentina e negativa para desmina. Todos os leiomiomas eram positivos para vimentina (em mais de 50% células) e para desmina (75% dos casos em mais de 50% das células e 25% em menos de 50%). Em um dos casos de leiomiossarcoma houve imunomarcação para vimentina e ausente para desmina e, no outro, ocorreu o oposto. Os fibroleiomiomas apresentaram marcação para vimentina em 94,4% (em mais de 50% células) e para desmina em 77,8% (64,3% em menos de 50% das células e 35,7% em mais de 50%).

Resumo: Neoplasmas em suínos são raros. Esse trabalho descreve os neoplasmas encontrados em suínos na rotina diagnóstica de um laboratório de patologia veterinária localizado na Região Central do Rio Grande do Sul. Durante um período de 49 anos, 2.266 casos de várias afecções em suínos foram encontrados, dos quais 37 (1,6%) eram neoplasmas. Em ordem decrescente de frequência, os seguintes neoplasmas foram encontrados: Linfoma (11/37), nefroblastoma (11/37), melanoma (8/37) e papiloma (2/37). Adicionalmente, um caso de cada um dos seguintes tumores foi observado: Adenoma hepatocelular, carcinoma hepatocelular, colangiocarcinoma, histiocitoma fibroso maligno e sarcoma granulocítico. O aspecto macroscópico e histológico desses tumores é descrito e a sua epidemiologia é comparada com os dados disponíveis na literatura para neoplasia suína.

Este trabalho objetivou avaliar os efeitos de métodos de superação de dormência sobre a germinação das sementes de Ipomoea grandifolia, I. hederifolia, I. nil, I. quamoclit, Merremia aegyptia e M. cissoides. Os tratamentos consistiram em ácido sulfúrico concentrado, nitrato de potássio, água quente e calor seco (50 ºC), lixa e fogo. Constatou-se que o ácido sulfúrico promoveu aumento na germinação das sementes de I. grandifolia (58, 37, 22 e 34%), I. hederifolia (76, 49, 82 e 55%), I. quamoclit (43, 33, 66 e 35%), I. nil (69, 79, 72 e 62%), M. cissoides (8, 19, 35 e 57%) e M. aegyptia (24, 64, 56 e 63%) após períodos de imersão de 5, 10, 15 e 20 minutos, respectivamente. A água (20 e 40 minutos), o calor seco (20 e 40 minutos) e o fogo melhoraram a germinação de I. grandifolia (68, 59, 62, 67 e 59%), M. cissoides (50, 52, 18, 25 e 46%) e M. aegyptia (54, 47, 21, 21 e 45%), respectivamente. O calor seco de 20 e 40 minutos melhorou a germinação de I. nil (49 e 36%), e o de 40 minutos, a de I. hederifolia (70%). O uso de nitrato de potássio e lixa não proporcionou melhoria na germinação das sementes.

A semeadura adensada do algodoeiro na safrinha é uma estratégia adotada pelos cotonicultores para reduzir os custos de produção e aumentar a rentabilidade dessa atividade. O objetivo deste trabalho foi determinar os períodos de convivência e controle das plantas daninhas no algodoeiro em semeadura adensada na safrinha. O experimento foi conduzido na Fazenda Pouso Frio, em Chapadão do Sul-MS. A semeadura do cultivar BRS 293 ocorreu no dia 07/02/2010, adotando-se espaçamento de 0,45 m entre linhas e a densidade de 260 mil sementes por hectare. Os tratamentos constaram de dois grupos. No primeiro, a cultura foi mantida por períodos iniciais crescentes em convivência com plantas daninhas, após sua emergência, para determinação do PAI, e no segundo grupo, para determinação do PTPI, o algodoeiro permaneceu por períodos iniciais livre da matocompetição. Foram analisados períodos de 0, 5, 10, 15, 22, 29, 36, 43, 50, 57, 64, 71 e 165 dias após emergência (DAE) de convivência ou controle inicial das plantas daninhas. As plantas daninhas de maior importância relativa foram Bidens pilosa, Amaranthus retroflexus, Digitaria horizontalis, Eleusine indica, Commelina benghalensis e Euphorbia heterophylla. Tolerando a redução da produtividade de até 5%, o PAI foi até 4 DAE; o PTPI, até 32 DAE; e o PCPI com duração de 28 dias, entre 4 e 32 DAE. Quando se admitiu a queda de produtividade equivalente ao desvio-padrão da média do tratamento mantido no limpo, estabeleceu-se o PAI de 1 DAE; PTPI, de 37 DAE; e PCPI durando 36 dias, entre 1 e 37 DAE.

The lognormal distribution model is frequently found in communities, especially those which are rich in species and influenced by many environmental factors, as those of the cerrado. We tested the hypothesis that the abundance distribution of woody plant species in a cerrado fragment fits the lognormal model. We placed 20 lines in a cerrado fragment and sampled, with the point-quarter method, 800 individuals with stem perimeter equal or larger than 3 cm. We plotted the abundance-class histogram of the species, verified its normality with the Kolmogorov-Smirnov test, and estimated the expected number of woody species for this community. Of the 63 obtained species, Anadenanthera falcata (with 185 species), Eriotheca gracilipes (43), Stryphnodendron obovatum (37), and Miconia albicans (36) were the most abundant ones. Twelve species were represented by only one individual. We did not reject the null hypotheses that the distribution of woody component species was normal and, thus, their abundances fitted the lognormal model. Therefore, with our work, we can predict that cerrado plant communities fit the lognormal model. If this pattern is maintained in other cerrado communities, there would be implications for the conservation of this vegetation type, because rare species are susceptible of extinction, and implications to their structure, because the dominant species may act as keystone species.

The human immunoglobulin lambda variable locus (IGLV) is mapped at chromosome 22 band q11.1-q11.2. The 30 functional germline v-lambda genes sequenced untill now have been subgrouped into 10 families (Vl1 to Vl10). The number of Vl genes has been estimated at approximately 70. This locus is formed by three gene clusters (VA, VB and VC) that encompass the variable coding genes (V) responsible for the synthesis of lambda-type Ig light chains, and the Jl-Cl cluster with the joining segments and the constant genes. Recently the entire variable lambda gene locus was mapped by contig methodology and its one- megabase DNA totally sequenced. All the known functional V-lambda genes and pseudogenes were located. We screened a human genomic DNA cosmid library and isolated a clone with an insert of 37 kb (cosmid 8.3) encompassing four functional genes (IGLV7S1, IGLV1S1, IGLV1S2 and IGLV5a), a pseudogene (VlA) and a vestigial sequence (vg1) to study in detail the positions of the restriction sites surrounding the Vl genes. We generated a high resolution restriction map, locating 31 restriction sites in 37 kb of the VB cluster, a region rich in functional Vl genes. To study the effect of halothane as a cardioplegic agent, ten Wistar rats were anesthetized by ether inhalation and their hearts were perfused in a Langendorff system with Krebs-Henseleit solution (36oC; 90 cm H2O pressure). After a 15-min period for stabilization the control values for heart rate, force (T), dT/dt and coronary flow were recorded and a halothane-enriched solution (same temperature and pressure) was perfused until cardiac arrest was obtained. The same Krebs-Henseleit solution was reperfused again and the parameters studied were recorded after 1, 3, 5, 10, 20 and 30 min. Cardiac arrest occurred in all hearts during the first two min of perfusion with halothane-bubbled solution. One minute after reperfusion without halothane, the following parameters reported in terms of control values were obtained: 90.5% of control heart rate (266.9 ± 43.4 to 231.5 ± 71.0 bpm), 20.2% of the force (1.83 ± 0.28 to 0.37 ± 0.25 g), 19.8% of dT/dt (46.0 ± 7.0 to 9.3 ± 6.0 g/s) and 90.8% of coronary flow (9.9 ± 1.5 to 9.4 ± 1.5 ml/min). After 3 min of perfusion they changed to 99.0% heart rate (261.0 ± 48.2), 98.9% force (1.81 ± 0.33), 98.6 dT/dt (45.0 ± 8.2) and 94.8% coronary flow (9.3 ± 1.4). At 5 min 100.8% (267.0 ± 40.6) heart rate, 105.0% (1.92 ± 0.29) force and 104.4% (48.2 ± 7.2) dT/dt were recorded and maintained without significant differences (P>0.01) until the end of the experiment. These data demonstrate that volatile cardioplegia with halothane is an effective technique for fast induction of and prompt recovery from normothermic cardiac arrest of the rat heart

In the present study we investigated the influence of methotrexate (MTX) and azathioprine (AZA) on the serum levels of the IgA-a1-antitrypsin (IgA-AT) complex in patients with the systemic form of juvenile chronic arthritis (JCA). Fifty-six JCA patients (22 treated with MTX, 18 treated with AZA, and 16 not treated with any immunosuppressive agent) were enrolled in the study. MTX dosage ranged from 0.3 to 0.5 mg kg-1 week-1, while AZA was given daily at an average dose of 1 mg/kg. MTX was given for 13 months (SD = 7 months) whereas AZA for 11 months (SD = 6 months). The average value of the complex was higher in JCA patients than in both control groups (0.74 ± 0.73 U vs 0.37 ± 0.13 U (control children), P<0.001 and vs 0.23 ± 0.12 U (control adults), P<0.001). Values exceeding the normal range were found in twenty-two JCA patients (39.4%). Serum IgA-AT level was lowest in the MTX group compared to AZA and non-treated patients (0.56 ± 0.24 U, 0.76 ± 0.43 U, 0.95 ± 0.52 U, respectively, P<0.05). IgA values exceeding normal levels for age were found in 14% of the patients. A correlation between the levels of the IgA-AT complex and C-reactive protein (r = 0.43, P<0.01), a1-acid-glycoprotein (r = 0.45, P<0.01), a1-antichymotrypsin (r = 0.52, P<0.01), a1-antitrypsin (r = 0.40, P<0.01) and IgA (r = 0.56, P<0.01) was established

HLA class II genes are strongly associated with susceptibility and resistance to insulin-dependent diabetes mellitus (IDDM). The present study reports the HLA-DRB1 genotyping of 41 IDDM patients and 99 healthy subjects from the Southeast of Brazil (Campinas region). Both groups consisted of an ethnic mixture of Caucasian, African Negro and Amerindian origin. HLA-DRB1*03 and *04 alleles were found at significantly higher frequencies among IDDM patients compared to the controls (DRB1*03: 48.8% vs 18.2%, P<0.005, RR = 4.27; DRB1*04: 43.9% vs 15.1%, P<0.008, RR = 4.37) and were associated with a susceptibility to the disease. DRB1*03/*04 heterozygosity conferred a strong IDDM risk (RR = 5.44). In contrast, the HLA-DRB1*11 allele frequency was lower among IDDM patients (7.3% vs 26.3% in controls), but the difference was not significant. These data agree with those described for other populations and allow genetic characterization of IDDM in Brazil

Thyroid hormone (T3) is essential to normal brain development. Previously, we have shown that T3 induces cerebellar astrocyte proliferation. This effect is accompanied by alteration in glial fibrillary acidic protein (GFAP) and fibronectin organization. In the present study, we report that the C6 glioma cell line, which expresses GFAP and is classified as an undifferentiated astrocytic cell type, is a target for T3 action. The C6 monolayers were treated with 50 nM T3 for 3 days, after which the cells were maintained for 2 days without medium changes. In C6 cells, T3 induced the expression of proteins of 107, 73 and 62 kDa. The hormone also up-regulated protein bands of 100 (+50%), 37 (+50%) and 25.5 kDa (+50%) and down-regulated proteins of 94 (-100%), 86.5 (-100%), 68 (-100%), 60 (-100%), 54 (-33%), 51 (-33%) and 43.5 kDa (-33%). We suggest, on the basis of molecular mass, that the 54-, 51- and 43.5-kDa proteins could be the cytoskeletal proteins vimentin, GFAP and actin, respectively. The down-regulation of these proteins may be involved in the effects of thyroid hormone on C6 differentiation.

Mother-pup interaction, as well as other behavioral reactions were studied during the lactation period in 24 litters of Wistar rats and their dams fed either a 16% (control - C; 12 litters) or a 6% (malnourished - M; 12 litters) protein diet. The diets were isocaloric. Throughout lactation there was a 36.4% weight loss of M dams and a 63% body weight deficit in the M pups when compared to control pups. During this period, half of the litters were exposed daily to additional tactile stimulation (CS or MS), while the other half were submitted to normal rearing conditions (CN or MN). The tactile stimulation of pups (handling) consisted of holding the animal in one hand and gently touching the dorsal part of the animal's body with the fingers for 3 min. A special camera and a time-lapse video were used to record litter behavior in their home cages. Starting at 6 p.m. and ending at 6 a.m., on days 3, 6, 12, 15, 18 and 21 of lactation, photos were taken at 4-s intervals. An increase in the frequency (154.88 ± 16.19) and duration (455.86 ± 18.05 min) of suckling was observed throughout the lactation period in all groups compared to birth day (frequency 24.88 ± 2.37 and duration 376.76 ± 21.01 min), but the frequency was higher in the C (84.96 ± 8.52) than in the M group (43.13 ± 4.37); however, the M group (470.2 ± 11.87 min) spent more time suckling as compared with the C group (393.67 ± 13.09 min). The M dams showed a decreased frequency of resting position throughout the lactation period (6.5 ± 2.48) compared to birth day (25.42 ± 7.74). Pups from the C group were more frequently observed separated (73.02 ± 4.38) and interacting (258.99 ± 20.61) more with their mothers than the M pups (separated 66.94 ± 5.5 and interacting 165.72 ± 12.05). Tactile stimulation did not interact with diet condition, showing that the kind of stimulation used in the present study did not lead to recovery from the changes induced by protein malnutrition. The changes in mother-pup interaction produced by protein malnutrition of both may represent retardation in neuromotor development and a higher dependence of the pups on their mothers. These changes may represent an important means of energy saving and heat maintenance in malnourished pups.

We examined some of the mechanisms by which the aspirin metabolite and the naturally occurring metabolite gentisic acid induced relaxation of the guinea pig trachea in vitro. In preparations with or without epithelium and contracted by histamine, gentisic acid caused concentration-dependent and reproducible relaxation, with mean EC50 values of 18 µM and Emax of 100% (N = 10) or 20 µM and Emax of 92% (N = 10), respectively. The relaxation caused by gentisic acid was of slow onset in comparison to that caused by norepinephrine, theophylline or vasoactive intestinal peptide (VIP). The relative rank order of potency was: salbutamol 7.9 > VIP 7.0 > gentisic acid 4.7 > theophylline 3.7. Gentisic acid-induced relaxation was markedly reduced (24 ± 7.0, 43 ± 3.9 and 78 ± 5.6%) in preparations with elevated potassium concentration in the medium (20, 40 or 80 mM, respectively). Tetraethylammonium (100 µM), a nonselective blocker of the potassium channels, partially inhibited the relaxation response to gentisic acid, while 4-AP (10 µM), a blocker of the voltage potassium channel, inhibited gentisic acid-induced relaxation by 41 ± 12%. Glibenclamide (1 or 3 µM), at a concentration which markedly inhibited the relaxation induced by the opener of ATP-sensitive K+ channels, levcromakalim, had no effect on the relaxation induced by gentisic acid. Charybdotoxin (0.1 or 0.3 µM), a selective blocker of the large-conductance Ca2+-activated K+ channels, caused rightward shifts (6- and 7-fold) of the gentisic acid concentration-relaxation curve. L-N G-nitroarginine (100 µM), a NO synthase inhibitor, had no effect on the relaxant effect of gentisic acid, and caused a slight displacement to the right in the relaxant effect of the gentisic acid curve at 300 µM, while methylene blue (10 or 30 µM) or ODQ (1 µM), the inhibitors of soluble guanylate cyclase, all failed to affect gentisic acid-induced relaxation. D-P-Cl-Phe6,Leu17[VIP] (0.1 µM), a VIP receptor antagonist, significantly inhibited (37 ± 7%) relaxation induced by gentisic acid, whereas CGRP (8-37) (0.1 µM), a CGRP antagonist, only slightly enhanced the action of gentisic acid. Taken together, these results provide functional evidence for the direct activation of voltage and large-conductance Ca+2-activated K+ channels, or indirect modulation of potassium channels induced by VIP receptors and accounts for the predominant relaxation response caused by gentisic acid in the guinea pig trachea.

Lead has been shown to produce cognitive and motor deficits in young rats that could be mediated, at least in part, by inhibition of the zinc-containing heme biosynthetic enzyme delta-aminolevulinate dehydratase (ALA-D). In the present study we investigated the effects of lead and/or zinc treatment during the second stage of rapid postnatal brain development on brain, kidney and blood ALA-D specific activity, as well as the negative geotaxis behavior of rats. Eight-day-old Wistar rats were injected intraperitoneally with saline, lead acetate (8 mg/kg) and/or zinc chloride (2 mg/kg) daily for five consecutive days. Twenty-four hours after treatment, ALA-D activity was determined in the absence and presence of DL-dithiothreitol (DTT). The negative geotaxis behavior was assessed in 9- to 13-day-old rats. Treatment with lead and/or zinc did not affect body, brain or kidney weights or brain- or kidney-to-body weight ratios of the animals. In spite of the absence of effect of any treatment on ALA-D specific activity in brain, kidney and blood, the reactivation index with DTT was higher in the groups treated with lead or lead + zinc than in the control group, in brain, kidney and blood (mean ± SEM; brain: 33.33 ± 4.34, 38.90 ± 8.24, 13.67 ± 3.41; kidney: 33.50 ± 2.97, 37.60 ± 2.67, 15.80 ± 2.66; blood: 63.95 ± 3.73, 56.43 ± 5.93, 31.07 ± 4.61, respectively, N = 9-11). The negative geotaxis response behavior was not affected by lead and/or zinc treatment. The results indicate that lead and/or zinc treatment during the second stage of rapid postnatal brain growth affected ALA-D, but zinc was not sufficient to protect the enzyme from the effects of lead in brain, kidney and blood.
