Foram estudados 17 indivíduos cujas idades variaram de 2 a 78 anos, procedentes da área endêmica de Jacarepaguá (RJ), com diagnóstico clínico, epidemiológico, imunológico e parasitológico de leishmaniose tegumentar americana. Todos foram tratados pelo antimoniato de N-metilglucamina na dose de 60 mg/kg/dia em três séries de 10 dias de duração, com 10 dias de intervalo entre cada série e submetidos à coleta e sangue venoso para a reação de imunofluorescência indireta (IF-IgG) antes, durante e após o medicamento. Estes 17 indivíduos foram reatores a intradermorreação de Montenegro (= 5 mm) e soro-reagentes (= 1:45) na IF-1gG, sendo que em 6 deles (31,7%) foi demonstrado ou isolado Leishmania braziliensis. A positividade da IF-IgG antes do tratamento foi de 76,4% (13 casos com título = 1:90). As médias geométricas das recíprocas dos títulos antes da primeira série do antimonial (89,9), durante o tratamento (63,6 a 29,3) e 10, 30 e 120 dias após medicação, mostraram uma graduação nitidamente decrescente (14,9;2,1 e 1,2), respectivamente. Todos tiveram suas lesões cicatrizadas ao final do tratamento sendo maior o número de cicatrizações após a 2°série. Somente 120 dias após a terapêutica, foram observados títulos abaixo de 1:45 na quase todalidade dos pacientes (16 casos - 94,1%) sugerindo que, na utilização da IF-IgG como controle de cura, faz-se necessário um acompanhamento sorológico de no mínimo 3 a 4 meses.

Com o intuito de se obter marcadores epidemiológicos, foram analisadas 240 amostras de S. agona isoladas de diferentes fontes (humana, alimentar e ambiental) oriundas de cinco Estados brasileiros (MG,SP,RJ,PE e RS). O sestudo da sensibilidade a 15 antimicrobianos e codificação numérica dos perfis de resistência propiciou o reconhecimento de 56 biotipos antimicrobianos, enquanto foram evidenciadas 40 amostras produtoras de colicina, pertencentes aos tipos: Ia (55%); B (32,5%), Ib (10%) e não tipável (2,5%). A aplicação desses elementos numa diferenciação intra-sorotipo é discutida.

A investigação teve por objetivo a avaliação de resistência a drogas dessa importante salmonela de distribuição cosmopolita. Nesse sentido, analisou-se a resistência a antibióticos e quimioterápicos de 240 amostras de S.agona isoladas de diferentes fontes (humana, alimentar e ambiental) provenientes de cinco estados brasileiros (MG, SP, RJ, PE e RS). Paralelamente, determinou-se a presença de fatores R em 26 estirpes representativas da amostragem.

Investigou-se a transferência de marcadores genéticos e a presença de DNA plasmidial em 240 culturas de Escherichia coli originárias de água de esgoto (afluente e fluentes) da Estação de Tratamento da Ilha do Governador, na cidade do Rio de Janeiro, RJ. Experimentos de conjugação com E. coli K 12 permitiram o isolamento de transconjugantes com resistência a antibióticos (Su, Sm, Tc, Cm e Ap); a metais pesados (Cu, Hg e Zn) e fatores colicinogênicos (Col Ia, Ib e V) principalmente para os coliformes isolados nos setores terminais da estação de tratamento. A distribuição de plasmídeos foi prevalente nas culturas de E. coli advindas dos efluentes, com percentuais superiores a 65.

Um total de 4.298 exemplares de Bradybaena similaris foram coletados na zona rural do município do Rio de Janeiro e nos municípios de Paracambi e Itaguai, RJ, entre outubro de 1983 e setembro de 1986. Destes caramujos, 2.005 apresentaram metacercárias livres na cavidade pericádica. As prevalências médias anuais foram de: 42,89% (primeiro ano), 43,42% (segundo ano) e 55,86% (terceiro ano). As metacercárias foram identificadas, através de infecções experimentais em pintos, como sendo de Posttharmostomum gallinum. A pouca variação de tamanho entre as metacercárias de um mesmo hospedeiro indica que não existe um recrutamento contínuo do parasito. Análise estatística mostrou que não existe correlação entre o tamanho do caramujo e o número de metacercárias presentes na cavidade pericárdica, e que a mortalidade dos caramujos, observada em laboratório, era devido à infecção parasitária.

The proteins of adults worms (male and female) of two isolates (BH and RJ) of Shistosoma mansoni were extracted using Triton X-114 phase separation. The SDS-polyacrilamide gel electrophoresis profiles of the three phases (detergent, aqueous and insoluble proteins) obtained were compared after Coomassie blue and silver staining, surface radioiodination and Western blotting. No major differences were detected between the 2 isolates. Of the 25 or more proteins which partitioned into the detergent phase, only about 8 proteins could be surface radiodinated on live adult worms. A comparison was also made between the profiles of mael and females worms, isolated from bisexually infected mice. Two major female-specific and one male-specific band were detected by silver and/or Coomassie staining. The female bands, 32 KDa and 18 KDa, partitioned into the detergent and aqueous phase, respectively. The male-specific band of 42 KDa remained in the insoluble phase. Antigenic differences between male and females protins were detected by Western vlotting using a sera from infected Nectomys squamipes.

The evaluation of the role of rodents as natural hosts of Schistosoma mansoni was studied at the Pamparrão Valley, Sumidouro, RJ, with monthly captures and examination of the animals. Twenty-three Nectomys squamipes and 9 Akodom arviculoides with a shistosomal infection rate of 56.5% and 22.2% respectively eliminated a great majority of viable eggs. With a strain isolated from one of the naturally infected N. squamipes, we infected 75% of simpatric Biomphalaria glabrata and 100% of albino Mus musculus mice. The adult worms, isolated from N. squamipes after perfusion were located mainly in the liver (91.5%) and the mesenteric veins (8.5%). The male/female proportion was 2:1. The eggs were distributed on small intestine segments (proximal, medial and distal portions) and the large intestine without any significant differences in egg concentration of these segments. In A. arviculoides, the few eggs eliminated by the stools were viable and there was litlle egg retention on intestinal segments. Considering the ease to complete S. mansoni biological cycle in the Nectomys/Biomphalaria/Nectomys system under laboratory conditions, probably the same is likely to occur in natural conditions. In support to this hypotesis there are also the facts that human mansonic shistosomiasis has a very low prevalence in Sumidouro and endemicity among the rodents has not changed even after repetead treatments of the local patients. Based on our experiments, we conclude that N. squamipes has become a natural host of S. mansoni and possibly may participate in keeping the cycle of schistosomiasis transmission at Pamparrão Valley.

Groups of 10 and 20 first instar larvae of Peckia chrysostoma (Wiedemann, 1830) were combined in a proteic source media with groups of the same number of first instar larvae of Adiscochaeta ingens (Walker, 1849) under the environmental conditions of Rio de Janeiro, RJ, Brasil. P. chrysostoma and A. ingens obtained average competitive potentials of 94.0 ± 2.0% and 31.0 ± 5.0% respectively. In the second experiment, larvae of P. chrysostoma were introduced approximately 15 hr after the introduction of A. ingens larvae (whose majority had already passed to the second instar) in the media. The corresponding average competitive potential of P. chrysostoma (82.0 ± 2.0%) was decreased when compared to the first experiment, but still greater than that of A. ingens (64.5 ± 9.5%). The competitive potential of A. ingens, however, increased significatively, demonstrating the influence of its previous colonization in the media for achieving a higher viability. In both experiments the competitive potential of P. chrysostoma was greater and similar to observations cited in the literature. Control-groups of each species were observed, individually, for the comparison. The mean value obtained for P. chrysostoma was 94.0 ± 3.7% (0.0% [experiment 1] and only 12.8% [experiment 2] greater than the average competitive factor). For A. ingens the average was 86.0 ± 7.3% (64.0% [experiment 1] and 25.0% [experiment 2] greater than average competitive factor).

Forty ducks, 18 Anas bahamensis (white-cheecked pintail) and 22 Amaxonetta brasiliensis (Brazilian duck) from Maricá and Guarapina lagoons, Maricá, RJ, Brazil, were examined. The prevalence of Philophthalmus gralli in A. bahamensis was 22.2% ans in A. brasiliensis was 27.27%. This is the first record of P. gralli in the Neotropical region, and A. bahamensis and A. brasiliensis are new host records for P. gralli.

In order to evaluate the permissiveness of Nectomys squamipes to Schistosoma mansoni and the influence of the albino mice on the morphological aspects of adult worms derived from a population isolated from N. squamipes, the morphology of adult S. mansoni Sambon, 1907 male worms was studied using a digital image analyser (MOP VIDEOPLAN) and light microscopy. Their sources were as follows: (1) recovered from the wild rodent N. squamipes Brants naturally infected from Sumidouro, RJ, Brazil; (2) recovered from albino mice experimentally infected with the strain derived from N. squamipes; (3) recovered after the isolation of a strain derived from aboriginal human infections in Sumidouro. Worms recovered from N. squamipes (group 1) showed body lenght and distance between suckers significantly bigger than those of the specimens maintained in mice (groups 2 and 3). The number of tests in group 1 was statistically less than of groups 2 and 3. Group 2 strains which were maintained in mice, presented the lenght of the worms as the only significant different character. Data show that: (1) N. squamipes is a more suitable host for the development of S. mansoni when compared to the albino mice; (2) a strain of S. mansoni isolated from a natural host undergoes morphological changes after its passage in the white mouse.

To characterize antibody binding to a panel of V3 loop peptides representing diverse HIV-1 neutralization epitopes, 149 HIV-1 infected individuals from Rio de Janeiro (RJ) were investigated. Results were analyzed with respect to risk factors for infection and other epidemiological and clinical data. Peptide reactivity was not associated with sex, clinical status, CD4 counts, antigenemia or ß2-microglobulin serum level. A segregation of peptide reactivity according to route of infection was encountered. This finding suggests that more then one viral strain may be circulating in RJ, in subjects with different risk factors for HIV-1 infection. An investigation of prevalent HIV-1 genotypes, serotypes and immunotypes may be of importance for the design and selection of potential vaccines to be used in Brazil as well as for the selection of populations to be included in future vaccine efficacy trials.

Worm burdens recovered from inbred mice strains, namely C57Bl/6, C57Bl/10, CBA, BALB/c, DBA/2 and C3H/He, conventionally maintained in two institutional animal houses in the State of Rio de Janeiro, RJ, Brazil, were analyzed and compared, regarding their prevalences and mean intensities.Three parasite species were observed: the nematodes Aspiculuris tetraptera, Syphacia obvelata and the cestode Vampirolepis nana. A modification of the anal swab technique is also proposed for the first time as an auxiliary tool for the detection of oxyurid eggs in mice

Thermophilic campylobacters were isolated from three sewage plants in Rio de Janeiro, RJ, Brazil and identified. Laboratory analysis of 390 sewage samples showed the presence of 169 thermophilic strains. The results demonstrated that human and animal pathogenic biotypes could be isolated from activated sludge during the initial processing steps. The aeration tank could be considered a barrier to Campylobacter survival. C. jejuni was the prevalent species isolated (40.8%).The most common biotypes were C. jejuni biotype I (21.3%), C. coli biotype I (16%) and C. jejuni biotype II ( 14.8%).

Between January and March 1998, a cross-sectional survey was carried out in four rural communities in Honduras, Central America. We examined the prevalence and intensity of Ascaris lumbricoides and Trichuris trichiura infections among 240 fecal specimens, and the association between selected socio-demographic variables and infection for 62 households. The overall prevalence of A. lumbricoides and T. trichiura was 45% (95% CI 39.0-51.9) and 38% (95% CI 31.8-44.4) respectively. The most intense infections for Ascaris and Trichuris were found in children aged 2-12 years old. By univariate analysis variables associated with infections of A. lumbricoides were: number of children 2-5 years old (p=0.001), level of formal education of respondents (p=0.01), reported site of defecation of children in households (p=0.02), households with children who had a recent history of diarrhea (p=0.002), and the location of households (p=0.03). Variables associated with both A. lumbricoides and T. trichiura infection included: number of children 6-14 years old (p=0.01, p=0.04, respectively), ownership of a latrine (p=0.04, p=0.03, respectively) and coinfection with either helminth (p=0.001, p=0.001, respectively). By multivariate analysis the number of children 2-5 years living in the household, (p=0.01, odds ratio (OR)=22.2), children with a recent history of diarrhea (p=0.0, OR=39.8), and infection of household members with T. trichiura (p=0.02, OR=16.0) were associated with A. lumbricoides infection. The number of children 6-14 years old in the household was associated with both A. lumbricoides and T. trichiura infection (p=0.04, p=0.01, OR=19.2, OR=5.2, respectively).

Five field populations of Musca domestica L. collected in poultry farms were bioassayed in order to detect possible resistance to the larvicide cyromazine in Brazil. The concentrations used were 0, 0.5, 0.1, 0.2, 0.4, 1, 2, 4 and 8 ppm. Three populations (Petrópolis, RJ, Montes Claros, MG and Promissão, SP) were resistant, while the other two populations (Ibiuna, SP and Monte Mor, SP) were more susceptible than the reference pathern used by the World Health Organization. The presence of three resistant house fly populations to cyromazine in Brazilian poultry farms strongly suggests that the operational aspects of larvicide use are important for the resistance development. Cyromazine is applied as a feed-through, both in Brazil and in the USA, where resistance has already been documented. However, in Denmark, where it was approved only as a topical manure spray, no case of resistance has yet been detected.
