The spatial and temporal retention of metals has been studied in water and sediments of the Gavião River, Anagé and Tremedal Reservoirs, located in the semi-arid region, Bahia - Brazil, in order to identify trends in the fluxes of metals from the sediments to the water column. The determination of metals was made by ICP OES and ET AAS. The application of statistical methods showed that this aquatic system presents suitable conditions to move Cd2+ and Pb2+ from the water column to the sediment.

The flavonoid fraction was purified by a combination of chromatography on Amberlite XAD-2 and preparative silica gel TLC. Morin (3, 5, 7, 2', 4'- pentahydroxyflavone) was the only flavonol found in honey from Brazilian Citrus sp.. The structure of morin was determined on the basis of UV and ¹H and 13C NMR spectral data together with literature references. This is the first report on the isolation of morin from Brazilian Citrus honey.

This study examined the spatial and temporal variations of six important parameters of the salt accumulation process in water samples collected along section urban of Contas River. The Na+, K+, Ca2+ and Mg2+ concentrations were determined by FAAS. The conductivity, total dissolved solids, Na+ and Ca2+ presented the largest seasonal and spatial variations in the urban area demonstrated that are appropriate indicators of urban contamination. The readily soluble salts in drainage urban, contribute for the degradation of the water of rivers located in semi-arid zones.

Alexandre Vandelli was the heir of two illustrious scientific traditions in the Luso-Brazilian world of the late eighteenth and early nineteenth centuries, that of his father, the celebrated Italian-Portuguese naturalist and chemist Domingos (Domenico) Vandelli, and that of his father-in-law, that protean figure in several scientific specialties as well as in politics, José Bonifácio de Andrada e Silva, who in later life was of paramount importance in the process of Brazilian independence from Portugal. The younger Vandelli followed their footsteps but soon engaged in a multiple career, at first in Portugal and later in Brazil, of which little is known and is here presented for the first time.

Mathematical models can help to prevent high levels of toxic substances in soil or fruits of plants treated with pesticides and indicate that such substances should be systematically monitored. The aim of this research was to study the kinetics of paclobutrazol biodegradation by soil native bacteria using mathematical models. Three models were used to assess the kinetics of paclobutrazol biodegradation obtained experimentally. Excellent fits were obtained using dual kinetic and logistic models. The use of glycerol as additional carbon source increased the biodegradation of PBZ and consequently decreased the time required for a given PBZ initial concentration be halved.

The influence of pre-processing of arabica coffee beans on the composition of volatile precursors including sugars, chlorogenic acids, phenolics, proteins, aminoacids, trigonelline and fatty acids was assessed and correlated with volatiles formed during roasting. Reducing sugars and free aminoacids were highest for natural coffees whereas total sugars, chlorogenic acids and trigonelline were highest for washed coffees. The highest correlation was observed for total phenolics and volatile phenolics (R= 0.999). Experimental data were evaluated by Principal Components Analysis and results showed that washed coffees formed a distinct group in relation to semi-washed and natural coffees.

Superabsorbent hydrogels based on poly (acrylamide-co-acrylate) and different kaolins, were prepared by free-radical aqueous copolymerization. FTIR and WAXS techniques were employed for characterization of a series of hydrogels, obtained by varying the percentage of clay, crosslinking and constitution of kaolin. The water absorbency at equilibrium (Weq) decreased with increasing clay content and the amount of crosslinking agent. Superabsorbent hydrogel (Weq > 1084 g H2O/g gel) was obtained as 10 wt% of white kaolin and 0.05 mol% of crosslinking agent were used. The hydrogel proved sensitive to pH variation and the presence of salts.

The use and characterization of the first Certified Reference Material (CRM) of Brazilian coal for analytical quality control are presented. All results were determined within the limits of repeatability and reproducibility allowed under the standards. Notable among the characterization parameters was the mineral matter content (45.39%) obtained by an alternative procedure showing a low relative error (-2.1%) compared to the standard technique (low ashing temperature). The possible expanded application of this CRM application in national laboratories, beyond the certificated and reference parameters, has advantages such as lower cost and easy availability, but round robin tests must be performed.

Um isolado do Southern bean mosaic virus (SBMV), gênero Sobemovirus, encontrado em feijoeiro (Phaseolus vulgaris) no Estado de São Paulo, foi purificado e algumas de suas propriedades moleculares determinadas. As partículas virais apresentam diâmetro de 28-30 nm e proteína capsidial com massa molecular estimada em 30 kDa. Das partículas virais foi extraído RNA de vários tamanhos (4,2 Kb, 3,1 Kb, 2,65 Kb, 2,15 Kb, 1,64 Kb, 1,36 Kb e 1,0 Kb) sendo aquele de 4,2 Kb o RNA genômico e o de 1,0 Kb supostamente um subgenômico que codifica para a proteína capsidial. Ácidos ribonucleicos de mesmo tamanho foram também detectados in vivo, indicando estar associados à replicação viral. Na análise do RNA de fita dupla (dsRNA), somente duas espécies foram detectadas (4,2 Kpb e 1,0 Kpb) correspondendo às formas replicativas do RNA genômico e do subgenômico para proteína capsidial. Os resultados indicam que somente estes dois RNA são replicados por meio de formas replicativas (RFs), enquanto os demais devem ser formados talvez por iniciação interna da fita negativa do RNA genômico. O SBMV-B SP apresentou propriedades moleculares análogas àquelas do SBMV descrito na América do Norte.

Oito espécies de Ravenelia descritas anteriormente foram revisadas e acrescentadas ilustrações inéditas das características morfológicas de alguns desses fungos causadores de ferrugem em Leguminosae. As espécies de Ravenelia estudadas foram: Ravenelia bezerrae sobre Enterolobium ellipticum ; R. densifera sobre Senna silvestris; R. dieteliana sobre Calliandra dysantha. var. dysantha; R. geminipora sobre Platymenia reticulata; R. lonchocarpi sobre Lonchocarpus campestris; R. mimosae-sensitivae sobre Mimosa radula var. imbricata; R. pileolarioides sobre Caesalpinia pyramydales e R. santos-costae, sobre Calliandra dysantha.

O presente trabalho caracteriza a região 5'-terminal de um isolado do Southern bean mosaic virus encontrado no Estado de São Paulo (SBMV-SP). O RNA foi extraído de partículas virais purificadas e submetido a RT-PCR usando oligonucleotídeos desenhados para amplificar cerca de 590 nt da região 5'-terminal do RNA viral. Foi obtido um fragmento de tamanho esperado que, após clonagem e seqüenciamento, mostrou a existência de uma região não codificadora com 92 nt e a primeira ORF, começando no primeiro AUG (posição 93) e terminando no códon UGA na posição 534. Na região não codificadora foi detectado um segmento parcialmente complementar ao RNA ribossomal 18S. A ORF1 codifica uma proteína de 147 aminoácidos com massa molecular estimada de 17080 Da. A extremidade 3' da ORF1 sobrepõe a extremidade 5' da ORF2 em 34 nucleotídeos. Os resultados obtidos indicam que a região 5'-terminal do RNA do SBMV-SP é similar ao isolado Arkansas (SBMV-ARK) descrito na América do Norte.

O presente trabalho caracteriza a região 3'-terminal do genoma de um isolado do Southern bean mosaic virus encontrado no Estado de São Paulo (SBMV-SP). O RNA foi extraído de partículas virais purificadas e submetido a RT-PCR usando oligonucleotídeos desenhados para amplificar 972 nt da região 3'-terminal do RNA viral. Foi obtido fragmento de tamanho esperado que inclui o gene da proteína capsidial e a região 3'-terminal não codificadora. O gene da proteína capsidial (ORF4) contém 801 nucleotídeos, incluindo-se o códon de terminação UGA, com seqüência deduzida de 266 aminoácidos e massa molecular estimada de 28.800 Da. Sessenta e um aminoácidos terminais da ORF2 estão sobrepostos na ORF4. O "sinal de localização nuclear", encontrado dentro do "Domínio R" na região 5'-terminal da ORF4 de alguns sobemovírus, não foi identificado no SBMV-SP. Esse dado pode explicar a ausência de partículas virais do SBMV-SP no núcleo celular. A seqüência do SBMV-SP apresentou identidade de nucleotídeos e aminoácidos de, respectivamente, 91% e 93% com o isolado "Arkansas" (SBMV-ARK) descrito nos EUA. Os resultados obtidos indicam que o SBMV-SP e o SBMV-ARK são isolados muito proximamente relacionados.

A ramulose do algodoeiro (Gossypium hirsutum) causada por Colletotrichum gossypii var. cephalosporioides é responsável por danos apreciáveis no rendimento de algodão. O patógeno ataca toda a parte aérea das plantas provocando manchas nas folhas e no colmo, e superbrotamento da planta. Plantas severamente infetadas normalmente mostram nanismo. Devido a carência de informação sobre o mecanismo de resistência, o processo de produção de novas cultivares com resistência a ramulose é algo prejudicado. O objetivo do presente trabalho foi compreender o mecanismo de herança a ramulose em duas cultivares resistentes (BRS ANTARES e IAC 23) quando cruzadas com uma cultivar suscetível (STO 474). As avaliações das populações F2 e das populações de retrocruzamento demonstraram em BRS ANTARES que a resistência a ramulose é condicionada por um gene dominante, enquanto que em IAC 23 a resistência é governada por dois genes dominantes independentes, e de efeito duplicado.

Re-finding of Balanoglossus gigas FR. MUULLER on the brasilian sea shore. Balanoglossus gigas, the giant Enteropneusta, has been described for the first time by Fritz Muller in his notes collected and published by Dr. HERMANN VON IHERING (1898, pg. 35). FR. MULLER found the Balanoglossus on the coats of the State of Santa Catarina in 1884. Once again in 1885 several specimens of that animal were captured by FR. MULLER in the same place. Since that time up to now no references on the occurrence of this animal have been found in the zoological bibliography. During the spring of 1948 Prof. W. BESNARD, Director of he Instituto Paulista de Oceanografia saw some signals indicating the existence of Balanoglossus at the beach of São Sebastião, State of São Paulo. From 1948 to now several attempts have been made to catch the animal alive and complete. On the last September during a shorts visit to the beach of São Sebastião one Balanoglossus was captured and brought to the Department of General and Animal Physiology in good conditions. The animal measured 1.80 m in length. It seems to be the largest Balanoglossus known. According to the descriptions of SPENGEL (1893, pg. 158), and VAN DER HORST (1939) pg. 717) it was possible to identify this Enteropneusta as Balanoglossus gigas FR. MULLER, refound at the Brazilian coast sixty six years after its discovery by FR. MÜLLER.
