Orally transmitted Chagas disease (ChD), which is a well-known entity in the Brazilian Amazon Region, was first documented in Venezuela in December 2007, when 103 people attending an urban public school in Caracas became infected by ingesting juice that was contaminated with Trypanosoma cruzi. The infection occurred 45-50 days prior to the initiation of the sampling performed in the current study. Parasitological methods were used to diagnose the first nine symptomatic patients; T. cruzi was found in all of them. However, because this outbreak was managed as a sudden emergency during Christmas time, we needed to rapidly evaluate 1,000 people at risk, so we decided to use conventional serology to detect specific IgM and IgG antibodies via ELISA as well as indirect haemagglutination, which produced positive test results for 9.1%, 11.9% and 9.9% of the individuals tested, respectively. In other more restricted patient groups, polymerase chain reaction (PCR) provided more sensitive results (80.4%) than blood cultures (16.2%) and animal inoculations (11.6%). Although the classical diagnosis of acute ChD is mainly based on parasitological findings, highly sensitive and specific serological techniques can provide rapid results during large and severe outbreaks, as described herein. The use of these serological techniques allows prompt treatment of all individuals suspected of being infected, resulting in reduced rates of morbidity and mortality.

Diarrhoeal disease is still considered a major cause of morbidity and mortality among children. Among diarrhoeagenic agents, Shigella should be highlighted due to its prevalence and the severity of the associated disease. Here, we assessed Shigella prevalence, drug susceptibility and virulence factors. Faeces from 157 children with diarrhoea who sought treatment at the Children's Hospital João Paulo II, a reference children´s hospital in Belo Horizonte, state of Minas Gerais, Brazil, were cultured and drug susceptibility of the Shigella isolates was determined by the disk diffusion technique. Shigella virulence markers were identified by polymerase chain reaction. The bacterium was recovered from 10.8% of the children (88.2% Shigella sonnei). The ipaH, iuc, sen and ial genes were detected in strains isolated from all shigellosis patients; set1A was only detected in Shigella flexneri. Additionally, patients were infected by Shigella strains of different ial, sat, sen and set1A genotypes. Compared to previous studies, we observed a marked shift in the distribution of species from S. flexneri to S. sonnei and high rates of trimethoprim/sulfamethoxazole resistance.

Lipophorin (Lp) is the main haemolymphatic lipoprotein in insects and transports lipids between different organs. In adult females, lipophorin delivers lipids to growing oocytes. In this study, the interaction of this lipoprotein with the ovaries of Rhodnius prolixus was characterised using an oocyte membrane preparation and purified radiolabelled Lp (125I-Lp). Lp-specific binding to the oocyte membrane reached equilibrium after 40-60 min and when 125I-Lp was incubated with increasing amounts of membrane protein, corresponding increases in Lp binding were observed. The specific binding of Lp to the membrane preparation was a saturable process, with a Kdof 7.1 ± 0.9 x 10-8M and a maximal binding capacity of 430 ± 40 ng 125I-Lp/µg of membrane protein. The binding was calcium independent and pH sensitive, reaching its maximum at pH 5.2-5.7. Suramin inhibited the binding interaction between Lp and the oocyte membranes, which was completely abolished at 0.5 mM suramin. The oocyte membrane preparation from R. prolixus also showed binding to Lp from Manduca sexta. When Lp was fluorescently labelled and injected into vitellogenic females, the level of Lp-oocyte binding was much higher in females that were fed whole blood than in those fed blood plasma.

The polymerase chain reaction (PCR)-based methods for the diagnosis of malaria infection are expected to accurately identify submicroscopic parasite carriers. Although a significant number of PCR protocols have been described, few studies have addressed the performance of PCR amplification in cases of field samples with submicroscopic malaria infection. Here, the reproducibility of two well-established PCR protocols (nested-PCR and real-time PCR for the Plasmodium 18 small subunit rRNA gene) were evaluated in a panel of 34 blood field samples from individuals that are potential reservoirs of malaria infection, but were negative for malaria by optical microscopy. Regardless of the PCR protocol, a large variation between the PCR replicates was observed, leading to alternating positive and negative results in 38% (13 out of 34) of the samples. These findings were quite different from those obtained from the microscopy-positive patients or the unexposed individuals; the diagnosis of these individuals could be confirmed based on the high reproducibility and specificity of the PCR-based protocols. The limitation of PCR amplification was restricted to the field samples with very low levels of parasitaemia because titrations of the DNA templates were able to detect < 3 parasites/µL in the blood. In conclusion, conventional PCR protocols require careful interpretation in cases of submicroscopic malaria infection, as inconsistent and false-negative results can occur.

In 2009, the World Health Organization (WHO) issued a new guideline that stratifies dengue-affected patients into severe (SD) and non-severe dengue (NSD) (with or without warning signs). To evaluate the new recommendations, we completed a retrospective cross-sectional study of the dengue haemorrhagic fever (DHF) cases reported during an outbreak in 2011 in northeastern Brazil. We investigated 84 suspected DHF patients, including 45 (53.6%) males and 39 (46.4%) females. The ages of the patients ranged from five-83 years and the median age was 29. According to the DHF/dengue shock syndrome classification, 53 (63.1%) patients were classified as having dengue fever and 31 (36.9%) as having DHF. According to the 2009 WHO classification, 32 (38.1%) patients were grouped as having NSD [4 (4.8%) without warning signs and 28 (33.3%) with warning signs] and 52 (61.9%) as having SD. A better performance of the revised classification in the detection of severe clinical manifestations allows for an improved detection of patients with SD and may reduce deaths. The revised classification will not only facilitate effective screening and patient management, but will also enable the collection of standardised surveillance data for future epidemiological and clinical studies.

The role played by different mammal species in the maintenance of Trypanosoma cruzi is not constant and varies in time and place. This study aimed to characterise the importance of domestic, wild and peridomestic hosts in the transmission of T. cruzi in Tauá, state of Ceará, Caatinga area, Brazil, with an emphasis on those environments colonised by Triatoma brasiliensis. Direct parasitological examinations were performed on insects and mammals, serologic tests were performed on household and outdoor mammals and multiplex polymerase chain reaction was used on wild mammals. Cytochrome b was used as a food source for wild insects. The serum prevalence in dogs was 38% (20/53), while in pigs it was 6% (2/34). The percentages of the most abundantly infected wild animals were as follows: Thrichomys laurentius 74% (83/112) and Kerodon rupestris 10% (11/112). Of the 749 triatomines collected in the household research, 49.3% (369/749) were positive for T. brasiliensis, while 6.8% were infected with T. cruzi (25/369). In captured animals, T. brasiliensis shares a natural environment with T. laurentius, K. rupestris, Didelphis albiventris, Monodelphis domestica, Galea spixii, Wiedomys pyrrhorhinos, Conepatus semistriatus and Mus musculus. In animals identified via their food source, T. brasiliensis shares a natural environment with G. spixii, K. rupestris, Capra hircus, Gallus gallus, Tropidurus oreadicus and Tupinambis merianae. The high prevalence of T. cruzi in household and peridomiciliar animals reinforces the narrow relationship between the enzootic cycle and humans in environments with T. brasiliensis and characterises it as ubiquitous.

Visceral leishmaniasis (VL) in Brazil is transmitted by the phlebotomine Lutzomyia longipalpis and in some midwestern regions by Lutzomyia cruzi. Studies of the phlebotomine fauna, feeding habits and natural infection rate by Leishmania contribute to increased understanding of the epidemiological chain of leishmaniases and their vectorial capacity. Collections were performed in Jaciara, state of Mato Grosso from 2010-2013, during which time 2,011 phlebotomines (23 species) were captured (68.70% Lu. cruzi and 20.52% Lutzomyia whitmani). Lu. cruzi females were identified by observing the shapes of the cibarium (a portion of the mouthpart) and spermatheca, from which samples were obtained for polymerase chain reaction to determine the rates of natural infection. Engorged phlebotomines were assessed to identify the blood-meal host by ELISA. A moderate correlation was discovered between the number of Lu. cruzi and the temperature and the minimum rate of infection was 6.10%. Twenty-two females were reactive to the antisera of bird (28%), dog (3.30%) and skunk (1.60%). We conclude that Lu. cruzi and Lu. whitmani have adapted to the urban environment in this region and that Lu. cruzi is the most likely vector of VL in Jaciara. Moreover, maintenance of Leishmania in the environment is likely aided by the presence of birds and domestic and synanthropic animals.

Orally transmitted Chagas disease has become a matter of concern due to outbreaks reported in four Latin American countries. Although several mechanisms for orally transmitted Chagas disease transmission have been proposed, food and beverages contaminated with whole infected triatomines or their faeces, which contain metacyclic trypomastigotes of Trypanosoma cruzi, seems to be the primary vehicle. In 2007, the first recognised outbreak of orally transmitted Chagas disease occurred in Venezuela and largest recorded outbreak at that time. Since then, 10 outbreaks (four in Caracas) with 249 cases (73.5% children) and 4% mortality have occurred. The absence of contact with the vector and of traditional cutaneous and Romana’s signs, together with a florid spectrum of clinical manifestations during the acute phase, confuse the diagnosis of orally transmitted Chagas disease with other infectious diseases. The simultaneous detection of IgG and IgM by ELISA and the search for parasites in all individuals at risk have been valuable diagnostic tools for detecting acute cases. Follow-up studies regarding the microepidemics primarily affecting children has resulted in 70% infection persistence six years after anti-parasitic treatment. Panstrongylus geniculatushas been the incriminating vector in most cases. As a food-borne disease, this entity requires epidemiological, clinical, diagnostic and therapeutic approaches that differ from those approaches used for traditional direct or cutaneous vector transmission.

In the present paper, we announce new draft genomes of four Leptospira interrogans strains named Acegua, RCA, Prea, and Capivara. These strains were isolated in the state of Rio Grande do Sul, Brazil, from cattle, dog, Brazilian guinea pig, and capybara, respectively.

O objetivo neste trabalho foi propor uma perspectiva de análise do ambiente propício à inovação com a incorporação da variável cultura organizacional. A base da análise recai sobre a adaptação da metodologia proposta por Van de Ven, Angle e Poole (2000). A contribuição proposta neste trabalho situa-se na adaptação da metodologia, retirando o foco do processo de inovação para o ambiente de inovação. Para que fosse contemplado todo o ambiente, foi necessária a inclusão de uma análise dos comportamentos coletivos organizacionais. Incorporaram-se questões que envolvem aspectos relacionados com a cultura organizacional, sob a perspectiva dos estudos de Bates et al. (1995). O questionário proposto pelo Minnesota Innovation Research Program (MIRP), com 45 questões adaptadas para o levantamento do ambiente de inovação, foi aplicado a 349 empregados de uma empresa de grande porte do setor metal-mecânico. Com a técnica de modelagem de equações estruturais (MEE), testou-se o modelo apresentado pelo MIRP, considerando as dimensões externas e internas ao grupo de inovação e os resultados decorrentes do ambiente de inovação. Em um segundo momento, um novo teste com a incorporação da variável cultura organizacional foi realizado. Os resultados apontaram que o segundo teste obteve um melhor ajuste, em torno de 27%, indicando que a variável impactou o resultado da modelagem. Convém ressaltar que o construto apresentado foi baseado em um levantamento teórico e para sua validação necessita de gama maior de resultados empíricos.

Neste trabalho, visa-se avaliar modelos organizacionais existentes e propostas para os perímetros públicos de irrigação (PPIs) no semiárido brasileiro. Em tempos de transformação na agricultura e na remodelação de investimentos públicos com o uso de mecanismos de parcerias público-privadas, alternativas quanto a arquiteturas organizacionais e PPIs começam a surgir. É utilizada a nova economia institucional e suas vertentes de economia dos custos de transação e de ambiente institucional como base de análise. Entrevistas com potenciais investidores selecionados são feitas e é sugerido que o modelo contratual de PPI tende a ser o mais viável tanto sob o ponto de vista conceitual como sob o resultante das entrevistas com a iniciativa privada. Todavia, são feitas ressalvas nas formas contratuais entre os agentes, como forma de assegurar o desenvolvimento dos perímetros e questões sobre o limite da ação pública.

A metodologia desenvolvida pelo Minnesota Innovation Research Program para avaliação dos processos de inovação, denominada Minnesota Innovation Survey (MIS) serviu de base para a proposição de um modelo de análise de ambiente propício ao desenvolvimento de inovações. A necessidade da proposição de um novo modelo surgiu a partir da observação dos resultados de pesquisas que utilizaram a mesma metodologia, em que questões e dimensões se apresentavam altamente correlacionadas, gerando redundância de medição de construtos e pouca precisão nos resultados. Com isso buscou-se, por meio de embasamento teórico e estatística multivariada, reduzir o número de construtos afins no intuito de evitar a multicolinearidade e os resultados dúbios. A metodologia utilizada para o desenvolvimento do modelo proposto caracteriza-se como descritiva com método quantitativo. Foram aplicados questionários da metodologia MIS a 349 empregados de uma empresa metal mecânica. A aplicação do modelo original obedeceu a todos os passos prescritos, mostrando a aderência por meio de modelagem de equações estruturais. Como resultado verificou-se que a redução de dados proveniente dos testes estatísticos impactou significantes alterações na metodologia de base, caracterizando o surgimento de uma nova metodologia de análise pela diminuição de 70% dos dados multivariados. Conclui-se que a nova metodologia, apesar de eliminar 65 questões do instrumento de coleta de dados, não reduz seu poder de explicação e eficácia quanto às relações dos ambientes organizacionais com os resultados da inovação. Em termos de execução de pesquisa, o novo modelo facilita a coleta de dados, minimiza a dispersão do respondente no momento do preenchimento do questionário, aumentando a confiabilidade dos dados, fornece informações acuradas sobre o ambiente inovador e garante robustez de análise. Empresas podem beneficiar-se desse instrumento de pesquisa por ser de fácil aplicação, entendimento e análise.

RESUMONeste artigo, analisa-se como os mercados diferenciados (status), o conselho de administração e as características organizacionais condicionam a reputação das empresas de capital aberto brasileiras. Com base em 1.396 observações coletadas entre os anos de 2002 e 2007 na BM&F Bovespa, utiliza-se um modelo de seleção em dois estágios para evidenciar que empresas que fazem parte do Novo Mercado, como do mercado norte-americano por meio de American Depositary Receipt (ADR), têm maiores chances de ser mais admiradas. Todavia, isso não influencia a diferenciação da reputação. Também se verifica que o capital social dos conselhos, por meio dos laços fracos, está situado em empresas com reputação destacada. Além disso, os resultados apontam que a rentabilidade e o tamanho da empresa também estão positivamente associados com a reputação. Diferentemente do que se hipotetizou, o coeficiente de agrupamento afeta de forma negativa a reputação, remetendo ao chamado lado obscuro das redes. Por fim, evidencia-se que a alavancagem financeira tem um efeito paradoxal: entre empresas mais admiradas, a alavancagem está positivamente relacionada ao aumento da reputação, mas entre aquelas menos admiradas, o endividamento apresenta efeito inverso.

RESUMOO objetivo neste trabalho é analisar se os Fundos Setoriais redefiniram o modelo de promoção de Ciência, Tecnologia e Inovação (CT&I), mudando de ofertista-linear para o modelo sistêmico, como previsto em sua concepção. Para a consolidação desse modelo, o setor produtivo precisa estar inserido no processo inovativo. Dessa forma, apoiou-se na análise da presença ou não da empresa em dois objetos dos Fundos Sociais. Os resultados apontam que nem as agências de fomento, nem os projetos aprovados têm buscado estimular a cooperação universidade-empresa. Assim, o que se verifica é que não houve uma redefinição para o modelo sistêmico de inovação, e sim que a política de promoção de CT&I continua a seguir um modelo linear, mas agora induzido pela demanda.
