Proteins are potential targets for singlet molecular oxygen (¹O2) oxidation. Damages occur only at tryptophan, tyrosine, histidine, methionine, and cysteine residues at physiological pH, generating oxidized compounds such as hydroperoxides. Therefore, it is important to understand the mechanisms by which ¹O2, hydroperoxides and other oxidized products can trigger further damage. The improvement and development of new tools, such as clean sources of ¹O2 and isotopic labeling approaches in association with HPLC/mass spectrometry detection will allow one to elucidate mechanistic features involving ¹O2-mediated protein oxidation.

Electrochemical sensors have attracted considerable attention in recent years because they provide data about the chemical state of our surroundings and the dynamics of the chemical transformations in the form a spatially resolved image. Particular interest has been directed to measurements in restricted-volume samples as new technologies enable the fabrication of miniaturized versions of sensors with reproducible characteristics. Taking these aspects into consideration, this review focuses on the use of electrodes of micrometer dimensions to acquire chemical information in microdomains in which concentrations may not be spatially homogeneous. This is possible because microelectrodes allow fast-response measurements with micrometer resolution to be performed. On the other hand, the use of microelectrodes as amperometric sensors presents an inherent drawback owing to the insufficient specificity toward the substrate of interest. Hence, some comments on strategies to enhance the selectivity of amperometric sensors are also made. Finally, recent applications of structurally microscopic electrodes as in vivo sensors are shown, as well as a prospect of the future trend in this field.

Gas chromatography coupled with mass spectrometry (GC-MS) is widely used for the characterization of volatile compounds. However, due to the complexity of the soluble coffee matrix, a complete identification of the components should not be based on mass spectra interpretation only. The linear index of retention (LRI) is frequently used to give support to mass spectra. The aim of this work is to investigate the characterization of the volatile compounds in soluble coffee samples by GC-MS using LRI values found with a HP-INNOWAX column. The method used allows a significant increase of the reliability of identifying compounds.

Ozonation tests with and without prior filtration by means of a 50 micron mesh cartridge filter were conducted with primary sanitary effluents. Filtration led to increased inactivation efficiencies with regard to total and thermotolerant coliforms but it did not seem to influence heterotrophic plate count (HPC) bacteria inactivation efficiencies significantly. Application of the Chick-Watson model to experimental data obtained in the situation of constant inactivation showed that the ozone dosage was more important to bacterial inactivation than the contact time with regard to the cases of thermotolerant coliform inactivation in filtered samples and HPC bacteria and total coliform inactivation in non-filtered samples.

The chemical role of iron-bearing compounds on the dynamics of phosphorus in selected Brazilian latosols was investigated. The iron oxides were characterized in an attempt to assess their main chemical-mineralogical properties influencing the ion sorption mechanisms in those pedosystems. It was found that increasing total iron contents tend to increase the phosphorus adsorption capacity in the selected soils. 110 K-Mössbauer data reveal that the dominant iron oxides are hematite and goethite. Particularly for the yellower soil samples some prominent doublets, more certainly due to superparamagnetic relaxation, may be assigned to corresponding fractions of relatively small-sized particles.

The present work aimed to characterize an aluminum industry by-product in natura (L.A. nat) and after phosphate and thermal pretreatments; evaluate the adsorption/desorption capacity of Cd and Pb by this L.A. nat form and after the aforementioned pretreatments, comparing them with an in natura iron mining by-product (L.F. nat). The L.A. nat presented a high pH as well as a high Na concentration and also an oxide-rich mineralogy. Pretreatment of the by-product had no significant effect upon Cd and Pd adsorption/desorption. The L.A. nat performed better than the L.F. nat as an Cd and Pb adsorbent.

Samplings of atmospheric particulate matter (PM) were carried out between the months of March and April of 2007, simultaneously in two areas of Londrina, an urban (Historical Museum) and other rural (Farm School-UEL). PM was collected using the cascade impactor consisting of four impaction stages (0.25 to 10 μm). The results indicated that the fine fraction (PM2.5) represented a significant portion of the mass of PM10 (70 and 67% in the urban and rural places, respectively). Cl-, NO3- and SO4(2-) were determined by ion chromatography and the size distribution is presented. Natural and anthropogenic sources were suggested to the ionic components in the fine and coarse mode of PM.

Magnetic soils forming on tuffite of the region of Alto Paranaíba, Minas Gerais, Brazil, usually contain iron-rich spinels exceptionally rich in magnesium and titanium. In this work, samples of the magnetically separated portion from the sand fraction of a Brunizém (Chernossolo) and from its mother-rock material were analyzed with synchrotron X-ray diffraction and 57Fe-Mössbauer spectroscopy. Magnesioferite (MgFe2O4) and maghemite (its pure non-stoichiometric spinel structure, Fe8/3 ⊕ 1/3 O4, where ⊕ = cation vacancy, corresponds to γFe2O3) were the magnetic iron oxides so identified. Basing on these data, a consistent chemical-mineralogical model is proposed for the main transformation steps involving these iron oxides in the pedosystem, starting on magnesioferrite to finally render hematite (αFe2O3), passing through maghemite as an intermediate specie.

Glass-ceramics are prepared by controlled separation of crystal phases in glasses, leading to uniform and dense grain structures. On the other hand, chemical leaching of soluble crystal phases yields porous glass-ceramics with important applications. Here, glass/ceramic interfaces of niobo-, vanado- and titano-phosphate glasses were studied by micro-Raman spectroscopy, whose spatial resolution revealed the multiphase structures. Phase-separation mechanisms were also determined by this technique, revealing that interface composition remained unchanged as the crystallization front advanced for niobo- and vanadophosphate glasses (interface-controlled crystallization). For titanophosphate glasses, phase composition changed continuously with time up to the equilibrium composition, indicating a spinodal-type phase separation.

This work evaluated the kinetics as well as the retention capacity of Cu, Zn, Cd, and Pb by arbuscular mycorrhizal fungi (AMF) mycelium. The metal retention is a fast process with Cu being retained 3, 30, and 60 times faster than Zn, Cd, and Pb, respectively. Metal retention capacity varied amongst the different tested AMF species and decreased in the following order: Cu>Zn>>Cd>Pb. The Glomus clarum mycelium showed the highest retention capacity for Cu, Cd and Pb, whereas Zn was mostly retained by Gigaspora gigantea mycelium. The simultaneous application of all tested metals in solution decreased Cu and Zn retention by AMF mycelium. The high retention capacity of Cu and Zn by mycelium of G. clarum and G. gigantea suggests a promising use of these isolates in phytoremediation.

The earth diatomite is a material used by the industries in the filtration process and clarification of the beer. This material presents a reduced useful life due to the blockages of their pores during the filtration process. The objective of this work was to reactivate the properties of filtration of the earth diatomite, saturated with organic matter during the filtration stage, starting from a controlled thermal treatment. The obtained results demonstrated that the earth diatomite saturated with organic matter submitted to a controlled thermal treatment has their filtration properties reactivated, could be reused in the beer production process.

The biodegradation of lignocellulosic materials is an important natural process because it is responsible for the carbon recycling. When induced under controlled conditions, this process can be used for technological applications such as biopulping, biobleaching of cellulosic pulps, pre-treatment for subsequent saccharification and cellulosic-ethanol production, and increase of the digestibility in agroindustrial residues used for animal feed. In the present work, the enzymatic and non-enzymatic mechanisms involved in the biodegradation of lignocellulosic materials by fungi were reviewed. Furthermore, the technological applications of these extracellular metabolites are presented and discussed.

Solutions of [hydroxy(tosyloxy)iodo]benzene (HTIB or Koser's reagent) in acetonitrile were analyzed using high resolution electrospray ionization mass spectrometry (ESI-MS) and electrospray ionization tandem mass spectrometry (ESI-MS/MS) under different conditions. Several species were characterized in these analyses. Based on these data, mechanisms were proposed for the disproportionation of the iodine(III) compounds in iodine(V) and iodine(I) species.

O presente trabalho testou quatro métodos de isolamento de Streptomyces spp. em tubérculos de batata (Solanum tuberosum) com sarna comum, superficial e profunda, caracterizando os isolados quanto a morfologia, serologia e patogenicidade. Para o isolamento foram testados: meio de cultura ágar-água a pH 10; meio contendo antibióticos; meio de asparagina e meio de quitina. O meio ágar-água pH 10 foi o mais eficaz no isolamento de Streptomyces spp. com média de 129 colônias/placa, além de ser de fácil preparo, menor custo e proporcionar melhor visualização das colônias. No meio de antibiótico verificou-se média de 54% dos fragmentos de tubérculos plaqueados com crescimento de Streptomyces spp. Já nos meios de asparagina e quitina a média foi de 36,3 e 2,5 colônias/placa, respectivamente. Para caracterização dos isolados obtidos utilizou-se o meio de extrato de levedura e malte. As colônias originadas apresentaram coloração que variou de cinza a marrom e de branco a creme, com ou sem produção de pigmento, com cadeias de esporos flexuosas ou espiraladas, de tamanho variável e produzindo ou não micélio aéreo em colônias com cadeias espiraladas. Dezenove isolados, representando esses diferentes tipos, foram inoculados na batata cv. Monalisa por infestação de solo autoclavado, antes da semeadura dos tubérculos-sementes. Sintomas típicos da doença foram verificados 14 semanas após a inoculação, para oito isolados. Anti-soros produzidos em coelhos contra três isolados fitopatogênicos apresentaram reação serológica (dupla difusão em gel-ágar de Ouchterlony) para os antígenos homólogos e para poucos antígenos heterólogos, porém os isolados de Streptomyces com patogenicidade confirmada não apresentaram antígenos em comum.

Compostos secundários presentes em plantas medicinais podem desempenhar funções importantes em interações planta-patógeno, através de ação antimicrobiana direta ou ativando mecanismos de defesa de outras plantas que venham a ser tratadas com esses compostos. Com o objetivo de verificar o potencial de eucalipto (Eucalyptus citriodora) no controle alternativo de antracnose em pepino (Cucumus sativus), extrato aquoso (EA) desta essência florestal, autoclavado ou não autoclavado, nas concentrações de 0,1, 1, 5, 10, 15, 20 e 25% foi utilizado nos seguintes ensaios: indução de fitoalexinas em mesocótilos estiolados de sorgo (Sorghum bicolor) e em cotilédones de soja (Glycine max); fungitoxicidade in vitro sobre conídios de Colletotricum lagenarium; indução de resistência local ou sistêmica em plantas de pepino. Água e Bion foram utilizados como tratamentos controle. Os resultados indicaram que os EAs autoclavados e não autoclavados induziram a síntese de fitoalexinas em sorgo a partir da concentração de 1%. Em soja ocorreu síntese de gliceolina a partir de 10% e 15% dos EAs autoclavados e não autoclavados, respectivamente. Houve inibição total na germinação de esporos e formação de apressórios de C. lagenarium em concentrações de 20% e 1% do EA autoclavado, respectivamente. Para o extrato não autoclavado houve 75% de inibição da germinação de esporos em 25% do EA e inibição total da formação de apressórios em 15% do EA. Baseado no parâmetro tamanho de lesão, o extrato aquoso de E. citriodora, não autoclavado, apresenta potencial para induzir resistência local em pepino contra C. lagenarium.
