In hospital-based series viral hepatitis B has been frequently described in association with schistosomiasis whilst in field-based studies the association has not been confirmed. The association between schistosomiasis and Salmonella bacteraemia has been well documented. More recently, acute schistosomiasis has been shown to be a facilitating factor in the genesis of pyogenic liver abscesses caused by Staphylococcus aureus. New evidences indicate an interaction between the acquired immunodeficiency syndrome (AIDS) and schistosomiasis. In this paper, data on the association of schistosomiasis with other infections are updated.

The essential oil (EO) of Ocimum gratissimum inhibited Staphylococcus aureus at a concentration of 0.75 mg/ml. The minimal inhibitory concentrations (MICs) for Shigella flexineri, Salmonella enteritidis, Escherichia coli, Klebsiella sp., and Proteus mirabilis were at concentrations ranging from 3 to 12 mg/ml. The endpoint was not reached for Pseudomonas aeruginosa (>=24 mg/ml). The MICs of the reference drugs used in this study were similar to those presented in other reports. The minimum bactericidal concentration of EO was within a twofold dilution of the MIC for this organism. The compound that showed antibacterial activity in the EO of O. gratissimum was identified as eugenol and structural findings were further supported by gas chromatography/mass spectra retention time data. The structure was supported by spectroscopic methods.

One hundred and thirty cases of diarrhea and 43 age-matched controls, 0 to 5 years old, were studied in a pediatric outpatient unit from a poor peri urban area of Porto Velho, Rondônia. Eighty percent of diarrheal cases were observed in the groups under 2 years of age. Rotavirus (19.2%) was the most frequent enteropathogen associated with diarrhea, followed by Shigella flexneri (6.15%) and S. sonnei (1.5%) and Salmonella sp. (6.9%). Four cases of E. coli enterotoxigenic infections (3.1%), E. coli enteropathogenic (EPEC)(2.3%) one case of E. coli enteroinvasive infection (0.8%) and one case of Yersinia enterocolitica (0.8%) were also identified. Mixed infections were frequent, associating rotavirus, EPEC and Salmonella sp. with Entamoeba histolytica and Giardia lamblia.

The antimicrobial activity of three different extracts (hexanic, ethyl acetate, methanol) obtained from Brazilian Drosera species (D. communis, D. montana var. montana, D. brevifolia, D. villosa var. graomogolensis, D. villosa var. villosa, Drosera sp. 1, and Drosera sp. 2 ) were tested against Staphylococcus aureus (ATCC 25923), Enterococcus faecium (ATCC23212), Pseudomonas aeruginosa (ATCC27853), Escherichia coli (ATCC11229), Salmonella choleraesuis (ATCC10708), Klebsiella pneumoniae (ATCC13883), and Candida albicans (a human isolate). Better antimicrobial activity was observed with D. communis and D. montana var. montana ethyl acetate extracts. Phytochemical analyses from D. communis, D. montana var. montana and D. brevifolia yielded 5-hydroxy-2-methyl-1,4-naphthoquinone (plumbagin); long chain aliphatic hydrocarbons were isolated from D. communis and from D. villosa var. villosa, a mixture of long chain aliphatic alcohols and carboxylic acids, was isolated from D. communis and 3b-O-acetylaleuritolic acid from D. villosa var. villosa.

Strains of enterotoxigenic Escherichia coli (ETEC) are responsible for significant rates of morbidity and mortality among children, particularly in developing countries. The majority of clinical and public health laboratories are capable of isolating and identifying Salmonella, Shigella, Campylobacter, and Escherichia coli O157:H7 from stool samples, but ETEC cannot be identified by routine methods. The method most often used to identify ETEC is polymerase chain reaction for heat-stable and heat-labile enterotoxin genes, and subsequent serotyping, but most clinical and public health laboratories do not have the capacity or resources to perform these tests. In this study, polyclonal rabbit and monoclonal mouse IgG2b antibodies against ETEC heat-labile toxin-I (LT) were characterized and the potential applicability of a capture assay was analyzed. IgG-enriched fractions from rabbit polyclonal and the IgG2b monoclonal antibodies recognized LT in a conformational shape and they were excellent tools for detection of LT-producing strains. These findings indicate that the capture immunoassay could be used as a diagnostic assay of ETEC LT-producing strains in routine diagnosis and in epidemiological studies of diarrhea in developing countries as enzyme linked immunosorbent assay techniques remain as effective and economical choice for the detection of specific pathogen antigens in cultures.

Acute schistosomiasis is a systemic hypersensitivity reaction against the migrating schistosomula and eggs. A variety of clinical manifestations appear during the migration of schistosomes in humans: cercarial dermatitis, fever, pneumonia, diarrhoea, hepatomegaly, splenomegaly, skin lesions, liver abscesses, brain tumours and myeloradiculopathy. Hypereosinophilia is common and aids diagnosis. The disease has been overlooked, misdiagnosed, underestimated and underreported in endemic areas, but risk groups are well known, including military recruits, some religious congregations, rural tourists and people practicing recreational water sports. Serology may help in diagnosis, but the finding of necrotic-exudative granulomata in a liver biopsy specimen is pathognomonic. Differentials include malaria, tuberculosis, typhoid fever, kala-azar, prolonged Salmonella bacteraemia, lymphoma, toxocariasis, liver abscesses and fever of undetermined origin. For symptomatic hospitalised patients, treatment with steroids and schistosomicides is recommended. Treatment is curative in those timely diagnosed.

Neuropathy and bone deformities, lifelong sequelae of leprosy that persist after treatment, result in significant impairment to patients and compromise their social rehabilitation. Phosphate-regulating gene with homologies to endopeptidase on the X chromosome (PHEX) is a Zn-metalloendopeptidase, which is abundantly expressed in osteoblasts and many other cell types, such as Schwann cells, and has been implicated in phosphate metabolism and X-linked rickets. Here, we demonstrate that Mycobacterium leprae stimulation downregulates PHEX transcription and protein expression in a human schwannoma cell line (ST88-14) and human osteoblast lineage. Modulation of PHEX expression was observed to a lesser extent in cells stimulated with other species of mycobacteria, but was not observed in cultures treated with latex beads or with the facultative intracellular bacterium Salmonella typhimurium. Direct downregulation of PHEX by M. leprae could be involved in the bone resorption observed in leprosy patients. This is the first report to describe PHEX modulation by an infectious agent.

Aiming to identify new sources of bioactive secondary metabolites, we isolated 82 endophytic fungi from stems and barks of the native Brazilian tree Caesalpinia echinata Lam. (Fabaceae). We tested their ethyl acetate extracts in several in vitro assays. The organic extracts from three isolates showed antibacterial activity against Staphylococcus aureus and Escherichia coli [minimal inhibitory concentration (MIC) 32-64 μg/mL]. One isolate inhibited the growth of Salmonella typhimurium (MIC 64 μg/mL) and two isolates inhibited the growth of Klebsiella oxytoca (MIC 64 μg/mL), Candida albicans and Candida tropicalis (MIC 64-128 μg/mL). Fourteen extracts at a concentration of 20 μg/mL showed antitumour activities against human breast cancer and human renal cancer cells, while two isolates showed anti-tumour activities against human melanoma cancer cells. Six extracts were able to reduce the proliferation of human peripheral blood mononuclear cells, indicating some degree of selective toxicity. Four isolates were able to inhibit Leishmania (Leishmania) amazonensis and one isolate inhibited Trypanosoma cruzi by at least 40% at 20 μg/mL. The trypanocidal extract obtained from Fusarium sp. [KF611679] culture was subjected to bioguided fractionation, which revealed beauvericin as the compound responsible for the observed toxicity of Fusarium sp. to T. cruzi. This depsipeptide showed a half maximal inhibitory concentration of 1.9 μg/mL (2.43 μM) in a T. cruzi cellular culture assay.

Nitroimidazoles exhibit high microbicidal activity, but mutagenic, genotoxic and cytotoxic properties have been attributed to the presence of the nitro group. However, we synthesised nitroimidazoles with activity against the trypomastigotes of Trypanosoma cruzi, but that were not genotoxic. Herein, nitroimidazoles (11-19) bearing different substituent groups were investigated for their potential induction of genotoxicity (comet assay) and mutagenicity (Salmonella/Microsome assay) and the correlations of these effects with their trypanocidal effect and with megazol were investigated. The compounds were designed to analyse the role played by the position of the nitro group in the imidazole nucleus (C-4 or C-5) and the presence of oxidisable groups at N-1 as an anion receptor group and the role of a methyl group at C-2. Nitroimidazoles bearing NO2 at C-4 and CH3 at C-2 were not genotoxic compared to those bearing NO2 at C-5. However, when there was a CH3 at C-2, the position of the NO2 group had no influence on the genotoxic activity. Fluorinated compounds exhibited higher genotoxicity regardless of the presence of CH3 at C-2 or NO2 at C-4 or C-5. However, in compounds 11 (2-CH3; 4-NO2; N-CH2OHCH2Cl) and 12 (2-CH3; 4-NO2; N-CH2OHCH2F), the fluorine atom had no influence on genotoxicity. This study contributes to the future search for new and safer prototypes and provide.

O objetivo deste trabalho foi avaliar aspectos fisiológicos e microbiológicos de beterrabas minimamente processadas. A taxa respiratória e a produção de etileno foram avaliadas até quatro horas após o processamento e durante 10 dias de armazenamento a 5ºC. As análises microbiológicas foram realizadas no dia do processamento e após 10 dias de conservação. Raízes intactas apresentaram a menor taxa respiratória (5 mL CO2 kg-1 h-1), enquanto beterrabas raladas e descascadas obtiveram taxa respiratória de 30 mL CO2 kg-1 h-1 após quatro horas do processamento. Beterrabas minimamente processadas apresentaram um pico respiratório no segundo dia de armazenamento (80 mL CO2 kg-1 h-1), estabilizando em 30 mL CO2 kg-1 h-1 no quarto dia. Não foi detectado etileno em beterrabas intactas e descascadas ao passo que em beterrabas minimamente processadas, a produção de etileno nas primeiras horas após o corte atingiu 0,90 µL kg-1 h-1, registrando-se pouca variação ao longo das quatro horas de avaliação e durante o período de armazenamento a 5ºC. As contagens de bactérias psicrotróficas e coliformes totais mantiveram-se dentro dos limites aceitáveis, e não foi detectada presença de coliformes fecais e Salmonella.

Resumo: O objetivo deste trabalho foi avaliar o efeito de temperaturas de desidratação por atomização sobre as características microbiológicas, físicas e químicas de gemas de ovos em pó e sobre o rendimento do processo. A desidratação por atomização foi realizada a 90, 120 e 150°C, com cinco repetições para cada tratamento. O rendimento foi avaliado pela relação entre a quantidade de gema em pó obtida e a quantidade de gema in natura utilizada na secagem. As gemas desidratadas foram analisadas quanto à composição centesimal, à cor objetiva e à rancidez. Para as análises microbiológicas, foi detectada a presença de estafilococos coagulase-positiva, pela contagem direta em placas; Salmonella spp., em amostra de 25 g; e coliformes, a 45°C. A temperatura de secagem por atomização influenciou a umidade das gemas em pó, sem interferir nos teores de proteínas, lipídeos e cinzas, nas características microbiológicas ou na rancidez dos produtos finais. As temperaturas de secagem mais elevadas proporcionam maior rendimento de produto, mas, a 150°C, ocorre escurecimento e diminuição na intensidade da coloração amarela das gemas em pó.

O processamento mínimo tem sido descrito como a manipulação, o preparo, embalagem e a distribuição de produtos agrícolas, através de procedimentos como seleção, limpeza, lavagem, descascamento e corte, que não afetem suas características organolépticas, agregando valor aos mesmos. Vários fatores limitam o consumo destes produtos, os quais estão envolvidos na sua vida útil, tais como: aumento da respiração e da produção de etileno, escurecimento enzimático, descoloração da superfície, perda de água e, sobretudo, alterações microbiológicas. No presente trabalho, o enfoque foi dado a este ultimo fator. Foram analisadas 20 amostras de goiaba, manga, melão japonês, mamão formosa e abacaxi minimamente processados e comercializados em supermercados da cidade de Fortaleza-CE, considerando-se a quantificação de coliformes totais, fecais, bolores e leveduras, e detecção de bactérias potencialmente patogênicas, tais como Salmonella sp e Staphylococcus coagulase positivo. Através dos resultados deste trabalho, permitiu-se sugerir a necessidade de controle de qualidade, observando as boas práticas de fabricação, para posterior implementação de um sistema preventivo de APPCC, análise de perigos e pontos críticos de controle, para se assegurar um produto saudável e seguro para o consumidor.

Objetivou-se desenvolver geleia tradicional de umbu-cajá, caracterizá-la quanto a parâmetros físicos, químicos, microbiológicos e sensoriais, e avaliar sua estabilidade durante o armazenamento por seis meses em condições ambientais. Para processamento, foram utilizados 44% de polpa diluída de umbu-cajá, 1% de pectina de alto teor de metoxilação (ATM) e 55% de açúcar cristal. A formulação foi submetida à cocção em tacho aberto de aço inoxidável até teor de sólidos solúveis totais de cerca de 68 ªBrix. A geleia foi envasada em recipientes de vidros transparentes, caracterizada e estocada em temperatura e umidade relativa médias de 23,25 ºC e 81%, respectivamente, com acompanhamento por meio de análises físicas e químicas a cada 30 dias de armazenamento. Os resultados da caracterização química evidenciaram produto com elevado teor de carboidratos, baixos conteúdos de cinzas e proteínas e valor calórico de 256 kcal/100g. Não foi verificado contagem dos microorganismos pesquisados (bolores e leveduras, coliformes totais e termotolerantes, Staphylococcus, bactérias mesófilas e Salmonella). Constatou-se alta aceitabilidade, com índices de aceitação superiores a 70% para todos os atributos sensoriais investigados (cor, aparência, aroma, consistência, sabor, doçura e impressão global) e intenção de compra de 67,5%, indicando potencial para industrialização e comercialização. O armazenamento promoveu aumento significativo nos valores de pH, sólidos solúveis totais (SST), relação SST/ATT e firmeza e reduções significativas na acidez total titulável (ATT), atividade de água, luminosidade, intensidades de vermelho e amarelo, croma, ângulo de tonalidade, extrusão e adesividade. Constatou-se tendência à estabilidade dos valores de umidade e de sólidos totais. O processamento do umbu-cajá para elaboração de geleia mostrou-se viável, apresentando-se como mais uma opção de renda para pequenos produtores do semiárido brasileiro.

This work is a literature review of PBTAs, the phenylbenzotriazoles generated from the reduction and chlorination of azo dyes. The PBTAs and the non- chlorinated PBTAs were isolated for the first time from river blue rayon organic extracts that showed high mutagenic activity in the Salmonella/microsome genotoxicity assay. To date, 8 PBTAs have been identified and beside their mutagenic activity in bacteria they cause genotoxic effects in fish and mammalian cell cultures. Due to the large number of textile dyeing facilities in Brazil, studies to determine them in the aquatic environment seem to be relevant.

Nitrite, which is present in preserved meat and can be produced in the oral cavity by reduction of nitrate taken from vegetables, could react in stomach with nitrosatable drugs, giving genotoxic-carcinogenic N-nitroso compounds (NOC). The mutagenicity of reaction mixtures formed by sodium nitrite and selected sulfa-drugs (sulfathiazole, HST; phtalylsulfathiazole, PhST; complex Co(II)-sulfathiazole, Co(II)-ST) in acidic medium was evaluated using the Salmonella typhimurium reverse mutation assay (Ames test), with TA98 and TA 100 strains. The reactions were carried out at room temperature, with a mole ratio [nitrite]/[sulfa-drug] > 1. The three reaction mixtures showed mutagenic effects in the considered range.
