Stenotrophomonas maltophilia is a multidrug-resistant nosocomial pathogen that is difficult to identify unequivocally using current methods. Accordingly, because the presence of this microorganism in a patient may directly determine the antimicrobial treatment, conventional polymerase chain reaction (PCR) and real-time PCR assays targeting 23S rRNA were developed for the specific identification of S. maltophilia. The PCR protocol showed high specificity when tested against other species of Stenotrophomonas, non-fermentative Gram-negative bacilli and 100 clinical isolates of S. maltophilia previously identified using the Vitek system.

Schistosomiasis is an endemic parasite disease and praziquantel is the only drug currently in use to control this disease. Experimental and epidemiological evidence strongly suggests that Microtus fortis( Mf) is a naturally resistant vertebrate host of Schistosoma japonicum. In the present study, we found that Mfserum albumin ( Mf-albumin) and the conditioned medium of pcDNA3.1- Mf-albumin caused 46.2% and 38.7% schistosomula death rates in 96 h, respectively, which were significantly higher than that of the negative control (p < 0.05). We also found that mice injected with Mf-albumin had a 43.5% reduction in worm burden and a 48.1% reduction in liver eggs per gram (p < 0.05) in comparison to the control animals. To characterise the mechanisms involved in clearance, schistosomula were incubated with fluorescein isothiocyanate-labelled Mf-albumin and fluorescent enrichment effects were found in the gut lumen of schistosomula after 48 h of incubation. Next, digestive tract excretions from schistosomula were collected and the sensitivity of Mf-albumin to digestive tract excretions was evaluated. The results indicated that schistosomula digestive tract excretions showed indigestibility of Mf-albumin. The death of schistosomula could be partially attributed to the lack of digestion of Mf-albumin by digestive tract excretions during the development of the schistosomula stage. Therefore, these data indicate the potential of Mf-albumin as one of the major selective forces for schistosomiasis.

The Firmicutes bacteria participate extensively in virulence and pathological processes. Enterococcus faecalis is a commensal microorganism; however, it is also a pathogenic bacterium mainly associated with nosocomial infections in immunocompromised patients. Iron-sulfur [Fe-S] clusters are inorganic prosthetic groups involved in diverse biological processes, whose in vivo formation requires several specific protein machineries. Escherichia coli is one of the most frequently studied microorganisms regarding [Fe-S] cluster biogenesis and encodes the iron-sulfur cluster and sulfur assimilation systems. In Firmicutes species, a unique operon composed of the sufCDSUB genes is responsible for [Fe-S] cluster biogenesis. The aim of this study was to investigate the potential of the E. faecalis sufCDSUB system in the [Fe-S] cluster assembly using oxidative stress and iron depletion as adverse growth conditions. Quantitative real-time polymerase chain reaction demonstrated, for the first time, that Gram-positive bacteria possess an OxyR component responsive to oxidative stress conditions, as fully described for E. coli models. Likewise, strong expression of the sufCDSUB genes was observed in low concentrations of hydrogen peroxide, indicating that the lowest concentration of oxygen free radicals inside cells, known to be highly damaging to [Fe-S] clusters, is sufficient to trigger the transcriptional machinery for prompt replacement of [Fe-S] clusters.

Bacillus thuringiensisis a ubiquitous Gram-positive and sporulating bacterium. Its crystals and secreted toxins are useful tools against larvae of diverse insect orders and, as a consequence, an alternative to recalcitrant chemical insecticides. We report here the draft genome sequence ofB. thuringiensis147, a strain isolated from Brazil and with high insecticidal activity. The assembled genome contained 6,167,994 bp and was distributed in seven replicons (a chromosome and 6 plasmids). We identified 12 coding regions, located in two plasmids, which encode insecticidal proteins.

Cystic fibrosis (CF) patients with Burkholderia cepacia complex (Bcc) pulmonary infections have high morbidity and mortality. The aim of this study was to compare different methods for identification of Bcc species isolated from paediatric CF patients. Oropharyngeal swabs from children with CF were used to obtain isolates of Bcc samples to evaluate six different tests for strain identification. Conventional (CPT) and automatised (APT) phenotypic tests, polymerase chain reaction (PCR)-recA, restriction fragment length polymorphism-recA, recAsequencing, and matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) were applied. Bacterial isolates were also tested for antimicrobial susceptibility. PCR-recA analysis showed that 36 out of the 54 isolates were Bcc. Kappa index data indicated almost perfect agreement between CPT and APT, CPT and PCR-recA, and APT and PCR-recA to identify Bcc, and MALDI-TOF and recAsequencing to identify Bcc species. The recAsequencing data and the MALDI-TOF data agreed in 97.2% of the isolates. Based on recA sequencing, the most common species identified were Burkholderia cenocepacia IIIA (33.4%),Burkholderia vietnamiensis (30.6%), B. cenocepaciaIIIB (27.8%), Burkholderia multivorans (5.5%), and B. cepacia (2.7%). MALDI-TOF proved to be a useful tool for identification of Bcc species obtained from CF patients, although it was not able to identify B. cenocepacia subtypes.

Acinetobacter baumannii, a strictly aerobic, non-fermentative, Gram-negative coccobacillary rod-shaped bacterium, is an opportunistic pathogen in humans. We recently isolated a multidrug-resistant A. baumannii strain KBN10P02143 from the pus sample drawn from a surgical patient in South Korea. We report the complete genome of this strain, which consists of 4,139,396 bp (G + C content, 39.08%) with 3,868 protein-coding genes, 73 tRNAs and six rRNA operons. Identification of the genes related to multidrug resistance from this genome and the discovery of a novel conjugative plasmid will increase our understanding of the pathogenicity associated with this species.

Realizou-se estudo retrospectivo do registro de 69 óbitos ocorridos em hospital pediátrico em 1993 para identificar a relação da infecção hospitalar com o óbito. As principais infecções diagnosticadas foram as pneumonias e infecções de corrente sangüínea com um predomínio de bactérias gram-negativas. Em 30,4% das crianças, a infecção hospitalar foi causa direta do óbito e em 50,8% foi contribuinte. A infecção hospitalar foi mais importante como causa de óbito nos pacientes com afecção classificada como não fatal à admissão.

Kits para aerossol são artigos utilizados na terapêutica de afecções do trato respiratório e requerem no mínimo desinfecção de nível intermediário para reuso. Os objetivos deste estudo foram verificar uma possível contaminação microbiana em kits para aerossol pós-reprocessamento e identificar os microrganismos isolados. Estudo transversal, experimental realizado na unidade pediátrica de um hospital em Goiânia-GO. Coletaram-se amostras de três segmentos (máscara, copo, interior da extensão) de 15 kits previamente desinfetados, que foram semeadas em diferentes meios de culturas e os microrganismos isolados foram identificados por provas bioquímicas. Dos 15 kits analisados, 13 copos, nove extensões e 13 máscaras estavam contaminados. Isolou-se no total 101 UFC, 39 provindos dos copos, 20 das extensões e 42 das máscaras. Dentre os patógenos isolados destaca-se: Staphylococcus coagulase positivo, Staphylococcus coagulase negativo, Bastonetes Gram negativo fermentadores, Bastonetes Gram negativo não fermentadores, micrococose leveduras. A detecção microbiana indica prováveis falhas no reprocessamento desses artigos.

The fall armyworm, Spodoptera frugiperda, is one of the major field pests for maize production. It is mainly controlled by means of synthetic, and more recently by resistant cultivar of maize expressing Bt toxins. The neem tree, Azadirachta indica, is a plant that can potentially control insects with the advantage of being food and environmental safe. The aim of this study was to assess the effect of neem oil on the development and survival of S. frugiperda caterpillars by assessing histological alterations caused on their midgut. Newly hatched caterpillars were submitted to three neem oil concentrations: 0.006; 0.05; 0.4%, which were added to their artificial diet. Ten 3rd instar caterpillars, taken from each treatment, were submitted to histological analysis. The alimentary canals from the specimens were fixed in Baker for 12 hours, desiccated and diaphanized in alcohol/xylol (1:1) and xylol. After placing the samples in paraffin, they were sliced in 8 µm sections and stained with hematoxylin-eosin stain. The neem oil added to the diet of S. frugiperda caused total mortality at dose of 0.4% whilst still in the first instars, prolonged the larval and pupal stages, and reduced the pupal weight. Histo-physiological alterations such as degeneration of the epithelial lining of the midgut and in the peritrophic matrix were found at all concentrations of neem oil.

As gramíneas, usadas comumente para reabilitação de áreas degradadas, podem formar associações em suas raízes com bactérias fixadoras de nitrogênio e, assim, contribuir para a sustentabilidade do ecossistema. Por outro lado, a diversidade microbiana exerce papel relevante na resiliência dos processos biológicos do solo, que incluem a fixação biológica de N2. O presente estudo teve como objetivo a caracterização fenotípica de 72 isolados de bactérias diazotróficas associativas Gram-negativas, oriundos de áreas sob diferentes estratégias de reabilitação, após a mineração da bauxita em Poços de Caldas (MG), por meio da inoculação de amostras de solo nos meios de cultivo NFb, Fam e JNFb. Estirpes tipo e referência de espécies de Azospirillum, Herbaspirillum e Burkholderia foram usadas para comparação, uma vez que são capazes de crescer nestes meios. O dendrograma de similaridade baseado em sete características culturais dos isolados em meio de cultivo GNA revelou grande diversidade, sendo obtidos 50 grupos a 81 % de similaridade. A tolerância a NaCl em meio de cultivo batata/sacarose/ácido málico variou de 0 a 50 g L-1, permitindo separar os isolados e estirpes tipo em cinco grupos. O diâmetro celular variou de 0,61 a 1,21 µm, e 13 isolados diferiram das estirpes tipo e referência. Os padrões de proteína total, obtidos por eletroforese em gel de poliacrilamida (SDS-PAGE), foram bastante diversos, possibilitando a formação de 15 grupos com similaridade a 75 %. Não houve relação entre os grupos formados pelas várias características estudadas, tampouco destes com as áreas de origem dos isolados. Os meios de cultivo Fam e JNFb detectaram, além das espécies-alvo, outras não identificadas. A alta dissimilaridade da maioria dos isolados em relação a estirpes tipo, principalmente com relação aos padrões eletroforéticos de proteína total, admite a possibilidade da presença de novas espécies entre eles.

Two bacterial strains that amplified part of the nifH gene, RP1p and RP2p, belonging to the genus Enterobacter and Serratia, were isolated from the rhizoplane of Lupinus albescens. These bacteria are Gram-negative, rod-shaped, motile, facultative anaerobic, and fast-growing; the colonies reach diameters of 3-4 mm within 24 h of incubation at 28 ºC. The bacteria were also able to grow at temperatures as high as 40 ºC, in the presence of high (2-3 % w/v) NaCl concentrations and pH 4 -10. Strain RP1p was able to utilize 10 of 14 C sources, while RP2p utilized nine. The isolates produced siderophores and indolic compounds, but none of them was able to solubilize phosphate. Inoculation of L. albescens with RP1p and RP2p strains resulted in a significant increase in plant dry matter, indicating the plant-growth-promoting abilities of these bacteria.

Diversos relatos evidenciam os benefícios de procariotos fixadores de nitrogênio atmosférico no crescimento e na nutrição de muitas espécies vegetais; entretanto, não há, até o momento, nenhum trabalho visando à prospecção desses microrganismos na rizosfera da seringueira (Hevea brasiliensis). Assim, os objetivos deste trabalho foram verificar a ocorrência de bactérias diazotróficas em solos sob plantio de seringueira, assim como em suas raízes, e isolar e caracterizar essas bactérias. Para essa finalidade, coletaram-se amostras de solo e de raízes finas de seringueiras cultivadas no Campus Experimental da Universidade Federal de Lavras (Lavras, MG) para inoculação em meios de cultura semissólidos sem N na forma combinada, de modo a favorecer o crescimento de algumas espécies de bactérias diazotróficas. Foram obtidos 19 isolados nas amostras de solo, e não houve crescimento de bactérias fixadoras de nitrogênio nas culturas com amostras de raízes. A caracterização celular e das colônias desses isolados indicou que 17 deles produzem grande quantidade de exopolissacarídeo elástico, algumas vezes cartilaginoso. Eles são todos Gram-negativos, com formato celular de bastonete, imóveis e com dois glóbulos de poli-β-hidroxibutirato (PBH), um em cada extremidade do bastonete. O sequenciamento do 16S rDNA e sua análise filogenética confirmaram que isolados representativos desse grupo pertencem ao gênero Beijerinckia (B. indica e B. derxii) e que os outros dois isolados Gram-positivos pertencem ao gênero Bacillus. A presença da nitrogenase - a enzima responsável pela fixação biológica do nitrogênio atmosférico (FBN) - foi confirmada por meio da técnica de redução do acetileno. Conclui-se que, no solo sob plantio de seringueira, houve predominância de diazotróficas de vida livre pertencentes ao gênero Beijerinckia (B. indica e B. derxii), não havendo indícios de bactérias endofiticas ou rizosféricas.

Sistemas conservacionistas em pomares de laranja podem aumentar a capacidade de suporte de carga do solo minimizando os seus riscos de compactação. O objetivo deste trabalho foi avaliar a capacidade de suporte de carga por meio da pressão de preconsolidação e sua dependência a conteúdo de água, densidade e carbono orgânico de um Argissolo Vermelho distrófico latossólico, após 18 anos de implantação de plantas de cobertura permanentes nas entrelinhas de um pomar de laranja. O delineamento experimental foi de blocos ao acaso, com três repetições. Foram estudados três tratamentos de manejo por meio de roçadas nas entrelinhas do pomar, desde o plantio do pomar em 1993: gramínea Paspalum notatum, leguminosa Arachis pintoi e vegetação espontânea. Cento e oito amostras indeformadas de solo foram coletadas em 2011 sob o rodado e entrerrodado das máquinas nas estrelinhas do pomar com textura arenosa nas camadas de 0,00-0,10 m (87 g kg-1 de argila) e 0,10-0,20 m (122 g kg-1 de argila). Determinaram-se as pressões de preconsolidação das curvas de compressão do solo (25, 50, 100, 200, 400, 800 e 1.600 kPa) em conteúdos de água sob três potenciais (-80, -330 e -1.000 hPa), a densidade do solo e o teor de carbono orgânico do solo. A pressão de preconsolidação não foi dependente do conteúdo de água, da densidade do solo e do teor de carbono orgânico do solo. A pressão de preconsolidação sob o rodado na camada de 0,00-0,10 m foi menor nos tratamentos leguminosa e vegetação espontânea. A manutenção permanente da gramínea manejada com roçadas nas entrelinhas do pomar de laranja proporcionou maior capacidade de suporte de carga do solo sob o rodado na camada superficial arenosa.

The objective of this work was to evaluate the effects of fire regimes and vegetation cover on the structure and dynamics of soil microbial communities, through phospholipid fatty acid (PLFA) analysis. Comparisons were made between native areas with different woody covers ("cerrado stricto sensu" and "campo sujo"), under different fire regimes, and a 20-year-old active palisadegrass pasture in the Central Plateau of Brazil. Microbial biomass was higher in the native plots than in the pasture, and the highest monthly values were observed during the rainy season in the native plots. No significant differences were observed between fire regimes or between communities from the two native vegetation types. However, the principal component (PC) analysis separated the microbial communities by vegetation cover (native x pasture) and season (wet x dry), accounting for 45.8% (PC1 and PC3) and 25.6% (PC2 and PC3), respectively, of the total PLFA variability. Changes in land cover and seasonal rainfall in Cerrado ecosystems have significant effects on the total density of soil microorganisms and on the abundance of microbial groups, especially Gram-negative and Gram-positive bacteria.

The objective of this work was to isolate strains of lactic acid bacteria with probiotic potential from the digestive tract of marine shrimp (Litopenaeus vannamei), and to carry out in vitro selection based on multiple characters. The ideotype (ideal proposed strain) was defined by the highest averages for the traits maximum growth velocity, final count of viable cells, and inhibition halo against nine freshwater and marine pathogens, and by the lowest averages for the traits duplication time and resistance of strains to NaCl (1.5 and 3%), pH (6, 8, and 9), and biliary salts (5%). Mahalanobis distance (D²) was estimated among the evaluated strains, and the best ones were those with the shortest distances to the ideotype. Ten bacterial strains were isolated and biochemically identified as Lactobacillus plantarum (3), L. brevis (3), Weissella confusa (2), Lactococcus lactis (1), and L. delbrueckii (1). Lactobacillus plantarum strains showed a wide spectrum of action and the largest inhibition halos against pathogens, both Gram-positive and negative, high growth rate, and tolerance to all evaluated parameters. In relation to ideotype, L. plantarum showed the lowest Mahalanobis (D²) distance, followed by the strains of W. confusa, L. brevis, L. lactis, and L. delbrueckii. Among the analyzed bacterial strains, those of Lactobacillus plantarum have the greatest potential for use as a probiotic for marine shrimp.
