Neste trabalho, caracterizou-se a morfo-anatomia do caule e da folha de Brachiaria brizantha e B. humidicola, em três estratos, objetivando diferenciar tais estratos e espécies, bem como justificar, com estes parâmetros, a diferença de consumo que ocorre nessas espécies com o envelhecimento da planta. O experimento foi conduzido em casa de vegetação, utilizando-se vasos plásticos com areia esterilizada e recebendo solução nutritiva. Aos 70 dias após o corte de uniformização, cada planta foi dividida em três partes (estratos), coletando-se a folha mediana de cada estrato, separando-a em limbo e bainha foliar, e também o entrenó recoberto pela referida bainha. Para o estudo morfológico, foram mensurados a altura total das plantas, número total de folhas, nós e perfilhos, comprimento e largura da lâmina e da bainha foliar, comprimento e diâmetro do entrenó. Para a caracterização anatômica, as amostras foram fixadas em FAA, emblocadas em GMA, seccionadas em micrótomo e coradas com fucsina básica e azul de Astra. Os estudos morfo-anatômicos indicaram parâmetros que podem interferir na digestibilidade de seus tecidos. Verificou-se que as espécies apresentaram diferenças quanto aos aspectos morfológicos, destacando em B. humidicola menores valores de comprimento e largura do limbo, o que pode dificultar a seleção das folhas inferiores, interferindo no consumo dessa forrageira. Constatou-se, também, que o caule foi a fração que mais variou entre as espécies, apresentando B. brizantha diâmetro do entrenó maior e "parede" do colmo mais espessa, o que, além de tornar o caule mais resistente à apreensão, sugere maior número de feixes vasculares e, conseqüentemente, porcentagem de tecidos lignificados. Observaram-se estruturas secretoras na base dos tricomas da bainha foliar de B. brizantha, sugerindo cavidades secretoras, bastante raras na família Poaceae, não havendo estudos de sua interferência no consumo e digestibilidade.

O estudo foi realizado em um fragmento de mata ciliar (42 ha) onde a espécie arbórea, secundária inicial e decídua Centrolobium tomentosum Guill. ex Benth. é dominante no estrato superior da floresta estacional semidecidual. A espécie apresentou grande produção de serapilheira (4,2 kg.ind-1) com um comportamento bimodal de deposição (janeiro/fevereiro _ 25,1% e julho/agosto _ 52,3%), e grande capacidade de transferência de nutrientes para o solo (g.ind-1.ano-1): 74,7 N; 6,1 P; 29,0 K; 73,9 Ca; 25,8 Mg e 14,1 S. A decomposição da fração foliar da serapilheira apresentou padrão sazonal, resultando em cerca de 19,5 meses para seu desaparecimento. A qualidade e quantidade da serapilheira produzida por C. tomentosum indicam um alto potencial de utilização dessa espécie em programas de reabilitação funcional e estrutural das matas ciliares no Estado de São Paulo.

The aim of this study was to investigate the photosynthetic performance in populations of two legume tree species, Stryphnodendron adstringens (Mimosoideae), typical from Cerrado, and Cassia ferruginea (Caesalpinoideae) from the Atlantic Rain Forest. The photosynthetic traits were assessed by measures of chlorophyll fluorescence in progenies of naturally pollinated plants from three populations of S. adstringens and a population of C. ferruginea. Plants of S. adstringens growing under similar conditions of C. ferruginea plants demanded higher light values for photosynthesis saturation, 600 µmol.m-2.s-1 and 350 µmol.m-2.s-1 respectively, and showed higher intrinsic photosynthetic efficiency of photosystem II, Fv/Fm of 0.814 versus 0.783 in C. ferruginea. The highest values of Fv/Fm observed in S. adstringens can explain the highest electron transport rates (ETR) obtained for this species. No significant differences were found among progenies from different C. ferruginea trees nor among populations of S. adstringens, and only in few cases, variation among progenies within populations were found for S. adstringens plants. The fact that fluorescence parameters distinguished species but not populations or most of progenies may be related to low intraspecific genetic variation of these chlorophyll fluorescence traits or due to lack of expression on genetic differences in plants under no stressful conditions.

O subgênero Chevaliera está representado por 21 espécies, das quais grande parte ocorre no Brasil, habitando diferentes ecossistemas. Com o objetivo de auxiliar a taxonomia das espécies, são apresentados dados da anatomia foliar de 11 espécies do subgênero. Entre os caracteres mais relevantes destacam-se: posição dos estômatos, câmara subestomática com células espessadas, hipoderme, número de camadas do parênquima aqüífero, o formato das células dos canais de aeração e distribuição das fibras no mesofilo.

Cotyledon mesophyll cell morphology and lipid and protein synthesis of T. grandiflorum, T. subincanum and T. bicolor were analyzed and compared with T. cacao. These species possess foliar cotyledons folded around the hypocotyl radicle axis, typical of Sterculiaceae. Fruit size, morphology and weight are very distinct amongst the four species and so are the respective seeds. The main axis of the T. grandiflorum and T. bicolor seeds measured about 30 mm, while T. subincanum and T. cacao seeds measured 17 mm and 26 mm respectively. The seed weights of T. grandiflorum, T. bicolor, T. subincanum and T. cacao were 11.6 g, 9.4 g, 2.1 g and 3.0 g, respectively. The cotyledon mesophylls of the four species contained mainly polysaccharides and lipid-protein reserve cells. Theobroma cacao, T. grandiflorum and T. subincanum were composed of greater than 50% lipids. For the four species, lipid globules gradually accumulated adjacent to the cell wall, and these globules measured from 1 to 3 µm. TEM showed low-density proteins inside the central vacuole of the young mesophyll cells of T. cacao. The protein reserves of the mature cells were densely scattered amongst the lipid bodies, and a few starch granules occurred together with the cotyledon mesophyll of the four species. Polyphenolic cells were found throughout the mesophyll cells or aligned with the respective vascular bundles. Immature cells demonstrated the capacity to synthesize all these reserves, but gradually the pre-determined cells produced mainly lipid-protein reserves. Besides the unique characteristics of the T. cacao products, the lipid-protein synthesis capacities of T. grandiflorum, T. subincanum and T. bicolor suggest various possibilities for new industrialized food, pharmaceutical and cosmetic products.

Modelos matemáticos baseados no conceito de graus dia ("thermal-time") e Ψw dia ("hydrotime") podem ser usados para a elaboração de modelos mais gerais sobre a germinação e emergência de plântulas no campo, podendo ser uma importante ferramenta para estudos sobre a biologia de plantas daninhas e seu controle. Neste trabalho, avaliou-se a germinação de sementes de D. cordata em resposta ao potencial hídrico (Ψw), usando-se o modelo Ψw dia. Tanto a germinabilidade como a velocidade de germinação decresceram linearmente com a redução do Ψw, atingindo valores próximos a zero em -0.8 MPa. Em temperatura ótima, a taxa de queda na germinação foi maior em comparação com as temperaturas sub- e supra-ótima. O Ψw base (Ψwb) mediano foi similar entre as temperaturas sub-ótima (19 ºC) e supra-ótima (32 ºC), mas foi maior (menos negativo) à temperatura ótima (25 ºC), mostrando que sementes de D. cordata são menos sensíveis à redução do potencial hídrico à 19 ºC do que à 25 ºC. O Ψw dia foi maior para sementes germinadas à temperatura sub-ótima do que à temperatura ótima, mostrando que a velocidade de germinação num dado potencial hídrico é maior em temperatura ótima. A quantidade de Ψw dia necessária para a germinação foi maior em temperatura supra-ótima do que em temperatura ótima, e menor em temperatura supra-ótima do que em sub-ótima. Em geral, Ψw dia foi relativamente constante entre as diferentes sub-populações. O modelo de Ψw dia pode descrever bem o efeito do potencial hídrico sobre as curvas de germinação (porcentagem acumulada x tempo), especialmente às temperaturas sub-ótima e supra-ótima.

Estudo taxonômico do gênero Micrasterias C. Agardh ex Ralfs realizado em duas Áreas de Proteção Ambiental (APA Rio Capivara e APA Lagoas de Guarajuba), ambas situadas no Município de Camaçari, Estado da Bahia, nordeste do Brasil. Os materiais estudados, de origem do plâncton e do perifíton, provieram de 96 amostras coletadas no verão (dezembro de 2006 a março de 2007) e no inverno (maio a agosto de 2007), em ambientes lóticos e lênticos. Foram identificados 14 táxons, dos quais M.americana (Ehrenb.) Ralfs var. bahiensis I. B. Oliveira, C. E. M. Bicudo & C. W. N. Moura foi descrita como nova para a ciência e 10 constituem citacões pioneiras para a desmidioflórula da Bahia, quais sejam: M. alata G. C. Wall., M. arcuata Bailey var.subpinnatifida West & G. S. West f. subpinnatifida, M. borgei H. Krieg. var. borgei, M. foliacea Bailey ex Ralfs var. foliacea, M. furcata C. Agardh ex Ralfs var. furcata f. furcata, M. laticeps Nordst. var. laticeps, M. laticeps Nordst. var.acuminata H. Krieg., M. mahabuleshwarensis J. Hobson var.ampullacea (W. M. Maskell) Nordst., M. mahabuleshwarensis J. Hobson var. mahabuleshwarensis f. mahabuleshwarensis, M. pinnatifida (Kütz.) Ralfs var. pinnatifida f. pinnatifida, M. radiosa Ralfs var. elegantior (G. S. West) Croasdale, M. rotata (Grev.) Ralfs var. rotata e M. truncata (Corda) Bréb. ex Ralfs var. pusilla G. S. West. As espécies mais bem representadas na área estudada foram M. alata e M. pinnatifida presentes, respectivamente, em 50% e 45,83% das amostras analisadas. Micrasteriaslaticeps var.acuminata e M.americana var. bahiensis, foram as menos representadas, por terem sido registradas apenas em 2,08% e 1,04% respectivamente.

Os efeitos da concentração de ágar no crescimento de explantes e na formação de calos foram avaliados em culturas axênicas de gametófitos femininos de morfos de coloração verde e vermelha de Gracilaria domingensis (Kützing) Sonder ex Dickie. Culturas unialgáceas foram mantidas em água do mar esterilizada (30-32 ups) enriquecida com 25% da solução de von Stosch (VSES 25%), 22 ± 2 °C, fotoperíodo de 14 h, irradiância de 50-80 µmol de fótons m-2 s-1. Para a obtenção de explantes axênicos, segmentos apicais e intercalares dos dois morfos foram cultivados por 48 h em meio VSES 25% com adição de uma solução antibiótica e antifúngica, e submetidos a uma lavagem com uma solução de água do mar esterilizada com 0,5% de hipoclorito de sódio e 200 µL L-1 de detergente por 20 segundos. Para avaliar os efeitos da concentração de ágar, os segmentos axênicos foram inoculados em meio ASP 12-NTA com concentrações distintas de ágar que variaram de zero a 1%. A adição de ágar no meio inibiu o crescimento dos segmentos apicais de ambos os morfos, bem como o crescimento de segmentos intercalares do morfo verde. Observou-se uma tendência geral no crescimento dos explantes, onde a taxa de crescimento foi inversamente proporcional à concentração de ágar. A adição de ágar no meio induziu a formação de três tipos de calo, denominados conforme a região do explante onde se originaram: calo apical, calo basal e calo intermediário. As concentrações de 0,5% e 0,7% de ágar foram as concentrações ótimas para indução de calos basais e calos intermediários no morfo verde, respectivamente. A presença de ágar foi essencial para a formação de calos intermediários e apicais. Os resultados indicam que o ágar apresenta um papel na regulação dos processos morfogenéticos em morfos pigmentares de G. domingensis.

(In vitro culture at low temperature and ex vitro acclimatization of Vriesea inflata an ornamental bromeliad). In vitro culture by seeds is a technique for preservation of threatened species because it may provide a large number of plants with genetic diversity. The bromeliad Vriesea inflata (Wawra) Wawra, an ornamental bromeliad, is extensively and illegally collected from the nature and must be preserved. It is possible to form plant threatened collections in vitro by reducing the temperature of culture, while occupying little space, with the consequent reduction of maintenance costs. This work evaluated the influence of temperature on in vitro growth and morphology of plants of V. inflata, with the aim of establishing a slow growth-rate and analyzing the ex vitro acclimatization. Seeds were germinated in vitro in Murashige and Skoog (MS) medium, with macronutrients reduced to 50% (MS/2). After three months the plants were transferred to flasks of new same medium and kept in two germination chambers with the temperature adjusted to 15 °C and to 28 °C. After 24 months the plants were subject to biometric, photosynthetic pigments content and anatomical analyses. Results showed that plants maintained at 15 °C were smaller than those at 28 °C. Nevertheless, there were no alterations in pigments content, anatomy. In both treatments there was a survival rate of 100%. This work showed that plants of this species can be kept in vitro at 15 °C with the aim of forming a slow-growth collection, thereby seeking its preservation, and can be transferred to growth at ex vitro condition to achieved 100% survival rate.

The present work characterized and compared the anatomical structures of the leaves of Bactris gasipaes (Arecaceae) plants grown under different cultivation conditions (in vitro, ex vitro and in vivo) with the goal of identifying the origins of the difficulties encountered in acclimatizing micro-plants. The Quant program was used to determine leaf tissue thicknesses and areas, and histochemical tests were performed on leaf sections and analyzed using light microscopy. Stomatal and trichome densities were determined using the epidermal impression method and by scanning electronic microscopy. Our results indicated that there were no discernible alterations of the anatomical characteristics of the leaves of micro-plants cultivated under differing conditions and that the thickening of the mesophyll and the vascular fibers indicated adaptive responses to ex vitro conditions. As such, the observed difficulties in acclimatizing peach palm micro-plants to ex vitro conditions cannot be attributed to plant anatomical characteristics acquired during in vitro cultivation.

Cytomegalovirus (CMV) is the single most important infectious agent affecting recipients of organ transplants. To evaluate the incidence and the clinical importance of CMV infection in renal transplants in Brazil, 37 patients submitted to renal allograft transplants were tested periodically for the presence of cytomegalovirus DNA in urine using the polymerase chain reaction (PCR), and for the presence of IgM and IgG antibodies against CMV by enzyme-linked immunosorbent assay (ELISA) and indirect immunofluorescence (IIF). The PCR-amplified products were detected by gel electrophoresis and confirmed by dot-blot hybridization with oligonucleotide probes. Thirty-two of the 37 patients (86.4%) were positive by at least one of the three methods. In six patients, PCR was the only test which detected the probable CMV infection. Ten patients had a positive result by PCR before transplantation. In general, the diagnosis was achieved earlier by PCR than by serologic tests. Active infection occurred more frequently during the first four months after transplantation. Sixteen of the 32 patients (50%) with active CMV infection presented clinical symptoms consistent with CMV infection. Five patients without evidence of active CMV infection by the three tests had only minor clinical manifestations during follow-up. Our results indicate that PCR is a highly sensitive procedure for the early detection of CMV infection and that CMV infection in renal transplant patients is a frequent problem in Brazil.

To assess the clinical relevance of a semi-quantitative measurement of human cytomegalovirus (HCMV) DNA in renal transplant recipients within the typical clinical context of a developing country where virtually 100% of both receptors and donors are seropositive for this virus, we have undertaken HCMV DNA quantification using a simple, semi-quantitative, limiting dilution polymerase chain reaction (PCR). We evaluated this assay prospectively in 52 renal transplant patients from whom a total of 495 serial blood samples were collected. The samples scored HCMV positive by qualitative PCR had the levels of HCMV DNA determined by end-point dilution-PCR. All patients were HCMV DNA positive during the monitoring period and a diagnosis of symptomatic infection was made for 4 of 52 patients. In symptomatic patients the geometric mean of the highest level of HCMV DNAemia was 152,000 copies per 106 leukocytes, while for the asymptomatic group this value was 12,050. Symptomatic patients showed high, protracted HCMV DNA levels, whereas asymptomatic patients demonstrated intermittent low or moderate levels. Using a cut-off value of 100,000 copies per 106 leukocytes, the limiting dilution assay had sensitivity of 100%, specificity of 92%, a positive predictive value of 43% and a negative predictive value of 100% for HCMV disease. In this patient group, there was universal HCMV infection but relatively infrequent symptomatic HCMV disease. The two patient groups were readily distinguished by monitoring with the limiting dilution assay, an extremely simple technology immediately applicable in any clinical laboratory with PCR capability.

A prospective study of cytomegalovirus (CMV) infection was carried out on 34 renal transplant recipients managed at a General Hospital in Ribeirão Preto, SP, Brazil. Serologic tests showed that all patients were infected with CMV before renal transplantation. Two nested-PCR techniques with primers that recognize sequences of the glycoprotein B (gB) and H (gH) genes were used for CMV detection in blood and urine samples during the post-transplantation period. CMV was detected more frequently in blood samples than in urine samples (P<0.001). Thirty-three patients had CMV detected at least once in blood and/or urine samples. Seven of these patients (21.2%) were diagnosed as having symptomatic CMV infection and showed a worse clinical outcome, with a higher death rate (P = 0.03). No association between CMV viremia and graft rejection was observed. Nested-PCR was not useful to identify patients at risk for symptomatic CMV infection since only 21.2% of the patients with CMV infection were symptomatic.

Fungal infection is one of the most important causes of morbidity and mortality in bone marrow transplant (BMT) recipients. The growing incidence of these infections is related to several factors including prolonged granulocytopenia, use of broad-spectrum antibiotics, conditioning regimens, and use of immunosuppression to avoid graft-versus-host disease (GvHD). In the present series, we report five cases of invasive mold infections documented among 64 BMT recipients undergoing fluconazole antifungal prophylaxis: 1) A strain of Scedosporium prolificans was isolated from a skin lesion that developed on day +72 after BMT in a chronic myeloid leukemic patient. 2) Invasive pulmonary aspergillosis (Aspergillus fumigatus) was diagnosed on day +29 in a patient with a long period of hospitalization before being transplanted for severe aplastic anemia. 3) A tumoral lung lesion due to Rhizopus arrhizus (zygomycosis) was observed in a transplanted patient who presented severe chronic GvHD. 4) A tumoral lesion due to Aspergillus spp involving the 7th, 8th and 9th right ribs and local soft tissue was diagnosed in a BMT patient on day +110. 5) A patient with a history of Ph1-positive acute lymphocytic leukemia exhibited a cerebral lesion on day +477 after receiving a BMT during an episode of severe chronic GvHD. At that time, blood and spinal fluid cultures yielded Fusarium sp. Opportunistic infections due to fungi other than Candida spp are becoming a major problem among BMT patients receiving systemic antifungal prophylaxis with fluconazole.

We have determined the number of circulating T, B and natural killer cells in renal transplant recipients in order to detect changes during cytomegalovirus (CMV) infections. Serial blood samples were taken from 61 patients on standard triple immunosuppression therapy (cyclosporin A, azathioprine and prednisone). Using two-color flow cytometry analysis, the absolute number of CD3+, CD4+, CD8+, CD19+, CD3+HLA-DR+ and CD16+56+ cells was determined. Forty-eight patients (78.7%) developed active CMV infection, and all of them subsequently recovered. Twenty of the infected patients (32.8%) presented symptoms compatible with CMV disease during the infectious process. The number of lymphocytes and their main subpopulations were normal before the onset of CMV disease. During the disease there was a decrease followed by a significant increase (P<0.005) in the number of CD3+, CD4+, CD8+ and CD3+HLA-DR+ cells. No significant changes were observed in natural killer cells or B lymphocytes during the disease. We conclude, as observed in all viremic patients recovering from infection, that recovery is associated with an increase in the number of T cell subsets. The monitoring of different lymphocyte subsets along with antigenemia can be extremely useful in the detection of patients at high risk of developing CMV symptoms, allowing the early introduction of antiviral therapy or the reduction of immunosuppression therapy.
