In an article recently published in Química Nova, entitled "Chemistry Without Borders" ("Química Sem Fronteiras") [Pinto, A. C.; Zucco, C.; Galembeck, F.; Andrade, J. B.; Vieira, P. C. Quim. Nova 2012, 35, 2092], the authors highlighted the important aspects of science and technology with special emphasis on the field of Chemistry and its contributions toward a more prosperous Brazil of future. As a second step in that direction, this article extends the discussion of a key issue for the country in the framework of the chemistry community through the so called position papers in strategic areas. This document is a part of the contribution of the Brazilian Chemical Society to the World Science Forum to be held in Rio de Janeiro in November 2013. In this context, the present paper provides a brief discussion on neglected tropical diseases (NTDs) with emphasis on the current challenges and opportunities towards the development and evolution of the field. NTDs leads to illness, long-term disability or death, and has severe social, economic and psychological consequences for millions of men, women, and children worldwide. In most cases, the available treatments are inadequate and extremely limited in terms of efficacy and safety, leading to an urgent demand for new drugs. In addition to the traditional challenges involved in any drug discovery process, it is widely recognized that there is an innovation gap and a lack of investment for research and development (R&D) in the area of NTDs. In the last few decades, methods toward combating, eradication, prevention, and treatment of NTDs have been repeatedly emphasized in the major international agendas. Developments in these strategies and alliances have continued to have an essential impact, particularly in the area of drug discovery, both in Brazil and globally and should be encouraged and supported. Several examples of international activities dedicated to the reduction of the devastating global impact of NTDs can be provided. Despite the beneficial developments in the past 30 years, NTDs continue to devastate poor communities in remote and vulnerable areas, in large part, due to market failures and public policies. Recent studies have shown that among 756 new drugs approved between 2000 and 2011, only four new chemical entities (NCEs) were identified for the treatment of malaria, while none were developed against NTDs or tuberculosis. Furthermore, only 1.4% of approximately 150,000 clinical trials were registered for neglected diseases, with a smaller number of trials for NCEs. Establishment and strengthening of global strategies involving the triad "government-academia-industry" is fundamental to the success in R&D of new drugs for NTDs. National and international public-private initiatives that aim to create, encourage, and invest in R&D projects have been implemented and therefore are of utmost importance to successfully integrate Brazil into this new paradigm. It is essential to lay the foundation for mechanisms that will intensify investments in infrastructure, training, and qualification of personnel with an ultimate strategic vision that foresees continuity. Our research group has made significant contributions to the development of this field with the goal of forging new frontiers while tackling both current and future challenges that include indispensable elements such as innovation and integration.

A new flavonoid, catechin-3-O-(3"-O-trans-cinnamoyl)-α-rhamnopyranoside, along with known compounds, catechin-3-O-α-rhamnopyranoside, 3-oxo-urs-12-en-28-oic acid, 2,4,6-trimethoxybenzoic acid, 2-butyl-D-fructofuranoside and 1-butyl-D-fructofuranoside, has been isolated from the stem bark of V. thyrsoidea. These compounds were assayed for inhibition of protease activity (cathepsins B and K) and against cancer cell lines. Catechin-3-O-(3"-O-trans-cinnamoyl)-α-rhamnopyranoside showed moderate inhibitory activity (IC50 = 62.02 µM) against cathepsin B while 2-butyl-D-fructofuranoside was the most potent against a strain of CNS (SF-295) and human leukemia (HL-60) with IC50 = 36.80 µM and IC50 = 25.37 µM, respectively.

This paper describes the evaluation of simple and fast solubilization methods for the determination of Ca, Mg, and K in glycerin samples from biodiesel production by atomic spectrometry. The solubilization in water was compared with two other methods: solubilization in formic acid and solubilization in ethanol. Using solubilization in water, determination of the three analytes was possible; the values of limits of detection for Ca, K, Mg were 0.31, 0.06, and 0.16 mg kg−1, respectively. Because no adequate reference material was available, the accuracy was evaluated by assessing the recoveries tests with both solubilization methods; the evaluation ranged from 90% to 115%, with values of relative standard deviation >8%, indicating good accuracy of the measure. Four crude glycerin samples obtained from biodiesel plants of Rio Grande do Sul were analyzed after treatment with the different methods of solubilization, and the obtained results of Ca, Mg, and K concentration were in agreement with the values obtained from both solubilization methods. Therefore, solubilization in water is concluded to be a simpler, faster, and viable method for sample preparation of glycerin.

Samples of whole fluid pasteurized, UHT and powdered milk were analyzed for the presence of organophosphorus pesticide residues (OPR) using the QuEChERS method with detection by GC-FPD and confirmation by UPLC-MS/MS. A total of 23% of fluid milk samples and 27% of powdered milk samples contained at least one organophosphorus residue. The OPR found in the samples showed these can reach the milk and remain even after pasteurization, sterilization and drying processes. Some domestic sanitary residues were also present in the powdered milk samples. These residues have no LMR in Brazilian food legislation and are therefore not taken into account in food intake evaluations. There was an unidentified chromatographic peak in 30% of all the liquid and powdered milk samples, indicating the presence of some other contaminant.

The increase in incidence of charcoal rot caused by Macrophomina phaseolina on soybeans (Glycine max) was followed four seasons in conventional and no-till cropping systems. In the 1997/98 and 2000/01 seasons, total precipitation between sowing and harvest reached 876.3 and 846.9 mm, respectively. For these seasons, disease incidence did not differ significantly between the no-till and conventional systems. In 1998/99 and 1999/00 precipitation totaled 689.9 and 478.3 mm, respectively. In 1998/99, in the no-till system, the disease incidence was 43.7% and 53.1% in the conventional system. In 1999/00 the final incidence was 68.7% and 81.2% for the no-till and conventional systems, respectively. For these two seasons, precipitation was lower than that required for soybean crops (840 mm), and the averages of disease incidence were significantly higher in the conventional system. The concentration of microsclerotia in soil samples was higher in samples collected in conventional system at 0 - 10 cm depth. However, analysis of microsclerotia in roots showed that in years with adequate rain no difference was detected. In dry years, however, roots from plants developed under the conventional system had significantly more microsclerotia. Because of the wide host range of M. phaseolina and the long survival times of the microsclerotia, crop rotation would probably have little benefit in reducing charcoal rot. Under these study conditions it may be a better alternative to suppress charcoal rot by using the no-till cropping system to conserve soil moisture and reduce disease progress.

The effectiveness of fungicides in controlling white mold (Sclerotinia sclerotiorum) of dry beans (Phaseolus vulgaris) was evaluated when they were applied through irrigation water directly onto the plants or only to the soil. Two field trials were installed in April 1998 and April 1999 in Viçosa, MG. Trials were conducted as a (2 x 3) + 1 factorial: two fungicides x three application modes + one untreated control. The fungicides were benomyl (1.0 kg a.i. ha-1) and fluazinam (0.5 l a.i. ha-1). The three application modes were: (a) by backpack sprayer (667 l ha-1), (b) by garden watering-cans simulating sprinkler irrigation with 35,000 l ha-1 of water, and (c) by garden watering-cans applying water between the rows and near the soil surface in 35,000 l ha-1 of water. In 1998, fungicides were applied at 43 and 54 days after emergence (DAE); in 1999, at 47 and 61 DAE. Both fungicides were similarly effective on white mold control when applied by either chemigation or backpack sprayer, resulting in yields 21% higher than untreated control. Only fluazinam provided disease control when applications were made only in soil. Chemigation provided white mold control equivalent to that of backpack sprayer in terms of incidence, severity and number of diseased pods. Consequently, yield differences between these application methods were not significant.

Macrophomina phaseolina has been considered one of the most prevalent soybean (Glycine max) pathogens in Brazil. No genetic resistance has been determined in soybean and very little is known about the genetic diversity of this pathogen in tropical and sub-tropical regions. Fifty-five isolates from soybean roots were collected in different regions and analyzed through RAPD for genetic diversity. The UPGMA cluster analysis for 74 loci scored permitted identification of three divergent groups with an average similarity of 99%, 92% and 88%, respectively. The three groups corresponded to 5.45%, 59.95% and 34.6%, respectively of all isolates used. A single plant had three different haplotypes, while 10.9% of the analyzed plants had two different haplotypes. In another study the genetic similarity was evaluated among isolates from different hosts [soybean, sorghum (Sorghum bicolor), sunflower (Helianthus annuus), cowpea (Vigna unguiculata), corn (Zea mays) and wheat (Triticum aestivum)] as well as two soil samples from native areas. Results showed that more divergent isolates originated from areas with a single crop. Isolates from areas with crop rotation were less divergent, showing high similarity values and consequently formed the largest group. Amplification of the ITS region using primers ITS1 and ITS4 produced only one DNA fragment of 620 bp. None of the isolates were differentiated through PCR-RFLP. Our results demonstrated genetic variability among Brazilian isolates of M. phaseolina and showed that one single root can harbor more than one haplotype. Moreover, cultivation with crop rotation tends to induce less specialization of the pathogen isolates. Knowledge of this variation may be useful in screening soybean genotypes for resistance to charcoal rot.

No seu labirinto literário-filosófico, especialmente por meio da ideia de destino, Borges explora elementos chave que se tornaram parte da noção de crítica que enfatiza as ideias de contingência e da impossibilidade de controle deliberado dos efeitos da ação humana em seu curso temporal.

O trabalho teve o objetivo de investigar, isolada e integradamente, o efeito do corte polar do coco verde, a atmosfera modificada, e a associação de produtos químicos, sob condições de câmara frigorífica no controle da podridão basal pós-colheita causada por Lasiodiplodia theobromae. Frutos provenientes de áreas infestadas foram cortados transversalmente, removendo-se as brácteas e, em seguida, foram submetidos aos tratamentos com ceras e fungicidas. Após o tratamento, esses frutos foram embalados em caixas de papelão e transferidos para a câmara frigorífica à ±12º C por 30 ou 35 dias, de acordo com o ensaio. As avaliações foram realizadas a cada dois dias. A exclusão do fungo por meio do corte do mesocarpo na região das brácteas do coco verde foi eficiente no controle da doença, e a proteção com a cera EF-1 foi excelente protetor contra o dano pelo frio e fungos deteriorantes nos frutos cortados. Também, verificou-se que a associação corte basal do fruto + emulsão de cera + fungicida foi uma excelente alternativa de conservação e controle da doença em estudo.

Ensaios foram conduzidos para verificar a presença, o número de cópias e de sítios de integração de pAN7-1 no genoma de mutantes de M. grisea I-22 com patogenicidade alterada em arroz. Foram analisados T41, T93, T251 (gerados por mutagênese REMI) e T108 (oriundo de mutagênese convencional), os quais exibiram diferentes fenótipos mutantes. O DNA total desses mutantes foi submetido à reação em cadeia de polimerase (PCR) e às análises de hibridização com o vetor (Southern blot). A presença de pAN7-1 no genoma de todos os mutantes foi confirmada por PCR. Segundo as análises de Southern blot, T41 exibiu duas integrações do vetor, ambas na forma de cópia única. No genoma de T93 também foram detectados dois sítios de inserção de pAN7-1, um dos quais envolvendo múltiplas cópias do vetor. Os resultados indicaram a presença de apenas uma cópia do vetor em um único sítio nos genomas de T108 e T251. O padrão de integração em T251 foi o único a sugerir a ocorrência de evento REMI. As diferenças quanto ao tamanho dos fragmentos com homologia a pAN7-1 refletiram a possível aleatoriedade dos eventos de integração no genoma de M. grisea. Os resultados evidenciaram o potencial de REMI para a mutagênese insercional de M. grisea, quando conduzida com pAN7-1 e HindIII

Sugarcane (Saccharum spp.) cultivation is one of the major agricultural activities in the Brazilian states. This study aimed to molecularly identify the pathogen associated with rust in sugarcane cultivars in the state of Rio de Janeiro and to suggest a control strategy. Among the 14 PCR-tested cultivars, Puccinia kuehnii infection was identified for RB947520, RB92606, RB835486, RB72454, SP89-11I5, SP83-2847, both from infected leaf sample and from urediniospores. Puccinia kuehnii was not detected by PCR for the cultivars RB955971, RB951541, RB92579, RB867515, RB855536, SP91-1049, SP80-3280, SP80-1816. This is the first molecular detection of this fungus in the state of Rio de Janeiro for six of the 14 analyzed cultivars.

A utilização de gramíneas forrageiras em sistemas de integração lavoura-pecuária tem sido cada vez mais frequente no Brasil. Entretanto, alguns genótipos forrageiros podem hospedar fitonematoides, contribuindo para manter populações elevadas destes organismos no solo. Objetivando-se avaliar a reação de acessos e cultivares de Brachiaria spp. (B. ruziziensis, B. brizantha cvs. BRS Piatã e BRS Paiaguás, B. humidicola cv. BRS Tupi, e os genótipos B4 e HBGC 331) e Panicum maximum (cvs. Tanzânia-1 e Massai e os genótipos PM32, PM36, PM45 e PM46) à Pratylenchus brachyurus, realizou-se este trabalho. Como testemunhas utilizaram-se o milho BRS 2020 (suscetível) e milheto ADR 300 (resistente) à P. brachyurus. Os experimentos foram conduzidos em casa de vegetação, na Embrapa Gado de Corte, Campo Grande-MS, em blocos casualizados com sete repetições, de maio a setembro de 2011 e de março a maio de 2012. Utilizaram-se cinco plantas por vaso, que foram inoculadas com 1.000 espécimes de P. brachyurus. Após 90 dias, avaliaram-se as populações de nematoides na raiz e no solo, com a finalidade de obtenção da população final dos nematoides e o fator de reprodução (FR). Determinou-se ainda a reação dos genótipos em relação à porcentagem de redução do FR. Com exceção de B. humidicola cv. BRS Tupi, com FR de 0,98 e 0,44, no primeiro e segundo experimentos, respectivamente, todos os materiais avaliados permitiram a multiplicação do nematoide. Em relação à porcentagem de redução do FR, apenas a B. humidicola cv. BRS Tupi e o milheto ADR 300 foram classificados como moderadamente resistentes, com reduções de, no máximo 90,58% e 94,73%, no primeiro e segundo experimento, respectivamente. Entre os genótipos de forrageiras estudados a maioria mostrou-se suscetível à P. brachyurus, apesar de variação do grau de suscetibilidade entre os mesmos. Dessa forma, em áreas com histórico de presença de P. brachyurus, a B. humidicola cv. BRS Tupi pode ser indicada nos sistemas de integração e/ou em rotação de culturas, como estratégia de manejo para a redução populacional de P. brachyurus.

Alecrim da Chapada (Lippia gracilis Schauer) é considerada uma planta rica em óleo essencial e possui atividade antimicrobiana comprovada, devido aos monoterpenos fenólicos carvacrol e timol. O objetivo deste trabalho foi avaliar o efeito in vitro do óleo essencial e dos extratos etanólicos de raízes e folhas de L. gracilis no controle do fungo Monosporasccus cannonballus, causador do colapso do meloeiro. O extratos foram testados nas concentrações de 2500, 5000 e 7500 ppm e o óleo essencial de L. gracilis foi avaliado nas concentrações 255, 340 e 425 ppm tendo também um controle positivo com o fungicida comercial Captan(r) e um negativo apenas com meio BDA (batata, dextrose, ágar). Discos de micélio com 3 mm de diâmetro foram inoculados no centro de placas de Petri e as medições do crescimento micelial do fungo foram realizadas 48 horas após a montagem do experimento. Verificou-se que o óleo essencial de L. gracilis nas três concentrações testadas foi eficiente para o controle do fungo, com percentuais de inibição de 100%, em comparação ao tratamento com o controle negativo (0%) que não apresentou inibição. O extrato radicular, nas concentrações de 5000 e 7500 ppm e o extrato foliar na concentração de 7500 ppm também proporcionaram um percentual de inibição de 100%.
