Stem cell factor (SCF) is a major mast cell growth factor, which could be involved in the local increase of mast cell number in the asthmatic airways. In vivo, SCF expression increases in asthmatic patients and this is reversed after treatment with glucocorticoids. In vitro in human lung fibroblasts in culture, IL-1beta, a pro-inflammatory cytokine, confirms this increased SCF mRNA and protein expression implying the MAP kinases p38 and ERK1/2 very early post-treatment, and glucocorticoids confirm this decrease. Surprisingly, glucocorticoids potentiate the IL-1beta-enhanced SCF expression at short term treatment, implying increased SCF mRNA stability and SCF gene transcription rate. This potentiation involves p38 and ERK1/2. Transfection experiments with the SCF promoter including intron1 also confirm this increase and decrease of SCF expression by IL-1beta and glucocorticoids, and the potentiation by glucocorticoids of the IL-1beta-induced SCF expression. Deletion of the GRE or kappaB sites abolishes this potentiation, and the effect of IL-1beta or glucocorticoids alone. DNA binding of GR and NF-kappaB are also demonstrated for these effects. In conclusion, this review concerns new mechanisms of regulation of SCF expression in inflammation that could lead to potential therapeutic strategy allowing to control mast cell number in the asthmatic airways.

Proteinase-activated receptor-2 (PAR2) belongs to a novel subfamily of G-protein-coupled receptors with seven-transmembrane domains. This receptor is widely distributed throughout the body and seems to be importantly involved in inflammatory processes. PAR2 can be activated by serine proteases such as trypsin, mast cell tryptase, and bacterial proteases, such as gingipain produced by Porphyromonas gingivalis. This review describes the current stage of knowledge of the possible mechanisms that link PAR2 activation with periodontal disease, and proposes future therapeutic strategies to modulate the host response in the treatment of periodontitis.

The intestinal epithelium plays a crucial role in providing a barrier between the external environment and the internal milieu of the body. A compromised mucosal barrier is characteristic of mucosal inflammation and is a key determinant of the development of intestinal diseases such as Crohn's disease and ulcerative colitis. The intestinal epithelium is regularly exposed to serine proteinases and this exposure is enhanced in numerous disease states. Thus, it is important to understand how proteinase-activated receptors (PARs), which are activated by serine proteinases, can affect intestinal epithelial function. This review surveys the data which demonstrate the wide distribution of PARs, particularly PAR-1 and PAR-2, in the gastrointestinal tract and accessory organs, focusing on the epithelium and those cells which communicate with the epithelium to affect its function. PARs have a role in regulating secretion by epithelia of the salivary glands, stomach, pancreas and intestine. In addition, PARs located on subepithelial nerves, fibroblasts and mast cells have important implications for epithelial function. Recent data outline the importance of the cellular site of PAR expression, as PARs expressed on epithelia may have effects that are countered by PARs expressed on other cell types. Finally, PARs and their ability to promote epithelial cell proliferation are discussed in terms of colon cancer.

Histological and ultrastructural alterations in lung tissue of BALB/c mice infected with dengue virus serotype 2 (non-neuroadapted), by intraperitoneal and intravenous routes were analyzed. Lung tissues were processed following the standard techniques for photonic and electron transmission microscopies. Histopathological and ultrastructural studies showed interstitial pneumonia, characterized by the presence of mononuclear cells. In the mouse model, the dengue virus serotype 2 seems to led to a transient inflammatory process without extensive damage to the interalveolar septa, but caused focal alterations of the blood-exchange barrier. Endothelial cells of blood capillaries exhibited phyllopodia suggesting activation by presence of dengue virus. Morphometrical analysis of mast cells showed an expressive increase of the number of these cells in peribronchiolar spaces and adjacent areas to the interalveolar septa. Alveolar macrophages showed particles dengue virus-like inside rough endoplasmic reticulum and Golgi complex, suggesting viral replication. The tissue alterations observed in our experimental model were similar to the observed in human cases of dengue fever and dengue hemorrhagic fever. Our results show that BALB/c mice are permissive host for dengue virus serotype 2 replication and therefore provides an useful model to study of morphological aspects of dengue virus infection.

In vertebrate animals, pleural and peritoneal cavities are repositories of milky spots (MS), which constitute an organised coelom-associated lymphomyeloid tissue that is intensively activated by Schistosoma mansoni infection. This study compared the reactive patterns of peritoneal MS to pleural MS and concluded from histological analysis that they represent independent responsive compartments. Whole omentum, lungs and the entire mediastinum of 54 S. mansoni-infected mice were studied morphologically. The omental MS of infected animals were highly activated, modulating from myeloid-lymphocytic (60 days of infection) to lymphomyeloid (90 days of infection) and lymphocytic or lymphoplasmacytic (160 days of infection) types. The non-lymphoid component predominated in the acute phase of infection and was expressed by monocytopoietic, eosinopoietic and neutropoietic foci, with isolated megakaryocytes and small foci of late normoblasts and mast cells. Nevertheless, pleural or thoracic MS of infected mice were monotonous, consisting of small and medium lymphocytes with few mast and plasma cells and no myeloid component. Our data indicate that compartmentalisation of the MS response is dependent on the lymphatic vascularisation of each coelomic cavity, limiting the effects or consequences of any stimulating or aggressive agents, as is the case with S. mansoni infection.

RotaTeq® (Merck & Company, Inc, Whitehouse Station, NJ, USA) is an oral pentavalent rotavirus vaccine (RV5) that has shown high and consistent efficacy in preventing rotavirus gastroenteritis (RGE) in randomised clinical trials previously conducted in industrialised countries with high medical care resources. To date, the efficacy and effectiveness data for RV5 are available in some Latin American countries, but not Brazil. In this analysis, we projected the effectiveness of RV5 in terms of the percentage reduction in RGE-related hospitalisations among children less than five years of age in four regions of Brazil, using a previously validated mathematical model. The model inputs included hospital-based rotavirus surveillance data from Goiânia, Porto Alegre, Salvador and São Paulo from 2005-2006, which provided the proportions of rotavirus attributable to serotypes G1, G2, G3, G4 and G9, and published rotavirus serotype-specific efficacy from the Rotavirus Efficacy and Safety Trial. The model projected an overall percentage reduction of 93% in RGE-related hospitalisations, with an estimated annual reduction in RGE-related hospitalisations between 42,991-77,383 in the four combined regions of Brazil. These results suggest that RV5 could substantially prevent RGE-related hospitalisations in Brazil.

We report two ciprofloxacin and ofloxacin-resistant Neisseria gonorrhoeae strains that were isolated from the urethral discharge of male patients at the sexually transmitted diseases outpatient clinic of the Alfredo da Matta Foundation (Manaus, state of Amazonas, Brazil). The gonococci displayed minimal inhibitory concentrations (> 32.00 µg/mL) and three mutations in the quinolone resistance-determining region (S91F and D95G in GyrA and S87R in ParC). Both isolates were genotyped using N. gonorrhoeae multi-antigen sequence typing and the analysis showed that the ST225 which represented an emerging widespread multi-resistant clone that has also been associated with reduced susceptibility to ceftriaxone. We recommend continued surveillance of this pathogen to assess the efficacy of anti-gonococcal antibiotics in Brazil.

External morphology of the immature stages of Neotropical heliconians: IX. Dione glycera (C. Felder & R. Felder) (Lepidoptera, Nymphalidae, Heliconiinae). The biology of the Andean silverspot butterfly Dione glycera (C. Felder & R. Felder, 1861) is still poorly known. This species is restricted to high elevations in the Andes, where the immature stages are found in close association with species of Passiflora belonging to the section Tacsonia (Juss.) Harms, especially P. tripartida var. mollissima (Kunth), which is grown for subsistence by villagers. Herein we describe and illustrate the external features of the egg, larva and pupa of D. glycera, based on light and scanning electron microscopy.

Adubações minerais e orgânicas promovem alterações nas condições físicas e químicas do solo, com conseqüente efeito na produtividade das culturas. Um experimento foi realizado no Município de Campos dos Goytacazes, RJ, de fevereiro de 2005 a julho de 2006, para comparar diferentes adubos orgânicos com a adubação mineral do maracujazeiro-amarelo quanto aos efeitos sobre as características químicas e físicas do solo adubado. O delineamento experimental foi em blocos casualizados, com quatro repetições e seis tratamentos, correspondentes às seguintes adubações por planta: (AM) adubação mineral = 100 g da fórmula NPK 20-5-20 + cobertura morta (CM); EB = 5 L de esterco bovino + CM; FOC = 500 g de farinha de ossos e carne + CM; RM = 5 L de raspa de mandioca + CM; TF C/CM = 5 L de torta de filtro + CM; TF S/CM = 5 L de torta de filtro - sem CM. A adubação mineral foi feita a cada 30 dias e as adubações orgânicas a cada 60 dias. Foram avaliados os atributos químicos: pH, condutividade elétrica, teores de P, K, Ca, Mg, Na, Al, H + Al, e matéria orgânica; e os físicos: granulometria, densidade do solo e das partículas, porosidade total, macro e microporosidade, capacidade de campo, ponto de murcha e água disponível em três diferentes profundidades (0-5, 5-10, 10-15 cm). Os adubos orgânicos aplicados no maracujazeiro promoveram mudanças significativas nas características químicas do solo, em comparação com a adubação mineral tradicional, em que houve aumento do pH e do H + Al em todas as profundidades, e redução dos teores de Al nas camadas mais profundas. Houve, ainda, aumento nos teores de nutrientes no solo e, por conseqüência, na soma de bases, principalmente na camada superior, sendo a torta de filtro o composto orgânico mais eficiente em promover tais melhorias, inclusive aumentando a CTC do solo. Apenas a torta de filtro promoveu alterações nas características físicas do solo, reduzindo as quantidades de areia grossa e aumentando as quantidades de silte, argila e matéria orgânica. As demais características físicas do solo não foram influenciadas pela adição de adubos orgânicos no maracujazeiro, em comparação com a adubação mineral tradicional.

Foram conduzidos estudos para seleção de temperatura, meio de cultura e tempo de incubação ideais para germinação de grãos de pólen do maracujá-amarelo (Passiflora edulis f. flavicarpa), com a finalidade de adequar o método de avaliação da interferência de agrotóxicos. A temperatura de 28 + 0,5ºC e o meio com 50 g/L de sacarose; 0,2 g/L de ácido bórico e 1,0 g/L de nitrato de cálcio forneceram as melhores condições para germinação dos grãos de pólen do maracujazeiro. Não detectou-se efeito significativo do tempo de incubação (17 e 48 horas) na germinação dos grãos de pólen. A percentagem de germinação dos grãos de pólen não foi prejudicada pelo acaricida Dicofol + Tetradifon e pelos inseticidas Cartap, Fenpropathrin e Abamectin. Os demais agrotóxicos afetaram a germinação dos grãos de pólen. Os inseticidas Malathion, Fenthion, Trichlorfon, Vamidothion, Deltamethrine, Parathion Methyl e o espalhante adesivo N-dodecil benzeno de sulfato de sódio reduziram moderadamente a germinação, enquanto o Ethion e o Lambdacyhalothrin interferiram severamente na germinação dos grãos de pólen.

With the aim of studying the organogenesis in vitro in Passiflora edulis Sims f. flavicarpa Deg., the passionfruit, leaf-derived explants were cultured on media containing NAA or BAP and incubated either in continuous darkness or in light. The histological events leading to de novo organ formation were evaluated. Darkness induces rhizogenesis in the presence of NAA, whereas direct shoot regeneration is stimulated by light and BAP. This latter condition is recommended for passionfruit micropropagation as several adventitious shoot buds were formed from meristemoids of parenchymal origin.

O trabalho objetivou avaliar as alterações físicas do maracujá-amarelo (Passiflora edulis f. flavicarpa Degener), produzido em diferentes épocas, no sul de Minas Gerais. O delineamento experimental usado foi o inteiramente casualizado, com três tratamentos (épocas): EP1 (maio a julho/1995), temperatura e precipitação baixas; EP2 (outubro a dezembro/1995), temperatura e precipitação moderadas, e EP3 (janeiro a março/1996), temperatura e precipitação elevadas, dez repetições e cinco frutos por parcela. Os frutos da EP2 foram significativamente melhores em massa, comprimento, diâmetro e número de sementes, e os da EP1 em espessura de casca e rendimento em suco.

Dentre os insetos que atacam o maracujazeiro, Dione juno juno (Lepidoptera: Heliconidae) é considerada a praga-chave. Estudou-se a seletividade dos inseticidas fentiom, cartape, malatiom e deltametrina a Dione juno juno, em relação às vespas predadoras Polybia fastidiosuscula, Polybia scutellaris e Protonectarina sylveirae (Hymenoptera: Vespidae). Estimaram-se as curvas concentração-mortalidade e mediante o uso da concentração letal do inseticida em 90% dos indivíduos (CL90) calcularam-se os índices de seletividade diferencial e índices de tolerância. A deltametrina foi seletiva à P. scutellaris e P. fastidiosuscula e medianamente seletiva à P. sylveirae e o cartape foi medianamente seletivo às três espécies de vespas predadoras. O malatiom foi seletivo a P. sylveirae e medianamente seletivo a P. fastidiosuscula. As vespas predadoras P. fastidiosuscula eP. scutellaris foram mais tolerantes a deltametrina e ao fentiom do que P. sylveirae, enquanto o P. fastidiosuscula e P. sylveirae toleraram mais o cartape do que P. scutellaris. O malatiom foi mais tolerado pela espécie P. sylveirae do que por P. fastidiosuscula e P. scutellaris.
