Com o objetivo de avaliar a deposição e distribuição de solução traçante em plantas de feijoeiro e capim-braquiária, foi conduzido experimento no NuPAM-FCA/ UNESP, campus de Botucatu-SP, utilizando um simulador de pulverização em ambiente controlado. Os tratamentos utilizados foram: bico jato plano XR Teejet (XR110.02VS) + surfactante Aterbane BR (0,5% v/v); bico jato cônico Conejet (TXVK-4) + surfactante Aterbane BR (0,5% v/v); bico jato plano XR Teejet (XR110.02VS); e bico jato cônico Conejet (TXVK-4). Em todos os tratamentos foi aplicada uma solução de NaCl (0,5% p/v) + corante Poliglow laranja (0,5% p/v) + mancozeb (0,5% p/v). As unidades experimentais constituíram-se de vasos com duas plantas-alvo de capim-braquiária posicionadas sob uma planta-alvo de feijoeiro. A visualização da distribuição das gotas nas folhas de capim-braquiária foi efetuada com auxílio de luz negra, e o depósito da calda na superfície das plantas foi quantificado através da condutividade elétrica da solução aplicada e coletada por meio de lavagem de ambos os alvos. Os bicos de pulverização, jato plano (XR Teejet) e cônico (Conejet), não apresentaram diferença no depósito nos folíolos totais de feijoeiro quando submetidos a mesma condição de calda de pulverização. No entanto, o bico jato plano XR Teejet (XR110.02VS) e o bico jato cônico Conejet (TXVK-4) proporcionaram aumento de 67,1 e 61,5% na deposição da calda em relação a área foliar e 106,4 e 66,9% para matéria seca, respectivamente, em relação ao bico jato plano XR Teejet (XR110.02VS) + surfactante Aterbane BR (0,5% v/v) e ao bico jato cônico Conejet (TXVK-4) + surfactante Aterbane BR (0,5% v/v). Para o capim-braquiária, o bico cônico Conejet proporcionou deposição superior e distribuição mais uniforme em relação ao jato plano XR Teejet.

O conhecimento de informações básicas sobre a biologia de plantas daninhas pode contribuir significativamente para o desenvolvimento de estratégias para seu manejo. Emilia sonchifolia é uma espécie de ampla disseminação em todo o território brasileiro e apresenta moderada agressividade, sendo encontrada em áreas agrícolas, terrenos baldios e áreas urbanas. O presente trabalho teve como objetivo avaliar a germinação das sementes de E. sonchifolia em diferentes condições ambientais. Foram realizados quatro experimentos: combinação de quatro temperaturas (20, 25, 30 e 35 ºC) em duas condições de luz (ausência e presença); sete níveis de restrição hídrica proporcionada por PEG6000 (0,0; -0,1; -0,2; -0,4; -0,6; -0,8; e -1,0 MPa); seis níveis de estresse salino proporcionado pelo NaCl (0,0; -0,2; -0,4; -0,6; -0,8; e -1,0 MPa) e seis níveis de pH (3,0; 5,0; 6,0; 7,0; 9,0; e 11,0) em solução tamponada. A germinação das sementes de E. sonchifolia foi maximizada na presença de luz, em temperaturas constantes de 25 e 30 ºC. Houve redução da germinação em ambiente com restrição hídrica a partir de -0,1 MPa, e para estresse salino, a partir de 0,2 MPa. E. sonchifolia é sensível à variação de pH, ocorrendo maior germinação das sementes apenas em pH 6,0.

O comportamento invasivo de Melaleuca quinquenervia em áreas úmidas deve-se à sua estratégia de regeneração agressiva, que está alicerçada na produção de sementes em massa. O conhecimento da fisiologia da germinação de sementes de plantas daninhas pode contribuir significativamente para o desenvolvimento de estratégias de manejo. O objetivo deste trabalho foi avaliar os possíveis efeitos dos estresses hídrico e salino na germinação de sementes de M. quinquenervia. A semeadura foi realizada com quatro repetições de 0,05 g de sementes em papel umedecido com soluções nos potenciais osmóticos de 0,0; -0,2; -0,4; e -0,8 MPa, induzidos com polietilenoglicol (PEG 6000) e NaCl. O teste de germinação foi conduzido a 25 ºC na presença de luz. Avaliou-se a primeira contagem do teste aos sete dias após a semeadura e, semanalmente, a germinação (plântulas normais) até os 28 dias. Foi calculado o índice de velocidade de germinação. A análise dos resultados permitiu a conclusão de que o estresse hídrico acarreta maior redução na velocidade de germinação e na germinação acumulada de sementes de M. quinquenervia do que o estresse salino e, independentemente da substância utilizada para indução do estresse, o limite para germinação está entre -0,4 e -0,8MPa.

Smellmelon, an annual invasive weed of soybean production fields in the north of Iran, reproduces and spreads predominately through seed production. This makes seed bank survival and successful germination essential steps in the invasive process. To evaluate the potential of Smellmelon to invade water-stressed environments, laboratory studies were conducted to investigate the effect of desiccation and salinity at different temperatures on seed germination and seedling growth of Cucumis melo. Seeds were incubated at 25, 30, 35 and 40 ºC in the darkness in a solution (0, -0.2, -0.4, -0.6, -0.8, 1 and 1.2 MPa) of a salt (NaCl), and in a solution (0, -2, -4, -6, -8, -10, -12 bar) of PEG-6000 (Polyethylene glycol), in two separate experiments. The results showed that the highest percentage and rate of germination occurred at 35 ºC in salt concentrations of 0, -0.2, -0.4 MPa and PEG concentrations of 0, -2, -4 bar. Increasing the concentration of salt (NaCl) and PEG limited germination, seedling growth and water uptake but increased the sodium content in the seedlings. No significant difference was observed among 0, -0.2 and -0.4 MPa of NaCl and among 0, -2 and -4 bar of PEG concentration at 35 ºC. The negative effects of PEG were more than those of NaCl on germination percentage and germination rate. Increased stress levels lead to reduction of root and shoot length, and SVL of seedlings. Na+ content of seedling decreased with limited seedling growth of C. melo.

Asphodelus tenuifolius is becoming a more common weed in rain-fed area in Pakistan. Laboratory and greenhouse experiments were conducted to determine the effect of different environmental factors on germination and emergence of A.tenuifolius. Results showed that A.tenuifolius can tolerate a wide range of varying environmental factors. Greatest percentage of germination (80%) was recorded at 15 ºC constant temperature; however, considerable germination occurred at 20 and 25 ºC. Light for 10 h photoperiod stimulate germination of Asphodelus tenuifolius compared with complete darkness. Germination was totally inhibited at osmotic stress higher than -0.8 MPa. There was no significant difference in germination at pH 6 to 8; however, there was a slightly decrease at pH 9, compared with distilled water. Asphodelus tenuifolius was very sensitive to salinity; however, a few seeds of A.tenuifolius were able to germinate even at 150 mM NaCl concentration. Greatest emergence occurred with seed placed at soil surface and emergence decreased with increase in burial depth. No emergence occurred from 4 cm or greater. This information may aid in developing tools and strategies for management.

T. absinthioides, Inuleae, Compositae, is a weedy species that is spreading in the irrigation area of the Colorado River, Argentina. This species can be found in normal and saline soils. Morphological and anatomical variables were measured with two salts, sodium chloride and sodium sulfate, with three levels of osmotic potential -0.4, -0.8 and -1.5 MPa, in hydroponic culture, using Hoagland solution as the cultivation media. The total diameter of the roots of plants growing in Na2SO4 and NaCl increased when the osmotic potential was -0.4 MPa. In plants growing in NaCl this may have resulted from the increase in the size of the cortical cells and in plants growing in Na2SO4, the diameter increased may be due to an increase in the cambial activity. The number and length of shoot internodes decreased with increasing salinity, even though this was not statistically significant. In comparison to the control, the total diameter of the shoot increased at -0.4 MPa and decreased with the reduction of the osmotic potential. In comparison to the control, the length of the leaves decreased at -0.4 MPa and the leaves width increased at the same concentration. The palisade parenchyma appeared less developed in saline conditions. In comparison to the control, the number of hairs increased at -0.4 MPa. T. absinthioides acts as a semihalophytic species, according to the salt ranks it tolerates. The mechanisms of adaptation to saline conditions are succulence in root and stunted growth if the salt in the media is NaCl, and the production of haloxeromorphic characters if the salt in the media is Na2 SO4

Os efeitos da salinidade no crescimento e nos teores de solutos orgânicos e inorgânicos foram analisados em sete cultivares de Vigna unguiculata. As plantas foram cultivadas em solução nutritiva sem NaCl (controle) e com NaCl a 75 mM (estresse salino), em casa de vegetação. Após 14 dias da semeadura, adicionou-se 25 mM de sal diariamente, durante três dias, perfazendo o total de 75 mM. A coleta foi realizada no 33º dia. A salinidade reduziu a matéria seca de todos os cultivares, sendo Pitiúba e Vita 5 os menos afetados e TVU o que sofreu maior redução. A suculência foliar e o índice de esclerofilia foram concordantes com o grau de tolerância, aumentando significativamente, em função da salinidade, apenas nos cultivares com menores reduções nas matérias secas. Contrariamente, a relação raiz/parte aérea teve maior aumento nos cultivares mais afetados. Os teores de Na+ e Cl- foliares aumentaram em resposta ao estresse salino, sendo o Na+ acumulado em níveis maiores nos cultivares mais afetados, especialmente no TVU. Os níveis de K+ foliar, que se mostraram mais elevados que os de Na+ e Cl-, foram poucos afetados pela salinidade, de modo que a relação Na+/K+ aumentou mais nos cultivares mais afetados e, possivelmente, se constituiu num fator importante para a maior redução em seus crescimentos. As variações nos teores de prolina, carboidratos solúveis e N-aminossolúveis em resposta à salinidade indicam uma falta de proporcionalidade entre seus níveis e o grau de tolerância à salinidade nos cultivares de feijão-de-corda estudados.

(Germinação de megásporos e desenvolvimento esporofítico inicial da filicínea Regnellidium diphylum (Marsileaceae) sob deficiências minerais). Regnellidium diphyllum Lindm. é uma filicínea heterosporada da família Marsileaceae, de distribuição restrita ao sul do Brasil e algumas localidades do Uruguai e da Argentina. A espécie se encontra na lista de espécies ameaçadas de extinção do Rio Grande do Sul, como vulnerável. No presente estudo, verificou-se o efeito da ausência de nutrientes minerais na germinação de megásporos e no desenvolvimento inicial de esporófitos de R. diphyllum in vitro. O efeito de soluções nutritivas de Meyer modificadas pela ausência de NH4NO3, NaCl, FeCl3·6H2O, MgSO4·7H2O, KH2PO4 ou CaCl2 foi analisado na germinação e na formação de esporófitos de R. diphyllum. A germinação dos megásporos foi reduzida na ausência de CaCl2 (52%), quando comparada ao controle (77,3%). Houve baixa porcentagem de gametófitos com formação de esporófitos na ausência de MgSO4·7H2O (38,6%) e de KH2PO4 (33,8%), sendo que na ausência de CaCl2 não houve formação de esporófitos. O comprimento da raiz primária e das folhas primária e secundária foi reduzido significativamente na ausência de MgSO4·7H2O e de KH2PO4, destacando-se que na ausência de KH2PO4 não ocorreu desenvolvimento da folha secundária após cinco semanas em cultura. Os demais tratamentos não comprometeram o processo de germinação dos megásporos e o desenvolvimento inicial dos esporófitos, demonstrando que a espécie pode se estabelecer mesmo sob baixas concentrações desses nutrientes.

Variation in salt tolerance of six natural populations of Stylosanthes humilis from three ecogeographic regions, Mata (wet tropical climate), Agreste and Sertão (semi-arid tropical climate) of Pernambuco State, Northeast Brazil, was evaluated on germination in 201 mM NaCl. There were significant differences among families of all populations for germination percentage and of five populations (except Tamandaré, from Mata) for germination rate. Populations from semi-arid regions presented high coefficients of genetic variation, those from Agreste being higher than those from Sertão. Populations from Mata showed low coefficients of genetic variation. The coefficients of genotypic determination were high for five populations, except Tamandaré, both for germination percentage ( > or = 0.89) and for germination rate ( > or = 0.79), indicating the possibility of selection for salt tolerance in these populations. An electrophoretic analysis of esterase and peroxidase isozymes was also performed in the six populations, and correlations were estimated between salt tolerance and allelic frequencies. The analysis of salt tolerant and salt sensitive families of populations from Agreste suggested an association of alleles of a peroxidase locus with salt tolerance during germination in the Caruaru population

We determined whether ANP (atrial natriuretic peptide) concentrations, measured by radioimmunoassay, in the ANPergic cerebral regions involved in regulation of sodium intake and excretion and pituitary gland correlated with differences in sodium preference among 40 Wistar male rats (180-220 g). Sodium preference was measured as mean spontaneous ingestion of 1.5% NaCl solution during a test period of 12 days. The relevant tissues included the olfactory bulb (OB), the posterior and anterior lobes of the pituitary gland (PP and AP, respectively), the median eminence (ME), the medial basal hypothalamus (MBH), and the region anteroventral to the third ventricle (AV3V). We also measured ANP content in the right (RA) and left atrium (LA) and plasma. The concentrations of ANP in the OB and the AP were correlated with sodium ingestion during the preceding 24 h, since an increase of ANP in these structures was associated with a reduced ingestion and vice-versa (OB: r = -0.3649, P<0.05; AP: r = -0.3291, P<0.05). Moreover, the AP exhibited a correlation between ANP concentration and mean NaCl intake (r = -0.4165, P<0.05), but this was not the case for the OB (r = 0.2422). This suggests that differences in sodium preference among individual male rats can be related to variations of AP ANP level. Earlier studies indicated that the OB is involved in the control of NaCl ingestion. Our data suggest that the OB ANP level may play a role mainly in day-to-day variations of sodium ingestion in the individual rat

The objectives of the present investigation were 1) to study the effect of bacterial lipopolysaccharide (LPS) on rat gastric emptying (GE) and 2) to investigate a possible involvement of the vagus nerve in the gastric action of LPS. Endotoxin from E. coli (strain 055:B5) was administered sc, ip or iv to male Wistar rats (220-280 g body weight) at a maximum dose of 50 µg/kg animal weight. Control animals received an equivalent volume of sterile saline solution. At a given time period after LPS administration, GE was evaluated by measuring gastric retention 10 min after the orogastric infusion of a test meal (2 ml/100 g animal weight), which consisted of 0.9% NaCl plus the marker phenol red (6 mg/dl). One group of animals was subjected to bilateral subdiaphragmatic vagotomy or sham operation 15 days before the test. A significant delay in GE of the test meal was observed 5 h after iv administration of the endotoxin at the dose of 50 µg/kg animal weight. The LPS-induced delay of GE was detected as early as 30 min and up to 8 h after endotoxin administration. The use of different doses of LPS ranging from 5 to 50 µg/kg animal weight showed that the alteration of GE was dose dependent. In addition, vagotomized animals receiving LPS displayed a GE that was not significantly different from that of the sham control group. However, a participation of the vagus nerve in LPS-induced delay in GE could not be clearly demonstrated by these experiments since vagotomy itself induced changes in this gastric parameter. The present study provides a suitable model for identifying the mechanisms underlying the effects of LPS on gastric emptying

The existence of a circadian rhythm of atrial natriuretic peptide (ANP) in humans is controversial. We studied the plasma ANP response to isotonic blood volume expansion in the morning and in the afternoon and its relationship with adrenocorticotropic hormone (ACTH)-cortisol diurnal variation in seven normal subjects. Basal plasma ANP level was similar in the morning (19.6 ± 2.4 pg/ml) and in the afternoon (21.8 ± 4.8 pg/ml). The ANP peak obtained with saline infusion (0.9% NaCl, 12 ml/kg) in the morning (49.4 ± 8 pg/ml) did not differ from that obtained in the afternoon (60.3 ± 10.1 pg/ml). There was no correlation between the individual mean cortisol and ACTH levels and the ANP peak obtained with saline infusion. These data indicate no diurnal variation in plasma ANP secretion induced by blood volume expansion and no relationship between plasma ANP peak and ACTH-cortisol diurnal variation

We determined the effect of acute extracellular fluid volume changes on saline flow through 4 gut segments (ileocolonic, ileal, ileocolonic sphincter and proximal colon), perfused at constant pressure in anesthetized dogs. Two different experimental protocols were used: hypervolemia (iv saline infusion, 0.9% NaCl, 20 ml/min, volume up to 5% body weight) and controlled hemorrhage (up to a 50% drop in mean arterial pressure). Mean ileocolonic flow (N = 6) was gradually and significantly decreased during the expansion (17.1%, P<0.05) and expanded (44.9%, P<0.05) periods while mean ileal flow (N = 7) was significantly decreased only during the expanded period (38%, P<0.05). Mean colonic flow (N = 7) was decreased during expansion (12%, P<0.05) but returned to control levels during the expanded period. Mean ileocolonic sphincter flow (N = 6) was not significantly modified. Mean ileocolonic flow (N = 10) was also decreased after hemorrhage (retracted period) by 17% (P<0.05), but saline flow was not modified in the other separate circuits (N = 6, 5 and 4 for ileal, ileocolonic sphincter and colonic groups, respectively). The expansion effect was blocked by atropine (0.5 mg/kg, iv) both on the ileocolonic (N = 6) and ileal (N = 5) circuits. Acute extracellular fluid volume retraction and expansion increased the lower gastrointestinal resistances to saline flow. These effects, which could physiologically decrease the liquid volume being supplied to the colon, are possible mechanisms activated to acutely balance liquid volume deficit and excess.

The effects of dorsomedial hypothalamic (DMH) nucleus lesion on body weight, plasma glucose levels, and the gastric emptying of a liquid meal were investigated in male Wistar rats (170-250 g). DMH lesions were produced stereotaxically by delivering a 2.0-mA current for 20 s through nichrome electrodes (0.3-mm tip exposure). In a second set of experiments, the DMH and the ventromedial hypothalamic (VMH) nucleus were lesioned with a 1.0-mA current for 10 s (0.1-mm tip exposure). The medial hypothalamus (MH) was also lesioned separately using a nichrome electrode (0.3-mm tip exposure) with a 2.0-mA current for 20 s. Gastric emptying was measured following the orogastric infusion of a liquid test meal consisting of physiological saline (0.9% NaCl, w/v) plus phenol red dye (6 mg/dl) as a marker. Plasma glucose levels were determined after an 18-h fast before the lesion and on the 7th and 15th postoperative day. Body weight was determined before lesioning and before sacrificing the rats. The DMH-lesioned rats showed a significantly faster (P<0.05) gastric emptying (24.7% gastric retention, N = 11) than control (33.0% gastric retention, N = 8) and sham-lesioned (33.5% gastric retention, N = 12) rats, with a transient hypoglycemia on the 7th postoperative day which returned to normal by the 15th postoperative day. In all cases, weight gain was slower among lesioned rats. Additional experiments using a smaller current to induce lesions confirmed that DMH-lesioned rats had a faster gastric emptying (25.1% gastric retention, N = 7) than control (33.4% gastric retention, N = 17) and VMH-lesioned (34.6% gastric retention, N = 7) rats. MH lesions resulted in an even slower gastric emptying (43.7% gastric retention, N = 7) than in the latter two groups. We conclude that although DMH lesions reduce weight gain, they do not produce consistent changes in plasma glucose levels. These lesions also promote faster gastric emptying of an inert liquid meal, thus suggesting a role for the DMH in the regulation of gastric motility

Bothrops erythromelas is responsible for many snake bites in northeastern Brazil. In the present study we determined the in vivo distribution of the venom following its subcutaneous injection into mice. B. erythromelas venom and albumin were labeled individually with 131I by the chloramine T method, and separated in a Sephacryl® S-200 column. The efficiency of labeling was 68%. Male Swiss mice (40-45 g), which had been provided with drinking water containing 0.05% KI over a period of 10 days prior to the experiment, were inoculated dorsally (sc) with 0.3 ml (2.35 x 105 cpm/mouse) of 131I-venom (N = 42), 131I-albumin or 131I (controls, N = 28 each). Thirty minutes and 1, 3, 6, 12, 18 and 24 h after inoculation, the animals were perfused with 0.85% NaCl and skin and various organs were collected in order to determine radioactivity content. There was a high rate of venom absorption in the skin (51%) within the first 30 min compared to albumin (20.1%) and free iodine (8.2%). Up to the third hour after injection there was a tendency for venom and albumin to concentrate in the stomach (3rd h), small intestine (3rd h) and large intestine (6th h). Both control groups had more radioactivity in the digestive tract, especially in the stomach, but these levels decreased essentially to baseline by 12-18 h postinjection. In the kidneys, the distribution profiles of venom, albumin and iodine were similar. Counts at 30 min postinjection were low in all three groups (1.37, 1.86 and 0.77, respectively), and diminished to essentially 0% by 12-18 h. Albumin tended to concentrate in muscle until the 3rd h postinjection (1.98%). There was a low binding of labeled venom in the liver (<0.54%), thyroid (<0.11%) and lungs (<0.08%), and no iodinated venom was detected in brain, heart, diaphragm, spleen or bladder. The low venom binding observed in most internal organs, comparable to that of albumin, suggests that B. erythromelas venom does not specifically target most internal organs. That is, the systemic effects of envenomation are mainly due to an indirect action
