The objective of this study was to establish critical values of the N indices, namely soil-plant analysis development (SPAD), petiole sap N-NO3 and organic N in the tomato leaf adjacent to the first cluster (LAC), under soil and nutrient solution conditions, determined by different statistical approaches. Two experiments were conducted in randomized complete block design with four repli-cations. Tomato plants were grown in soil, in 3 L pot, with five N rates (0, 100, 200, 400 and 800 mg kg-1) and in solution at N rates of 0, 4, 8, 12 and 16 mmol L-1. Experiments in nutrient solution and soil were finished at thirty seven and forty two days after transplanting, respectively. At those times, SPAD index and petiole sap N-NO3 were evaluated in the LAC. Then, plants were harvested, separated in leaves and stem, dried at 70ºC, ground and weighted. The organic N was determined in LAC dry matter. Three statistical procedures were used to calculate critical N values. There were accentuated discrepancies for critical values of N indices obtained with plants grown in soil and nutrient solution as well as for different statistical procedures. Critical values of nitrogen indices at all situations are presented.

O objetivo deste trabalho foi avaliar a eficiência da técnica de miniestaquia seriada em quatro clones de Eucalyptus grandis, quanto à sobrevivência, ao enraizamento e ao crescimento em altura e diâmetro de colo das mudas. Foram realizados sete subcultivos pela miniestaquia seriada, por enraizamento sucessivo de brotações de minicepas, em casa de vegetação durante 25 dias, seguido de aclimatação por dez dias em casa de sombra e avaliação final das mudas aos 50 dias de idade, em pleno sol. A miniestaquia seriada mostrou efeito positivo, principalmente em relação aos clones que apresentaram menor potencial de enraizamento.

Even though much improvement has been made in plant transformation methods, the screening of transgenic plants is often a laborious work. Most approaches for detecting the transgene in transformed plants are still timeconsuming, and can be quite expensive. The objective of this study was to search for a simpler method to screen for transgenic plants. The infiltration of kanamycin (100 mg/mL) into tobacco leaves resulted in conspicuous chlorotic spots on the non-transgenic plant leaves, while no spots were seen on the leaves of transformed plants. This reaction occurred regardless of age of the tested plants, and the method has proven to be simple, fast, non-destructive, relatively cheap, and reliable. These results were comparable to those obtained by the polymerase chain reaction (PCR) amplification of the transgene using specific primers.

The objective of this work was to characterize the genetic variability of phytoplasma and Spiroplasma kunkelii isolated from maize plants showing symptoms of stunt collected from different Brazilian geographic regions. A DNA fragment of 500 base pairs (bp) was amplified from the spiralin gene in S. kunkelii and one fragment of 1,200 bp was generated from 16S rDNA gene in phytoplasma. The partial sequences of the spiralin gene showed similarity of 98% among the isolates of S. kunkelii analyzed. These sequences were compared with the sequence of the spiralin gene from other Spiroplasma species deposited in the GenBank, resulting in a similarity varying from 76.9% to 88.1%. The 16S rDNA sequence from the phytoplasma were completely similar within the Brazilian isolates and showed up to 98% of the similarity with sequences already found from other phytoplasmas. A very narrow genetic variability was detected by these gene fragments within phytoplasma and Spiroplasma analyzed. However, other genomic regions with higher polymorphic levels shall be identified in order to better evaluate the genetic diversity within these microorganisms population.

In vitro regeneration of Arachis retusa was examined for the purpose of germplasm renewal and conservation. Random amplified polymorphic DNA (RAPD) fingerprinting was used to evaluate the genetic stability of plants derived from embryo axes and apical segments. Ten arbitrary decamer primers were screened and five of them were selected. Ninety genomic regions were evaluated, with an average of 18 loci per clone. All amplified segments were monomorphic. The results indicate that recovered plants are genetically stable at the assessed genomic regions and that both regeneration processes are suitable for in vitro germplasm preservation of Arachis species.

O objetivo deste trabalho foi avaliar os efeitos dos reguladores de crescimento TDZ [1-fenil-3-(1,2,3-tia-diazol-5-il)uréia], BAP (6-benzilaminopurina) e ANA (ácido naftalenoacético) no desempenho da propagação in vitro por organogênese de explante foliar de três clones híbridos de Eucalyptus grandis x Eucalyptus urophylla. Houve resposta diferenciada dos clones quanto a intensidade, textura e coloração dos calos, em razão dos tratamentos com os reguladores de crescimento. Os melhores resultados de calejamento dos três genótipos foram observados nos tratamentos com a combinação dos reguladores de crescimento TDZ (0,5 mg L-1) e ANA (0,1 mg L-1), obtendo-se 100% de calejamento no explante foliar. Os piores resultados de calejamento foram observados nos tratamentos com a combinação dos reguladores de crescimento BAP (0,1 mg L-1) e ANA (0,1 mg L-1). Em relação à regeneração, a melhor resposta foi obtida com 1,0 mg L-1 BAP em que 8% dos calos formados a partir de explantes foliares regeneraram gemas, com número médio destas formadas por calo igual a 4,2.

Este trabalho teve como objetivo avaliar a deposição de serapilheira e a transferência de N associada às folhas e folíolos desse resíduo em plantios puros e consorciado de guachapele (Pseudosamanea guachapele) e eucalipto (Eucalyptus grandis), aos sete anos de idade, em Seropédica, RJ. No ano de avaliação (2000/2001) a deposição anual de serapilheira nos diferentes plantios não diferiu significativamente 12,75 Mg ha-1 (guachapele); 11,84 Mg ha-1 (eucalipto) e 12,44 Mg ha-1 (consorciado). Mesmo com pequena participação na composição da serapilheira em plantio consorciado (11% do total depositado), a leguminosa proporciona redução no tempo médio de residência dos resíduos no solo de 0,3 ano, comparativamente ao plantio puro de eucalipto.

The aim of this study was to characterize gas exchange responses of young cashew plants to varying photosynthetic photon flux density (PPFD), temperature, vapor-pressure deficit (VPD), and intercellular CO2 concentration (Ci), under controlled conditions. Daily courses of gas exchange and chlorophyll a fluorescence parameters were measured under natural conditions. Maximum CO2 assimilation rates, under optimal controlled conditions, were about 13 mmol m-2 s-1 , with light saturation around 1,000 mmol m-2 s-1. Leaf temperatures between 25ºC and 35ºC were optimal for photosynthesis. Stomata showed sensitivity to CO2, and a closing response with increasing Ci. Increasing VPD had a small effect on CO2 assimilation rates, with a small decrease above 2.5 kPa. Stomata, however, were strongly affected by VPD, exhibiting gradual closure above 1.5 kPa. The reduced stomatal conductances at high VPD were efficient in restricting water losses by transpiration, demonstrating the species adaptability to dry environments. Under natural irradiance, CO2 assimilation rates were saturated in early morning, following thereafter the PPFD changes. Transient Fv/Fm decreases were registered around 11h, indicating the occurrence of photoinhibition. Decreases of excitation capture efficiency, decreases of effective quantum yield of photosystem II, and increases in non-photochemical quenching were consistent with the occurrence of photoprotection under excessive irradiance levels.

O objetivo deste trabalho foi avaliar a eficiência da enxertia in vitro na propagação de dois clones híbridos de Eucalyptus urophylla e E. grandis. Foram utilizados porta-enxertos juvenis obtidos de sementes de E. grandis e E. urophylla germinadas in vitro. As plantas enxertadas apresentaram pegamento de até 93% aos 50 dias de idade para as combinações de enxerto e porta-enxerto. Quanto ao crescimento em altura, verificaram-se melhores resultados em relação ao porta-enxerto E. urophylla. A análise histológica das plantas enxertadas comprovou a eficiência do processo de cicatrização do calo formado na região de conexão, seguida da reconstituição vascular.

Recent studies on coffee (Coffea arabica L.) cultivation in agroforestry systems in Southern Brazil have shown the potential of partial shading to improve management of this crop. The objective of this work was to evaluate microclimatic conditions and their effects on coffee production of plants shaded with pigeon pea (Cajanus cajan) in comparison to unshaded ones, from May 2001 to August 2002 in Londrina, State of Paraná, Brazil. The appraised microclimatic characteristics were: global radiation, photosynthetic and radiation balance; air, leaf and soil temperatures; and soil humidity. Shading caused significant reduction in incident global solar radiation, photosynthetically active radiation and net radiation, and attenuated maximum leaf, air and soil temperatures, during the day. Shade also reduced the rate of cooling of night air and leaf temperatures, especially during nights with radiative frost. Soil moisture at 0-10 cm depth was higher under shade. The shaded coffee plants produced larger cherries due to slower maturation, resulting in larger bean size. Nevertheless, plants under shade emitted less plagiotropic branches, with smaller number of nodes per branch, and fewer nodes with fruits, resulting in a large reduction in coffee production. These results show the need to find an optimal tree density and management that do not compromise coffee production and protect against extreme temperatures.

Studies on the impact of Eucalyptus spp. on Brazilian soils have focused on soil chemical properties and isolating interesting microbial organisms. Few studies have focused on microbial diversity and ecology in Brazil due to limited coverage of traditional cultivation and isolation methods. Molecular microbial ecology methods based on PCR amplified 16S rDNA have enriched the knowledge of soils microbial biodiversity. The objective of this work was to compare and estimate the bacterial diversity of sympatric communities within soils from two areas, a native forest (NFA) and an eucalyptus arboretum (EAA). PCR primers, whose target soil metagenomic 16S rDNA were used to amplify soil DNA, were cloned using pGEM-T and sequenced to determine bacterial diversity. From the NFA soil 134 clones were analyzed, while 116 clones were analyzed from the EAA soil samples. The sequences were compared with those online at the GenBank. Phylogenetic analyses revealed differences between the soil types and high diversity in both communities. Soil from the Eucalyptus spp. arboretum was found to have a greater bacterial diversity than the soil investigated from the native forest area.

The objective of this work was to evaluate the use of basic density and pulp yield correlations with some chemical parameters, in order to differentiate an homogeneous eucalyptus tree population, in terms of its potential for pulp production or some other technological applications. Basic density and kraft pulp yield were determined for 120 Eucalyptus globulus trees, and the values were plotted as frequency distributions. Homogenized samples from the first and fourth density quartiles and first and fourth yield quartiles were submitted to total phenols, total sugars and methoxyl group analysis. Syringyl/guaiacyl (S/G) and syringaldehyde/vanillin (S/V) ratios were determined on the kraft lignins from wood of the same quartiles. The results show the similarity between samples from high density and low yield quartiles, both with lower S/G (3.88-4.12) and S/V (3.99-4.09) ratios and higher total phenols (13.3-14.3 g gallic acid kg-1 ). Woods from the high yield quartile are statistically distinguished from all the others because of their higher S/G (5.15) and S/V (4.98) ratios and lower total phenols (8.7 g gallic acid kg-1 ). Methoxyl group and total sugars parameters are more adequate to distinguish wood samples with lower density.

O objetivo deste trabalho foi avaliar os efeitos do estímulo com benzilaminopurina (BAP) na multiplicação in vitro de Eucalyptus grandis. Foram avaliadas as interações entre concentrações de BAP (0, 200, 400 e 600 mg L-1), tempo de exposição (1, 2 e 3 horas), pH (3 e 5,8) da solução e alterações morfológicas dos explantes. Semanalmente, foi determinada a massa da matéria fresca dos explantes. Aos 21 dias de cultura, foram avaliados: o número de brotações por tratamento, o número de brotações obtidas por explante (taxa de multiplicação), e foi realizado o resgate das plântulas para avaliação histológica. O pH não apresentou interação com os demais fatores estudados. Os tratamentos com BAP a 200 mg L-1, durante 1 e 2 horas, apresentaram-se como os mais indicados na multiplicação do E. grandis. Houve intensificação da divisão celular, no parênquima cortical e no procâmbio, representada pelo surgimento de meristemóides, em resposta aos tratamentos com 200 mg L-1 de BAP, durante 1 e 2 horas. O estímulo com BAP na multiplicação in vitro de E. grandis é viável.

This work aimed to analyse Eucalyptus grandis W.Hill ex Maiden seed behaviour, under controlled deterioration, and to estimate viability equation constants for the species. Seeds were harvested in the growing season of 1999, and the moisture contents were adjusted from 11.3% to a range between 1.2 and 18.1% at 25ºC. The subsamples were sealed into laminate aluminium-foil packets, for storage in incubators at 40, 50 and 65±0.5ºC. The seeds presented orthodox performance, in which the constants for predicting seed longevity of E. grandis were K E = 9.661, C W = 6.467, C H = 0.03498 and C Q = 0.0002330. The usual and inverse relationship between water content and seed longevity was also observed. The lowest moisture content limit for application of the viability equation at 65ºC was 4.9%, estimated under hygroscopic equilibrium with 23% of relative humidity in the storage environment.

The objective of this work was to evaluate the efficiency of ectomycorrhizal isolates on root colonization, phosphorus uptake and growth of Eucalyptus dunnii seedlings. Inocula of ten ectomycorrhizal isolates of Chondrogaster angustisporus, Hysterangium gardneri, Pisolithus spp., and Scleroderma spp. were aseptically produced in a peat-vermiculite mixture supplemented with liquid culture medium. Plants grew in a similar substrate supplemented with macro-and micro-nutrients; treatments were randomly distributed in a greenhouse. After three months, seedlings inoculated with three isolates - UFSC-Sc68 (Scleroderma sp.), UFSC-Ch163 (Chondrogaster angustisporus), and UFSC-Pt188 (Pisolithus microcarpus) - had a phosphorus shoot content and a shoot dry matter higher or equivalent to those of noninoculated controls which had been fertilized with a 16-fold phosphorus amount. These isolates were selected for new studies for establishing inoculum production techniques, in order to be applied in reforestation programmes under nursery and field conditions.
