Domiciliated Rhodnius prolixus and sylvatic R. colombiensis were analyzed in order to confirm their genetic divergence and verify the risk that the latter represents in the domiciliation process, and to provide tools for identifying the sources of possible reinfestation by triatomines in human dwellings allowing control programs to be undertaken. Comparison of random amplified polymorphic DNA amplification patterns and cluster analysis suggests reproductive discontinuity between the two species. The calculated statistical F value of 0.24 and effective migration rate of 0.6 individuals per generation are insufficient to maintain genetic homogeneity between them and confirm the absence of present genetic flow. R. colombiensis presents higher intrapopulation variability. Polymerase chain reaction of ribosomal DNA supports these findings. The low genetic flow between the two species implies that R. colombiensis do not represent an epidemiological risk for the domiciliary transmission of Trypanosoma cruzi in the Tolima Department. The lower variability of the domiciliated R. prolixus could result in greater susceptibility to the use of pesticides in control programs.

The aim of the present study was to examine genetic variability in populations of An. cruzii by employing PCR-RAPD and PCR-RFLP markers. All analyses were carried out using individuals of the F1 generation of wild caught females obtained in Santa Catarina State (Florianópolis and São Francisco do Sul), Paraná State (Morretes, Paranaguá and Guaratuba) and São Paulo State (Cananéia). In the PCR-RAPD experiments, seven primers were used for comparisons within and among populations. The restriction profile of the ITS2 including a fragment of both 5.8S and 28S regions of the rDNA was obtained with the enzymes BstUI, HaeIII, TaqI, HhaI, Sau96I, HinfI, HincII and NruI. The PCR-RAPD method detected a large number of polymorphic bands. Genetic distance among populations of An. cruzii varied from 0,0214 to 0,0673, suggesting that all individuals used in the analyses belong to a single species. The number of migrants per generation (Nm) was 4.3, showing the existence of gene flow among populations. The restriction profile of the ITS2, 5.8S and 28S gene regions was similar in all An. cruzii samples, whereas the results obtained by using HhaI and NruI are indicative that the individuals analyzed have nucleotide sequences distinct from those of An. cruzii samples from Peruíbe and Juquiazinho deposited in GenBank.

The emergence of host-races within aphids may constitute an obstacle to pest management by means of plant resistance. There are examples of host-races within cereals aphids, but their occurrence in Rose Grain Aphid, Metopolophium dirhodum (Walker, 1849), has not been reported yet. In this work, RAPD markers were used to assess effects of the hosts and geographic distance on the genetic diversity of M. dirhodum lineages. Twenty-three clones were collected on oats and wheat in twelve localitites of southern Brazil. From twenty-seven primers tested, only four primers showed polymorphisms. Fourteen different genotypes were revealed by cluster analysis. Five genotypes were collected only on wheat; seven only on oats and two were collected in both hosts. Genetic and geographical distances among all clonal lineages were not correlated. Analysis of molecular variance showed that some molecular markers are not randomly distributed among clonal lineages collected on oats and on wheat. These results suggest the existence of host-races within M. dirhodum, which should be further investigated using a combination of ecological and genetic data.

Random amplified polymorphic DNA markers (RAPD) were used to estimate the variability of 14 genotypes of Brazilian wheat (Triticum aestivum L.), using a set of 50 random 10mer primers. A total of 256 reproducibly scorable DNA amplification products were obtained from 48 of the primers, 83% of which were polymorphic. Genetic distances among genotypes were calculated and a dendrogram and a principal coordinates analysis showing the genetic relationships among them were obtained. Despite the low variability found (average genetic distance of 27%), two groups of genotypes could be identified, which probably reflect how they were formed. Studies such as this one may be important in the planning and development of future improvement programs for this plant species.

Recentemente, foi descoberta uma raça do nematóide-de-cisto-da-soja (NCS; Heterodera glycines) que apresentou a capacidade de quebrar a resistência da cultivar Hartwig, até então considerada resistente a todas as raças conhecidas do nematóide. Essa população foi coletada no Município de Sorriso, Estado do Mato Grosso, e foi caracterizada como raça 4. Para verificar a diversidade genética entre esta e outras populações pertencentes às raças 4 e 9, foi feita uma caracterização molecular pela técnica de marcadores moleculares RAPD. Foram utilizadas nove populações do NCS, das quais quatro apresentavam a capacidade de parasitar 'Hartwig'. Foi verificado que as populações capazes de parasitar 'Hartwig' foram bastante diferentes das demais. Por meio de análise de agrupamento, com base nas distâncias genéticas encontradas, foram obtidos três grupos: o primeiro, constituído por indivíduos classificados como raça 4, mas que não parasitam 'Hartwig'; o segundo, constituído por quatro populações capazes de parasitar 'Hartwig', e o terceiro, por apenas uma população, classificado como raça 9, e que também não parasita 'Hartwig'. Este estudo confirmou que a população de NCS, encontrada em Sorriso, é geneticamente distinta das demais populações da raça 4 encontradas e constitui uma nova raça, denominada 4+.

A aceroleira (Malpighia emarginata DC.) é uma cultura em expansão no Brasil, principalmente, por causa do elevado teor de vitamina C de seus frutos; contudo, ainda são escassas as informações sobre a espécie. Este trabalho teve o objetivo de estudar a atividade e o polimorfismo de alguns sistemas isozimáticos de uma coleção de aceroleiras. Foram estudadas as isozimas IDH, MDH, EST, ACP, GOT, PGM, PGI e POD. Utilizou-se o sistema de eletroforese horizontal em gel de amido e diferentes tampões de gel e cuba. Identificou-se o número de locos e alelos envolvidos no controle genético das enzimas. IDH demonstrou ser dimérica, e foi constatado um loco e dois alelos. PGM apresentou dois locos diméricos, com dois e quatro alelos. EST e POD demonstraram ser monoméricas, com um e dois locos, respectivamente, todos com dois alelos. MDH apresentou um loco onde o seu comportamento é monomérico e outro dimérico, ambos com dois alelos. GOT foi polimórfico mas não foi possível inferir sobre seu controle genético. O polimorfismo revelado pelos sistemas IDH, MDH, POD, EST, GOT e PGM demonstra o potencial de utilização das isozimas como marcadores genéticos em estudos de conservação e melhoramento de aceroleira.

The objective of this work was to study the genetic variability of the grasshopper Rhammatocerus schistocercoides (Orthoptera: Acrididae) using RAPD analysis among individuals from three populations, one from Colombia and two from Brazil (Goiás and Mato Grosso States). Ninety scorable binary markers were obtained by fingerprinting with 11 oligonucleotide primers. Most of the polymorphism was attributed to 42 markers with variable frequency among the different populations. Although the existence of significant difference among populations (P<0.0001), most of the genetic variability was found within populations (87.7% of total variation). Pairwise distances between Colombian and Brazilian populations were 0.12 (P<0.0001) and 0.18 (P<0.0001) for Goiás and Mato Grosso, respectively. The pairwise distance between Goiás and Mato Grosso populations was 0.06 (P<0.0001). These data indicated that the phenotypic differences among populations are associated mainly with the geographical distances between the Brazilian and Colombian populations.

Este trabalho teve como objetivo a obtenção de híbridos somáticos entre o limão 'Cravo' (Citrus limonia Osbeck) e a tangerina 'Cleópatra' (Citrus reshni Hort.), para serem usados como porta-enxertos de citros. O limão 'Cravo' é atualmente o principal porta-enxerto comercial utilizado no Brasil, em virtude de suas boas qualidades agronômicas. Entretanto, é suscetível ao "declínio" dos citros, doença responsável pela eliminação anual de milhões de plantas cítricas no Brasil. A tangerina 'Cleópatra' é uma espécie bastante utilizada como porta-enxerto em outros países e tem sido descrita na literatura como tolerante ao "declínio". Protoplastos de suspensões celulares embriogênicas e protoplastos derivados de tecidos foliares foram utilizados para fusão com solução de PEG (50%) e posterior cultivo em agarose. A porcentagem de obtenção de células híbridas interespecíficas logo após a fusão, variou entre 5,1% e 6,8%. Foram obtidas mais de 500 plantas a partir de produtos de fusão cultivados em gotas de agarose. No total de 180 plantas avaliadas, 11 híbridos somáticos foram discriminados e confirmados, utilizando-se marcadores moleculares RAPD e isoenzimáticos (sistemas PO, IDH e PGI). Seis plantas foram aclimatizadas, plantadas no solo e estão sendo multiplicadas por estaquia para avaliação futura como porta-enxertos de citros.

O aumento do número de plantas daninhas resistentes aos herbicidas inibidores da enzima acetolactato sintase é um tema abordado com freqüência por produtores e comunidade científica. No Brasil, nove espécies já foram documentadas por apresentarem tal problema. O objetivo deste trabalho foi determinar a diversidade genética de populações de leiteira (Euphorbia heterophylla L.) resistentes aos herbicidas inibidores da enzima acetolactato sintase. Quarenta populações de plantas oriundas de sementes coletadas em áreas do Estado do Rio Grande do Sul, Brasil, com suspeita de resistência, foram selecionadas, a partir da aplicação prévia de herbicidas com este mecanismo de ação em casa de vegetação. Vinte plantas de cada população serviram de amostra para a extração de DNA. Trinta marcadores de polimorfismo de DNA amplificado ao acaso (RAPD) foram selecionados, cada um com 10 oligonucleotídeos de seqüência arbitrária. Na análise de agrupamento, cujo coeficiente médio de similaridade foi de 40%, as populações foram separadas em sete grupos. As populações dos municípios de Pontão, Augusto Pestana e Não-me-Toque foram consideradas geneticamente diferentes. Há variabilidade genética relacionada à resistência do herbicida entre as populações de E. heterophylla que ocorrem no planalto do Estado do Rio Grande do Sul.

The objective of this work was to evaluate the processes of selection in a citrus hybrid population using segregation analysis of RAPD markers. The segregation of 123 RAPD markers between 'Cravo' mandarin (Citrus reticulata Blanco) and 'Pêra' sweet orange (C. sinensis (L.) Osbeck) was analysed in a F1 progeny of 94 hybrids. Genetic composition, diversity, heterozygosity, differences in chromosomal structure and the presence of deleterious recessive genes are discussed based on the segregation ratios obtained. A high percentage of markers had a skeweness of the 1:1 expected segregation ratio in the F1 population. Many markers showed a 3:1 segregation ratio in both varieties and 1:3 in 'Pêra' sweet orange, probably due to directional selection processes. The distribution analysis of the frequencies of the segregant markers in a hybrid population is a simple method which allows a better understanding of the genetics of citrus group.

The objective of this work was to construct linkage maps of 'Pêra' sweet orange [Citrus sinensis (L.) Osbeck] and 'Cravo' mandarin (Citrus reticulata Blanco) using RAPD markers and the pseudo-testcross strategy. The parents were chosen according to the resistance/susceptibility to citrus variegate chlorosis (CVC). The segregation of 176 markers was analyzed in 94 progeny of F1 hybrids, which were obtained from controlled crossings. The linkage map of 'Pêra' sweet orange had 117 markers defined by 12 linkage groups, which spanned 612.1 cM. Only six markers could not be linked to the linkage group and 48.7% of the markers showed segregation distortion. The linkage map of 'Cravo' mandarin had 51 markers defined by 12 linkage groups, which spanned 353.3 cM. Only two markers did not link to the groups and 15.7% showed segregation distortion. The construction of linkage maps is relevant to future mapping studies of the inheritance of CVC, citrus canker and leprosis.

In vitro regeneration of Arachis retusa was examined for the purpose of germplasm renewal and conservation. Random amplified polymorphic DNA (RAPD) fingerprinting was used to evaluate the genetic stability of plants derived from embryo axes and apical segments. Ten arbitrary decamer primers were screened and five of them were selected. Ninety genomic regions were evaluated, with an average of 18 loci per clone. All amplified segments were monomorphic. The results indicate that recovered plants are genetically stable at the assessed genomic regions and that both regeneration processes are suitable for in vitro germplasm preservation of Arachis species.

Fatores ambientais contribuem para a produção de sementes de soja com características diferentes do padrão da variedade e a análise visual para determinação de pureza varietal não é suficiente. Marcadores moleculares de DNA podem auxiliar na identificação da pureza genética de sementes, durante o processo de certificação de sementes, uma vez que não sofrem influências ambientais. O objetivo deste trabalho foi avaliar um método de análise da pureza genética de sementes de soja, utilizando marcadores microssatélites. Amostras de DNA de sementes de soja, consideradas atípicas pelo método visual, foram analisadas com microssatélites e comparadas ao padrão da variedade. As amostras de DNA foram analisadas em bulk, o que permitiu diminuir os custos, com a mesma precisão da análise individual. De onze lotes de sementes avaliados como impuros na análise visual, e portanto, eliminados do processo de certificação, apenas quatro apresentaram número de sementes de outras cultivares acima do limite tolerado.

The objectives of this work were to evaluate the frequency of polyembryony, and to identify zygotic and nucellar seedlings of Citrus volkameriana using RAPD. Twenty-five polyembryonic and eight monoembryonic seeds were cultivated in vitrofor six months. DNA from seedlings was extracted and used in combination with five RAPD primers to identify zygotic or nucellar origin of the seedlings. Environmental conditions of the year affected significantly (P<0.05) the morphological characteristics of fruitsand the number ofembryos per seed. Polyembryonic seeds ranged from 30.9%, 44.8% to 54.4% over three years. Morphological characteristic was not correlated with polyembryony. In vitro culture enable all embryos of each seed to grow, favoring the percentage of seedlings identified as zygotic. In polyembryonic and monoembryonic seeds, 25.9% and 87.5% of the seedlings, respectively, were sexually originated. In polyembryonic seeds, not all zygotic seedlings were produced by small embryos located at the micropyle.

The objective of this study was to verify the genetic diversity between and within seven populations of Moxotó goat (n = 264) from the States of Pernambuco, Paraíba and Rio Grande do Norte, using RAPD (Random Amplified Polymorphic DNA). Moxotó, as well as other naturalized breeds, suffers genetic losses due to the indiscriminate miscegenation with breeds raised in the Northeast Region of Brazil. The genetic characterization of these genetic resources is essential to conservation and breeding programs. DNA was extracted from lymphocytes using a non-organic protocol. The 16 primers used were selected from 120 decamer oligonucleotide primers and generated 56 polymorphic bands. The analysis of molecular variance (AMOVA) showed that the greater part of total genetic variability (71.55%) was due to differences between individuals within populations, while 21.21% was among populations. The analysis of variance among the pairs of populations demonstrated that the populations located in Floresta, PE x Angicos, RN presented a smaller value of intrapopulational differentiation (8.9%), indicating low genetic variability among them. Nei's genetic distances varied between 0.0546 and 0.1868 in the populations. The dendrogram generated showed that the Canindé breed, used as outgroup, clustered with the populations of Moxotó, indicating a possible common origin of the naturalized goat breeds.
