The cytotoxicity of three extracts (petroleum ether, ethyl acetate and n-butanol) from a plant used in folk medicine, Marchantia convoluta, to human non-small cell lung carcinoma (H1299) and liver carcinoma (HepG2) cell lines was tested. After 72-h incubation of lung and liver cancer cell cultures with varying concentrations of extracts (15 to 200 µg/mL), cytotoxicity was determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and reported in terms of cell viability. The extracts that showed a significant cytotoxicity were subjected to gas chromatography-mass spectrometry analysis to identify the components. The ethyl acetate, but not the petroleum ether or n-butanol extract, had a significant cytotoxicity against lung and liver carcinoma cells with IC50 values of 100 and 30 µg/mL, respectively. A high concentration of ethyl acetate extract (100 µg/mL) rapidly reduced the number of H1299 cells. At lower concentrations of ethyl acetate extract (15, 30, and 40 µg/mL), the numbers of HepG2 cells started to decrease markedly. Gas chromatography-mass spectrometry analysis of the ethyl acetate extract revealed the presence of several compounds such as phytol (23.42%), 1,2,4-tripropylbenzene (13.09%), 9-cedranone (12.75%), ledene oxide (7.22%), caryophyllene (1.82%), and caryophyllene oxide (1.15%). HPLC analysis result showed that there were no flavonoids in ethyl acetate extract, but flavonoids are abundant in n-butanol extract. Further studies are needed regarding the identification, toxicity, and mechanism of action of active compounds.

The objective of the present study was to investigate factors associated with cesarean sections in two cities located in different regions of Brazil and to determine factors that explain the higher cesarean section rate in the more developed city, Ribeirão Preto, compared to the less developed one, São Luís. Data from two cohort studies comprising 2846 women in Ribeirão Preto in 1994, and 2443 women in São Luís in 1997/1998 were used. Adjusted and non-adjusted risk estimates were calculated using a Poisson regression model. The cesarean section rate was 33.7% in São Luís and 50.8% in Ribeirão Preto. Adjusted analysis in a joint sequential model revealed a 51% higher risk of cesarean section in Ribeirão Preto compared to São Luís (prevalence rate ratio (PRR) = 1.51). Adjustment for category of hospital admission reduced the PRR to 1.09, i.e., this variable explained 82% of the difference in the cesarean section rate between the two cities. Adjustment for the variable "the same physician for prenatal care and delivery" reduced the PRR to 1.07, with the "physician" factor explaining 86% of the difference between rates. When simultaneously adjusted for the two variables, the PRR decreased to 1.05, with these two variables explaining 90% of the difference in the cesarean section rate between the two cities, and the difference was no longer significant. The difference in the cesarean section rate between the two Brazilian cities, one more and one less developed, was mainly explained by the physician factor and, to a lesser extent, by the category of hospital admission.

The pathogenesis of chagasic cardiomyopathy is not completely understood, but it has been correlated with parasympathetic denervation (neurogenic theory) and inflammatory activity (immunogenic theory) that could affect heart muscarinic acetylcholine receptor (mAChR) expression. In order to further understand whether neurogenic and/or immunogenic alterations are related to changes in mAChR expression, we studied two models of Trypanosoma cruzi infection: 1) in 3-week-old male Sprague Dawley rats chronically infected with T. cruzi and 2) isolated primary cardiomyocytes co-cultured with T. cruzi and peripheral blood mononuclear cells (PBMC). Using [³H]-quinuclidinylbenzilate ([³H]-QNB) binding assays, we evaluated mAChR expression in homogenates from selected cardiac regions, PBMC, and cultured cardiomyocytes. We also determined in vitro protein expression and pro-inflammatory cytokine expression in serum and cell culture medium by ELISA. Our results showed that: 1) mAChR were significantly (P < 0.05) up-regulated in right ventricular myocardium (means ± SEM; control: 58.69 ± 5.54, N = 29; Chagas: 72.29 ± 5.79 fmol/mg, N = 34) and PBMC (control: 12.88 ± 2.45, N = 18; Chagas: 20.22 ± 1.82 fmol/mg, N = 19), as well as in cardiomyocyte transmembranes cultured with either PBMC/T. cruzi co-cultures (control: 24.33 ± 3.83; Chagas: 43.62 ± 5.08 fmol/mg, N = 7 for both) or their conditioned medium (control: 37.84 ± 3.84, N = 4; Chagas: 54.38 ± 6.28 fmol/mg, N = 20); 2) [³H]-leucine uptake was increased in cardiomyocytes co-cultured with PBMC/T. cruzi-conditioned medium (Chagas: 21,030 ± 2321; control 10,940 ± 2385 dpm, N = 7 for both; P < 0.05); 3) plasma IL-6 was increased in chagasic rats, IL-1β, was increased in both plasma of chagasic rats and in the culture medium, and TNF-α level was decreased in the culture medium. In conclusion, our results suggest that cytokines are involved in the up-regulation of mAChR in chronic Chagas disease.

The aim of the present study was to evaluate the effect of joint immobilization on morphometric parameters and glycogen content of soleus muscle treated with clenbuterol. Male Wistar (3-4 months old) rats were divided into 4 groups (N = 6 for each group): control, clenbuterol, immobilized, and immobilized treated with clenbuterol. Immobilization was performed with acrylic resin orthoses and 10 µg/kg body weight clenbuterol was administered subcutaneously for 7 days. The following parameters were measured the next day on soleus muscle: weight, glycogen content, cross-sectional area, and connective tissue content. The clenbuterol group showed an increase in glycogen (81.6%, 0.38 ± 0.09 vs 0.69 ± 0.06 mg/100 g; P < 0.05) without alteration in weight, cross-sectional area or connective tissue compared with the control group. The immobilized group showed a reduction in muscle weight (34.2%, 123.5 ± 5.3 vs 81.3 ± 4.6 mg; P < 0.05), glycogen content (31.6%, 0.38 ± 0.09 vs 0.26 ± 0.05 mg/100 mg; P < 0.05) and cross-sectional area (44.1%, 2574.9 ± 560.2 vs 1438.1 ± 352.2 µm²; P < 0.05) and an increase in connective tissue (216.5%, 8.82 ± 3.55 vs 27.92 ± 5.36%; P < 0.05). However, the immobilized + clenbuterol group showed an increase in weight (15.9%; 81.3 ± 4.6 vs 94.2 ± 4.3 mg; P < 0.05), glycogen content (92.3%, 0.26 ± 0.05 vs 0.50 ± 0.17 mg/100 mg; P < 0.05), and cross-sectional area (19.9%, 1438.1 ± 352.2 vs 1724.8 ± 365.5 µm²; P < 0.05) and a reduction in connective tissue (52.2%, 27.92 ± 5.36 vs 13.34 ± 6.86%; P < 0.05). Statistical analysis was performed using Kolmogorov-Smirnov and homoscedasticity tests. For the muscle weight and muscle glycogen content, two-way ANOVA and the Tukey test were used. For the cross-sectional area and connective tissue content, Kruskal-Wallis and Tukey tests were used. This study emphasizes the importance of anabolic pharmacological protection during immobilization to minimize skeletal muscle alterations resulting from disuse.

Reproductive fish behavior is affected by male-female interactions that stimulate physiological responses such as hormonal release and gonad development. During male-female interactions, visual and chemical communication can modulate fish reproduction. The aim of the present study was to test the effect of visual and chemical male-female interaction on the gonad development and reproductive behavior of the cichlid fish Nile tilapia, Oreochromis niloticus (L.). Fifty-six pairs were studied after being maintained for 5 days under one of the four conditions (N = 14 for each condition): 1) visual contact (V); 2) chemical contact (Ch); 3) chemical and visual contact (Ch+V); 4) no sensory contact (Iso) - males and females isolated. We compared the reproductive behavior (nesting, courtship and spawning) and gonadosomatic index (GSI) of pairs of fish under all four conditions. Visual communication enhanced the frequency of courtship in males (mean ± SEM; V: 24.79 ± 3.30, Ch+V: 20.74 ± 3.09, Ch: 0.1 ± 0.07, Iso: 4.68 ± 1.26 events/30 min; P < 0.05, two-way ANOVA with LSD post hoc test), induced spawning in females (3 spawning in V and also 3 in Ch+V condition), and increased GSI in males (mean ± SEM; V: 1.39 ± 0.08, Ch+V: 1.21 ± 0.08, Ch: 1.04 ± 0.07, Iso: 0.82 ± 0.07%; P < 0.05, two-way ANOVA with LSD post hoc test). Chemical communication did not affect the reproductive behavior of pairs nor did it enhance the effects of visual contact. Therefore, male-female visual communication is an effective cue, which stimulates reproduction among pairs of Nile tilapia.

The health-promoting effects of exercise training (ET) are related to nitric oxide (NO) production and/or its bioavailability. The objective of this study was to determine whether single nucleotide polymorphism of the endothelial NO synthase (eNOS) gene at positions -786T>C, G894T (Glu298Asp) and at the variable number of tandem repeat (VNTR) Intron 4b/a would interfere with the cardiometabolic responses of postmenopausal women submitted to physical training. Forty-nine postmenopausal women were trained in sessions of 30-40 min, 3 days a week for 8 weeks. Genotypes, oxidative stress status and cardiometabolic parameters were then evaluated in a double-blind design. Both systolic and diastolic blood pressure values were significantly reduced after ET, which was genotype-independent. However, women without eNOS gene polymorphism at position -786T>C (TT genotype) and Intron 4b/a (bb genotype) presented a better reduction of total cholesterol levels (-786T>C: before = 213 ± 12.1, after = 159.8 ± 14.4, Δ = -24.9% and Intron 4b/a: before = 211.8 ± 7.4, after = 180.12 ± 6.4 mg/dL, Δ = -15%), and LDL cholesterol (-786T>C: before = 146.1 ± 13.3, after = 82.8 ± 9.2, Δ = -43.3% and Intron 4b/a: before = 143.2 ± 8, after = 102.7 ± 5.8 mg/dL, Δ = -28.3%) in response to ET compared to those who carried the mutant allele. Superoxide dismutase activity was significantly increased in trained women whereas no changes were observed in malondialdehyde levels. Women without eNOS gene polymorphism at position -786T>C and Intron 4b/a showed a greater reduction of plasma cholesterol levels in response to ET. Furthermore, no genotype influence was observed on arterial blood pressure or oxidative stress status in this population.

We investigated the association between pulse wave velocity (PWV) and HIV infection, antiretroviral treatment-related characteristics, viral load, immune status, and metabolic changes in a cross-sectional study nested in a cohort of HIV/AIDS patients who have been followed for metabolic and cardiovascular changes since 2007. The study included patients recruited from the cohort (N = 261) and a comparison group (N = 82) of uninfected individuals, all enrolled from April to November 2009. Aortic stiffness was estimated using the carotid-femoral PWV (Complior-Artech, Paris, France). The groups were similar with respect to age, metabolic syndrome, diabetes mellitus, Framingham score, and use of antihypertensive and hypolipidemic medications. Hypertension was more frequent among the controls. Individuals with HIV had higher triglyceride, glucose and HDL cholesterol levels. Among individuals with HIV/AIDS, those with a nadir CD4+ T-cell count <200 cells/mm³ had a higher PWV (P = 0.01). There was no statistically significant difference when subjects were stratified by gender. Heart rate, age, male gender, and blood pressure were independently correlated with PWV. Nadir CD4+ T-cell count did not remain in the final model. There was no significance difference in PWV between HIV-infected individuals and uninfected controls. PWV was correlated with age, gender, and blood pressure across the entire population and among those infected with HIV. We recommend cohort studies to further explore the association between inflammation related to HIV infection and/or immune reconstitution and antiretroviral use and PWV.

Blood pressure (BP) and physical activity (PA) levels are inversely associated. Since genetic factors account for the observed variation in each of these traits, it is possible that part of their association may be related to common genetic and/or environmental influences. Thus, this study was designed to estimate the genetic and environmental correlations of BP and PA phenotypes in nuclear families from Muzambinho, Brazil. Families including 236 offspring (6 to 24 years) and their 82 fathers and 122 mothers (24 to 65 years) were evaluated. BP was measured, and total PA (TPA) was assessed by an interview (commuting, occupational, leisure time, and school time PA). Quantitative genetic modeling was used to estimate maximal heritability (h²), and genetic and environmental correlations. Heritability was significant for all phenotypes (systolic BP: h² = 0.37 ± 0.10, P < 0.05; diastolic BP: h² = 0.39 ± 0.09, P < 0.05; TPA: h² = 0.24 ± 0.09, P < 0.05). Significant genetic (r g) and environmental (r e) correlations were detected between systolic and diastolic BP (r g = 0.67 ± 0.12 and r e = 0.48 ± 0.08, P < 0.05). Genetic correlations between BP and TPA were not significant, while a tendency to an environmental cross-trait correlation was found between diastolic BP and TPA (r e = -0.18 ± 0.09, P = 0.057). In conclusion, BP and PA are under genetic influences. Systolic and diastolic BP share common genes and environmental influences. Diastolic BP and TPA are probably under similar environmental influences.

A atividade antimicrobiana dos ácidos anacárdicos do óleo da casca da castanha de caju (CNSL) Anacardium occidentale (Anacardiaceae) foi estudada sobre os microrganismos da cavidade bucal Streptococcus mutans ATCC 25175, Staphylococcus aureus ATCC 12598, Candida albicans ATCC 10231 e Candida utilis. Os ácidos anacárdicos obtidos dos extratos etílicos do CNSL apresentaram atividade antibacteriana contra os microganismos citados, porém a maior atividade inibitória ocorreu sobre a bactéria Gram positiva Streptococcus mutans, considerada predominante na cárie dentária. As cáries dentárias são uma das mais freqüentes doenças infecciosas nos países em desenvolvimento. Os elementos que influenciam na cárie dentária incluem o estado nutricional, a ingestão de açúcar e a presença da microbiota cariogênica.

O objetivo desse trabalho foi avaliar os efeitos de métodos de abate (MA) (humanitário (MH) e por tiro (MT)) e sexo na composição centesimal, perfil de ácidos graxos (AG) e colesterol da carne de capivara (Hydrochaeris hydrochaeris L. 1766). Foram abatidos 20 animais (13 machos (M) e 7 fêmeas (F)) com peso médio de 45,71kg. No músculo longissimus dorsi (LD) foram determinados: umidade, lipídios totais (LT), proteína e cinzas e no semimembranosus (SM), o colesterol por colorimetria e o perfil de AG por cromatografia gasosa. O LD apresentou: 75,87% de umidade, 1,37% de LT, 22,11% de proteínas e 1,09% de cinzas. Houve diferença (p<0,01) nos teores de LT de M (1,75%) e F (0,98%). Os fatores sexo e MA não influenciaram: umidade, proteína e cinzas. Os AG encontrados em maior proporção (%) foram: C16:0 (29,57); C18:1w9 (27,87); C18:2w6 (19,19); C18:0 (6,57); C18:3w3 (4,97); C14:0 (3,64); C20:4w6 (3,45); C18:1w7 (3,31) e C16:1w7 (1,90). A relação AG poliinsaturados/saturados foi 0,82. As médias de AG w6 e w3 foram 23,41 e 5,63%. Os fatores sexo e MA não influenciaram os poliinsaturados e colesterol (28,11mg/100g). A carne de capivara apresentou teores elevados de proteína e valores reduzidos de lipídios totais e de colesterol e relação w6/w3 nutricionalmente adequada.

O vongole (Anomalocardia brasiliana) tem importância socioeconômica para grande parte da população da região da Baía de Todos os Santos, na costa da Bahia, por fazer parte da dieta familiar e ser uma das principais fontes de renda. Para ampliar a comercialização deste molusco foi desenvolvido um processo de obtenção de marinado, envolvendo acidificação do produto com vinagre (pH<4,5) e tratamento térmico convencional (banho-maria) durante 30 minutos. O Índice de Aceitabilidade do marinado foi de 78-82%, em relação à aparência, cor, aroma, sabor e textura. Sob os pontos-de-vista microbiológico, físico-químico, químico e sensorial, o marinado de vongole manteve-se estável durante 240 dias de armazenamento, a temperatura ambiente. O processo proposto atende a possibilidade de um maior e melhor aproveitamento comercial do vongole.

Propriedades termofísicas, densidade e viscosidade de soluções-modelo similares a sucos foram determinadas experimentalmente. Os resultados foram comparados aos preditos por modelos matemáticos (STATISTICA 6.0) e obtidos da literatura em função da sua composição química. Para definição das soluções-modelo, foi realizado um planejamento estrela, mantendo-se fixa a quanti-dade de ácido (1,5%) e variando-se a água (82-98,5%), o carboidrato (0-15%) e a gordura (0-1,5%). A densidade foi determinada em picnômetro. A viscosidade foi determinada em viscosímetro Brookfield modelo LVF. A condutividade térmica foi calculada com o conhecimento das propriedades difusividade térmica e calor específico (apresentados na Parte I deste trabalho MOURA [7]) e da densidade. Os resultados de cada propriedade foram analisados através de superfícies de respostas. Foram encontrados resultados significativos para as propriedades, mostrando que os modelos encontrados representam as mudanças das propriedades térmicas e físicas dos sucos, com alterações na composição e na temperatura.

Devido a necessidade da presença de cobre nos alambiques e as exigências cada vez mais acirradas com relação ao seu teor no destilado, é de interesse o desenvolvimento de metodologias para o controle deste metal nos destilados e/ou a sua remoção até níveis aceitáveis, em conformidade com as legislações nacional e internacional. Este trabalho investigou a potencialidade de dois sólidos: sílica modificada com trimetóxisili(propil)etilenodiamina (Si-Dia), e sílica-titânia, também modificada com trimetóxisili(propil)etilenodiamina (Si-Ti-Dia), como agentes removedores de íons Cu2+. Foram empregadas tanto soluções padrão de Cu2+, com teor alcoólico que simulavam aguardentes, como amostras reais de destilados. Os resultados indicam que um grama das matrizes Si-Dia e Si-Ti-Dia adsorvem 1,39 e 73,5mmol de Cu2+, respectivamente, a partir das soluções padrão, nas seguintes condições: tempo de contato 22h, pH 4,0; temperatura 25 ± 1,0°C e agitação constante. Para amostras reais, sob as mesmas condições de análises, o sólido Si-Dia reduziu as concentrações dos íons divalentes Cu, Zn e Fe em, respectivamente, 82, 83 e 64%, enquanto o Si-Ti-Dia, reduziu os mesmos íons em 88, 31 e 22%, respectivamente.

Entre as hortaliças minimamente processadas, a cenoura é uma das mais populares, sendo comercializada de várias maneiras: raladas, cortadas em fatias, palitos, e ainda apresentadas na forma de mini-cenoura (baby carrot). O objetivo deste estudo foi determinar as taxas respiratórias de cenouras (Daucus carota) da cultivar Nantes minimamente processadas. O armazenamento foi realizado nas temperaturas de 1 °C, 5 °C e 11 °C, e 90% UR. A taxa respiratória foi determinada usando um fluxo contínuo de ar. O teor de CO2 e etileno foi medido por um cromatógrafo a gás. A taxa de respiração para as cenouras fatiadas foi mais alta do que para os produtos inteiros. Os valores da energia de ativação obtidos para as cenouras fatiadas e inteiras foi de 69,82 kJmol-1 e 54,60 kJmol-1, respectivamente. A produção de etileno foi insignificante para as cenouras durante os 14 dias de armazenamento.

O objetivo deste trabalho foi estudar o desempenho zootécnico da ema (Rhea americana), rendimentos do abate, composição química e análise sensorial da carne. Onze animais foram submetidos a um sistema de confinamento, recebendo ração balanceada à vontade e com acesso a um piquete. Antes do abate as emas permaneceram em jejum hídrico por 12 h, foram pesadas e submetidas ao processamento: insensibilização, sangria, depenagem manual, esfola e evisceração. As emas apresentaram peso vivo médio, no momento do abate, de 19,393 kg, desempenho médio de 529,82 de consumo de ração/dia, 53,20 g de ganho de peso/dia e 9,44 de conversão alimentar. Em relação ao peso de carcaça, mostraram rendimento médio de 63,33% de carne, 5,09% de gordura e 22% de ossos. A composição centesimal média da carne de ema foi de 22,81% de proteína, 1,59% de lipídios, 1,62% de cinzas e de 74,72% de umidade. O conteúdo de colágeno variou de 0,94% a 1,12% e o teor de colesterol foi de 65,44 mg por 100 g de carne. Pela análise sensorial, verificou-se que a carne de ema processada foi bem aceita pelos provadores. Assim, admite-se que a carne de ema pode ser consumida como fonte de proteína animal com baixo teor de lipídios.
