The aim of this study was to determine the weed strip control (WSC) required for adequate coffee growth after transplanting. A non-irrigated, field-planted (spaced 3.80 x 0.70 m) crop was used. The experimental design was a randomized block, with four replicates. The treatments were arranged in a 9 x 18 split-plot design to test the WSC of 0, 15, 30, 45, 60, 90, 120, 150, and 190 cm, which involved continuously hand-weeding at each side of the coffee row, and 18 coffee growth measurements. Multiple regression analyses were carried out relating to growth-variables as a function of both WSC and growth-evaluation times. Brachiaria decumbens was the main weed accomplishing 88.5% of the total weed dry mass. The minimum width of the WSC increases as the crop ages after transplanting. Assuming reductions of 2% and 5% in the maximum coffee growth, the recommended WSC was 75 and 52 cm at 4 months after transplanting (MAT), 104 and 85 cm at 6 MAT, 123 and 105 cm at 9 MAT, 134 and 116 cm at 12 MAT, 142 and 124 cm at 15 MAT, and 148 and 131 cm at 18 MAT, respectively. It was concluded that integrated weed management in young coffee crops must focus on the weed control only in a minimum range along coffee rows, which increases with coffee plant age, keeping natural vegetation in the inter-rows.

O objetivo deste estudo foi avaliar o impacto da interferência de Euphorbia heterophylla (EPHHL) em diferentes densidades e épocas de semeadura sobre o rendimento do feijoeiro e seus componentes e o nível de dano econômico (NDE). O experimento foi conduzido em campo em delineamento de blocos ao acaso, em parcelas subdivididas, com quatro repetições. Nas parcelas principais foram alocadas oito densidades de EPHHL (0, 6, 10, 17, 29, 49, 83 e 142 pl m-2) e, nas subparcelas, duas épocas de semeadura da infestante (12 dias antes da semeadura do feijoeiro e simultaneamente à cultura). As perdas de rendimento da cultura do feijão foram diretamente proporcionais à densidade de plantas de EPHHL. A implantação simultânea das espécies cultivada e daninha resultou em perda máxima de 48% no rendimento de grãos de feijão, enquanto a semeadura de EPHHL 12 dias antes do feijoeiro resultou em perda de 60% no rendimento de grãos. O número de legumes por planta foi o principal componente do rendimento afetado pela competição. Cada planta de EPHHL reduziu em 2,4 e 5,5% o rendimento de grãos de feijoeiro, para as situações de implantação simultânea e 12 dias antes da semeadura da cultura, respectivamente. Análise de sensibilidade indicou que os valores de NDE dependeram da época de implantação da planta daninha, da expectativa de rendimento da cultura, da eficiência do herbicida, do valor do produto colhido e do custo do controle. Entretanto, em todas as simulações, o NDE foi muito baixo (<3,3 plantas m-2), enfatizando a necessidade de herbicidas residuais para evitar perdas econômicas na cultura.

Foram realizados estudos da composição florística e fitossociologia de 1 ha de floresta ciliar do rio Iapó (bacia do rio Tibagi), município de Tibagi, PR (24º31S e 50º25W) utilizando-se 100 parcelas contíguas de 10 x 10 m, tendo-se como critério de inclusão um diâmetro à altura do peito (DAP) mínimo de 5 cm. Para cada espécie amostrada foram estimados parâmetros relativos à freqüência, densidade e dominância, além do índice do valor de importância (IVI) e índice do valor de cobertura (IVC). O levantamento resultou em 1594 indivíduos pertencentes a 127 espécies, 81 gêneros e 43 famílias. As espécies mais importantes em IVI e IVC foram Eugenia blastantha, Faramea porophylla, Casearia obliqua, Nectandra grandiflora, Sebastiania commersoniana, Casearia sylvestris e Actinostemon concolor. As três famílias com maior IVI foram Myrtaceae, Lauraceae e Euphorbiaceae, sendo que Lauraceae possui 15 espécies e 142 indivíduos, Myrtaceae, 14 espécies e 280 indivíduos e Euphorbiaceae, cinco espécies e 274 indivíduos. O índice de diversidade de Shannon-Weaver encontrado foi de 3,67.

Através da análise de 18 locos isoenzimáticos polimórficos, foram estimadas as freqüências alélicas referentes a 214 indivíduos adultos de quatro populações naturais de Cryptocarya moschata de duas regiões do estado de São Paulo. Com base nas heterozigosidades observadas e esperadas, foram obtidas estimativas das estatísticas F de Wright. Para fins de comparação, utilizou-se também o método da análise da variância para estimação dos parâmetros correspondentes F = FIT, qP= FST e f = FIS. Os dois métodos forneceram resultados bastante concordantes: IT = 0,142; ST = 0,140; IS = 0,002 e = 0,116; P = 0,123 e = 0,008, indicando que os indivíduos dentro das populações devem ser panmíticos e que a diversidade entre populações é bastante alta, sendo similar à que se espera para famílias com estruturação de meios-irmãos. Calculando P com as populações tomadas duas a duas, notou-se tendência de P crescer com a distância geográfica o que sugere tendência de isolamento pela distância. O fluxo gênico foi estimado em 0,9 indivíduos por geração, o que corrobora a pronunciada diferenciação populacional encontrada. Devido ao valor negligível encontrado de IS, o tamanho efetivo de variância de cada população é equivalente ao número de indivíduos amostrados. As estratégias de manejo e conservação necessárias para a preservação da alta variabilidade genética intrapopulacional de C. moschata implicam na manutenção de populações com número grande de indivíduos. Além disso, para a preservação da espécie como um todo, a manutenção de muitas populações provavelmente é necessária.

O objetivo deste trabalho foi investigar a taxa de imigração e a distância de dispersão de pólen e sementes em uma pequena população fragmentada de Copaifera langsdorffii Desf., com base no polimorfismo de oito locos microssatélites. Foram mapeadas e genotipadas todas as 47 árvores adultas e 65 subadultas existentes no fragmento. Adultos tinham um total número menor de alelos (142) do que subadultos (164), sendo 20 alelos exclusivos aos adultos e 41 aos subadultos. Adultos tinham significativamente menor diversidade genética e maior endogamia (Â = 17,75 ± 0,51; Âe = 8,97 ± 0,27; ^F = 0,191 ± 0,017) do que subadultos (Â = 20,5 ± 0,65; Âe = 10,86 ± 0,29; ^F = 0,119 ± 0,013). O poder de exclusão do primeiro parente foi alto para o conjunto de locos nos adultos (P1º Parente = 0,9994), mostrando que estes oito locos microssatélites têm alto poder de resolver testes de parentescos. Dos 65 subadultos, foi encontrado o parental materno para 12 e o materno e paterno para apenas dois indivíduos, o que indica uma taxa de imigração de sementes e pólen na população de 81% (m sementes) e 97% (m pólen), respectivamente. Estes resultados sugerem uma alta taxa de fluxo gênico no passado. A distância média de dispersão de sementes foi de 38,4 m. Os resultados evidenciam que a população apresenta altos níveis de diversidade genética, devido a uma intensa taxa de imigração de sementes e pólen.

The aims of this study were to investigate the mating system of a fragmented population of the dioecious tropical tree Myracrodruon urundeuva Allemão, using five microsatellite loci and the mixed mating and correlated mating models. The study was conducted in the Estação Ecológica de Paulo de Farias (436 ha), where the population occupies about 142 ha. The mating system was estimated using 514 open-pollinated offspring, collected from 30 seed-trees. Estimates of the multilocus outcrossing rate confirm that the species is dioecious (t m = 1.0). Low levels of mating among relatives were detected in the population (1 - t s = 0.020). The estimate of paternity correlation (r p(m)) indicated that offsprings were composed of mixtures of half-sibs and full-sibs, with the latter occurring at a low frequency (average of 0.148). The estimated coancestry coefficient within families (Θ = 0.147) was larger and the effective population size (Ne(v)) was lower (Ne(v) = 2.98) than expected in progenies from panmictic populations (Θ = 0.125, Ne(v) = 4, respectively). In terms of conservation, the results indicate that to retain an effective population size of 150, is necessary to collect seeds from at least 50 seed-trees.

The influence of afterload on the rate of force generation by the myocardium was investigated using two types of preparations: the in situ dog heart (dP/dt) and isolated papillary muscle of rats (dT/dt). Thirteen anesthetized, mechanically ventilated and thoracotomized dogs were submitted to pharmacological autonomic blockade (3.0 mg/kg oxprenolol plus 0.5 mg/kg atropine). A reservoir connected to the left atrium permitted the control of left ventricular end-diastolic pressure (LVEDP). A mechanical constriction of the descending thoracic aorta allowed to increase the systolic pressure in two steps of 20 mmHg (conditions H1 and H2) above control values (condition C). After arterial pressure elevations (systolic pressure C: 119 ± 8.1; H1: 142 ± 7.9; H2 166 ± 7.7 mmHg; P<0.01), there were no significant differences in heart rate (C: 125 ± 13.9; H1: 125 ± 13.5; H2: 123 ± 14.1 bpm; P>0.05) or LVEDP (C: 6.2 ± 2.48; H1: 6.3 ± 2.43; H2: 6.1 ± 2.51 mmHg; P>0.05). The values of dP/dt did not change after each elevation of arterial pressure (C: 3,068 ± 1,057; H1: 3,112 ± 996; H2: 3,086 ± 980 mmHg/s; P>0.05). In isolated rat papillary muscle, an afterload corresponding to 50% and 75% of the maximal developed tension did not alter the values of the maximum rate of tension development (100%: 78 ± 13; 75%: 80 ± 13; 50%: 79 ± 11 g mm-2 s-1, P>0.05). The results show that the rise in afterload per se does not cause changes in dP/dt or dT/dt

The clinical spectrum of leishmaniasis and control of the infection are influenced by the parasite-host relationship. The role of cellular immune responses of the Th1 type in the protection against disease in experimental and human leishmaniasis is well established. In humans, production of IFN-g is associated with the control of infection in children infected by Leishmania chagasi. In visceral leishmaniasis, an impairment in IFN-g production and high IL-4 and IL-10 levels (Th2 cytokines) are observed in antigen-stimulated peripheral blood mononuclear cells (PBMC). Moreover, IL-12 restores IFN-g production and enhances the cytotoxic response. IL-10 is the cytokine involved in down-regulation of IFN-g production, since anti-IL-10 monoclonal antibody (mAb) restores in vitro IFN-g production and lymphoproliferative responses, and IL-10 abrogates the effect of IL-12. In cutaneous and mucosal leishmaniasis, high levels of IFN-g are found in L. amazonensis-stimulated PBMC. However, low or absent IFN-g levels were observed in antigen-stimulated PBMC from 50% of subjects with less than 60 days of disease (24 ± 26 pg/ml). This response was restored by IL-12 (308 ± 342 pg/ml) and anti-IL-10 mAb (380 ± 245 pg/ml) (P<0.05). Later during the disease, high levels of IFN-g and TNF-a are produced both in cutaneous and mucosal leishmaniasis. After treatment there is a decrease in TNF-a levels (366 ± 224 pg/ml before treatment vs 142 ± 107 pg/ml after treatment, P = 0.02). Although production of IFN-g and TNF-a might be involved in the control of parasite multiplication in the early phases of Leishmania infection, these cytokines might also be involved in the tissue damage seen in tegumentary leishmaniasis

Thiobarbituric acid reactant substances (TBARs) content, and the activities of glucose-6-phosphate dehydrogenase (G6PDh), citrate synthase (CS), Cu/Zn- and Mn-superoxide dismutase (SOD), catalase, and glutathione peroxidase (GPX) were measured in the lymphoid organs (thymus, spleen, and mesenteric lymph nodes (MLN)) and skeletal muscles (gastrocnemius and soleus) of adrenodemedullated (ADM) rats. The results were compared with those obtained for sham-operated rats. TBARs content was reduced by adrenodemedullation in the lymphoid organs (MLN (28%), thymus (40%) and spleen (42%)) and gastrocnemius muscle (67%). G6PDh activity was enhanced in the MLN (69%) and reduced in the spleen (28%) and soleus muscle (75%). CS activity was reduced in all tissues (MLN (75%), spleen (71%), gastrocnemius (61%) and soleus (43%)), except in the thymus which displayed an increment of 56%. Cu/Zn-SOD activity was increased in the MLN (126%), thymus (223%), spleen (80%) and gastrocnemius muscle (360%) and was reduced in the soleus muscle (31%). Mn-SOD activity was decreased in the MLN (67%) and spleen (26%) and increased in the thymus (142%), whereas catalase activity was reduced in the MLN (76%), thymus (54%) and soleus muscle (47%). It is particularly noteworthy that in ADM rats the activity of glutathione peroxidase was not detectable by the method used. These data are consistent with the possibility that epinephrine might play a role in the oxidative stress of the lymphoid organs. Whether this fact represents an important mechanism for the establishment of impaired immune function during stress remains to be elucidated.

The most important component of the upper esophageal sphincter (UES) is the cricopharyngeal muscle. During the measurement of sphincter pressure the catheter passed through the sphincter affects the pressure value. In Chagas' disease and primary achalasia there is an esophageal myenteric plexus denervation which may affect UES pressure. We measured the UES pressure of 115 patients with Chagas' disease, 28 patients with primary achalasia and 40 healthy volunteers. We used a round manometric catheter with continuous perfusion and the rapid pull-through method, performed in triplicate during apnea. Pressures were measured in four directions, and the direction with the highest pressure (anterior/posterior) and the average of the four directions were measured. The highest UES pressure in Chagas' disease patients without abnormalities upon radiologic esophageal examination (N = 63) was higher than in normal volunteers (142.8 ± 47.4 mmHg vs 113.0 ± 46.0 mmHg, mean ± SD, P<0.05). There was no difference in UES pressure between patients with primary achalasia and patients with Chagas' disease and similar esophageal involvement and normal volunteers (P>0.05). There was no difference between patients with or without esophageal dilation. In the group of subjects less than 50 years of age the UES pressure of primary achalasia (N = 21) was lower than that of Chagas' disease patients with normal radiologic esophageal examination (N = 41), measured at the site with the highest pressure (109.3 ± 31.5 mmHg vs 149.6 ± 45.3 mmHg, P<0.01) and as the average of the four directions (64.2 ± 17.1 mmHg vs 83.5 ± 28.6 mmHg, P<0.05). We conclude that there is no difference in UES pressure between patients with Chagas' disease, primary achalasia and normal volunteers, except for patients with minor involvement by Chagas' disease, for whom the UES pressure at the site with the highest pressure was higher than the pressure of normal volunteers and patients with primary achalasia.

In the present study we investigated the effect of salt intake on myenteric neuron size of the colon of adult male Wistar rats. The animals were placed on either a high-salt (HS; 8%; 12 animals) or a low-salt diet (LS; 0.15%; 12 animals) for 15 or 52 weeks and blood pressure was measured. The sizes of myenteric neurons of the distal colon from both groups were measured. No difference in neuron size was observed between the HS and LS groups after 15 weeks. After 52 weeks on HS, neuron size was increased (P<0.005) when compared with the LS group. The rats also presented hypertension, which was significantly different at 52 weeks (142 ± 11 vs 119 ± 7 mmHg). These results suggest that a long time on an HS diet can significantly increase myenteric nerve cell size.

Previous genetic association studies have overlooked the potential for biased results when analyzing different population structures in ethnically diverse populations. The purpose of the present study was to quantify this bias in two-locus association studies conducted on an admixtured urban population. We studied the genetic structure distribution of angiotensin-converting enzyme insertion/deletion (ACE I/D) and angiotensinogen methionine/threonine (M/T) polymorphisms in 382 subjects from three subgroups in a highly admixtured urban population. Group I included 150 white subjects; group II, 142 mulatto subjects, and group III, 90 black subjects. We conducted sample size simulation studies using these data in different genetic models of gene action and interaction and used genetic distance calculation algorithms to help determine the population structure for the studied loci. Our results showed a statistically different population structure distribution of both ACE I/D (P = 0.02, OR = 1.56, 95% CI = 1.05-2.33 for the D allele, white versus black subgroup) and angiotensinogen M/T polymorphism (P = 0.007, OR = 1.71, 95% CI = 1.14-2.58 for the T allele, white versus black subgroup). Different sample sizes are predicted to be determinant of the power to detect a given genotypic association with a particular phenotype when conducting two-locus association studies in admixtured populations. In addition, the postulated genetic model is also a major determinant of the power to detect any association in a given sample size. The present simulation study helped to demonstrate the complex interrelation among ethnicity, power of the association, and the postulated genetic model of action of a particular allele in the context of clustering studies. This information is essential for the correct planning and interpretation of future association studies conducted on this population.

Hydration is recommended in order to decrease the overload on the cardiovascular system when healthy individuals exercise, mainly in the heat. To date, no criteria have been established for hydration for hypertensive (HY) individuals during exercise in a hot environment. Eight male HY volunteers without another medical problem and 8 normal (NO) subjects (46 ± 3 and 48 ± 1 years; 78.8 ± 2.5 and 79.5 ± 2.8 kg; 171 ± 2 and 167 ± 1 cm; body mass index = 26.8 ± 0.7 and 28.5 ± 0.6 kg/m²; resting systolic (SBP) = 142.5 and 112.5 mmHg and diastolic blood pressure (DBP) = 97.5 and 78.1 mmHg, respectively) exercised for 60 min on a cycle ergometer (40% of VO2peak) with (500 ml 2 h before and 115 ml every 15 min throughout exercise) or without water ingestion, in a hot humid environment (30ºC and 85% humidity). Rectal (Tre) and skin (Tsk) temperatures, heart rate (HR), SBP, DBP, double product (DP), urinary volume (Vu), urine specific gravity (Gu), plasma osmolality (Posm), sweat rate (S R), and hydration level were measured. Data were analyzed using ANOVA in a split plot design, followed by the Newman-Keuls test. There were no differences in Vu, Posm, Gu and S R responses between HY and NO during heat exercise with or without water ingestion but there was a gradual increase in HR (59 and 51%), SBP (18 and 28%), DP (80 and 95%), Tre (1.4 and 1.3%), and Tsk (6 and 3%) in HY and NO, respectively. HY had higher HR (10%), SBP (21%), DBP (20%), DP (34%), and Tsk (1%) than NO during both experimental situations. The exercise-related differences in SBP, DP and Tsk between HY and NO were increased by water ingestion (P < 0.05). The results showed that cardiac work and Tsk during exercise were higher in HY than in NO and the difference between the two groups increased even further with water ingestion. It was concluded that hydration protocol recommended for NO during exercise could induce an abnormal cardiac and thermoregulatory responses for HY individuals without drug therapy.
