This paper reports the isolation of St. Louis encephalitis virus (SLEV) from a febrile human case suspected to be dengue, in São Pedro, São Paulo State. A MAC-ELISA done on the patient's acute and convalescent sera was inconclusive and hemagglutination inhibition test detected IgG antibody for flaviviruses. An indirect immunofluorescent assay done on the C6/36 cell culture inoculated with the acute serum was positive for flaviviruses but negative when tested with dengue monoclonal antibodies. RNA extracted from the infected cell culture supernatant was amplified by RT-PCR in the presence of NS5 universal flavivirus primers and directly sequenced. Results of BLAST search indicated that this sequence shares 93% nucleotide similarity with the sequence of SLEV (strain-MSI.7), confirmed by RT-PCR performed with SLEV specific primers. Since SLEV was identified as the cause of human disease, it is necessary to improve surveillance in order to achieve early detection of this agent in the state of São Paulo and in Brazil. This finding is also an alert to health professionals about the need for more complete clinical and epidemiological investigations of febrile illnesses as in the reported case. SLEV infections can be unrecognized or confused with other ones caused by an arbovirus, such as dengue.

RESUMO A espécie Anacardium humile (St. Hilaire) é uma fruteira nativa do Cerrado, utilizada tradicionalmente, tanto no consumo in natura como na forma de sucos, doces e geleias, mas ainda pouco explorada economicamente. Em uma área de cerrado campo sujo, com transição para cerrado campo rupestre, localizada no município de Patrocínio, Minas Gerais, Brasil, 38 genótipos de uma população dessa espécie foram estudados; com o objetivo de avaliar a diversidade fenotípica, estimada por análise multivariada, bem como características morfológicas promissoras para programas de melhoramento e conservação. Assim, o estudo mostrou variabilidade genética satisfatória dentro da população estudada, destacando-se os genótipos 6, 10, 24, 36 e 38 por elevada produção de frutos e maiores pedúnculos e por se apresentarem distantes geneticamente, sendo indicados para futuros trabalhos de melhoramento genético, para serem multiplicados e utilizados em plantios comerciais e de conservação da espécie.

Foi elaborado um instrumento de medida baseado na Técnica Q de W. Stephenson, cujas operações exigidas são semelhantes às primeiras etapas de construção de uma Escala Thurstone, pelo método de intervalos aparentemente iguais. Utilizando-se desse instrumento, foi realizado estudo sobre a "Percepção da Função do Educador de Saúde Pública, na Equipe de Saúde", desenvolvido em duas fases: a primeira, compreendendo a construção do Instrumento, propriamente dito, e a segunda, a execução de uma pesquisa exploratória fazendo uso do referido instrumento. São descritos todos os passos da primeira fase, que compreende: a coleta de informações sobre as atribuições da função do educador de saúde pública; listagem das mesmas, convertendo-as em afirmações transcritas em cartões: seleção de uma amostra de 70 cartões, por um corpo de juizes altamente especializado; tabulação dos dados obtidos e a definição do instrumento definitivo. Para preencher os requisitos de validade, a distribuição escalar deveria apresentar um certo equilíbrio entre as afirmações não pertinentes, intermediárias e pertinentes. Para a conveniência estatística e para garantir este equilíbrio, cada juiz foi instruído a colocar números variados de cartões em várias pilhas, fazendo de sua totalidade um continuum que se aproximasse de uma distribuição normal ou quase normal. Este equilíbrio foi alcançado e a distribuição de freqüência das afirmações amostradas pelos juizes demonstrou ótima concordância interna, o que permitiu considerar satisfatório este estudo. As 70 afirmações foram confirmadas como componentes definitivo do instrumento de medida PF-ESP-Percepção da Função do Educador de Saúde Pública, para a utilização na segunda fase desse estudo.

Los autores hacen una revisión de la problemática referida a la fiebre Q, tanto desde el punto de vista clínico como epidemiológico. El primer caso nacional se comunicó en el año 1956, en un adulto, obrero de frigorífico. En 1966 se comunica el primer caso en un niño. A partir de esa fecha se realizan varias encuestas serológicas en humanos y en animales. En seres humanos los hallazgos varían entre 4.2% y 5.5% según el año estudiado. En animales se hallaron valores que oscilaron entre 0.9% (animal para abasto) y 30% (animal tipo industria) para bovinos. En ovinos, en el único estudio se halló un 10.3% de positivos. En equinos los valores de positividad variaron de 5.5% a 21.7%. En suinos se refiere un porcentaje de positividad del 21.2% en la primera encuesta, sendo en la más reciente del 0.0%. En aves y cobayos no se evidenciaron sueros positivos. Entre 1975 y 1985 los autores estudiaron 14 brotes en seres humanos en esta-blecimentos de procesamiento de carne, con un total de 1358 casos estudiados, habiéndose confirmado el 60% de ellos. Pasan revista a la sintomatologia común, así como el tipo de tarea de los pacientes. Para los estudios serológicos, como los diagnósticos de los casos clínicos, los autores utilizaron la fijación del complemento, la aglutinación capilar y la microa-glutinación en lámina.

An in house indirect immmunofluorescence assay ( IFA ) in relation to neutralization (NT) reference test, was assessed as a fast and cheap method to carry out serological surveys for St. Louis Encephalitis virus (SLE). Sera obtained from 213 blood donors were analyzed by both tests. The prevalence of seropositivity obtained with IFA was lower than (30.98%) that observed on NT (41.78%). The relative specificity rate of IFA was 96.77% whereas its relative sensitivity rate was 69.66%. Kappa index showed a good correlation between both tests. The results indicate that neutralization assay is still the serological test with the highest sensitivity and specificity relative rates for detecting antibodies against SLE virus. Nevertheless, the IFA could be useful as an alternative test in order to learn the circulation of the Flavivirus genus in a certain area.

A serological screening was performed in 615 individuals aged 0-87 years, living in the city of Cordoba, Argentina to study the relationship between antibody prevalence for the SLE virus and age. A 13.98% prevalence of neutralizing antibodies was obtained and its relation to age was significantly high (p = 0.045). The highest seroprevalence was noted on individuals over 60 years old (>20%), whereas no subject under 10 was seropositive for this virus. Our results confirm that the agent is endemic in this area and neurological pathology studies should be performed on those individuals aged 60 since they represent the most susceptible group to SLE virus.

Q fever has been considered non-existing in Brazil where reports of clinical cases still cannot be found. This case-series of 16 patients is a result of a systematic search for such illness by means of clinical and serologic criteria. Serologic testing was performed by the indirect microimmunofluorescence technique using phase I/II C. burnetii antigens. Influenza-like syndrome was the most frequent clinical form (eight cases - 50%), followed by pneumonia, FUO (fever of unknown origin), mono-like syndrome (two cases - 12.5% each), lymphadenitis (one case - 6.3%) and spondylodiscitis associated with osteomyelitis (one case - 6.3%). The ages varied from four to 67 years old with a median of 43.5. All but one patient had positive serologic tests for phase II IgG whether or not associated with IgM positivity compatible with acute infection. One patient had both phase I and phase II IgG antibodies compatible with chronic Q fever. Seroconvertion was detected in 10 patients. Despite the known limitations of serologic diagnosis, the cases here reported should encourage Brazilian doctors to include Q fever as an indigenous cause of febrile illness.

Objective: To assess quantitative real-time polymerase chain reaction (q-PCR) for the sputum smear diagnosis of pulmonary tuberculosis (PTB) in patients living with HIV/AIDS with a clinical suspicion of PTB.Method: This is a prospective study to assess the accuracy of a diagnostic test, conducted on 140 sputum specimens from 140 patients living with HIV/AIDS with a clinical suspicion of PTB, attended at two referral hospitals for people living with HIV/AIDS in the city of Recife, Pernambuco, Brazil. A Löwenstein-Jensen medium culture and 7H9 broth were used as gold standard.Results: Of the 140 sputum samples, 47 (33.6%) were positive with the gold standard. q-PCR was positive in 42 (30%) of the 140 patients. Only one (0.71%) did not correspond to the culture. The sensitivity, specificity and accuracy of the q-PCR were 87.2%, 98.9% and 95% respectively. In 39 (93%) of the 42 q-PCR positive cases, the CT (threshold cycle) was equal to or less than 37.Conclusion: q-PCR performed on sputum smears from patients living with HIV/AIDS demonstrated satisfactory sensitivity, specificity and accuracy, and may therefore be recommended as a method for diagnosing PTB.

Apresenta-se, pela primeira vez, o estudo fitoquímico das frações larvicidas, isoladas da Magonia pubescens, monitorado pelo estudo de eficácia sobre larvas de 3º estádio de Aedes aegypti, na busca de alternativas para o controle desse mosquito e obtenção de estruturas químicas passíveis de aprimoramento da atividade pela via sintética de outros derivados. As frações bioativas foram monitoradas quimicamente através de cromatografia de camada delgada, utilizando como revelador uma solução ácida de vanilina, e analisadas por ressonância magnética nuclear de hidrogênio e espectrometria de massas. Os bioensaios com as frações foram realizados em quintuplicata, à temperatura de 28±1ºC, 80±5% de umidade relativa e fotofase de 12 horas. As concentrações letais encontradas da fração MP-9, que apresentou o maior potencial larvicida, CL50 e CL90, foram de 3,1 e 36,6ppm, respectivamente. Todos os experimentos foram acompanhados por uma série controle, contendo o mesmo número de larvas.

A febre Q é uma zoonose de distribuição mundial causada por Coxiella burnetii, sendo raros os registros da doença no Brasil. Estudos soroepidemiológicos mostraram uma freqüência relativamente elevada de anticorpos contra Coxiella burnetii em populações com exposição ocupacional. Em humanos, pode se manifestar clinicamente como doença aguda ou crônica, sendo que a endocardite é a forma crônica mais freqüente da febre Q e de maior morbi-mortalidade. Relatamos um caso grave de endocardite por Coxiella burnetii adquirida no Brasil com desfecho fatal, apesar de antibioticoterapia adequada e tratamento cirúrgico valvar.

INTRODUCTION: Fatty acids are abundant in vegetable oils. They are known to have antibacterial and antifungal properties. METHODS: Antifungal susceptibility was evaluated by broth microdilution assay following CLSI (formerly the NCCLS) guidelines against 16 fungal strains of clinical interest. RESULTS: In this work, fatty acid methyl esters (FAME) was able to inhibit 12 clinical strains of the pathogenic fungus Paracoccidioides brasiliensis and were also active in the bioautographic assay against Cladosporium sphaerospermum. CONCLUSIONS: FAME was a more potent antifungal than trimethoprim-sulfamethoxazole against P. brasiliensis under the experimental conditions tested.

Introduction Over the last recent years, the number of Q fever cases have has increased throughout the world. An epidemiological investigation was performed in the area in which the first molecular documentation of Q fever in Brazil was previously reported. Methods Indirect immunofluorescence assay (IFA) and PCR of Coxiella burnetii targeting the htpAB gene were performed in samples from 14 dogs (blood); 1 cat (blood); 10 goats (blood, milk, vaginal swab and anal swab); 3 sheep (blood); and 2 horses (blood). Results Two dogs, two sheep and five goats were seroreactive. DNA was amplified from 6 milk and 2 blood samples from goats and from dogs, respectively. The sequence of the amplicons exhibited 99% sequence similarity with the homologous sequence of the htpAB gene of C. burnetii RSA 331 (GenBank - CP000890). Conclusions The results confirm C. burnetii infection in animals in Rio de Janeiro and reinforce the need for the surveillance of Q fever in Brazil.

Introduction Triatoma sordida is the most captured Triatomine species in the Brazilian artificial environment. In 2008, the discovery of three Triatomine populations with altered susceptibilities to deltamethrin highlighted the importance of investigating the genetic potential for resistance in triatomines. The purpose of this study was to characterize the susceptibility to deltamethrin of peridomestic T. sordida populations in Minas Gerais, Brazil. Methods A susceptibility reference lineage derived from Uberaba, Minas Gerais, Brazil was used. Serial dilutions of deltamethrin were prepared and applied to the dorsal abdomen of first instar nymphs. The control group received only pure acetone. Mortality was evaluated after 72h. Qualitative tests assessed mortality in response to a diagnostic dose of 1xLD99 of the susceptibility reference lineage. Results Susceptibility profile characterization of T. sordida populations revealed resistance ratios (RR50s) ranging from 0.42 to 3.94. The percentage mortality in response to the diagnostic dose varied from 70% to 100%. A comparison of the results obtained in the quantitative and qualitative assays demonstrated a lack of correspondence for some populations. Conclusions We demonstrated that only T. sordida populations that present a RR50>1.0 have altered susceptibility, and the execution of simultaneous field and laboratory tests is required to understand the actual effect of vector control. A possible cause of the observed resistance ratios might be the continuous use of pyrethroids in Brazil since the 1980s.
