INTRODUÇÃO: A leptospirose é uma zoonose endêmica, mundialmente distribuída, causada por bactérias do gênero Leptospira. Este gênero compreende espécies patogênicas e saprofíticas, com mais de 200 sorovares distintos, dificultando sua caracterização. A técnica de pulsed field gel electrophoresis tem sido empregada como uma ferramenta para auxiliar nesta caracterização. Os objetivos deste trabalho foram padronizar a técnica de PFGE, determinar os perfis moleculares das cepas de referência utilizadas pelo Laboratório de Referência Nacional para Leptospirose/Centro Colaborador da Organização Mundial de Saúde para Leptospirose e criar um banco de dados com estes perfis. MÉTODOS: Foram analisadas, por PFGE, dezenove cepas utilizando a enzima de restrição NotI. RESULTADOS: Cada cepa apresentou um perfil único que pode ser considerado como uma identidade genômica específica, com exceção dos sorovares Icterohaemorrhagiae e Copenhageni, cujos perfis foram indistinguíveis. CONCLUSÕES: Dessa forma, foi possível a criação de um banco de perfis moleculares que está sendo utilizado no Laboratório para a comparação e identificação de cepas isoladas de quadros clínicos.

Group B streptococci (GBS) infections occur worldwide. Although serotyping has been used for epidemiologic purposes, this does not accurately characterize enough members of a genetically heterogeneous bacterial population. The aims of this work were to evaluate the genetic diversity of 45 type Ia GBS strains isolated in Brazil by pulsed-field gel electrophoresis as well as to evaluate antimicrobial susceptibility profiles and identify virulence genes. Twenty-four strains were assigned to cluster A. All strains under study contained the hylB and scpB genes. The bca gene was detected in only 10 strains and none of the streptococci carried the bac gene. Thirty-nine strains were resistant to tetracycline.

The epidemiology of urinary tract infections (UTI) by Staphylococcus saprophyticus has not been fully characterised and strain typing methods have not been validated for this agent. To evaluate whether epidemiological relationships exist between clusters of pulsed field gel-electrophoresis (PFGE) genotypes of S. saprophyticus from community-acquired UTI, a cross-sectional surveillance study was conducted in the city of Rio de Janeiro, Brazil. In total, 32 (16%) female patients attending two walk-in clinics were culture-positive for S. saprophyticus. Five PFGE clusters were defined and evaluated against epidemiological data. The PFGE clusters were grouped in time, suggesting the existence of community point sources of S. saprophyticus. From these point sources, S. saprophyticus strains may spread among individuals.
LOW COST ANALYZER FOR THE DETERMINATION OF PHOSPHORUS BASED ON OPEN-SOURCE HARDWARE AND PULSED FLOWS

The need for automated analyzers for industrial and environmental samples has triggered the research for new and cost-effective strategies of automation and control of analytical systems. The widespread availability of open-source hardware together with novel analytical methods based on pulsed flows have opened the possibility of implementing standalone automated analytical systems at low cost. Among the areas that can benefit from this approach are the analysis of industrial products and effluents and environmental analysis. In this work, a multi-pumping flow system is proposed for the determination of phosphorus in effluents and polluted water samples. The system employs photometric detection based on the formation of molybdovanadophosphoric acid, and the fluidic circuit is built using three solenoid micropumps. The detection is implemented with a low cost LED-photodiode photometric detection system and the whole system is controlled by an open-source Arduino Uno microcontroller board. The optimization of the timing to ensure the color development and the pumping cycle is discussed for the proposed implementation. Experimental results to evaluate the system behavior are presented verifying a linear relationship between the relative absorbance and the phosphorus concentrations for levels as high as 50 mg L-1.

Objetivou-se desenvolver metodologias para avaliação da qualidade de sementes de quixabeira que se encontra ameaçada de extinção, pelo fato de as cascas da árvore serem usadas na medicina popular. Foram realizados testes de secagem em laboratório e casa-de-vegetação por 0, 24, 48, 72, 96, 120, 144, 168 e 196 h de superação de dormência (escarificação com lixa; imersão em ácido sulfúrico por um, três, cinco e sete min; escarificação com lixa, seguida de embebição em água destilada por 12, 24, 36 e 48 h; embebição em água destilada por 12, 24, 36 e 48 h e testemunha) e com substratos (sobre e entre-areia, vermiculita, papel, bioplant®, plantmax e rolo de papel germitest) em diferentes temperaturas (20-30, 30 e 35 °C). A secagem em casa de vegetação prejudicou a qualidade das sementes. A escarificação com lixa é o tratamento mais eficiente para superação da dormência. As temperaturas de 30 °C com o substrato entre papel e 35 ºC com o substrato entre vermiculita são adequadas para testes de germinação com sementes de quixabeira.

Left ventricular diastolic dysfunction plays an important role on heart failure progression. In order to obtain additional reference values of left ventricular diastolic parameters and investigate influence of common variables, peak E wave (peak E), peak A wave (peak A), E/A ratio (E/A), E wave deceleration time (EDT) and isovolumic relaxation time (IRVT) were studied in 40 clinically healthy dogs, by pulsed wave Doppler. The following values were obtained: peak E = 0.747 ± 0.117 m/s, peak A = 0.487 ± 0.062 m/s, E/A = 1.533 ± 0.198, EDT = 88.7 ± 9.2 ms and IRVT = 0.080 ± 0.009 s. Some parameters were influenced by heart rate (peak E, peak A and IRVT), by age (peak A and E/A) and by body weight (TRIV). Gender influence was absent. Values obtained can be used as reference for canine specimens but its interpretation should consider on the influence of related variables.

The purpose of the present paper is to review work that has been done on the pulsed wire anemometer technique and also suggest further developments that could be made in its range of application. The aper discusses the three types of probes that have been used in pulsed wire anemometry: the crossed wire velocity probe, the parallel wire wall shear stress probe and the parallel wire velocity probe. The work shows that the crossed wire and the parallel wire techniques can be used to make velocity, turbulence and wall shear stress measurements in highly turbulent flows without any upper restriction on turbulence level. Comments are also made on the potential of a parallel wire probe for use in highly turbulent flows that would enable higher order velocity cross-product terms to be measured.

Polymerase chain reaction (PCR) with JB1 or REP consensus oligonucleotides and pulsed field gel electrophoresis (PFGE) were used to study genomic DNA extracted from 31 strains of enterococci. Eleven ATCC strains, representative of 11 species of Enterococcus, were initially tested by JB1-PCR, revealing that Enterococcus malodoratus and Enterococcus hirae presented identical banding patterns. Eight Enterococcus faecium isolates from Stanford University and 12 from São Paulo Hospital were studied by JB1-PCR, REP-PCR 1/2R and PFGE. Among the isolates from Stanford University, 5 genotypes were defined by JB1-PCR, 7 by REP-PCR 1/2R and 4 by PFGE. Among the isolates from São Paulo Hospital, 9 genotypes were identified by JB1-PCR, 6 by REP-PCR and 5 by PFGE. The three methods identified identical genotypes, but there was not complete agreement among them.

We studied the effect of pulsed ultrasound therapy (UST) and antibothropic polyvalent antivenom (PAV) on the regeneration of mouse extensor digitorum longus muscle following damage by Bothrops jararacussu venom. Animals (Swiss male and female mice weighing 25.0 ± 5.0 g; 5 animals per group) received a perimuscular injection of venom (1 mg/kg) and treatment with UST was started 1 h later (1 min/day, 3 MHz, 0.3 W/cm², pulsed mode). Three and 28 days after injection, muscles were dissected and processed for light microscopy. The venom caused complete degeneration of muscle fibers. UST alone and combined with PAV (1.0 mL/kg) partially protected these fibers, whereas muscles receiving no treatment showed disorganized fascicules and fibers with reduced diameter. Treatment with UST and PAV decreased the effects of the venom on creatine kinase content and motor activity (approximately 75 and 48%, respectively). Sonication of the venom solution immediately before application decreased the in vivo and ex vivo myotoxic activities (approximately 60 and 50%, respectively). The present data show that UST counteracts some effects of B. jararacussu venom, causing structural and functional improvement of the regenerated muscle after venom injury.

OBJETIVO: Investigar os efeitos causados pela poluição atmosférica na morbidade por pneumonia e por gripe em idosos entre 1996 e 1998. MÉTODOS: Foram obtidos dados diários de atendimentos por pneumonia e gripe para idosos em pronto-socorro médico de um hospital-escola de referência no Município de São Paulo, SP, Brasil. Os níveis diários de CO, O3, SO2, NO2 e PM10 foram obtidos na Companhia de Tecnologia de Saneamento Ambiental, e os dados diários de temperatura e umidade relativa do ar foram obtidos no Instituto Astronômico e Geofísico da USP. Para verificar a relação existente entre pneumonia e gripe e poluição atmosférica, utilizou-se o modelo aditivo generalizado de regressão de Poisson, tendo como variável dependente o número diário de atendimentos por pneumonia e gripe e como variáveis independentes as concentrações médias diárias dos poluentes atmosféricos. A análise foi ajustada para sazonalidade de longa duração (número de dias transcorridos), sazonalidade de curta duração (dias da semana), temperatura mínima, umidade média, períodos de rodízio e os atendimentos por doenças não-respiratórias em idosos. RESULTADOS: O3 e SO2 estão diretamente associados à pneumonia e à gripe, independentemente das variáveis de controle. Porém, na análise conjunta, eles perdem sua significância estatística. Pôde-se observar que um aumento interquartil (25%-75%) para o O3 (38,80 mig/m³) e SO2 (15,05 mig/m³) levaram a um acréscimo de 8,07% e 14,51%, respectivamente, no número de atendimentos por pneumonia e gripe em idosos. CONCLUSÕES: Os resultados sugerem que a poluição atmosférica promove efeitos adversos para a saúde de idosos.

OBJETIVOS: A presença de micotoxinas em alimentos tem sido correlacionada a várias patologias humanas, e as autoridades de saúde no mundo todo têm implementado ações para diminuir a ingestão desses compostos pela dieta. Realizou-se pesquisa para analisar os níveis de aflatoxinas e ocratoxina A de alimentos para consumo e avaliar o potencial de risco da exposição humana a essas micotoxinas. MÉTODOS: Foram analisadas 366 amostras de alimentos consumidos no Distrito Federal, no período de julho de 1998 a dezembro de 2001, como amendoim e derivados, castanhas, milho, produtos de trigo e/ou aveia, arroz e feijão. As amostras foram processadas, e as micotoxinas extraídas, detectadas e quantificadas por fluorescência após separação em cromatografia camada delgada. RESULTADOS: Foram detectadas aflatoxinas em 19,6% das amostras, em amendoim cru e derivados, milho de pipoca, milho em grão e castanha-do-pará (>2 mig/kg). Amendoim e derivados apresentaram maior incidência de contaminação por aflatoxinas (34,7%) com amostras contendo até 1.280 mig/kg de AFB1+AFG1 e 1.706 mig/kg de aflatoxinas totais. Das amostras positivas, AFB1 estava presente em 98,5%, AFB2 em 93%, AFG1 em 66,7% e AFG2 em 65,4%. A ocratoxina A não foi detectada (<25 mig/kg) em nenhuma amostra analisada. CONCLUSÃO: Os níveis de contaminação encontrados em amendoim e derivados ultrapassaram os níveis máximos permitidos pela legislação brasileira, podendo significar fator de risco para a população que os consome regularmente. A conscientização dos produtores de alimentos e as ações de vigilância sanitária permanentes são essenciais para diminuir a exposição humana a esses compostos e prevenir doenças crônicas advindas dessa exposição.

Yeasts of the genus Candida have been recognized as important microorganisms responsible for nosocomial fungemia. Six blood-stream and two intravenous central catheter C. albicans strains were isolated from eight patients and studied by electrophoretic karyotyping of chromosomal DNA by pulsed-field gel electrophoresis. Seven chromosomal DNA profiles were identified. Two patients showed isolates with the same profile, suggesting nosocomial transmission. Karyotyping of C. albicans revealed an excellent discriminatory power among the isolates and may therefore be useful in the study of nosocomial candidemia.

Salmonella Infantis has been the second most common serovar in Argentina in the last two years, being isolated mostly from paediatric hospitalised patients. In order to determine the clonal relationship among Salmonella Infantis strains, we examined 15 isolates from paediatric patient faeces in Argentina (12 geographically related and 3 geographically non-related) by using antimicrobial susceptibility, plasmid profiling, repetitive extragenic palindromic (REP) PCR, enterobacterial repetitive intergenic consensus (ERIC) PCR, and low-frequency restriction analysis of chromosomal DNA by pulsed field gel electrophoresis (PFGE). Four Spanish strains were included as controls of clonal diversity in molecular techniques. Antibiotype and plasmid profile was not useful as epidemiological tools. PFGE and REP-PCR were able to discriminate between Argentinean and Spanish isolates of Salmonella Infantis allowing to detect genetically related strains in three different cities. This finding indicates that a possible spread of a clone of this serovar in the North-eastern Region of Argentina has taken place in 1998.

In the present study, a total of 455 enterococcal isolates, recovered from patients living in the city of Porto Alegre, State of Rio Grande do Sul, Brazil, during the period from July 1996 to June 1997, were identified to the species level by conventional biochemical and microbiological tests, and assayed for their susceptibilities to antimicrobial agents. The genetic diversity of antimicrobial resistant strains was evaluated by pulsed-field gel electrophoresis (PFGE) analysis of SmaI restricted chromosomal DNA. The most frequent species was Enterococcus faecalis (92.8%). Other species identified were: E. faecium (2.9%), E. gallinarum (1.5%), E. avium (1.1%), E. hirae (0.7%), E. casseliflavus (0.4%), E. durans (0.4%) and E. raffinosus (0.2%). The overall prevalence of isolates with high-level resistance (HLR) to aminoglycosides was 37.8%. HLR to gentamicin was found in 24.8%. No strains with acquired resistance to vancomycin were found. PFGE analysis showed the predominance of clonal group A, comprising strains isolated from different clinical specimens obtained from patients in three hospitals. These results suggest intra and inter-hospital dissemination of one predominant clonal group of E. faecalis isolates with HLR to gentamicin in the hospitals included in this study.
