Araucaria angustifolia apresenta redução da taxa de crescimento durante o ano, podendo ser resposta às condições ambientais desfavoráveis, como ocorre nas fruteiras de clima temperado, que apresentam dormência de gemas como forma de sobrevivência. O objetivo deste trabalho foi avaliar a dinâmica da atividade respiratória de meristemas apicais de ramos plagiotrópicos de Araucaria angustifolia. Foram coletadas amostras de 0,4 g de brotações terminais de ramos plagiotrópicos de plantas jovens e adultas, as quais foram mantidas em 5 mL de solução de cloreto de 2,3,5 trifeniltetrazólio (1,2%) em sala de crescimento a 25°C. Em seguida, foram mantidas em 4 mL de álcool etílico absoluto para leitura por espectrofotometria da absorbância a 560 nm. A atividade respiratória de meristemas apicais de ramos plagiotrópicos de Araucaria angustifolia é variável durante o ano. A maior atividade respiratória ocorre na metade da primavera e a menor atividade respiratória ocorre no inverno, em plantas jovens e adultas. Plantas adultas permanecem em alta atividade respiratória por um período maior, até o início do verão.

Os métodos tradicionais para a quantificação de clorofilas implicam na destruição das folhas, além de serem demorados e dispendiosos. Uma alternativa aos métodos destrutivos é o uso de medidores portáteis, dentre eles o SPAD 502, que mede a intensidade da cor verde das folhas, resultando no índice SPAD (Soil Plant Analysis Development). No entanto, o índice SPAD deve ser ajustado para o teor de clorofilas, conforme a espécie de interesse. O objetivo do presente trabalho foi calibrar o índice SPAD para a quantificação de clorofilas em folhas de plantas de vime ( Salix viminalis ). Folhas desta espécie, com tonalidade variando de verde-amarelada (clorótica) a verde-escura, foram avaliadas individualmente com o SPAD-502, seguido de quantificações destrutivas dos teores de clorofilas a, b e totais, expressos em unidade de área e massa fresca foliar. Houve elevado coeficiente de determinação (R²) entre os valores de índice SPAD e os teores de clorofila a, b e totais nas folhas, expressos em μg cm-2 de área foliar (R² de 0,86; 0,88 e 0,93, respectivamente) e entre os valores de índice SPAD e os teores de clorofilas b e totais, expressos em μg g-1 de massa fresca (R² 0,79 e 0,81, respectivamente). Os resultados mostram que existe viabilidade no uso do clorofilômetro SPAD 502, como alternativa aos métodos destrutivos, para a quantificação de clorofilas (em unidade de área; μg cm-2) em folhas de vimeiro.

Purpose: To investigate the antioxidant and anticancer activities of phenolics from the leaf extract of Toona sinensis (TS). Methods: Acetone leaf extract of TS was screened for total phenolic and flavanoid contents, and the flanonoids were subjected to high performance liquid chromatographic (HPLC) analysis. Antioxidant properties were assessed via oxygen radical absorbance capacity (ORAC), peroxyl radical scavenging capacity (PSC) and cellular antioxidant activity (CAA), while anti-proliferative activity ins HepG2 cell line was assessed using methylene blue assay. Results: The extract contained 36.02 ± 0.24 mg of gallic acid equiv/g dry weight (DW) and 20.24 ± 1.73 mg of catechin equiv/g DW of total phenolic and total flavonoid, respectively. The levels of rutin and quercitrin were 0.51 and 19.55 mg/g, respectively. Epicatechin, gallic acid, quercitin, isoquercetin were not detected. The extract showed significant antioxidant potential and high anti-proliferation capacity with low cytotoxicity against HepG2 cell in vitro. The underlying mechanism of anti-proliferative effect was induction of apoptosis. Conclusion: TS leaf extract possesses significant in vitro antioxidant properties and anti-proliferative effect against HepG2 cells, which make it a potential anticancer drug source.

Purpose: To investigate the effect of withaferin A (WFA) on the proliferation and migration of brain endothelial cells. Methods: BALB-5023 mouse microvascular cells were treated with a range of withaferin A (WFA) concentrations from 10 to 100 ng/mL. Dojindo’s CCK-8 cell proliferation kit was used for the analysis of cell proliferation. Transwell cell culture inserts were used to determine the migration potential of WFAtreated endothelial cells. Absorbance was measured at 450 nm on an enzyme-linked immunosorbent (ELISA) reader. Results: The results revealed a significant increase in the proliferation and migration of endothelial cells following treatment with a low concentration (30 ng/mL) of WFA compared with the higher concentration (> 10 ng/mL). The effect was further enhanced when WFA was used in combination with soluble Fas ligand (sFasL). Autocrine signaling of vascular endothelial growth factor (VEGF) by endothelial cells was significantly increased following treatment with WFA or in combination with sFasL. WFA increased the expression of Fas on endothelial cells, suggesting the involvement of sFasL in the proliferation and migration of brain endothelial cells. Conclusion: Thus, WFA promotes the proliferation and migration of endothelial cells through increase in the expression of Fas and secretion of VEGF.

Purpose: To evaluate the antibacterial, enzyme-inhibitory and hemolytic activities of Salkylated/ aralkylated 2-(1H-indol-3-ylmethyl)-1,3,4-oxadiazole-5-thiol derivatives. Methods: Antibacterial activities of the compounds were evaluated using broth dilution method in 96 well plates. Enzyme inhibitory activities assays were investigated against α-glucosidase, butyrylcholinesterase (BchE) and lipoxygenase (LOX) using acarbose, eserine and baicalien as reference standards, respectively. A mixture of enzyme, test compound and the substrate was incubated and variation in absorbance noted before and after incubation. In tests for hemolytic activities, the compounds were incubated with red blood cells and variations in absorbance were used as indices their hemolytic activities. Results: The compounds were potent antibacterial agents. Five of them exhibited very good antibacterial potential similar to ciprofloxacin, and had minimum inhibitory concentrations (MIC) of at least 9.00 ± 4.12 μM against S. aureus, E.coli, and B. subtilis. One of the compounds had strong enzyme inhibitory potential against α-glucosidase, with IC50 of 17.11 ± 0.02 μg/mL which was better than that of standard acarbose (IC50 38.25 ± 0.12 μg/mL). Another compound had 1.5 % hemolytic activity. Conclusion: S-Alkylated/aralkylated 2-(1H-indol-3-ylmethyl)-1,3,4-oxadiazole-5-thiol deviratives with valuable antibacterial, anti-enzymatic and hemolytic activities have been successfully synthesized. These compounds may be useful in the development of pharmaceutical products.
