Microstructure manipulation is a fundamental process to the study of biology and medicine, as well as to advance micro- and nano-system applications. Manipulation of microstructures has been achieved through various microgripper devices developed recently, which lead to advances in micromachine assembly, and single cell manipulation, among others. Only two kinds of integrated feedback have been demonstrated so far, force sensing and optical binary feedback. As a result, the physical, mechanical, optical, and chemical information about the microstructure under study must be extracted from macroscopic instrumentation, such as confocal fluorescence microscopy and Raman spectroscopy. In this research work, novel Micro-Opto-Electro-Mechanical-System (MOEMS) microgrippers are presented. These devices utilize flexible optical waveguides as gripping arms, which provide the physical means for grasping a microobject, while simultaneously enabling light to be delivered and collected. This unique capability allows extensive optical characterization of the structure being held such as transmission, reflection, or fluorescence. The microgrippers require external actuation which was accomplished by two methods: initially with a micrometer screw, and later with a piezoelectric actuator. Thanks to a novel actuation mechanism, the "fishbone", the gripping facets remain parallel within 1 degree. The design, simulation, fabrication, and characterization are systematically presented. The devices mechanical operation was verified by means of 3D finite element analysis simulations. Also, the optical performance and losses were simulated by the 3D-to-2D effective index (finite difference time domain FDTD) method as well as 3D Beam Propagation Method (3D-BPM). The microgrippers were designed to manipulate structures from submicron dimensions up to approximately 100 μm. The devices were implemented in SU-8 due to its suitable optical and mechanical properties. This work demonstrates two practical applications: the manipulation of single SKOV-3 human ovarian carcinoma cells, and the detection and identification of microparts tagged with a fluorescent "barcode" implemented with quantum dots. The novel devices presented open up new possibilities in the field of micromanipulation at the microscale, scalable to the nano-domain.

This dissertation evaluated the feasibility of using commercially available immortalized cell lines in building a tissue engineered in vitro blood-brain barrier (BBB) co-culture model for preliminary drug development studies. Mouse endothelial cell line and rat astrocyte cell lines purchased from American Type Culture Collections (ATCC) were the building blocks of the co-culture model. An astrocyte derived acellular extracellular matrix (aECM) was introduced in the co-culture model to provide a novel in vitro biomimetic basement membrane for the endothelial cells to form endothelial tight junctions. Trans-endothelial electrical resistance (TEER) and solute mass transport studies were engaged to quantitatively evaluate the tight junction formation on the in-vitro BBB models. Immuno-fluorescence microscopy and Western Blot analysis were used to qualitatively verify the in vitro expression of occludin, one of the earliest discovered tight junction proteins. Experimental data from a total of 12 experiments conclusively showed that the novel BBB in vitro co-culture model with the astrocyte derived aECM (CO+aECM) was promising in terms of establishing tight junction formation represented by TEER values, transport profiles and tight junction protein expression when compared with traditional co-culture (CO) model setups and endothelial cells cultured alone. Experimental data were also found to be comparable with several existing in vitro BBB models built from various methods. In vitro colorimetric sulforhodamine B (SRB) assay revealed that the co-cultured samples with aECM resulted in less cell loss on the basal sides of the insert membranes than that from traditional co-culture samples. The novel tissue engineering approach using immortalized cell lines with the addition of aECM was proven to be a relevant alternative to the traditional BBB in vitro modeling.

Fusarium oxysporum forma specialis cubense is a soilborne phytopathogen that infects banana. The true evolutionary identity of this so called species, Fusarium oxysporum, is still unknown. Many techniques have been applied in order to gain insight for the observed genetic diversity of this species. The current classification system is based on vegetative compatibility groups (VCG's). Vegetative compatibility is a self non-self recognition system in which only those belonging to a VCG can form stable heterokaryons, cells containing two distinct nuclei. Heterokaryons in turn, are formed from hypha! anastomosis, the fusion of two hyphae. Furthermore, subsequent to heterokaryon formation potential mechanisms exist which may generate genetic variability. One is through viral transfer upon hyphal anastomosis. The other mechanism is a form of mitotic recombination referred to as the parasexual cycle. Very little research has been performed to directly obser.ve the cellular events; hypha! anastomosis, heterokaryon formation, and the parasexual cycle in Fusarium oxysporum f. sp. cubense. The purpose of this research was to design and use methods which would allow for the detection of hypha! anastomosis and heterokaryon formation, as well as any characteristics surrounding this event, within and between VCG's in Foe. First, some general growth properties were recorded: the number of nuclei per hypha, the size ofthe hyphal tip cell, the size of the cell adjacent to the hypha! tip (pre-tip) cell, and the number of cells to the first branch point. Second, four methods were designed in order to assay hyphal anastomosis and heterokaryon formation: 1) pairings on membrane: phase or brightfield microscopy, 2) pairings on membrane: fluorescence microscopy, 3) spore crosses: fluorescence microscopy, and 4) double picks in fractionated MMA.
