DNA serves as a target molecule for several types of enzymes and may assume a wide variety of structural motifs depending upon the local sequence. The BssHII restriction site (GC)3 resides in a 9bp region of alternating pyrimidine and purine residues within the &phis;X174 genome. Such sequences are known to demonstrate non-canonical helical behavior under the appropriate conditions. The kinetics of BssHII cleavage was investigated in supercoiled and linear plasmid DNA, and in a 323bp DNA fragment obtained via amplification of &phis;X174. The rate of enzyme cleavage was enhanced in the supercoiled form and in the presence of 50μM cobalt hexamine. Similarly, cobalt hexamine was also found to enhance TaqI activity directly adjacent to the (GC)3 region. ^ Initial DNA polymerase I binding studies (including a gel mobility shift assay and a protection assay) indicated a notable interaction between DNA polymerase I and the BssHII site. An in-depth study revealed that equilibrium binding of DNA polymerase I to the T7 RNA polymerase promoter was comparable to that of the (GC)3 site, however the strongest interaction was observed with a cruciform containing region. Increasing the ionic strength of the solution environment, including the addition of DNA polymerase I reaction buffer significantly decreased the equilibrium dissociation constant values. ^ It is suggested that the region within or around the BssHII site experiences a conformational change generating a novel structure under the influence of supercoiled tension or 50μM cobalt hexamine. It is proposed that this transition may enhance enzyme activity and binding by providing an initial enzyme-docking site—the rate-limiting step in restriction enzyme kinetics. The high binding potential of DNA polymerase I for each of the motifs described, is hypothesized to be due to recognition of the structural DNA anomalies by the 3′–5′ exonuclease domain. ^

Coral reefs are in decline worldwide and coral disease is a significant contributing factor. However, etiologies of coral diseases are still not well understood. In contrast with the Caribbean, extremely little is known about coral diseases in the Philippines. In 2005, off Southeast Negros Island, Philippines, I investigated relationships between environmental parameters and prevalence of the two most common coral diseases, ulcerative white spot (UWS) and massive Porites growth anomalies (MPGAs). Samples were collected along a disease prevalence gradient 40.5 km long. Principal component analyses showed prevalence of MPGAs was positively correlated with water column nitrogen, organic carbon of surface sediments, and colony density. UWS was positively correlated with water column phosphorus. This is the first quantitative evidence linking anthropogenically-impacted water and sediment to a higher prevalence of these diseases. Histological and cytological alterations were investigated by comparing tissues from two distinct types of MPGA lesions (types 1 and 2) and healthy coral using light and electron microscopy. Skeletal abnormalities and sloughing, swelling, thinning, and loss of tissues in MPGAs resembled tissues exposed to bacterial or fungal toxins. Both lesion types had decreases in symbiotic zooxanthellae, which supply nutrients to corals. Notable alterations included migrations of chromophore cells (amoebocytes) (1) nocturnally to outer epithelia to perform wound-healing, including plugging gaps and secreting melanin in degraded tissues, and (2) diurnally to the interior of the tissue possibly to prevent shading zooxanthellae in order to maximize photosynthate production. Depletion of melanin (active in wound healing) in type 2 lesions suggested type 2 tissues were overtaxed and less stable. MPGAs contained an abundance of endolithic fungi and virus-like particles, which may result from higher nutrient levels and play roles in disease development. Swollen cells and mucus frequently blocked gastrovascular canals (GVCs) in MPGAs. Type 1 lesions appeared to compensate for impeded flow of wastes and nutrients through these canals with proliferation of new GVCs, which were responsible for the observed thickened tissues. In contrast, type 2 tissues were thin and more degraded. Dysplasia and putative neoplasia were also observed in MPGAs which may result from the tissue regeneration capacity being overwhelmed.

The purpose of this research was to investigate the influence of elevation and other terrain characteristics over the spatial and temporal distribution of rainfall. A comparative analysis was conducted between several methods of spatial interpolations using mean monthly precipitation values in order to select the best. Following those previous results it was possible to fit an Artificial Neural Network model for interpolation of monthly precipitation values for a period of 20 years, with input values such as longitude, latitude, elevation, four geomorphologic characteristics and anchored by seven weather stations, it reached a high correlation coefficient (r=0.85). This research demonstrated a strong influence of elevation and other geomorphologic variables over the spatial distribution of precipitation and the agreement that there are nonlinear relationships. This model will be used to fill gaps in time-series of monthly precipitation, and to generate maps of spatial distribution of monthly precipitation at a resolution of 1km2.

This study examined how different rainfall regimes affect a set of leaf functional traits related to plant stress and forest structure in tropical dry forest (TDF) species on limestone substrate. One hundred fifty eight individuals of four tree species were sampled in six ecological sites in south Florida and Puerto Rico, ranging in mean annual rainfall from 858 to 1933 mm yr-1. Leaf nitrogen content, specific leaf area (SLA), and N:P ratio of evergreen species, but not deciduous species, responded positively to increasing rainfall. Phosphorus content was unaffected in both groups. Canopy height and basal area reached maxima of 10.3 m and 31.4 m2 ha-1, respectively, at 1168 mm annual rainfall. Leaf traits reflected soil properties only to a small extent. This led us to the conclusion that water is a major limiting factor in TDF and some species that comprise TDF ecosystems are limited by nitrogen in limestone sites with less than ~1012 mm rainfall, but organismal, biological and/or abiotic forces other than rainfall control forest structure in moister sites.

Tropical Rainfall Measuring Mission (TRMM) rainfall retrieval algorithms are evaluated in tropical cyclones (TCs). Differences between the Precipitation Radar (PR) and TRMM Microwave Imager (TMI) retrievals are found to be related to the storm region (inner core vs. rainbands) and the convective nature of the precipitation as measured by radar reflectivity and ice scattering signature. In landfalling TCs, the algorithms perform differently depending on whether the rainfall is located over ocean, land, or coastal surfaces. Various statistical techniques are applied to quantify these differences and identify the discrepancies in rainfall detection and intensity. Ground validation is accomplished by comparing the landfalling storms over the Southeast US to the NEXRAD Multisensor Precipitation Estimates (MPE) Stage-IV product. Numerous recommendations are given to algorithm users and developers for applying and interpreting these algorithms in areas of heavy and widespread tropical rainfall such as tropical cyclones.

This study computed trends in extreme precipitation events of Florida for 1950-2010. Hourly aggregated rainfall data from 24 stations of the National Climatic Data Centre were analyzed to derive time-series of extreme rainfalls for 12 durations, ranging from 1 hour to 7 day. Non-parametric Mann-Kendall test and Theil-Sen Approach were applied to detect the significance of trends in annual maximum rainfalls, number of above threshold events and average magnitude of above threshold events for four common analysis periods. Trend Free Pre-Whitening (TFPW) approach was applied to remove the serial correlations and bootstrap resampling approach was used to detect the field significance of trends. The results for annual maximum rainfall revealed dominant increasing trends at the statistical significance level of 0.10, especially for hourly events in longer period and daily events in recent period. The number of above threshold events exhibited strong decreasing trends for hourly durations in all time periods.
