A detailed knowledge of the mapping between sequence and structure spaces in populations of RNA molecules is essential to better understand their present-day functional properties, to envisage a plausible early evolution of RNA in a prebiotic chemical environment and to improve the design of in vitro evolution experiments, among others. Analysis of natural RNAs, as well as in vitro and computational studies, show that certain RNA structural motifs are much more abundant than others, pointing out a complex relation between sequence and structure. Within this framework, we have investigated computationally the structural properties of a large pool (10 molecules) of single-stranded, 35 nt-long, random RNA sequences. The secondary structures obtained are ranked and classified into structure families. The number of structures in main families is analytically calculated and compared with the numerical results. This permits a quantification of the fraction of structure space covered by a large pool of sequences. We further show that the number of structural motifs and their frequency is highly unbalanced with respect to the nucleotide composition: simple structures such as stem-loops and hairpins arise from sequences depleted in G, while more complex structures require an enrichment of G. In general, we observe a strong correlation between subfamilies-characterized by a fixed number of paired nucleotides-and nucleotide composition. Our results are compared to the structural repertoire obtained in a second pool where isolated base pairs are prohibited. © 2008 Elsevier Ltd. All rights reserved.

INTRODUCTION: Bipolar disorder requires long-term treatment but non-adherence is a common problem. Antipsychotic long-acting injections (LAIs) have been suggested to improve adherence but none are licensed in the UK for bipolar. However, the use of second-generation antipsychotics (SGA) LAIs in bipolar is not uncommon albeit there is a lack of systematic review in this area. This study aims to systematically review safety and efficacy of SGA LAIs in the maintenance treatment of bipolar disorder. METHODS AND ANALYSIS: The protocol is based on Preferred Reporting Items for Systematic reviews and Meta-Analyses (PRISMA) and will include only randomised controlled trials comparing SGA LAIs in bipolar. PubMed, EMBASE, CINAHL, Cochrane Library (CENTRAL), PsychINFO, LiLACS, http://www.clinicaltrials.gov will be searched, with no language restriction, from 2000 to January 2016 as first SGA LAIs came to the market after 2000. Manufacturers of SGA LAIs will also be contacted. Primary efficacy outcome is relapse rate or delayed time to relapse or reduction in hospitalisation and primary safety outcomes are drop-out rates, all-cause discontinuation and discontinuation due to adverse events. Qualitative reporting of evidence will be based on 21 items listed on standards for reporting qualitative research (SRQR) focusing on study quality (assessed using the Jadad score, allocation concealment and data analysis), risk of bias and effect size. Publication bias will be assessed using funnel plots. If sufficient data are available meta-analysis will be performed with primary effect size as relative risk presented with 95% CI. Sensitivity analysis, conditional on number of studies and sample size, will be carried out on manic versus depressive symptoms and monotherapy versus adjunctive therapy.

For the first time we report full numerical NLSE-based modeling of generation properties of random distributed feedback fiber laser based on Rayleigh scattering. The model which takes into account the random backscattering via its average strength only describes well power and spectral properties of random DFB fiber lasers. The influence of dispersion and nonlinearity on spectral and statistical properties is investigated. The evidence of non-gaussian intensity statistics is found. © 2013 Optical Society of America.

Astrocytes in the rat thalamus display spontaneous [Ca2+]i oscillations that are due to intracellular release, but are not dependent on neuronal activity. In this study we have investigated the mechanisms involved in these spontaneous [Ca2+]i oscillations using slices loaded with Fluo-4 AM (5 μM) and confocal microscopy. Bafilomycin A1 incubation had no effect on the number of spontaneous [Ca2+]i oscillations indicating that they were not dependent on vesicular neurotransmitter release. Oscillations were also unaffected by ryanodine. Phospholipase C (PLC) inhibition decreased the number of astrocytes responding to metabotropic glutamate receptor (mGluR) activation but did not reduce the number of spontaneously active astrocytes, indicating that [Ca2+]i increases are not due to membrane-coupled PLC activation. Spontaneous [Ca2+]i increases were abolished by an IP3 receptor antagonist, whilst the protein kinase C (PKC) inhibitor chelerythrine chloride prolonged their duration, indicating a role for PKC and inositol 1,4,5,-triphosphate receptor activation. BayK8644 increased the number of astrocytes exhibiting [Ca2+]i oscillations, and prolonged the responses to mGluR activation, indicating a possible effect on store-operated Ca2+ entry. Increasing [Ca2+]o increased the number of spontaneously active astrocytes and the number of transients exhibited by each astrocyte. Inhibition of the endoplasmic reticulum Ca2+ ATPase by cyclopiazonic acid also induced [Ca2+]i transients in astrocytes indicating a role for cytoplasmic Ca2+ in the induction of spontaneous oscillations. Incubation with 20 μM Fluo-4 reduced the number of astrocytes exhibiting spontaneous increases. This study indicates that Ca2+ has a role in triggering Ca2+ release from an inositol 1,4,5,-triphosphate sensitive store in astrocytes during the generation of spontaneous [Ca2+]i oscillations

Dual-polarization multi-band orthogonal frequency-division multiplexing (DP-MB-OFDM) signals are optimized at data rates up to 500 Gb/s in dispersion compensation fiber (DCF)-free long-haul links. We compare different polarization crosstalk compensation techniques and explore the DP-MB-OFDM transmission distance limitations. In addition, the impact of the OFDM subcarrier number and high-level modulation format on the transmission performance are also investigated. Finally, a comparison is made between DP-MB-OFDM and the industrial solution of single-carrier DP quaternary phase-shift keying (DP-QPSK) at 1000 km.

Multiwavelength lasing in the random distributed feedback fiber laser is demonstrated by employing an all fiber Lyot filter. Stable multiwavelength generation is obtained, with each line exhibiting sub-nanometer line-widths. A flat power distribution over multiple lines is also obtained, which indicates the contribution of nonlinear wave mixing towards power redistribution and equalization in the system. The multiwavelength generation is observed simultaneously in first and second Stokes waves. © 2014 SPIE.

The simplest model for a description of the random distributed feedback (RDFB) Raman fiber laser is a power balance model describing the evolution of the intensities of the waves over the fiber length. The model predicts well the power performances of the RDFB fiber laser including the generation threshold, the output power and pump and generation wave intensity distributions along the fiber. In the present work, we extend the power balance model and modify equations in such a way that they describe now frequency dependent spectral power density instead of integral over the spectrum intensities. We calculate the generation spectrum by using the depleted pump wave longitudinal distribution derived from the conventional power balance model. We found the spectral balance model to be sufficient to account for the spectral narrowing in the RDFB laser above the threshold of the generation. © 2014 SPIE.

We demonstrate a random fiber laser of ultimate efficiency. More than 2 Watts are generated from 0.5W of pump excess over the generation threshold. At higher power, an optical efficiency corresponds to the quantum limit.

In this letter, the polarization properties of a random fiber laser operating via Raman gain and random distributed feedback owing to Rayleigh scattering are investigated for the first time. Using polarized pump, the partially polarized generation is obtained with a generation spectrum exhibiting discrete narrow spectral features contrary to the smooth spectrum observed for the depolarized pump. The threshold, output power, degree of polarization and the state of polarization (SOP) of the lasing can be significantly influenced by the SOP of the pump. Fine narrow spectral components are also sensitive to the SOP of the pump wave. Furthermore, we found that random lasing's longitudinal power distributions are different in the case of polarized and depolarized pumping that results in considerable reduction of the generation slope efficiency for the polarized radiation. Our results indicate that polarization effects play an important role on the performance of the random fiber laser. This work improves the understanding of the physics of random lasing in fibers and makes a step forward towards the establishment of the vector model of random fiber lasers.

A localized method to distribute paths on random graphs is devised, aimed at finding the shortest paths between given source/destination pairs while avoiding path overlaps at nodes. We propose a method based on message-passing techniques to process global information and distribute paths optimally. Statistical properties such as scaling with system size and number of paths, average path-length and the transition to the frustrated regime are analyzed. The performance of the suggested algorithm is evaluated through a comparison against a greedy algorithm. © 2014 IOP Publishing Ltd and SISSA Medialab srl.

Random distributed feedback (DFB) fiber lasers have attracted a great attention since first demonstration [1]. Despite big advance in practical laser systems, random DFB fiber laser spectral properties are far away to be understood or even numerically modelled. Up to date, only generation power could be calculated and optimized numerically [1,2] or analytically [3] within the power balance model. However, spectral and statistical properties of random DFB fiber laser can not be found in this way. Here we present first numerical modelling of the random DFB fiber laser, including its spectral and statistical properties, using NLSE-based model. © 2013 IEEE.

We present a comprehensive study of power output characteristics of random distributed feedback Raman fiber lasers. The calculated optimal slope efficiency of the backward wave generation in the one-arm configuration is shown to be as high as ∼90% for 1 W threshold. Nevertheless, in real applications a presence of a small reflection at fiber ends can appreciably deteriorate the power performance. The developed numerical model well describes the experimental data. © 2012 Optical Society of America.

Narrow-band generation is achieved in random distributed feedback (RDFB) fiber laser by using narrow-band filters in the center of a distributed cavity. The resulting line-width of ∼0.1 nm is 10 times less than line-width in classical random distributed feedback fiber laser. Spectral properties can be optimized further. © 2012 OSA.

We experimentally demonstrate a Raman fiber laser based on multiple point-action fiber Bragg grating reflectors and distributed feedback via Rayleigh scattering in an ∼22-km-long optical fiber. Twenty-two lasing lines with spacing of ∼100 GHz (close to International Telecommunication Union grid) in the C band are generated at the watt level. In contrast to the normal cavity with competition between laser lines, the random distributed feedback cavity exhibits highly stable multiwavelength generation with a power-equalized uniform distribution, which is almost independent on power. © 2011 Optical Society of America.

We demonstrate a CW random distributed feedback Raman fiber laser operating in a 1.2 μm spectral band. The laser generates up to 3.8 W of the quasi-CW radiation at 1175 nm with the narrow spectrum of 1 nm. Conversion efficiency reaches 60%. Up to 1 W is generated at the second Stokes wavelength of 1242 nm. It is shown that the generation spectrum of RDFB Raman fiber laser is much narrower than the spectrum in the system without a weak random feedback. © 2011 Optical Society of America.
