The skin immune system is believed to be a crucial site of contact between immunocompetent cells and invading organisms. A novel T cell component of murine epidermis is the Thy-1$\sp+$ dendritic epidermal cell (Tdec). To assess the immunocompetence of Tdec, the ability of Tdec to induce immune responses was tested. Tdec were unable to induce positive immune responses in three models of immunocompetence. Subsequent studies were designed to test the hypothesis that Tdec are involved in the down-regulation of cell-mediated immunity against cutaneous antigens. Cultured Tdec lines were conjugated in vitro with the hapten, fluorescein isothiocyanate (FITC). The intrafootpad (ifp.) or intravenous (i.v.) injection of FTIC-conjugated Tdec induced immunologic tolerance to subsequent epicutaneous sensitization with FITC. This induction of tolerance was antigen-specific, and injection of unconjugated Tdec had no effect on the contact hypersensitivity response to FITC. Tolerance was not H-2-restricted, since it could be induced in both syngeneic and allogeneic recipients of FITC-conjugated Tdec. No suppressive activity could be detected in lymphoid organs of animals tolerized by the ifp. injection of hapten-conjugated Tdec. In contrast, suppressor T cells were present in the spleens of mice injected i.v. with hapten-conjugated Tdec. These results indicate that Ts cells are not involved in the induction of tolerance by the ifp. injection of hapten-conjugated Tdec. To investigate the mechanism by which the ifp. injection of hapten-conjugated Tdec induced tolerance to contact sensitization, the activity of these cells was measured in vitro. The addition of hapten-conjugated Tdec inhibited the proliferation of Con A-stimulated lymphocytes. In addition, FITC-conjugated Tdec abrogated the proliferation of normal lymphocytes in response to FITC-labeled stimulator cells. These studies suggest that specific T cell-mediated immunity is the target of the inhibitory effect of Tdec in vitro. In summary, these results demonstrate that while Tdec are unable to induce positive immune responses, they can produce a state of specific immunologic tolerance when injected ifp. or i.v. These results also suggest that the induction of immunologic tolerance by hapten-conjugated Tdec may occur through the inactivation or elimination of activated T lymphocytes resulting in down-regulation of cell-mediated immunity against cutaneous antigens. ^

Purpose. Fluorophotometry is a well validated method for assessing corneal permeability in human subjects. However, with the growing importance of basic science animal research in ophthalmology, fluorophotometry’s use in animals must be further evaluated. The purpose of this study was to evaluate corneal epithelial permeability following desiccating stress using the modified Fluorotron Master™. ^ Methods. Corneal permeability was evaluated prior to and after subjecting 6-8 week old C57BL/6 mice to experimental dry eye (EDE) for 2 and 5 days (n=9/time point). Untreated mice served as controls. Ten microliters of 0.001% sodium fluorescein (NaF) were instilled topically into each mouse’s left eye to create an eye bath, and left to permeate for 3 minutes. The eye bath was followed by a generous wash with Buffered Saline Solution (BSS) and alignment with the Fluorotron Master™. Seven corneal scans using the Fluorotron Master were performed during 15 minutes (1 st post-wash scans), followed by a second wash using BSS and another set of five corneal scans (2nd post-wash scans) during the next 15 minutes. Corneal permeability was calculated using data calculated with the FM™ Mouse software. ^ Results. When comparing the difference between the Post wash #1 scans within the group and the Post wash #2 scans within the group using a repeated measurement design, there was a statistical difference in the corneal fluorescein permeability of the Post-wash #1 scans after 5 days (1160.21±108.26 vs. 1000.47±75.56 ng/mL, P<0.016 for UT-5 day comparison 8 [0.008]), but not after only 2 days of EDE compared to Untreated mice (1115.64±118.94 vs. 1000.47±75.56 ng/mL, P>0.016 for UT-2 day comparison [0.050]). There was no statistical difference between the 2 day and 5 day Post wash #1 scans (P=.299). The Post-wash #2 scans demonstrated that EDE caused a significant NaF retention at both 2 and 5 days of EDE compared to baseline, untreated controls (1017.92±116.25, 1015.40±120.68 vs. 528.22±127.85 ng/mL, P<0.05 [0.0001 for both]). There was no statistical difference between the 2 day and 5 day Post wash #2 scans (P=.503). The comparison between the Untreated post wash #1 with untreated post wash #2 scans using a Paired T-test showed a significant difference between the two sets of scans (P=0.000). There is also a significant difference between the 2 day comparison and the 5 day comparison (P values = 0.010 and 0.002, respectively). ^ Conclusion. Desiccating stress increases permeability of the corneal epithelium to NaF, and increases NaF retention in the corneal stroma. The Fluorotron Master is a useful and sensitive tool to evaluate corneal permeability in murine dry eye, and will be a useful tool to evaluate the effectiveness of dry eye treatments in animal-model drug trials.^

T cell activation and expansion is essential for immune response against foreign antigens. However, uncontrolled T cell activity can be manifested as a number of lymphoid derived diseases such as autoimmunity, graft versus host disease, and lymphoma. The purpose of this research was to test the central hypothesis that the Jak3/Stat5 pathway is critical for T cell function. To accomplish this objective, two novel Jak3 inhibitors, AG490 and PNU156804, were identified and their effects characterized on Jak3/Stat5 activation and T cell growth. Inhibition of Jak3 selectively disrupted primary human T lymphocyte growth in response to Interleukin-2 (IL-2), as well as other γ c cytokine family members including IL-4, IL-7, IL-9, and IL-15. Inhibition of Jak3 ablated IL-2 induced Stat5 but not TNF-α mediated NF-κβ DNA binding. Loss of Jak3 activity did not affect T cell receptor mediated signals including activation of p56Lck and Zap70, or IL-2 receptor a chain expression. To examine the effects of Jak3/Stat5 inhibition within a mature immune system, we employed a rat heart allograft model of Lewis (RT1 1) to ACI (RT1a). Heart allograft survival was significantly prolonged following Jak3/Stat5 inhibition when rats were treated with AG490 (20mg/kg) or PNU156804 (80mg/kg) compared to non-treated control animals. This effect was synergistically potentiated when Jak3 inhibitors were used in combination with a signal 1/2 disrupter, cyclosporine, but only additively potentiated with another signal 3 inhibitor, rapamycin. This suggested that sequential inhibition of T cell function is more effective. To specifically address the role of Stat5 in maintaining T cell activity, novel Stat5 antisense oligonucleotides were synthesized and characterized in vitro. Primary human T cells and T-cell tumor lines treated with Stat5 antisense oligonucleotide (7.5 μM) rapidly underwent apoptosis, while no changes in cell cycle were observed as measured by FACS analysis utilizing Annexin-V-Fluorescein and Propidium iodide staining. Evidence is provided to suggest that caspase 8 and 9 pathways mediate this event. Thus, Stat5 may act rather as a negative regulator of apoptotic signals and not as a positive regulator of cell cycle as previously proposed. We conclude that the Jak3/Stat5 pathway is critical for γc cytokine mediated gene expression necessary for T cell expansion and normal immune function and represents an therapeutically relevant effector pathway to combat T cell derived disease. ^
