Resumo:
The solution phase parallel synthesis involving reactions of Baylis-Hillman products of 3-substituted-5-isoxazolecarbaldehydes with nucleophiles and their in vivo antithrombotic evaluations are described along with the results of in vitro platelet aggregation inhibition assay of a few compounds. Results of the detailed evaluation of one of the compounds as an inhibitor of platelet aggregation are also presented.

A mild and convenient synthesis of substituted α-methylene--valerolactones was achieved by SN2 nucleophilic substitution of the acetates of the Baylis-Hillman adducts with acetyl acetone followed by one-pot saponification of the ester, reduction of the keto group and subsequent intramolecular ring closure in aqueous medium.

An alternate approach to densely substituted quinolines from the products of SN2 nucleophilic substitution reaction between the acetyl derivatives of the Baylis-Hillman adducts obtained from 2-nitrobenzaldehydes and the carbonyl group containing carbon nucleophiles is described. Treatment of these compounds with SnCl2, trigger a tandem reaction wherein reduction of the nitro group is followed by a remarkably regioselective intramolecular cyclization and subsequent dehydrogenation to afford 4-(substituted vinyl)-quinolines.

A simple and convenient synthesis of 3-methylene-4-aryl-1,3,4,5-tetrahydro-benzo[b][1,4] diazepin-2-ones was accomplished by the SN2 nucleophilic substitution of the acetates of Baylis-Hillman adducts of acrylate with 1,2-phenylenediamines followed by base-mediated intramolecular cyclization. On the other hand similar substrates derived from the Baylis-Hillman adducts of acrylonitrile via Pinner’s reaction leads to 3-arylmethylene-4,5-dihydro-3H-benzo[b][1,4]diazepin-2-ylamines in good yields..

Acetylation and formylation of 3-phenyl-cycl[3.2.2]azine derivatives, in the presence of Lewis acids, have been Investigated. It has been found that the orientation of substitution in 2-carbomethoxy- 3-phenyl-cycl[3.2.2]azine for these two reactions, under Identical conditions, is different.
