There is increasing recognition that transdisciplinary approaches are needed to create suitable knowledge for sustainable water management. However, there is no common understanding of what transdisciplinary research may be and there is very limited debate on potentials and challenges regarding its implementation. Against this background, this paper presents a conceptual framework for transdisciplinary co-production of knowledge in water management projects oriented towards more sustainable use of water. Moreover, first experiences with its implementation are discussed. In so doing, the focus lies on potentials and challenges related to the co-production of systems, target and transformation knowledge by researchers and local stakeholders.

BACKGROUND: The sensory drive hypothesis predicts that divergent sensory adaptation in different habitats may lead to premating isolation upon secondary contact of populations. Speciation by sensory drive has traditionally been treated as a special case of speciation as a byproduct of adaptation to divergent environments in geographically isolated populations. However, if habitats are heterogeneous, local adaptation in the sensory systems may cause the emergence of reproductively isolated species from a single unstructured population. In polychromatic fishes, visual sensitivity might become adapted to local ambient light regimes and the sensitivity might influence female preferences for male nuptial color. In this paper, we investigate the possibility of speciation by sensory drive as a byproduct of divergent visual adaptation within a single initially unstructured population. We use models based on explicit genetic mechanisms for color vision and nuptial coloration. RESULTS: We show that in simulations in which the adaptive evolution of visual pigments and color perception are explicitly modeled, sensory drive can promote speciation along a short selection gradient within a continuous habitat and population. We assumed that color perception evolves to adapt to the modal light environment that individuals experience and that females prefer to mate with males whose nuptial color they are most sensitive to. In our simulations color perception depends on the absorption spectra of an individual's visual pigments. Speciation occurred most frequently when the steepness of the environmental light gradient was intermediate and dispersal distance of offspring was relatively small. In addition, our results predict that mutations that cause large shifts in the wavelength of peak absorption promote speciation, whereas we did not observe speciation when peak absorption evolved by stepwise mutations with small effect. CONCLUSION: The results suggest that speciation can occur where environmental gradients create divergent selection on sensory modalities that are used in mate choice. Evidence for such gradients exists from several animal groups, and from freshwater and marine fishes in particular. The probability of speciation in a continuous population under such conditions may then critically depend on the genetic architecture of perceptual adaptation and female mate choice.

Understanding factors driving the ecology of N cycling microbial communities is of central importance for sustainable land use. In this study we report changes of abundance of denitrifiers, nitrifiers and nitrogen-fixing microorganisms (based on qPCR data for selected functional genes) in response to different land use intensity levels and the consequences for potential turnover rates. We investigated selected grassland sites being comparable with respect to soil type and climatic conditions, which have been continuously treated for many years as intensely used meadows (IM), intensely used mown pastures (IP) and extensively used pastures (EP), respectively. The obtained data were linked to above ground biodiversity pattern as well as water extractable fractions of nitrogen and carbon in soil. Shifts in land use intensity changed plant community composition from systems dominated by s-strategists in extensive managed grasslands to c-strategist dominated communities in intensive managed grasslands. Along the different types of land use intensity, the availability of inorganic nitrogen regulated the abundance of bacterial and archaeal ammonia oxidizers. In contrast, the amount of dissolved organic nitrogen determined the abundance of denitrifiers (nirS and nirK). The high abundance of nifH carrying bacteria at intensive managed sites gave evidence that the amounts of substrates as energy source outcompete the high availability of inorganic nitrogen in these sites. Overall, we revealed that abundance and function of microorganisms involved in key processes of inorganic N cycling (nitrification, denitrification and N fixation) might be independently regulated by different abiotic and biotic factors in response to land use intensity.

OBJECTIVES Evidence increases that cognitive failure may be used to screen for drivers at risk. Until now, most studies have relied on driving learners. This exploratory pilot study examines self-report of cognitive failure in driving beginners and error during real driving as observed by driving instructors. METHODS Forty-two driving learners of 14 driving instructors filled out a work-related cognitive failure questionnaire. Driving instructors observed driving errors during the next driving lesson. In multiple linear regression analysis, driving errors were regressed on cognitive failure with the number of driving lessons as an estimator of driving experience controlled. RESULTS Higher cognitive failure predicted more driving errors (p < .01) when age, gender and driving experience were controlled in analysis. CONCLUSIONS Cognitive failure was significantly associated with observed driving errors. Systematic research on cognitive failure in driving beginners is recommended.

In the present study, the tetracycline-off and Cre/loxP systems were combined to gain temporal and spatial control of transgene expression. Mice were generated that carried three transgenes: Tie2-tTA, tet-O-Cre and either the ZEG or ZAP reporter. Tie2-tTA directs expression of tetracycline-controlled transactivator (tTA) in endothelial and hematopoietic cells under the control of the Tie2 promoter. Tet-O-Cre produces Cre recombinase from a minimal promoter containing the tet-operator (tetO). ZEG or ZAP contains a strong promoter and a loxP-flanked stop sequence, followed by an enhanced green fluorescence protein (EGFP) or human placental alkaline phosphatase (hPLAP) reporter. In the presence of tetracycline, the tTA transactivator produced by Tie-2-tTA is disabled and Cre is not expressed. In the absence of tetracycline, the tTA binds tet-O-Cre to drive the expression of Cre, which recombines the loxP sites of the ZEG or ZAP transgene and results in reporter gene expression. In the present study, the expression of the ZEG or ZAP reporter genes in embryos and adult animals with and without tetracycline treatment was examined. In the presence of tetracycline, no reporter gene expression was observed. When tetracycline was withdrawn, Cre excision was activated and the reporter genes were detected in endothelial and hematopoietic cells. These results demonstrate that this system may be used to bypass embryonic lethality and access adult phenotypes.

EINLEITUNG Anhand eines Pelvitrainer Modells wurde ein sogenannter „Handheld Roboter“ (Kymerax© Precision- Drive Articulating Surgical System von Terumo©) mit konventionellen laparoskopischen Instrumenten verglichen. Das Kymerax© System verfügt über eine Instrumentenspitze, welche durch Knöpfe am Handgriff zusätzlich abgewinkelt und rotiert werden kann. METHODE 45 Probanden wurden in 2 Erfahrungsgruppen aufgeteilt: 20 ExpertInnen (mehr als 50 selbstständig durchgeführte laparoskopische Operationen pro Jahr) und 25 StudentInnen (keine Erfahrung in der Laparoskopie). Sie führten 6 standardisierte Übungen durch, wobei die ersten beiden Übungen jeweils nur der Instrumenteninstruktion dienten und nicht ausgewertet wurden. In den restlichen 4 Übungen wurden Zeit, Fehleranzahl und Präzision erfasst. Es wurde in 2 Gruppen randomisiert. Eine Gruppe führte die Übungen zuerst mit dem konventionellen System und dann mit dem Kymerax© System durch. Bei der anderen Gruppe erfolgten die Übungen in umgekehrter Reihenfolge. Am Ende beantworteten die Teilnehmer Fragen zu den Übungen und den Operationssystemen. Die Daten wurden mittels Varianzanalyse ausgewertet. RESULTATE In allen 4 gemessenen Übungen brauchten die Probanden mit Kymerax© signifikant mehr Zeit (20%-40%). Vorteile des Kymerax© Systems waren eine bessere Nadelkontrolle bei einer auf den Operateur gerichteten Stichrichtung, eine geringere Abweichung beim Schneiden einer graden Linie, sowie ein geringeres Ausfransen der Schnittlinie beim graden wie beim runden Schneiden. Im Gegensatz zu den Experten kamen Studenten, welche das Kymerax© System in der zweiten Runde verwendeten, besser mit diesem zu Recht, als Ihre Studentenkollegen, die das Kymerax© System in der ersten Runde verwendeten. In der Befragung gaben über 90% der Teilnehmer an, dass das Kymerax© System bei der Durchführung der Übungen einen Vorteil bringt. Die Probanden empfanden jedoch die Bedienung als gewöhnungsbedürftig und erschöpften mit dem Kymerax© System schneller. Bemängelt wurde beim Kymerax© System die nicht freie Rotation, die eingeschränkte Abwinklung, die Sichteinschränkung durch den 7mm Schaft sowie die Ergonomie des Handgriffs. DISKUSSION Das Kymerax© System bringt Vorteile bei gewissen komplexen laparoskopischen Aufgaben. Der Preis hierfür ist die langsamere Durchführung der Aufgaben, die längere Angewöhnungszeit an das Instrument sowie die schnellere Ermüdung des Benutzers. Das System zeigt ein grosses Potential für die laparoskopische Chirurgie, jedoch sind weitere Verbesserungen notwendig. Von der Firma Terumo© wurde zwischenzeitlich das Operationssystem vom Markt genommen.
