In order to determine the extent and timing of dyke formation in the Ladakh Batholith we examined about 30 mostly andesitic dykes intruding the Ladakh batholith in a ca. 50 km wide area to the west of Leh (NW India). The dykes in the east of the area trend E-NE and those in the west trend N-NW. The difference in orientation is also evident in the petrography and isotopic signatures. The eastern dykes contain corroded quartz xenocrysts and show negative ε0(Nd) and positive ε0(Sr) values, where as the western dykes do not contain quartz xenocrysts and exhibit positive ε0(Nd) and near-zero ε0(Sr) values. The variability in Sr-Nd isotopes (ε0(Nd) = 3.6 to −9.6, ε0(Sr) = 0.4 to 143) and the quartz xenocrysts can best be explained by (differing degrees of) crustal assimilation of the parent magma of the dykes. Separated minerals from five dykes were dated by 40Ar-39Ar incremental heating: amphibole ages range between 50 and 54 Ma, and one biotite dated both by Rb-Sr and by 40Ar-39Ar gave an age of 45 Ma. One dated pseudotachylyte sample attests to brittle faulting at ca. 54 Ma. The combination of structural field evidence with petrographic, isotopic and geochronological analyses demonstrates that the dykes did not form from a single, progressively differentiating magma chamber, despite having formed in the same tectonic setting around the same time, and that processes such as crustal assimilation and magma mixing/mingling also played a significant role in magma petrogenesis.

Palaeoclimatic stability is regarded as an important factor in explaining patterns of endemism in the Azorean flora. However, modelling simulations and quantitative reconstructions for the last 6000 years suggest considerable palaeoclimatic variability. Here we explore the link between Holocene palaeoclimate and palaeovegetation on the islands of Flores and Pico. Modern pollen assemblages indicate that most major plant communities can be detected using pollen analysis and that, in some cases, the pre-colonisation vegetation was quite similar to present-day relict vegetation. A 200–500-year pollen record from Alagoinha, a low-elevation mire in western Flores, shows that Juniperus brevifolia-dominated communities were widespread at lower elevations prior to large-scale deforestation. Today these communities are generally restricted to higher elevations. While our results are preliminary, there appears to be a weak link between palaeovegetation (which was primarily influenced by volcanism, soil formation and human impact) and palaeoclimatic changes detected through geochemical proxies. Even if the Azorean palaeoclimate varied substantially, its impact on the pristine vegetation, at least in terms of pollen production, was relatively small.

The tropical montane forests of the E Andean cordillera in Ecuador receive episodic Sahara-dust inputs particularly increasing Ca deposition. We added CaCl2 to isolate the effect of Ca deposition by Sahara dust to tropical montane forest from the simultaneously occurring pH effect. We examined components of the Ca cycle at four control plots and four plots with added Ca (2 × 5 kg ha–1 Ca annually as CaCl2) in a random arrangement. Between August 2007 and December 2009 (four applications of Ca), we determined Ca concentrations and fluxes in litter leachate, mineral soil solution (0.15 and 0.30 m depths), throughfall, and fine litterfall and Al concentrations and speciation in soil solutions. After 1 y of Ca addition, we assessed fine-root biomass, leaf area, and tree growth. Only < 3% of the applied Ca leached below the acid organic layer (pH 3.5–4.8). The added CaCl2 did not change electrical conductivity in the root zone after 2 y. In the second year of fertilization, Ca retention in the canopy of the Ca treatment tended to decrease relative to the control. After 2 y, 21% of the applied Ca was recycled to soil with throughfall and litterfall. One year after the first Ca addition, fine-root biomass had decreased significantly. Decreasing fine-root biomass might be attributed to a direct or an indirect beneficial effect of Ca on the soil decomposer community. Because of almost complete association of Al with dissolved organic matter and high free Ca2+ : Al3+ activity ratios in solution of all plots, Al toxicity was unlikely. We conclude that the added Ca was retained in the system and had beneficial effects on some plants.

Neuronal precursor cell-expressed developmentally down-regulated 4 (Nedd4) proteins are ubiquitin ligases, which attach ubiquitin moieties to their target proteins, a post-translational modification that is most commonly associated with protein degradation. Nedd4 ubiquitin ligases have been shown to down-regulate both potassium and sodium channels. In this study, we investigated whether Nedd4 ubiquitin ligases also regulate Ca(v) calcium channels. We expressed three Nedd4 family members, Nedd4-1, Nedd4-2, and WWP2, together with Ca(v)1.2 channels in tsA-201 cells. We found that Nedd4-1 dramatically decreased Ca(v) whole-cell currents, whereas Nedd4-2 and WWP2 failed to regulate the current. Surface biotinylation assays revealed that Nedd4-1 decreased the number of channels inserted at the plasma membrane. Western blots also showed a concomitant decrease in the total expression of the channels. Surprisingly, however, neither the Ca(v) pore-forming α1 subunit nor the associated Ca(v)β and Ca(v)α(2)δ subunits were ubiquitylated by Nedd4-1. The proteasome inhibitor MG132 prevented the degradation of Ca(v) channels, whereas monodansylcadaverine and chloroquine partially antagonized the Nedd4-1-induced regulation of Ca(v) currents. Remarkably, the effect of Nedd4-1 was fully prevented by brefeldin A. These data suggest that Nedd4-1 promotes the sorting of newly synthesized Ca(v) channels for degradation by both the proteasome and the lysosome. Most importantly, Nedd4-1-induced regulation required the co-expression of Ca(v)β subunits, known to antagonize the retention of the channels in the endoplasmic reticulum. Altogether, our results suggest that Nedd4-1 interferes with the chaperon role of Ca(v)β at the endoplasmic reticulum/Golgi level to prevent the delivery of Ca(v) channels at the plasma membrane.

A number of mathematical models for predicting growth and final height outcome have been proposed to enable the clinician to 'individualize' growth-promoting treatment. However, despite optimizing these models, many patients with isolated growth hormone deficiency (IGHD) do not reach their target height. The aim of this study was to analyse the impact of polymorphic genotypes [CA repeat promoter polymorphism of insulin-like growth factor-I (IGF-I) and the -202 A/C promoter polymorphism of IGF-Binding Protein-3 (IGFBP-3)] on variable growth factors as well as final height in severe IGHD following GH treatment. DESIGN, PATIENTS AND CONTROLS: One hundred seventy eight (IGF-I) and 167 (IGFBP-3) subjects with severe growth retardation because of IGHD were studied. In addition, the various genotypes were also studied in a healthy control group of 211 subjects.
