The norepinephrine transporter inhibitor reboxetine reduces stimulant effects of MDMA ("ecstasy") in humans

This study assessed the pharmacodynamic and pharmacokinetic effects of the interaction between the selective norepinephrine (NE) transporter inhibitor reboxetine and 3,4-methylenedioxymethamphetamine (MDMA, "ecstasy") in 16 healthy subjects. The study used a double-blind, placebo-controlled crossover design. Reboxetine reduced the effects of MDMA including elevations in plasma levels of NE, increases in blood pressure and heart rate, subjective drug high, stimulation, and emotional excitation. These effects were evident despite an increase in the concentrations of MDMA and its active metabolite 3,4-methylenedioxyamphetamine (MDA) in plasma. The results demonstrate that transporter-mediated NE release has a critical role in the cardiovascular and stimulant-like effects of MDMA in humans.

Duloxetine inhibits effects of MDMA ("ecstasy") in vitro and in humans in a randomized placebo-controlled laboratory study

This study assessed the effects of the serotonin (5-HT) and norepinephrine (NE) transporter inhibitor duloxetine on the effects of 3,4-methylenedioxy-methamphetamine (MDMA, ecstasy) in vitro and in 16 healthy subjects. The clinical study used a double-blind, randomized, placebo-controlled, four-session, crossover design. In vitro, duloxetine blocked the release of both 5-HT and NE by MDMA or by its metabolite 3,4-methylenedioxyamphetamine from transmitter-loaded human cells expressing the 5-HT or NE transporter. In humans, duloxetine inhibited the effects of MDMA including elevations in circulating NE, increases in blood pressure and heart rate, and the subjective drug effects. Duloxetine inhibited the pharmacodynamic response to MDMA despite an increase in duloxetine-associated elevations in plasma MDMA levels. The findings confirm the important role of MDMA-induced 5-HT and NE release in the psychotropic effects of MDMA. Duloxetine may be useful in the treatment of psychostimulant dependence.

Sozialismus ohne Wachstum und Technologie? Die Linke in der Schweiz und die Umweltfrage 1968–1990

Die Arbeiterbewegung hat sich ursprünglich der sozioökonomischen Schattenseite der Industrialisierung verschrieben. Für die ökologischen Nebenfolgen der technoökonomischen Entwicklung haben die linken Gesellschaftskritiker insofern kein Gespür gezeigt, als der historische Naturschutz bis in die 1970er Jahre eine traditionelle Domäne der Konservativen war. Die Linke verstand sich selbst als „progressiv“ und glaubte, einzig mit dem Fortschreiten von Wachstum und Technologie die Utopie einer sozialistischen Zukunft realisieren zu können. Dann aber – im Zuge der ökologischen Zäsur Ende der 1960er Jahre – wurde das Verhältnis zur Natur aus einer neuen Perspektive betrachtet. Die sich für die Umwelt zuständig fühlenden Kräfte gruppierten sich um. Das Umweltthema geriet in das Blickfeld der Arbeiterparteien, die sich mit dem eigenen Fortschrittsmythos auseinander zu setzen hatten. Wie haben die „Progressiven“ die Diskussion um die Grenzen der wirtschaftlichen und technologischen Entwicklung geführt? Welche traditionellen Denkmuster wurden im Zuge der Lernprozesse ökologisch aufgebrochen? Welchen Einfluss hatte der Umweltdiskurs der Linken auf das gesamtgesellschaftliche Reden und Denken über die Umwelt?

The matrix metalloproteinase inhibitor RS-130830 attenuates brain injury in experimental pneumococcal meningitis.

BACKGROUND Pneumococcal meningitis (PM) is characterized by high mortality and morbidity including long-term neurofunctional deficits. Neuropathological correlates of these sequelae are apoptosis in the hippocampal dentate gyrus and necrosis in the cortex. Matrix metalloproteinases (MMPs) play a critical role in the pathophysiology of PM. RS-130830 (Ro-1130830, CTS-1027) is a potent partially selective inhibitor of MMPs of a second generation and has been evaluated in clinical trials as an anti-arthritis drug. It inhibits MMPs involved in acute inflammation but has low activity against MMP-1 (interstitial collagenase), MMP-7 (matrilysin) and tumour necrosis factor α converting enzyme (TACE). METHODS A well-established infant rat model of PM was used where live Streptococcus pneumoniae were injected intracisternally and antibiotic treatment with ceftriaxone was initiated 18 h post infection (hpi). Treatment with RS-130830 (75 mg/kg bis in die (bid) i.p., n = 40) was started at 3 hpi while control littermates received the vehicle (succinylated gelatine, n = 42). RESULTS Cortical necrosis was significantly attenuated in animals treated with RS-130830, while the extent of hippocampal apoptosis was not influenced. At 18 hpi, concentrations of interleukin (IL)-1β and IL-10 were significantly lower in the cerebrospinal fluid of treated animals compared to controls. RS-130830 significantly reduced weight loss and leukocyte counts in the cerebrospinal fluid of survivors of PM. CONCLUSION This study identifies MMP inhibition, specifically with RS-130830, as an efficient strategy to attenuate disease severity and cortical brain injury in PM.

Activation of the epidermal growth factor receptor (EGFR) by a novel metalloprotease pathway

Neutrophil Elastase (NE) is a pro-inflammatory protease present at higher than normal levels in the lung during inflammatory disease. NE regulates IL-8 production from airway epithelial cells and can activate both EGFR and TLR4. TACE/ADAM17 has been reported to trans-activate EGFR in response to NE. Here, using 16HBE14o-human bronchial epithelial cells we demonstrate a new mechanism by which NE regulates both of these events. A high molecular weight soluble metalloprotease activity detectable only in supernatants from NE-treated cells by gelatin and casein zymography was confirmed to be meprin alpha by Western immunoblotting. In vitro studies demonstrated the ability of NE to activate meprin alpha, which in turn could release soluble TGFalpha and induce IL-8 production from 16HBE14o- cells. These effects were abrogated by actinonin, a specific meprin inhibitor. NE-induced IL-8 expression was also inhibited by meprin alpha siRNA. Immunoprecipitation studies detected EGFR/TLR4 complexes in NE-stimulated cells overexpressing these receptors. Confocal studies confirmed colocalization of EGFR and TLR4 in 16HBE14o- cells stimulated with meprin alpha. NFkappaB was also activated via MyD88 in these cells by meprin alpha. In bronchoalveolar lavage fluid from NE knock-out mice infected intra-tracheally with Pseudomonas aeruginosa meprin alpha was significantly decreased compared with control mice, and was significantly increased and correlated with NE activity, in bronchoalveolar lavage fluid from individuals with cystic fibrosis but not healthy controls. The data describe a previously unidentified lung metalloprotease meprin alpha, and its role in NE-induced EGFR and TLR4 activation and IL-8 production.

Aphrodite zählt zu den bekanntesten und interessantesten mythischen Frauengestalten der Antike. Als Göttin der Liebe und der Schönheit nahm sie eine Schlüsselrolle im Trojanischen Krieg ein; als Göttin in Waffen besaß sie aber auch selbst kriegerische Charakterzüge. Das Buch geht der Frage nach, wie der Aphrodite-Kult entstand und sich verbreitete. Dabei entdecken die Autoren z. B. Einflüsse von weiblichen Kriegsgottheiten aus dem alten Orient. Auf der Insel Zypern lässt sich die Verehrung der Aphrodite bis in römische Zeit nachvollziehen. Und so ist diese Göttin nicht nur eine immer noch populäre Gestalt der Mythologie, sondern zugleich ein Beispiel für den kulturellen Austausch zwischen Ost und West.

Wahrheitskommissionen als Instrument der Transitional Justice. Genese, Entwicklung, Verbreitung

Wahrheitskommissionen haben in den vergangenen rund dreißig Jahren eine bemerkenswerte internationale Karriere aufzuweisen gehabt. Sie haben sich dabei von einer der wichtigsten Innovationen im gesellschaftlichen Umgang mit historischem Unrecht zu einem globalen Aufarbeitungsstandard entwickelt. Bis in die Gegenwart sind Dutzende Wahrheitskommissionen auf fünf Kontinenten eingesetzt worden. Diese Entwicklung ist eng mit derjenigen des Feldes der Transitional Justice verbunden gewesen. Der Beitrag betrachtet die Genese, die Entwicklung und die Verbreitung der Wahrheitskommissionen als zentrales Instrument von Transitional Justice.
