Orphan- or understudied-crops are mostly staple food crops in developing world. They are broadly classified under cereals, legumes, root crops, fruits and vegetables. These under-researched crops contribute to the diet of a large portion of resource-poor consumers and at the same time generate income for small-holder farmers in developing countries, particularly in Africa. In addition, they perform better than major crops of the world under extreme soil and climatic conditions. However, orphan crops are not without problems. Due to lack of scientific investigation, most of them produce low yields while others have a variety of toxins that affect the health of consumers. Here, we present some highlights on the status and future perspectives of the Tef Biotechnology Project that employs modern improvement technique in order to genetically improve tef (Eragrostis tef), one of the most important orphan crop in Africa. A reverse genetics approach known as TILLING (Targeting Induced Local Lesions IN Genome) is implemented in order to tackle lodging, the major yield limiting factor in tef.Key words: Orphan crops, underresearched crops, Eragrostis tef, TILLING, semi-dwarf.

The SLC43 family is composed of only three genes coding for the plasma membrane facilitator system l amino acid transporters LAT3 (SLC43A1; TC 2.A.1.44.1) and LAT4 (SLC43A2; TC 2.A.1.44.2), and the orphan protein EEG1 (SLC43A3; TC 2.A.1.44.3). Besides the known mechanism of transport of LAT3 and LAT4, their physiological roles still remain quite obscure. Morphants suggested a role of LAT3 in renal podocyte development in zebrafish. Expression in liver and skeletal muscle, and up-regulation by starvation suggest a role of LAT3 in the flux of branched-chain amino acids (BCAAs) from liver and skeletal muscle to the bloodstream. Finally, LAT3 is up-regulated in androgen-dependent cancers, suggesting a role in mTORC1 signaling in this type of tumors. In addition, LAT4 might contribute to the transfer of BCAAs from mother to fetus. Unfortunately, the EEG1 mouse model (EEG1(Y221∗)) described here has not yet offered a clue to the physiological role of this orphan protein.

Arzneimittel für seltene Krankheiten (orphan drugs) lassen sich unter normalen Marktbedingungen kaum kostendeckend erforschen, entwickeln und vermarkten, was zu einem Mangel an geeigneten Therapien und zugelassenen Arzneimitteln für seltene Krankheiten führt. Um den fortdauernden Ausschluss von betroffenen Patienten vom medizinischen Fortschritt zu verhindern und die Defizite in der Erforschung und Entwicklung von orphan drugs zu beseitigen, schuf die Europäische Union – nach US-amerikanischem Vorbild – ein Anreizsystem. Der vorliegende Beitrag erläutert die regulatorischen Rahmenbedingungen für orphan drugs in der EU und zeigt auf, dass die Schweiz in der Schaffung günstiger Rahmenbedingungen für Patientinnen und Patienten, die an einer seltenen Krankheit leiden, anderen Ländern um Jahre hinter her hinkt, sich aber allmählich etwas tut.

While the safety and availability of medicinal products for the majority of adult patients has steadily improved in recent decades, for children and people suffering from rare diseases (orphan diseases) there is a lack of approved medicinal products for these patient populations. Since the research and development of medicinal products is associated with high costs, the costs for paediatric medicinal products and medicines for rare diseases (orphan drugs) may barely be covered under normal market conditions due to the small patient populations. In order to prevent the continued exclusion of children and persons suffering from rare diseases from medical progress and to eliminate the deficits in the research and development of medicinal products for these patient groups, the European Union created, along the lines of the U.S. model, a system of incentives and constraints. Since 2000, under Regulation (EC) No. 141/2000 (Orphan Drug Regulation) there has been an incentive system to encourage the research and development of orphan drugs. With the goal of improving the health of children in Europe, Regulation (EC) No. 1901/2006 (Paediatric Regulation) combines economic incentives with the requirement to conduct paediatric studies. This article explains and comments on the specific regulatory framework for orphan drugs and paediatric medicinal products in the European Union.

The paper will focus on basic ways of communication between the actors in the house and home and their direct social environments (esp. neighbourhood) during the early modern period. Such ways of communication were established in and through work relations, sociability, social control and certain liminal rites. So far underestimated, the neighbourhood was both helpful and inevitable to keep house and household running. A typical aspect of the practice of communication was the importance of repetitive performative events in everyday life. In order to establish and maintain social relations, the honour of the ‘house’ as such and fundamental roles like housefather and housemother had to be performed under the eyes of neighbours and other actors. Thus, empirical evidence reveals the house and home as a specific kind of stage. In contrast to the outdated concept of ‘das ganze Haus’ (the whole house) by Otto Brunner and also a reduced socioeconomic understanding of household, the openness of the house proves to be a highly relevant feature of early modern society. This openness refers to accessibility, visibility and control. The paper will explain the proposed concept and analyse concrete examples from work and wedding.
