The production, segregation and migration of melt and aqueous fluids (henceforth called liquid) plays an important role for the transport of mass and energy within the mantle and the crust of the Earth. Many properties of large-scale liquid migration processes such as the permeability of a rock matrix or the initial segregation of newly formed liquid from the host-rock depends on the grain-scale distribution and behaviour of liquid. Although the general mechanisms of liquid distribution at the grain-scale are well understood, the influence of possibly important modifying processes such as static recrystallization, deformation, and chemical disequilibrium on the liquid distribution is not well constrained. For this thesis analogue experiments were used that allowed to investigate the interplay of these different mechanisms in-situ. In high-temperature environments where melts are produced, the grain-scale distribution in “equilibrium” is fully determined by the liquid fraction and the ratio between the solid-solid and the solid-liquid surface energy. The latter is commonly expressed as the dihedral or wetting angle between two grains and the liquid phase (Chapter 2). The interplay of this “equilibrium” liquid distribution with ongoing surface energy driven recrystallization is investigated in Chapter 4 and 5 with experiments using norcamphor plus ethanol liquid. Ethanol in contact with norcamphor forms a wetting angle of about 25°, which is similar to reported angles of rock-forming minerals in contact with silicate melt. The experiments in Chapter 4 show that previously reported disequilibrium features such as trapped liquid lenses, fully-wetted grain boundaries, and large liquid pockets can be explained by the interplay of the liquid with ongoing recrystallization. Closer inspection of dihedral angles in Chapter 5 reveals that the wetting angles are themselves modified by grain coarsening. Ongoing recrystallization constantly moves liquid-filled triple junctions, thereby altering the wetting angles dynamically as a function of the triple junction velocity. A polycrystalline aggregate will therefore always display a range of equilibrium and dynamic wetting angles at raised temperature, rather than a single wetting angle as previously thought. For the deformation experiments partially molten KNO3–LiNO3 experiments were used in addition to norcamphor–ethanol experiments (Chapter 6). Three deformation regimes were observed. At a high bulk liquid fraction >10 vol.% the aggregate deformed by compaction and granular flow. At a “moderate” liquid fraction, the aggregate deformed mainly by grain boundary sliding (GBS) that was localized into conjugate shear zones. At a low liquid fraction, the grains of the aggregate formed a supporting framework that deformed internally by crystal plastic deformation or diffusion creep. Liquid segregation was most efficient during framework deformation, while GBS lead to slow liquid segregation or even liquid dispersion in the deforming areas.

In order to eliminate the de Gennes packing problem, which usually limits the attainable size of dendrimers, a new branching unit containing para-tetraphenylene ethynyl arms has been synthesized and utilized in the preparation of dendrimers of the Müllen type. The divergent principle of synthesis, based on the Dilthey reaction, could be carried up to sixth generation which contains 2776 benzene rings and possesses a diameter in the 27 nm range ("exploded dendrimer"). Monodispersity and dimensions of this and the lower generation species have been studied by MALDI-TOF MS (including the very recent superconducting tunnel junction detector), by size-exclusion chromatography, dynamic light scattering, transmission electron microscopy, and atomic force microscopy. Interesting features, apart from the huge dimension, are the low density and high porosity of these giant molecules which cause extensive aggregation in the gas phase, flattening on solid support (AFM) and the ready incorporation of guest molecules in the condensed phase. Since the synthesis of the para-tetraphenylene arms is quite elaborate, similar dendrimers containing para-terphenylene arms have been prepared; they are accessible more economically ("semi-exploded dendrimers"). It has been shown that they in several aspects mimic the features of the "exploded dendrimers". In order to take advantage of the presence of large internal cavities in this dendrimer type, dendrons containing -C≡C- triple bonds have also been incorporated. Surprisingly, they are readily hydrogenated under the condition of heterogeneous catalysis (Pd/C) which demonstrates the large size of the cavities. As revealed by a quartz microbalance study the post-hydrogenation dendrimers are less prone to incorporate guest molecules than before hydrogenation. Obviously, the more flexible nature of the former reduces porosity, it also leads to significant shrinkage. An interesting perspective is the use of homogeneous hydrogenation catalysts of variable size with the aim of determining the dimension of internal free space.

Disruption of the blood-brain barrier (BBB) results in cerebral edema formation, which is a major cause for high mortalityrnafter traumatic brain injury (TBI). As anesthetic care is mandatory in patients suffering from severe TBI it may be importantrnto elucidate the effect of different anesthetics on cerebral edema formation. Tight junction proteins (TJ) such as zonularnoccludens-1 (ZO-1) and claudin-5 (cl5) play a central role for BBB stability. First, the influence of the volatile anestheticsrnsevoflurane and isoflurane on in-vitro BBB integrity was investigated by quantification of the electrical resistance (TEER) inrnmurine brain endothelial monolayers and neurovascular co-cultures of the BBB. Secondly brain edema and TJ expression ofrnZO-1 and cl5 were measured in-vivo after exposure towards volatile anesthetics in native mice and after controlled corticalrnimpact (CCI). In in-vitro endothelial monocultures, both anesthetics significantly reduced TEER within 24 hours afterrnexposure. In BBB co-cultures mimicking the neurovascular unit (NVU) volatile anesthetics had no impact on TEER. In healthyrnmice, anesthesia did not influence brain water content and TJ expression, while 24 hours after CCI brain water contentrnincreased significantly stronger with isoflurane compared to sevoflurane. In line with the brain edema data, ZO-1 expressionrnwas significantly higher in sevoflurane compared to isoflurane exposed CCI animals. Immunohistochemical analysesrnrevealed disruption of ZO-1 at the cerebrovascular level, while cl5 was less affected in the pericontusional area. The studyrndemonstrates that anesthetics influence brain edema formation after experimental TBI. This effect may be attributed tornmodulation of BBB permeability by differential TJ protein expression. Therefore, selection of anesthetics may influence thernbarrier function and introduce a strong bias in experimental research on pathophysiology of BBB dysfunction. Futurernresearch is required to investigate adverse or beneficial effects of volatile anesthetics on patients at risk for cerebral edema.

Organophosphate können chronische Lungenerkrankungen und Vergiftungen hervorrufen. Bei der Vergiftung erfolgt eine Immunreaktion, welche noch nicht erforscht ist. In dieser Arbeit wurden Toxizitätsstudien an dendritischen Zellen und einem bronchialen Triple-Kultur-Modell durchgeführt. Dendritische Zellen spielen bei der ersten Immunabwehr in der Lunge eine große Rolle. Aus der Zell-Linie THP-1 und primären Monozyten wurden reife dendritische Zellen differenziert und mittels Durchflusszytometrie und Immunfluoreszenz auf spezifische Zellmarker, wie zum Beispiel CD11c, CD83 oder auch CD209, charakterisiert und etabliert. Durch die Vergiftung der Zellen mit Dimethoat und Chlorpyrifos konnte eine Erhöhung des Zelltodes, die Sekretion von proinflammatorischen Mediatoren, Veränderungen in der Morphologie der Zellen und ein Effekt auf den Proteinkinase-Signalweg festgestellt werden. Spezifische dendritische Zellmarker (CD83, CD209) wurden inhibiert und die Dendriten der Dendritischen Zellen kürzer und beschädigt. Die Schädigung von Chlorpyrifos war erheblich größer, als die bei Dimethoat.rnDie weiteren Toxizitätsstudien wurden an einem bronchialen Triple-Kultur-Modell durchgeführt. Hierzu wurden auf Transwell-Filtermembranen bronchiale Epithelzellen, Fibroblastenzellen und Dendritische Zellen verwendet. Die bronchialen Epithelzellen und Fibroblastenzellen waren hier physiologisch voneinander getrennt, konnten aber durch Poren in der Membran miteinander interagieren. Die Etablierung des Triple-Kultur-Modells erfolgte durch die Untersuchung von Entzündungsprozessen, durch Stimulation mit LPS, TNF-alpha und Interferon-gamma. In der Ko-Kultur konnten Zell-Zell-Kontakt Schädigungen und Erhöhung von proinflammatorischen Markern, wie zum Beispiel IL-1ß, IL-6 oder auch IL-8 gemessen werden. Versuche in der Triple-Kultur zeigten den positiven Effekt von Dendritischen Zellen. Bei höheren Konzentrationsbereichen von Dimethoat und Chlorpyrifos konnte ein Wandern der Zellen zu den geschädigten Zell-Zell-Kontakten nachgewiesen werden. Die Ausschüttung der proinflammatorischen Mediatoren wurde inhibiert, vor allem bei IL-10 war eine deutliche Reduktion, um mehr als 70% messbar. Ebenso konnten Veränderungen in dem Apoptose-Signalblick festgestellt werden. Vor allem anti-apoptotische Proteine wurden nach einer Vergiftung der Triple-Kultur induziert. Interventionsstudien mit Vitamin C zeigten allerdings keinen positiven Effekt.

Measurements of the self coupling between bosons are important to test the electroweak sector of the Standard Model (SM). The production of pairs of Z bosons through the s-channel is forbidden in the SM. The presence of physics, beyond the SM, could lead to a deviation of the expected production cross section of pairs of Z bosons due to the so called anomalous Triple Gauge Couplings (aTGC). Proton-proton data collisions at the Large Hadron Collider (LHC) recorded by the ATLAS detector at a center of mass energy of 8 TeV were analyzed corresponding to an integrated luminosity of 20.3 fb-1. Pairs of Z bosons decaying into two electron-positron pairs are searched for in the data sample. The effect of the inclusion of detector regions corresponding to high values of the pseudorapidity was studied to enlarge the phase space available for the measurement of the ZZ production. The number of ZZ candidates was determined and the ZZ production cross section was measured to be: rn7.3±1.0(Stat.)±0.4(Sys.)±0.2(lumi.)pb, which is consistent with the SM expectation value of 7.2±0.3pb. Limits on the aTGCs were derived using the observed yield, which are twice as stringent as previous limits obtained by ATLAS at a center of mass energy of 7 TeV.
