Education is generally perceived as a public good which should be provided by the state. In Egypt, free and equal access to education has been guaranteed to all citizens since President Nasser’s socialist reforms in the 1950s. However, due to high population growth rates and a lack of financial resources, the public education system has been struggling to accommodate rapidly increasing numbers of students. While enrolment rates have risen steadily, the quality of state-provided services has deteriorated. Teachers and students have to cope with high class densities, insufficient facilities, a rigid syllabus and a centralized examination system. Today, teaching is among the lowest-paying occupations in the public sector. One strategy to cope with this situation is the widespread practice of private tutoring, which usually takes place at students’ homes or in commercial tutoring centers. Based on research carried out in Cairo in 2004/05 and 2006, I use an actor-centered approach to analyze the motivations of Egyptian teachers and students for participating in private tutoring and the impact that this practice has on the relationship between teachers and students. Students of all socio-economic backgrounds resort to tutoring in order to succeed in a highly competitive and exam-oriented education system. However, the form and quality of tutoring that can be accessed depends on the financial means of the family. For teachers, tutoring provides a good opportunity not only to supplement their income, but also, in the case of renowned “star teachers”, to improve their professional status and autonomy. On the informal “market of education” that has developed in Egypt during the last decades, the educational responsibilities of the state are increasingly being taken over by private actors, i.e. the process of teaching and learning is dissociated from the direct control of the state and from school as an institution. At the same time, education is turned into a marketable commodity. Despite the government’s efforts to provide free education to all citizens, the quality of social services that can be accessed in Egypt, thus, depends mainly on the financial means of the individual or the family.

The global mid-ocean ridge system creates oceanic crust and lithosphere that covers more than two-thirds of the Earth. Basalts are volumetrically the most important rock type sampled at mid-ocean ridges. For this reason, our present understanding of upper mantle dynamics and the chemical evolution of the earth is strongly influenced by the study of mid-ocean ridge basalts (MORB). However, MORB are aggregates of polybarically generated small melt increments that can undergo a variety of physical and chemical processes during their ascent and consequently affect their derivative geochemical composition. Therefore, MORB do not represent “direct” windows to the underlying upper mantle. Abyssal peridotites, upper mantle rocks recovered from the ocean floor, are the residual complement to MORB melting and provide essential information on melt extraction from the upper mantle. In this study, abyssal peridotites are examined to address these overarching questions posed by previous studies of MORB: How are basaltic melts formed in the mantle, how are they extracted from the mantle and what physical and chemical processes control mantle melting? The number of studies on abyssal peridotites is small compared to those on basalts, in part because seafloor exposures of abyssal peridotites are relatively rare. For this reason, abyssal peridotite characteristics need to be considered in the context of subaerially exposed peridotites associated with ophiolites, orogenic peridotite bodies and basalt-hosted xenoliths. However, orogenic peridotite bodies are mainly associated with passive continental margins, most ophiolites are formed in supra-subduction zone settings, and peridotite xenoliths are often contaminated by their host magma. Therefore, studies of abyssal peridotites are essential to understanding the primary characteristics of the oceanic upper mantle free from the influence of continental rifting, subduction and tectonic emplacement processes. Nevertheless, numerous processes such as melt stagnation and cooling-induced, inter-mineral exchange can affect residual abyssal peridotite compositions after the cessation of melting. The aim of this study is to address these post-melting modifications of abyssal peridotites from a petrological-geochemical perspective. The samples in this study were dredged along the axis of the ultraslow-spreading Gakkel Ridge in the Arctic Ocean within the “Sparsely Magmatic Zone”, a 100 km ridge section where only mantle rocks are exposed. During two expeditions (ARK XVII-2 in 2001 and ARK XX-2 in 2004), exceptionally fresh peridotites were recovered. The boulders and cobbles collected cover a range of mantle rock compositions, with most characterized as plagioclase-free spinel peridotites or plagioclase- spinel peridotites. This thesis investigates melt stagnation and cooling processes in the upper mantle and is divided into two parts. The first part focuses on processes in the stability field of spinel peridotites (>10 kb) such as melt refertilization and cooling related trace element exchange, while the second part investigates processes in the stability field of plagioclase peridotites (< 10 kb) such as reactive melt migration and melt stagnation. The dissertation chapters are organized to follow the theoretical ascent of a mantle parcel upwelling beneath the location where the samples were collected.

Die Metalloprotease Ovastacin, ein Vertreter der Astacin-Familie, wurde erstmals 2004 beschrieben. Im Ovar von Säugetieren ist Ovastacin-mRNA im Zeitfenster vom Stadium der Sekundärfollikel bis kurz nach der Befruchtung der Eizelle zu finden. Der Expressionsort und -zeitpunkt sowie die Sequenzähnlichkeit von über 60% mit sogenannten „Schlüpfenzymen“ (engl. hatching enzymes), die man in den Eizellen und Zygoten niederer Wirbeltiere und Wirbelloser gefunden hatte, ließen die Vermutung aufkommen, es könnte sich hier um das Säugerhomolog dieser Proteasen handeln. Generell lösen hatching Enzyme die derben embryonalen Hüllstrukturen (bei Säugern die Zona pellucida, ZP) beim Schlüpfvorgang auf. Die essentielle Bedeutung des Ovastacins für die Befruchtung wird durch die um ca. 30% reduzierte Fruchtbarkeit von Ovastacin defizienten Mäusen belegt. Hochinteressant war in diesem Zusammenhang die Entdeckung des Ovastacins in den Cortikalgranula der Oocyten sowie seine Fähigkeit, das Zona pellucida Protein 2 zu schneiden. Die dadurch bewirkte Verhärtung der Zona pellucida verhindert das Eindringen weiterer Spermien, das heißt sie baut eine Barriere gegen Polyspermie auf. Ziel dieser Arbeit war es, Belege für die physiologische Funktion des Ovastacins zu finden. Vor allem galt es, potentielle Aktivatoren zu identifizieren, da das Enzym wie alle Astacine als inaktive Vorstufe gebildet wird, die proteolytisch aktiviert werden muss. Zu diesem Zweck exprimierte ich rekombinantes Pro-Ovastacin in Insektenzellen. Aktivierungsstudien in vitro zeigten, dass ein saures Milieu zu einer Aktivierung führt, ohne die Abspaltung des Propeptids zu bewirken. Sequenzalignments und ein homologes Strukturmodell des Ovastacins wiesen auf Trypsin- oder Elastase-ähnliche Serinproteasen als potentielle Aktivierungsenzyme hin. Tatsächlich konnte mit diesen beiden Proteasetypen zum ersten Mal aktives Ovastacin aus Pro-Ovastacin erzeugt werden. Trypsin kommt als physiologischer Aktivator allerdings nicht in Betracht, da es bisher in keinem der Gewebe nachgewiesen werden konnte, in dem Ovastacin exprimiert wird. Die neutrophile Elastase dagegen konnte in der Leber, im Herz sowie im Blutplasma nachgewiesen werden. Mit Hilfe spezifischer Antikörper konnte das Herz als Expressionsort für Ovastacin bestätigt werden. Somit wäre Elastase ein potentieller physiologischer Aktivator von Ovastacin. Die Identifikation des Ovastacins in Geweben wie Leber, Herz, Nabelschnur und im Blutplasma weist auf eine Rolle der Protease in proteolytischen Netzwerken außerhalb der Spermien-Ei-Interaktion hin. Die Bedeutung der biologischen Kontrolle des Ovastacins bei der Befruchtung der Säugereizelle wird durch die Beobachtung untermauert, dass das Leberprotein Fetuin B als physiologischer Ovastacininhibitor fungiert und dadurch eine vorzeitige Verhärtung der Zona pellucida verhindert, die andernfalls die Penetration von Spermien prinzipiell verhindern würde.
