Die neuronale Signalübertragung beruht auf dem synaptischen Vesikelzyklus, der durch das koordinierte Zusammenspiel von circa 400 verschiedenen Proteinen reguliert wird. Eines der Hauptproteine des synaptischen Vesikels ist Synaptophysin (SYP), das zu den tetraspan vesicle membrane proteins (TVPs) gehört. Es wird vermutet, dass es zahlreiche Funktionen der Exo- und Endozytose moduliert, wenngleich die zugrunde liegenden molekularen Mechanismen bisher größtenteils unverstanden sind. Ziel der Arbeit war daher die Identifizierung von Interaktionspartnern von SYP, um zum Verständnis der vielen ungeklärten Prozesse im synaptischen Vesikelzyklus beizutragen. Mit dem Split-Ubiquitin Yeast Two-Hybrid System, das eine direkte in vivo Interaktion von Membranproteinen erlaubt, konnten in der vorliegenden Arbeit bekannte, aber auch neue SYP-Bindungspartner identifiziert werden. Ein bekannter Interaktionspartner war Synaptobrevin2 (SYB2), das zu den stärksten im Split-Ubiquitin Y2H System identifizierten Bindeproteinen zählt. Zu den neuen starken SYP-Interaktionspartnern gehören die TVPs Synaptogyrin3 (SYNGR3) und SCAMP1. Somit konnten erstmals heterophile Interaktionen zwischen den verschiedenen TVP-Genfamilien nachgewiesen werden, die für eine universelle Funktion der TVPs sprechen. Die Validierung der im Split-Ubiquitin Y2H System ermittelten Interaktionspartner wurde auf eine Auswahl von Proteinen beschränkt, die vermutlich am synaptischen Vesikelzyklus beteiligt sind. Dabei konnte eine immunhistologische Kolokalisierung von SYP mit SYB2, SYNGR3, SCAMP1, Stathmin-like3 (STMN3), Rho family GTPase2 (RND2), Phospholipid transfer protein, Vesicle transport through interaction with t-SNAREs 1B homolog, Arfaptin2 und Profilin1 in den Synapsen-reichen Schichten der Retina beobachtet werden. Die SYP/SYB2- und SYP/SYNGR3-Komplexe konnten zudem sowohl aus Synaptosomen-Lysat als auch aus cDNA-transfizierten Epithelzellen koimmunpräzipitiert werden, wohingegen dies für die anderen Interaktionspartner nicht gelang. Da Koimmunpräzipitation die Struktur der Proteine durch Solubilisierung mit Detergenzien beeinflusst, wurden die in der Hefe beobachteten Interaktionen noch mittels Fluoreszenz-Resonanz-Energie-Transfer überprüft, mit dem Proteinwechselwirkungen in der nativen Umgebung nachgewiesen werden können. Ein positives FRET-Signal konnte für SYP mit SYB2, SYP, SYNGR3, SCAMP1, STMN3, RND2 und Arfaptin2 detektiert werden, lediglich für SYP mit Phospholipase D4 (PLD4) gelang dieser Nachweis nicht. Ferner zeigten FRET-Analysen von Synaptophysin-Mutanten, dass der zytoplasmatische C-Terminus für die Interaktion mit zytoplasmatischen und membranassoziierten Proteinen benötigt wird. Durch in vivo FRET-Studien mit der SH2-Domäne der Src-Kinase, die an phosphorylierte Tyrosine bindet, konnte eine Tyrosin-Phosphorylierung des zytoplasmatischen C-Terminus von Synaptophysin und von Synaptogyrin3 detektiert werden. Viele der neu identifizierten Synaptophysin-Interaktionspartner sind im Lipid-Metabolismus involviert. Vermutlich rekrutiert der zytoplasmatische und durch Phosphorylierung modifizierbare C-Terminus diese Partner in spezifische Lipoproteindomänen, die an der Feinabstimmung der synaptischen Vesikelendo- und -exozytose beteiligt sind.

Studies of organic fluorescent dyes are experiencing a renaissance related to the increasing demands posed by new microscopy techniques for high resolution and high sensitivity. While in the last decade single molecule equipment and methodology has significantly advanced and in some cases reached theoretical limits (e.g. detectors approaching unity quantum yields) unstable emission from chromophores and photobleaching become more and more the bottleneck of the advancement and spreading of single-molecule fluorescence studies. The main goal of this work was the synthesis of fluorophores that are water-soluble, highly fluorescent in an aqueous environment, have a reactive group for attachment to a biomolecule and posses exceptional photostability. An approach towards highly fluorescent, water-soluble and monofunctional perylene-3,4,9,10-tetracarboxdiimide and terrylene-3,4:11,12-tetra carboxidiimide chromophores was presented. A new synthetic strategy for the desymmetrization of perylenetetracarboximides was elaborated; water-solubility was accomplished by introducing sulfonyl substituents in the phenoxy ring. Two strategies have been followed relying on either non-specific or site specific labeling. For this purpose a series of new water-soluble monofunctional perylene and terrylene dyes, bearing amine or carboxy group were prepared. The reactivity and photophysical properties of these new chromophores were studied in aqueous medium. The most suitable chromophores were further derivatized with amine or thiol reactive groups, suitable for chemical modification of proteins. The performance of the new fluorescent probes was assessed by single molecule enzyme tracking, in this case phospholipase acting on phospholipid supported layers. Phospholipase-1 (PLA-1) was labeled with N-hydroxysuccinimide ester functionalized perylene and terrylene derivatives. The purification of the conjugates was accomplished by novel convenient procedure for the removal of unreacted dye from labeled enzymes, which involves capturing excess dye with a solid support. This novel strategy for purification of bioconjugates allows convenient and fast separation of labeled proteins without the need for performing time consuming chromatographic or electrophoretic purification steps. The outstanding photostability of the dyes and, associated therewith, the extended survival times under strong illumination conditions allow a complete characterization of enzyme action on its natural substrates and even connecting enzyme mobility to catalytic activity. For site-specific attachment of the rylene dyes to proteins the chromophores were functionalized with thioesters or nitrilotriacetic acid groups. This allowed attachment of the emitters to the N-terminus of proteins by native chemical ligation or complexation with His-tagged polypeptides at the N- or C-termini, respectively. The synthesis of a water-soluble perylenebis (dicarboximide) functionalized with a thioester group was presented. This chromophore exhibits an exceptional photostability and a functional unit for site-specific labeling of proteins. The suitability of the fluorophore as a covalent label was demonstrated via native chemical ligation with protein containing N-terminal cystein residue. We exploited also oligohisitidine sequences as recognition elements for site-selective labeling. The synthesis of a new water-soluble perylene chromophore, containing a nitrilotriacetic acid functional group was demonstrated, using solution-phase and solid-phase approaches. This chromophore combines the exceptional photophysical properties of the rylene dyes and a recognition unit for site-specific labeling of proteins. An important feature of the label is the unchanged emission of the dye upon complexation with nickel ions.

A novel nanosized and addressable sensing platform based on membrane coated plasmonic particles for detection of protein adsorption using dark field scattering spectroscopy of single particles has been established. To this end, a detailed analysis of the deposition of gold nanorods on differently functionalized substrates is performed in relation to various factors (such as the pH, ionic strength, concentration of colloidal suspension, incubation time) in order to find the optimal conditions for obtaining a homogenous distribution of particles at the desired surface number density. The possibility of successfully draping lipid bilayers over the gold particles immobilized on glass substrates depends on the careful adjustment of parameters such as membrane curvature and adhesion properties and is demonstrated with complementary techniques such as phase imaging AFM, fluorescence microscopy (including FRAP) and single particle spectroscopy. The functionality and sensitivity of the proposed sensing platform is unequivocally certified by the resonance shifts of the plasmonic particles that were individually interrogated with single particle spectroscopy upon the adsorption of streptavidin to biotinylated lipid membranes. This new detection approach that employs particles as nanoscopic reporters for biomolecular interactions insures a highly localized sensitivity that offers the possibility to screen lateral inhomogeneities of native membranes. As an alternative to the 2D array of gold nanorods, short range ordered arrays of nanoholes in optically transparent gold films or regular arrays of truncated tetrahedron shaped particles are built by means of colloidal nanolithography on transparent substrates. Technical issues mainly related to the optimization of the mask deposition conditions are successfully addressed such that extended areas of homogenously nanostructured gold surfaces are achieved. Adsorption of the proteins annexin A1 and prothrombin on multicomponent lipid membranes as well as the hydrolytic activity of the phospholipase PLA2 were investigated with classical techniques such as AFM, ellipsometry and fluorescence microscopy. At first, the issues of lateral phase separation in membranes of various lipid compositions and the dependency of the domains configuration (sizes and shapes) on the membrane content are addressed. It is shown that the tendency for phase segregation of gel and fluid phase lipid mixtures is accentuated in the presence of divalent calcium ions for membranes containing anionic lipids as compared to neutral bilayers. Annexin A1 adsorbs preferentially and irreversibly on preformed phosphatidylserine (PS) enriched lipid domains but, dependent on the PS content of the bilayer, the protein itself may induce clustering of the anionic lipids into areas with high binding affinity. Corroborated evidence from AFM and fluorescence experiments confirm the hypothesis of a specifically increased hydrolytic activity of PLA2 on the highly curved regions of membranes due to a facilitated access of lipase to the cleavage sites of the lipids. The influence of the nanoscale gold surface topography on the adhesion of lipid vesicles is unambiguously demonstrated and this reveals, at least in part, an answer for the controversial question existent in the literature about the behavior of lipid vesicles interacting with bare gold substrates. The possibility of formation monolayers of lipid vesicles on chemically untreated gold substrates decorated with gold nanorods opens new perspectives for biosensing applications that involve the radiative decay engineering of the plasmonic particles.

The multiligand Receptor for Advanced Glycation End products (RAGE) is involved in various pathophysiological processes, including diabetic inflammatory conditions and Alzheimers disease. Full-length RAGE, a cell surface-located type I membrane protein, can proteolytically be converted by metalloproteinases ADAM10 and MMP9 into a soluble RAGE form. Moreover, administration of recombinant soluble RAGE suppresses activation of cell surface-located RAGE by trapping RAGE ligands. Therefore stimulation of RAGE shedding might have a therapeutic value regarding inflammatory diseases. We aimed to investigate whether RAGE shedding is inducible via ligand-induced activation of G protein-coupled receptors (GPCRs). We chose three different GPCRs coupled to distinct signaling cascades: the V2 vasopressin receptor (V2R) activating adenylyl cyclase, the oxytocin receptor (OTR) linked to phospholipase Cβ, and the PACAP receptor (subtype PAC1) coupled to adenylyl cyclase, phospholipase Cβ, calcium signaling and MAP kinases. We generated HEK cell lines stably coexpressing an individual GPCR and full-length RAGE and then investigated GPCR ligand-induced activation of RAGE shedding. We found metalloproteinase-mediated RAGE shedding on the cell surface to be inducible via ligand-specific activation of all analyzed GPCRs. By using specific inhibitors we have identified Ca2+ signaling, PKCα/PKCβI, CaMKII, PI3 kinases and MAP kinases to be involved in PAC1 receptor-induced RAGE shedding. We detected an induction of calcium signaling in all our cell lines coexpressing RAGE and different GPCRs after agonist treatment. However, we did not disclose a contribution of adenylyl cyclase in RAGE shedding induction. Furthermore, by using a selective metalloproteinase inhibitor and siRNAmediated knock-down approaches, we show that ADAM10 and/or MMP9 are playing important roles in constitutive and PACAP-induced RAGE shedding. We also found that treatment of mice with PACAP increases the amount of soluble RAGE in the mouse lung. Our findings suggest that pharmacological stimulation of RAGE shedding might open alternative treatment strategies for Alzheimers disease and diabetes-induced inflammation.
